      • Generation of human induced pluripotent stem cells from osteoarthritis patient–derived synovial cells

        <P><B>Abstract</B></P><P><B>Objective</B></P><P>This study was undertaken to generate and characterize human induced pluripotent stem cells (PSCs) from patients with osteoarthritis (OA) and to examine whether these cells can be developed into disease‐relevant cell types for use in disease modeling and drug discovery.</P><P><B>Methods</B></P><P>Human synovial cells isolated from two 71‐year‐old women with advanced OA were characterized and reprogrammed into induced PSCs by ectopic expression of 4 transcription factors (Oct‐4, SOX2, Klf4, and c‐Myc). The pluripotency status of each induced PSC line was validated by comparison with human embryonic stem cells (ESCs).</P><P><B>Results</B></P><P>We found that OA patient–derived human synovial cells had human mesenchymal stem cell (MSC)–like characteristics, as indicated by the expression of specific markers, including CD14−, CD19−, CD34−, CD45−, CD44+, CD51+, CD90+, CD105+, and CD147+. Microarray analysis of human MSCs and human synovial cells further determined their unique and overlapping gene expression patterns. The pluripotency of established human induced PSCs was confirmed by their human ESC–like morphology, expression of pluripotency markers, gene expression profiles, epigenetic status, normal karyotype, and in vitro and in vivo differentiation potential. The potential of human induced PSCs to differentiate into distinct mesenchymal cell lineages, such as osteoblasts, adipocytes, and chondrocytes, was further confirmed by positive expression of markers for respective cell types and positive staining with alizarin red S (osteoblasts), oil red O (adipocytes), or Alcian blue (chondrocytes). Functional chondrocyte differentiation of induced PSCs in pellet culture and 3‐dimensional polycaprolactone scaffold culture was assessed by chondrocyte self‐assembly and histology.</P><P><B>Conclusion</B></P><P>Our findings indicate that patient‐derived synovial cells are an attractive source of MSCs as well as induced PSCs and have the potential to advance cartilage tissue engineering and cell‐based models of cartilage defects.</P>

        A Review of Fermented Foods with Beneficial Effects on Brain and Cognitive Function

        Around the world, fermentation of foods has been adopted over many generations, primarily due to their commercial significance with enriched flavors and high-profile nutrients. The increasing application of fermented foods is further promoted by recent evidence on their health benefits, beyond the traditionally recognized effects on the digestive system. With recent advances in the understanding of gut-brain interactions, there have also been reports suggesting the fermented food’s efficacy, particularly for cognitive function improvements. These results are strengthened by the proposed biological effects of fermented foods, including neuroprotection against neurotoxicity and reactive oxygen species. This paper reviews the beneficial health effects of fermented foods with particular emphasis on cognitive enhancement and neuroprotective effects. With an extensive review of fermented foods and their potential cognitive benefits, this paper may promote commercially feasible applications of fermented foods as natural remedies to cognitive problems.

