        마우스 섬유아세포(3T3 fibroblast cells)에서 Insulin-like Growth Factor-I(IGF-I) 및 IGF Binding Protein-3 (IGFBP-3)이 세포증식에 미치는 영향

        연구배경: 세포성장은 세포증식과 세포죽음의 균형에 의해 이루어 진다. 세포성장에 관여하는 여러 growth factor증 IGF-I은 IGF-IR와 결합하여 세포증식을 유발하는 mitogen으로 알려져 있다. 또한 IGF-I에 결합하는 IGFBPs중에 IGFBP-3는 혈액내에 가장 많은 carrier protein으로, IGF-I과 결합하여 IGF-I의 세포증식 효과를 증가 혹은 억제시킨다. 방 법: 3T3 fibroblast 세포를 이용하여 IGF-I과 IGF-IR transcripts를 northern blot으로 확인하고, IGF-I에 의한 mitogenic effect를 MTT assay 및 $^3H$-thymidine incorporation test로 관찰하고, IGF-I의 receptor인 IGF-IR의 활성화를 보기 위해 intracellular $\beta$-subunit의 tyrosine kinase domain의 phosphorylation을 western blot으로 관찰하였다. 또한 IGFBP-3가 3T3 세포에서 mitogenic effect에 미치는 영향을 보기 위해 anti-IGFBP-3와 ${\alpha}IR_3$을 단독 및 병용투여하여 관찰하였다. 결 과: 3T3 세포는 IGF-I와 IGF-IR의 mRNA expression을 보였으며, IGF-I을 투여시 IGF-IR의 intracelluar cytoplasmic protein인 $\beta$-subunit의 tyrosine kinase domain을 phosphorylation시켜 활성화시키며, 5%, 1% serum-containing media에서 세포증식을 보였으나, serum-free media에서는 세포증식을 보이지 않았다. 또한 anti-IGFBP-3 투여와 ${\alpha}IR_3$과 anti-IGFBP-3를 병용투여시는 세포종식이 각각 2배이상 증가하였으나, ${\alpha}IR_3$을 4시간 전처치후 ${\alpha}IR_3$과 anti-IGFBP-3를 병용투여시는 anti-IGFBP-3에 의한 세포종식이 보이지 않은 것으로 보아 IGF -I/IGFBP-3 결합에서 분리되는 free IGF-I어l 의해 세포증식이 유도된 것으로 생각된다. 결 론: IGF-I은 IGF-IR를 phosphorylation시켜 mitogenic effect를 보이며, IGFBP-3는 IGF-I의 mitogenic effect를 억제하는 것으로 생각된다. Background: Cell growth is a balance between cell proliferation and cell death. Insulin-like growth factor-I(IGF-I), which binds IGF-I receptor(IGF-IR), mediates cellular proliferation as a potent mitogen. IGF binding protein-3(IGFBP-3) as a circulating major IGFBP can inhibit or enhance the effects of IGF-I on cellular growth by binding IGFs. Methods: We investigated the expressions of mRNA of IGF-I and IGF-IR by northern blot and phosphorylation of IGF-IR with the treatment of IGF-I by western blot in 3T3 fibroblast cells. The cellular proliferations of 3T3 cells with the treatments of IGF-I were evaluated using $^3H$-thymidine incorporation and MTT assay. Also to observe the effect of IGFBP-3 on cellular proliferation, 3T3 cells were treated with anti-IGFBP-3 and ${\alpha}IR_3$(monoclonal antibody to IGF-IR) alone or in combination. Results: Our results demonstrated that 3T3 cells showed mRNA expressions of IGF-I and IGF-IR and the IGF-I increased phosphorylation of IGF-IR. The treatments of 3T3 cells with IGF-I increased cellular proliferation in 5 % and 1 % seruma-containing media, not in serum-free media. The addition of anti-IGFBP-3 to neutralize IGFBP-3 showed 2-fold increase of cellular proliferation, and also co-incubation of anti-IGFBP-3 and ${\alpha}IR_3$ together showed similar increase of cellular proliferation in 3T3 cells. Interestingly, when the cells were pretreated with ${\alpha}IR_3$ for 4 hr, prior to the simultaneous addition of ${\alpha}IR_3$ and anti-IGFBP-3, anti-IGFBP-3-mediated cellular proliferation was decreased to control level. All of these results suggest that free IGF-I released from IGF-I/IGFBP-3 complex would be involved in the cellular proliferation. Conclusion: IGF-I is a mitogen through the activation of IGF-IR in 3T3 cells, and IGFBP-3 could be a potent inhibitor for IGF-I action by binding IGF-I.

