To improve the seed purity management system of Korean wheat cultivars, 50 Korean wheat cultivars were subjected to chemicalassays for grain color, genotyping of grain weight-related genes, and grain image analysis. The tested cultivars were primarily classified byNaOH and ninhydrin tests as white (26%) and red (74%) cultivars, as well as high PPO activity (48%), and low PPO activity (52%) cultivars,respectively. The allelic variations of Tamyb10 gene revealed Tamyb-A1a/Tamyb-B1a/Tamyb-D1a as the major allelic combination in whitewheat and five different Tamyb10 genotypes (i.e., aba, abb, baa, bba, and bbb) in red wheat. Those cultivars with high PPO activity possessedthe Ppo-A1a/Ppo-B1b/Ppo-D1b genotype, while those with low PPO activity possessed the Ppo-A1b/Ppo-B1a/Ppo-D1a genotype. In the grainimage analysis, long grain cultivars displayed increased grain width, circularity, and area. Based on cluster analysis of grain traits, the Koreanwheat cultivars were classified into two groups - 1) large red grain cultivars released before 2000, and 2) small red grain cultivars and whitewheat cultivars released after 2000. Further research is required to determine the effects of grain filling conditions on the grain characteristicsof Korean wheat cultivars and to develop efficient and reliable molecular markers for an improved seed purity management system.
Multiplex STS-SSR 마커를 활용한 국산밀 품종 판별

This study aimed to develop an agarose gel-based multiplex PCR assay using sequence-tagged site (STS) and simple sequencerepeat (SSR) markers that can differentiate Korean wheat cultivars. Forty-nine Korean wheat cultivars were primarily classified based on seedcoat color into red (36) and white (13) groups. Red wheat cultivars were further differentiated by three multiplex PCRs using molecular markersfor Ppo-A1/Vrn-D1a/Rht-B1b, Glu-A3ac/TaCwi-A1b/Lr34, and Glu-A1ac/Glu-B1b/KWSM003/TaSE96. Similarly, white wheat cultivars werefurther differentiated using two multiplex PCRs using the molecular markers for Ppo-A1/Vrn-D1a/Pina-D1a and Ppo-B1/Glu-B3h. A multiplexPCR assay using molecular markers for Glu-A1b/Glu-D1d/Wx-B1 was developed to differentiate four Korean wheat cultivars used as governmentcertified seeds: Baekkang, Hwanggeumal, Keumkang, and Saekeumkang. A multiplex PCR assay using molecular markers for Glu-B3h andPin-D1a was used for colored wheat cultivars, Arijinheuk, Ariheuk, and Chinese colored wheat. The multiplex PCR assays developed in thisstudy can provide useful molecular tools for differentiating Korean wheat cultivars, developing wheat seed management systems, and guaranteeingwheat seeds in Korea.

Background: Despite the popularity of the study of borderline personality disorder, definition of “Chronic Emptiness” were applied to define the theoretical concept and never met an agreement among evaluators. Methods: First, the preliminary questionnaire was prepared based on the data collected through the in-depth interviews with the previous studies and the people with borderline personality disorder, and the final 18 items were developed through the item analysis. In order to verify the reliability and validity of the emptiness scale, 526 adults from 18 to 40 years of age were tested for internal consistency coefficient, and factor analysis and validity test were conducted. Results: The internal consistency coefficient of the emptiness scale was very good. As a result of the factor analysis, it was confirmed that it has a single structure. As a result of validity test, emptiness was highly correlated with borderline personality disorder, depression, and suicide ideation. Conclusions: Emptiness is associated with identity problems, and that adolescents tend to do NSSI to deal with emptiness related to identity formation. Until now, the terms of loneliness, solitude, and emptiness have not been sufficiently distinguished, but the distinction between the different terms has become possible through the emptiness measure The implications and limitations of this research and subsequent studies discussed. 경계선 성격장애의 진단기준 중‘만성적 공허감’은 개념적 정의가 모호하고, 평정자간 일치도가 낮아 반구조화된 면담으로 평정하는데 제한이 있다. 이에 본 연구는 경계선 성격장애의 공허감을 측정하기 위한 척도를 개발하는데 목적을 두었다. 먼저 선행연구들과 경계선 성격장애를 가지고 있는 사람들과의 심층면접을 통해 수집된 자료를 토대로 예비 문항을 작성한 후, 문항분석을 거쳐 최종 18문항의 공허감 척도를 개발하였다. 공허감 척도의 신뢰도와 타당도를 검증하기 위해 만 18세부터 40세까지의 수도권 및 지방 대학에 재학 중인 학생과 직장인 526명을 대상으로 내적 일치도 계수를 산출하고 요인분석과 준거 타당도 검증을 실시하였다. 공허감 척도의 내적 일치도 계수는 매우 양호한 수준이었다. 요인분석 결과 단일 구조를 가지고 있는 것으로 확인되었다. 준거타당도 검증 결과 공허감은 경계선 성격장애, 우울, 자살사고와 높은 상관을 보였다. 이러한 결과를 바탕으로 본 연구의 시사점 및 제한점을 논의하였다.