        레딩스(B. Readings)의 ‘수월성(excellence) 대학 담론’에 대한 비판적 논의

        근대 이후의 대학은 교양을 연구하고 전수하여 국민 국가의 시민을 양성하는 것을 목적으로 하였지만, 오늘날의 대학은 국가의 통제를 뛰어넘은 자본주의의 영향으로 인하여 대대적인 변화를 맞이하게 되었다. 이러한 대학의 변화는 대학의 위치와 방향성에 대하여 논의할 수 있는 다양한 관점이 요구된다. 현대 대학을 다양한 관점에서 논의하는 담론들 중 이데올로기의 혐의가 있는 수월성 개념을 중심으로 대학 담론의 관점을 확장시킨 레딩스의 『폐허의 대학』은 중요한 텍스트로 간주될 만하다. 레딩스는 경제적 지구화로 인한 자본 재생산의 기반인 국민국가의 쇠퇴는 대학의 지향점을 변경하고 대학을 “초국적인 관료적 기업”으로 변모시켰다고 주장한다. 이로써 전통적인 인문학 또는 교양(culture)은 대학의 중심에서 벗어나 ‘수월성’에게 그 자리를 내주었다는 것이다. 레딩스에 따르면 수월성 개념이 사실상 텅 빈 개념으로서 비(非)지시적 성격을 가졌기에 대학이 이데올로기로부터는 벗어났지만, 자본주의에 무의식적으로 순응하면서 대학을 운영할 수밖에 없다. 이러한 예리한 지적에도 불구하고 레딩스는 경제적 논리를 우선할 수밖에 없는 현실을 인정하면서 수월성 대학에 대한 강력한 비판을 하지 못한다. 더욱이 레딩스의 대학 담론의 치명적인 약점은 대학이 교육기관임에도 불구하고 교육과 관련된 수월성 담론을 다루지 않았다는 것이다. 이러한 점에서 ‘폐허의 대학에 머물면서 진정한 사유(Thought)를 가능하게 해줄 수도 있다’는 레딩스의 제안은 여전히 논란의 여지가 있다. Although modern universities aimed to cultivate the citizens of the nation-state by studying and teaching the culture, today's universities have undergone a great change due to influences of capitalism beyond the control of the nation-state. Among the discourses that debate current universities from various viewpoints, Readings’ book "The University in Ruins", which has expanded the view of university discourse around the concept of excellence applied as an ideological mark in current universities, is regarded as one of the most important texts. As a result of the economic globalization and the decline of the nation-state which is the basis of capitalist reproduction, the university becomes a "transnational bureaucratic corporation" and traditional humanities or cultures have moved from the center of universities to the edge. However, because of the non-referential concept of excellence, the university has escaped from ideology, but that concept has enabled the university to operate unconsciously in compliance with capitalism. On the other hand, Readings failed to make a strong criticism on the university of excellence because he admitted the fact that economic logic should be a priority. In addition, despite the fact that university is an educational institute, there was no discourse on excellence related to education in Readings’ discourse on universities. Therefore, Readings’ proposal, ‘the true thinking by staying at the ruins of university’ may remain in debatable question.

        식생활 라이프스타일 유형이 다이어트 도시락 선택속성의 중요도에 미치는 영향

        The study subjects were 302 adult males and females aged more than 20 years living in the metropolitan area of South Korea. This study was conducted to obtain baseline data to establish proper development and marketing strategies by examining the effects of food-related lifestyles on the importance of diet, purchasing behavior towards diet lunch boxes, and their selected attributes such as menu, packaging, and services. With respect to food-related lifestyle, a cluster analysis was performed by using five factors such as convenience factor, health factor, safety factor, taste factor, and economy factor obtained from factor analysis to derive the economy type, the taste and economy type, the convenience type, the safety type, and the health type. As a result, the respondents regarded 'food hygiene (4.59)', 'freshness (4.47)', 'taste (4.28)', and 'nutrient balance (4.19)' as the selected attributes of diet lunch box menus. Moreover, the importance of diet lunch box menus (β=0.179) was increased with increasing safety orientation. 'Shelf life label (4.42)' was the most important selected attribute of diet lunch boxes, followed by 'ingredient label (4.19)', 'nutrition facts label (4.16)', and 'indication of origin (4.15)'. In particular, the importance of packaging for diet lunch boxes (β=0.203) was increased with increasing safety orientation. With respect to the selected attributes of services in purchasing diet lunch boxes, 'provision of personalized menus (4.07)' was the most important, and the importance of services for diet lunch box (β=0.160) was increased with increasing taste and economy orientation. Based on the above results, the respondents gave importance to the selected attributes related to food safety and health such as hygiene and, freshness. In addition, they also placed emphasis on hygiene and safe factors such as shelf life, ingredients, and nutrition facts labels. Therefore, it is considered necessary to develop diet lunch boxes by taking these factors into account. Furthermore, in services for diet lunch boxes, it is considered necessary to establish a service system capable of providing consumers with specialized menu or nutrition counseling according to the food-related lifestyle for their proper health management. Particularly, because consumers place emphasis on both food hygiene and safety, and health, it is considered necessary to thoroughly manage hygiene, safety, and nutrition in menu or packaging so that it is possible to enhance customer satisfaction by considering these selected attributes in greater detail.

