Clinical Aspect of MicroRNA in Lung Cancer

MicroRNAs (miRNAs) are a class of small noncoding RNAs that modulate target gene activity, and are aberrantly expressed in most types of cancer as well in lung cancer. A miRNA can potentially target a diverse set of mRNAs; further, it plays a critical role in lung tumorigenesis as well as affects patient outcome. Previous studies focused mainly on abnormal miRNAs expressions in lung cancer tissues. Interestingly, circulating miRNAs were identified in human plasma and serum in 2008. Since then, considerable effort has been directed to the study of circulating miRNAs as one of the biomarkers of lung cancer. miRNAs expression of tissues and blood in lung cancer patients is being analyzed by more researchers. Recently, to overcome the high false-positivity of low-dose chest computed tomography scan, miRNAs in lung cancer screening are being investigated. This article summarizes the recent researches regarding clinical applications of miRNAs in the diagnosis and management of lung cancer.

Introduction: MicroRNAs (miRNAs) play important role in development of cancer and metastasis. Metastatic cancer cells are regarded as having characteristics of cancer stem cells (CSCs). Recently, there were studies that spheroid-forming cells among lung cancer cells have trait of CSCs. Objective: To investigate expressions of miRNAs in spheroid-forming subpopulation of the HCC1588 human lung squamous cell carcinoma cell lines. Methods: Spheroid-forming cells were cultured and isolated among HCC1588 cells in condition of serum free media. We checked the makers of stemness and epithelial mesenchymal transition (EMT) by RT-PCR in cancer cells. Differently expressed miRNAs were analyzed between spheroid-forming cells and parental cells of HCC1588 by miRNA microarray and RT-PCR. Results: There were increased expression of ALDH1, CD133, and EMT in spheroid-forming cells compared with parental cells of HCC1588. We identified 86 differentially expressed miRNAs in the spheroid-forming subpopulation of HCC1588 cells, 44 of which were significantly over-expressed, including miR-1290, miR-210, miR-4455, and miR-203a, and 42 of which were significantly down-regulated, including miR-34c-5p, miR-18a-5p, and miR-200c-5p. Conclusion: Our results suggest that dysregulation of miRNAs may play a role in the stem cell-like properties of lung squamous cell carcinoma stem cells.Introduction: MicroRNAs (miRNAs) play important role in development of cancer and metastasis. 증례 : 호흡기 ; 객혈과 폐종괴로 내원한 amiodarone 유인성 폐독성 1예

Amiodarone은 부정맥의 치료에 효과적으로 사용되고 있지만 폐독성의 발생빈도가 약물 복용자의 1~15%로 보고되어 있어 사용에 주의를 요하는 약물이다. 종괴상 병변으로 amiodarone 유인성 폐독성이 나타난 예는 드물며 이는 폐의 악성 신생물, 결핵 등 타 질환과의 감별을 요하기 때문에 중요하다고 할 수 있다. 저자 등은 객혈로 내원한 환자에서 흉부 방사선 검사상 좌폐상엽의 종괴가 관찰되어 흉부 신생물로 오인될 수 있었던 amiodarone 유인성 폐독성의 1예를 경험하였으며 약물 중단 이후 객혈 및 종괴상 병변의 호전을 관찰하여 이에 보고하는 바이다. Amiodarone is an antiarrhythmic drug that has adverse effects involving the lungs and other organs. Amiodarone-induced pneu-monitis is a distinct, not uncommon form of drug-induced lung injury. By contrast, amiodarone-induced pulmonary toxicity presenting with hemoptysis and a lung mass is rare. We report the case of a 67-year-old woman with hemoptysis and a lung mass who had taken amiodarone for 6 years to control a ventricular arrhythmia. After drug withdrawal without any additional treatment, clinical and radiological improvement was observed. (Korean J Med 77:S1202-S1205, 2009)
