These studies were undertaken to investigate the stability of lysinomethylene ampicillin in various solution such as pH solutions made of 0.1N NaOH∼0.1N HCl, 0.1N Na₂HPO₄∼0.1N Citric acid and aqueous solution, artificial gastric juices, intestinal juices, used of spectrophotometer at 320㎚. The results were as follows; 1. Lysinomethylene ampicillin in the various pH solution made of 0.1N NaOH 0.1N HCl (pH1∼10) was decomposed below to pH 1 and showed the best stability at pH 5. 2. In the various pH solutions with 0.1N Na₂HPO₄∼0.1N Citric acid (pH 2-9) lysinomethylene ampicillin was decomposed below to pH 4, and showed the best stability at pH 7. 3. Lysinomethylene ampicillin the various pH solution with the artificial gastric juices and intestinal juices was more stabled than that of ampicillin.
RT-PCR에 의한 박 종자의 오이녹반모자이크바이러스 검정

Cucumber green mottle mosaic virus (CGMMV) was a major pathogen of watermelon and had affected seriously to watermelon production in Korea. Rapid and sensitive detection method of CGMMV associated with bottle gourd (Lagenaria siceraria) seeds was developed by using RT-PCR in this study. A pair of primer, Wmf1 and Wmr1, specific for CGMMV was designed from coat protein gene sequences of CGMMV-W and used for amplifying 420 bp product in RT-PCR. To simplify the virus extraction procedure and reduce an inhibitor from the extract for the RT-PCR, some methods using ethanol precipitation, double filtration, polyethylene glycol precipitation and phenol/chloroform/ isoamyl alcohol extraction procedure were compared and the phenol/chloroform/isoamyl alcohol extraction procedure was selected by its enhanced sensitivity. This detection method using the selected extraction step and the primers for RT-PCR could reliably detect an infected level of one CGMMV-infested seed in 1,000 seeds. This rapid and sensitive RT-PCR assay provides a useful tool for the specific detection of CGMMV in bottle gourd seed samples containing high levels of background inhibitors.

Background: Malassezia is considered as major factor in dandruff of human scalp. Objective: In order to develop an antimicrobial agent, bamboo oil was extracted by high temperture suction from dried bamboo truk abd then antimicrobial activities against Malassezia are investigated. Methods: Minimum inhibitory concentration and antimicrobial activity were measured in Malassezia species. Results: 1. Minimum inhibitory concentration of the Bamboo (Phyllosrachys bambusoides) Essential Oil Malassezia furfur standard, Malassezia furfur patient, Malassezia sympodialis standard, Malassezia sympodialis patient, Malassezia dermatis standard, Malassezia dermatis patient were 10 μl/ml, 5 μl/ml,5 μl/ml, 10 μl/ml, 5 μl/ml and 10 μl/ml respectively. 2. Minimum inhibitory concentration of the Itraconazole Malassezia furfur standard, Malassezia furfur patient, Malassezia sympodialis standard,Malassezia sympodialis patient, Malassezia dermatis standardntia, Malassezia dermatis patient were 10μg/ml, 10 μg/ml, 10 μg/ml, 0.1 μg/ml, 1 μg/ml, and 0.01 μg/ml, respectively. 3. Minimum inhibitory concentration of the ketoconazole Malassezia furfur standard, Malassezia furfur patient, Malassezia sympodialis standard, Malassezia sympodialis patient, Malassezia dermatis standard, Malassezia dermatis patient were 0.01 μg/ml, 10 μg/ml, 10 μg/ml, 0.1 μg/ml, 0.01 μg/ml, and 0.01 μg/ml,respectively. 4. Malassezia furfur standard, Malassezia furfur patient, Malassezia sympodialis patient and Malassezia dermatis patient showed the strongest antimicrobial effect on bamboo oil > ketoconazole > itraconazole. 5. Malassezia sympodialis standard, Malassezia sympodialis patient and Malassezia dermatis standard strongest antimicrobial effect on ketoconazole > bamboo oil > itraconazole.Conclusion: Bamboo oil might be applied as antidandruff treatment modality by its anti-malassezial effect.
녹차와 구기자 물 추출물이 분말멸치의 저장 안전성에 미치는 영향

The storage stability from powdered anchovy and its preparation cooperated with three additives (water extract from green tea and boxthorn and BHT) was studied. The results are as follows: 1. When the additives were added to powdered anchovy, the storage stability was improved. The antioxidative activities of water extract from green tea and boxthorn were considerably higher than BHT. The antioxidative activities were decreased in the rank order boxthorn $gt; green tea $gt; BHT. 2. The more the concentration of natural additives was increased, the more the storage stability was improved. The rank order was 60° $gt; 40° $gt; 20° Bx. 3. The AV and POV of powdered anchovy were increased in the rank order to 6 $lt; 12 $lt; 18 $lt; 24 $lt; 30 months by storage term. 4. This clearly suggested that water extract at 20° Bx level from boxthorn was possible utilization as a new natural antioxidant for storage stability of powdered anchovy.

According to previous paper, the hardness of semi-solid as ointments and suppositories causes the tough or skin, the pain and connected with absorption, the state of spread or the efficiency of drug. When the hardening agent or the softening agent is added to ointment whild, is registered on the pharmacopeia of the republic of Korea, the experimental data by apparent logarithmic hardness are shown is Table 1. The results are as the following. 1) In case of the softening agent which is added to ointment base, each has shown two different equation with differented slope as k₁, and k₂, value determined (Fig. 1, 2). 2) When the hardening agent is added to same ointment base, these studies indicated that there is a direct correlation between k₁ and k₂. However, in case of softening agent k₁ and k₁ are not correlated as indicated in Fig. 3∼8. 3) On condition of same ointment base, the critical poit is proportional to k₁ 4) The second effect is at least 3∼17 times more sensitive than the first effect or in average 8.3 times more sensitive, Therefore, the results presented in this paper. Suggest that, when the drug is added to a certain ointment base, it should be added within this first effect before the critical point is reached.

Apoptosis serves to protect normal cells during the processes of development, aging, tissue homeostasis, immunity, and inflammation. Molecules such as caspases, the Bcl-2 family proteins, cytochrome c, APAF-1, and apoptotic endonucleases are concerned with and regulate different stages of apoptosis. DNA fragmentation factor (DFF) is a key final molecule in chromosomal DNA fragmentation during the last step of apoptosis, and is a heterodimeric complex consisting of alpha/inhibitor of caspase-activated DNase (DFFA/ICAD) and beta/caspaseactivated DNase (DFFB/CAD) subunits. DFFB/CAD is normally combined with the inhibitor, DFFA/ICAD, in healthy cells. However, after cleavage of the DFF complex by effector caspases, DFFB/CAD acts as an apoptotic nuclease and induces DNA fragmentation in apoptotic cells. The study of DFF proteins is focused on the roles of the apoptotic pathway in mammals. However, gene expression and functions under immune stimulation are not well known in other species. In the present study, we first characterized teleost DFFA/ICAD and DFFB/CAD from rock bream at the molecular level and analyzed the transcriptional expression pattern under immune challenges, to understand their role in immunity. These putative proteins have shown conserved domains and interaction sites orthologous to those of other species. High mRNA expression of both genes was found in the gills and blood of tissues involved in the immune response. The increased expression of DFF genes upon bacterial and viral stimulation was confirmed by qPCR analysis. DFFA/ICAD and DFFB/CAD of rock bream may play a pivotal role in apoptosis and may be involved in host immunity against bacterial and viral infection.