        푸코의 에피스테메(episteme) 구분에 따른 코메니우스『세계도회』의 유형 분석

        푸코는 『말과 사물』에서 에피스테메(episteme)를 특정한 시대의 학문분야나 지식을 대상으로 추출되는 ‘담론의 질서’ 또는 사상사의 ‘선험적 여건’이라고 규정하였다. 에피스테메는 특정한 방식으로 사물들에 질서를 부여하는 ‘무의식적인 기초’라는 점에서 불연속적인 것으로 특징지어진다. 이에 근거하여 푸코는 유럽의 정신사적 시대를 르네상스, 고전주의, 근대라는 단절된 층으로 구분한다. 논문은 이러한 에피스테메의 구분에 의거하여 교육학의 고전인 코메니우스의『세계도회』의 유형이 두 가지 방식으로 분석 가능함을 탐색하고 있다. 하나는 이 교육학 고전이 전형적인 고전주의적 에피스테메인 재현의 방식에 따라 기술되고 있다는 해석이고, 다른 하나는 르네상스로부터 고전주의로 넘어가는 과도기의 성격을 지니고 있다는 해석이다. 재현이라는 고전주의 에피스테메를 반영하고 있다는 근거는 사물들을 재현한 150개 범주의 서술방식이 말과 사물의 일치를 의도했다는 점이다. 반면에 코메니우스의 범지론적 우주상이 언어와 사물의 유비적인 대응관계에 기초한 세계질서를 보여주고 있지만, 동시에 사물과 유리된 언어와 같은 기호로의 전환을 드러내는 재현의 에피스테메를 반영하고 있다는 해석도 가능하다. 논문에서는 이러한 과도기적 유형화 가능성을 논하면서 일본 교육학자들의 주장을 검토하였다. In his book The Order of Things (Les Mots et les choses)(1966) M. Foucault defined ‘episteme’ as a ‘transcendental condition’ of thought history or as an ‘order of discourse`` whose object extracts the disciplinary domain or knowledge in the certain epoch. The episteme has a discontinuous characteristic as far as it is a ‘unconscious fundament`` which gives world of things an order in a particular way. On a basis of such a definition Foucault assorts the periods of European thought history as Renaissance, classical, and modern. This article tries to analyze Comenius`` Orbis Sensualium Pictus(1631) on the basis of episteme typology of Foucault. There can be two possibilities of its analysis: One is an interpretative mode that the Comenius`` book would reflect the one of representation as the typical episteme in the classical epoch, and the other interpretation shows that his work would have the transitional characteristic from the Renaissance into the classical. The ground for the representation episteme in the classical epoch lies in the fact that the description method of 150 categories represented through things would intend to correspond between words and things. On the other hand the stereotyping as the transitional episteme is grounded upon the fact that the cosmological picture consisting of Comenius`` pansophism shows the world order based upon the analogous correspondent relationship between language and things, as well as moves into the space of representation, in which the transition into the symbol such as language detached from things comes out. Especially in order to discuss the possibility of transitional stereotyping, this paper examined the several advanced researches from Japanese community of pedagogy.

        Epidermis of building becomes target of design as component of space, as no more secondary element of space in modern society which is governed by speed and image. Especially, epidermis design in interior space enables users to communicate more smoothly with space by providing various narrative as it provides abundant visual perception experiences Patterns mean regular shape or form or type. In other words, it means design which appears repetitive regular shape. Patterns are important design elements in fashion and textile design, also modern space designers are using fully patterns on surface of building. As interior surface design element, pattern becomes one of components of space through supplementation in lack of space building, communication method between space and users, beyond just simple decoration. Through case studies, we can know, patterns in interior space are used two-dimensional and three-dimensional way, come out visual type like "objectization", "spacization", "framization".

      • Effective suppression of nitric oxide production by HX106N through transcriptional control of heme oxygenase-1.

        <P>Heme oxygenase-1 (HO-1) has been suggested to be a key neuroprotective enzyme because of its widespread distribution in the brain as well as its strong antioxidative effects. HX106N, a water-soluble botanical formulation, has previously been demonstrated to prevent amyloid β-induced memory impairment and oxidative stress in mice by upregulating HO-1 levels. In this study, the underlying molecular mechanisms of HX106N-induced HO-1 expression were investigated using BV-2 cells, a murine microglial cell line, and primary microglia. Treatment with HX106N induced the expression of HO-1 at the transcriptional level through the stress-responsive element-containing enhancer present in the ho-1 promoter. Nuclear factor E2-related factor 2 (Nrf2) was activated in cells treated with HX106N. The results from knockdown assay showed that small interfering RNA of Nrf2 attenuated HX106N-mediated HO-1 expression. Pharmacological inhibitors of p38 and JNK mitogen-activated protein kinases suppressed the HX106N-mediated induction of HO-1. The NF-κB signaling pathway was activated by HX106N and played a role in HX106N-induced HO-1 expression. Furthermore, HO-1 and one of its by-products during the enzymatic degradation of heme, CO, were found to be involved in HX106N-mediated suppression of NO production. Taken together, these data indicate that HX106N exerts potent antioxidative effects by increasing the expression of HO-1 through multiple signaling pathways, leading to the suppression of NO production.</P>