        Profile Index에 따른 Panelipse Ⅱ 방사선 상층의 변화

        This research was made to investigate the change of the image layer and of the vertical and horizontal magnification on the Panelipse radiographic image by the control of profile index. Using the Panelipse, a series of 60 exposures were taken with the steel balls placed in the holes of the plastic model plate, and then evaluated by 4 observers. Two points were assigned for a reading of sharp, one for middling sharp, and zero for unsharp. Each ball image then could be given a total reader score of 0 to 8. The author analyzed the image layer as defined by a sharpness score of 6 or more. The results obtained were as follows: As the profile index was increased, the shape of the image layer was not changed, and the width of the image layer was increased, and the position of the layer shifted away from the rotation center. As the profile index was increased, the ranges of vertical and horizontal magnification was increased, especially the ranges of horizontal magnification was greater than that of vertical magnification. 저자는 Panelipse Ⅱ의 상층 및 상의 확대변화를 관찰하기 위하여 모형판에 금속구를 위치시켜 3가지 Profile Index에서 각기 20장씩 방사선사진을 촬영하여 관찰한 결과를 다음과 같이 요약할 수 있었다. Profile Index의 증가에 따른 Panelipse Ⅱ 방사선상의 상층에 있어 모양의 변화는 없었고, 폭은 더 넓어졌으며, 회전축중심에서 외측으로 상층은 이동하였다. 방사선상에서 수평 수직확대는 공히 Profile Index가 증가함에 따라 그 범위가 커졌으며 이는 수평확대에 있어 수직확대보다 현저하였다.

        중소병원 간호조직의 변혁적 리더십이 임파워먼트와 조직효과성에 미치는 영향

        This study was designed to explore relationship among transformational leadership, empowerment, job satisfaction, and customer orientation of care service organization working at small medium size hospital. The main purpose of this study is to obtain an organizational effectiveness in order to offer better nursing service through reasonable human resource management. This study has some important implications and they are as follows. Firstly, sub factors of transformational leadership were empirically turned out to divided into three factors such as charisma, individual consideration, inspirational motivation, intelligent stimulus and impact. Secondary, sub factor of empowerment of social welfare organization comprise three factors such as self-decision, ability and impact. Thirdly, positive relationships were found among transformational leadership, empowerment, job satisfaction and customer orientation except for relationship between transformational leadership vs. customer orientation and empowerment vs customer orientation, and that assist result of previous studies. This study has some limitations in relative to specific survey region and classification according to job diversity.

        의료서비스품질이 고객만족, 관계품질 및 재이용의도에 미치는 영향 -대구지역 소규모 병의원을 중심으로-

        These days, medical service quality and relationship quality are regarded as important factor in successful hospital management. Generally, service quality affects relationship quality, customer satisfaction and reuse intent in diverse service industries, and most of researchers agree to this conclusion. This study is designed to explore medical service quality in Small-size Medical Clinic and relationship among service quality, customer satisfaction, relationship quality and reuse intention. Additionally, we explored roles of relationship quality as moderating valuables in causal relationship. Through conclusion of this study, we could find that medical specialized knowledge, kindness, and good manners affected customer satisfaction, relationship quality and reuse intent directly and indirectly. Moreover we found that relationship quality was playing an important role as connecting bridge between customer satisfaction and reuse intent.

        한방병원의 이미지 및 관계성 향상을 위한 의료서비스생산성 전략연구

        This study provides strategic implications that can be used in Korean medicine hospitals through analysis of causal relationship among factors focusing on service quality, image improvement and relationship productivity and quality building. As a result of this study, it has some theoretical implications that are as follows. First, service quality of Korean medical hospital could be divided into four factors such as medical staff, administration service, medical facilities, and subsidiary facilities. Second, medical service quality has positive effects on image, but it never has direct effects on customer satisfaction. Third, image had a positive effect on customer satisfaction. Fourth, image and customer satisfaction had positive effect on relationship quality, but it did not have an effect on reuse intention. They could have indirect effects on reuse intention just only through relationship quality. Fifth, in the viewpoint of total effect, medical staff and medical facilities in the medical service quality had positive effects on customer satisfaction, relationship quality, and reuse intention. Lastly, based on previous study and analysis result, we presented that relationship quality played a mediating role between image and reuse intention, and between customer satisfaction and reuse intention. This study suggests some practical implications that are as follows. First, it is necessary to maintain continuous improvement activities to medical service quality and productivity. Because medical service quality has meaningful effect on customer satisfaction, image improvement, relationship building. Second, hospital medical staff should treat patients kindly and respond their needs promptly. In order to increase better image, it is evitable to keep sufficient and kind explanation, and clean and proper medical examination environment. Third, it is necessary to plot plans to strengthen relationship formation with customers. This is because the proper attachment of factors such as satisfaction, image and quality of relationship is relatively stronger. Lastly, there is a correlation between relationship immersion with customer and loyalty intention. Therefore, it is important to make an effort for customer to have attachment for oriental medicine by means of public relations and marketing activities. The study has some limitations that need additional research in the future. First, the present research was based on the result of questionnaire survey, and it is necessary to research focusing on strategic medical service. Third, the following research should include direct factors for the associated customers of oriental medicine hospitals.

      • 상사와의 관계증진이 업무태도에 미치는 영향 : 대구지역 노인요양병원의 간호사를 중심으로

        본 연구는 노인요양병원의 간호단위에서 조직특성인 리더-구성원 교환관계, 구성원의 행동양식을 결정하는 조직몰입을 선행변수로 조직시민행동에 미치는 영향을 연구하고자 하였다. 연구결과를 토대로 실무적 시사점은 다음과 같다. 첫째, 노인요양병원의 간호조직에서 상사와 부하간의 관계증진 활동은 직무만족, 조직시민행동 같은 조직유효성을 효과적으로 높일 수 있다. 둘째, 노인요양병원의 간호조직의 조직몰입의 형태는 설문내용으로 판단해 보면 정서적 몰입의 형태보다는 유지적 몰입의 형태를 가진다. 셋째, 특히 본 연구에서는 LMX가 직접적으로 조직몰입에 유의한 영향을 미치지 못하고 간접적으로 직무만족을 통하여 조직몰입에 영향을 미치고 있다. 이는 요양병원의 간호조직의 직무만족은 상사와의 관계의 질에서 출발하며 이를 통하여 유지적 직무몰입에 영향을 주고 있음을 보여주고 있다. 즉 상사와 부하 간의 관계의 질이 노인요양병원의 간호조직에서 매우 중요한 사안임을 보여주고 있다. From this study, we intended to find the causal relationship among LMX, job satisfaction, organizational commitment, and organizational citizenship behavior in nurse organizational unit of silver hospital. Practical implications according to result of this study are as follows. First, activities for promotion of relationship between higher office and follower can efficiently improve organizational effect such as job satisfaction, organizational citizen behavior in nurse organization of silver hospital. Second, style of organizational commitment of nurse organization turned out sustainable commitment. Third, especially, LMX didn't directly have influence on organizational commitment, but indirectly have influence on it via job satisfaction. It shows that job satisfaction of nurse organization in silver hospital comes from LMX, it also have effect on sustainable organizational commitment. In a word, LMX is very important issue in nurse organization of silver hospital.

        한말 철도의 인식 변화와 정치경제적 이해관계

        이 연구는 왜 한말에 한반도 철도의 자율성을 상실하게 되었는가를 한말 철도의 인식을 중심으로 분석하고자 한다. 이를 위해서 이 연구는 19세기 제국주의 시대에서 한국은 한말 철도에 대하여 어떠한 인식을 가졌는가 또한 이러한 한말 철도의 부설과정에서 어떠한 정치경제적 이해관계가 나왔는가를 알아보고자 한다. 특히 한말 철도의 자율성 상실을 인식, 정책, 국제체계라는 3가지 차원에서 각각 분석하고, 이를 토대로 향후 남북한 철도 연계를 비롯한 한국종단철도와 동북아 철도네트워크 형성을 위한 역사적 교훈과 정책적 함의를 도출하고 있다. 19세기 제국주의 시대 상황 속에서 철도에 대한 인식 미흡과 동북아 질서의 역학관계로 인하여 한반도 철도의 자율성을 상실하게 되었다는 점에 주목하였다. 또한 한반도 종단철도와 동아시아 철도네트워크에 대한 정세 파악과 철도의 역할에 대한 인식을 보다 명확하게 할 필요가 있다는 역사적 교훈을 얻었다. 또한, 상충되는 동북아 관련국의 정치경제적 이해관계를 타협과 협상을 통한 갈등 해소의 제도화를 준비하여야 할 것이라는 정책적 함의를 얻었다. This study is focused to analyze why the Late Joseon Dynasty and Dahanjeguk could not maintain the right of managing the railroad system on the Korea peninsula Also It is aimed to find out political and economic interests emerged in the process of laying the Korean railroad system. In particular, the loss of autonomy of Korean-Korean railways is analyzed in the three dimensions of recognition, policy, and international system, and based on this, historical lessons and policy implications of Korean railways for the future railroad linkage between South and North Korea and Northeast Asia are derived. As results of this study, we knew that in the imperialism of the 19th century, the lack of proper perceptions on railroads and the dynamics of the Northeast Asian order led to a loss of autonomy of railroads on the Korean peninsula. We learned a historical lesson that it is necessary to do it. In addition, there was a policy agreement that the conflicting political and economic interests of Northeast Asian countries should be prepared for institutionalization of conflict resolution through compromise and negotiation.
