The hematopoietic cell malignancy is one of the most prevalent type of cancer and the disease has multiple pathologic molecular signatures. Research on the origin of hematopoietic cancer stem cells and the mode of subsequent maintenance and differentiation needs robust animal models that can reproduce the transformation and differentiation event in vivo. Here, we show that co-trans-duction of MYC and PIM2 proto-oncogenes into mouse bone marrow cells readily establishes permanent cell lines that can induce lethal myeloid sarcoma in vivo. Unlike the previous doubly transgenic mouse model in which co-expression of MYC and PIM2 transgenes exclusively induced B cell lymphoma, we were able to show that the same combination of genes can also transform primary bone marrow myeloid cells in vitro resulting in permanent cell lines which induce myeloid sarcoma upon in vivo transplantation. By inducing cancerous transformation of fresh bone marrow cells in a controlled environment, the model we established will be useful for detailed study of the molecular events involved in initial transformation process of primary myeloid bone marrow cells and provides a model that can give insight to the molecular pathologic characteristics of human myeloid sarcoma, a rare presentation of solid tumors of undifferentiated myeloid blast cells associated with various types of myeloid leukemia.
DRG2 Regulates G2/M Progression via the Cyclin B1-Cdk1 Complex

Developmentally regulated GTP-binding protein 2 (DRG2) plays an important role in cell growth. Here we explored the linkage between DRG2 and G2/M phase checkpoint function in cell cycle progression. We observed that knockdown of DRG2 in HeLa cells affected growth in a wound-healing assay, and tumorigenicity in nude mice xenografts. Flow cytometry assays and [3H] incorporation assays indicated that G2/M phase arrest was responsible for the decreased proliferation of these cells. Knockdown of DRG2 elicited down-regulation of the major mitotic promoting factor, the cyclin B1/Cdk1 complex, but up-regulation of the cell cycle arresting proteins, Wee1, Myt1, and p21. These findings identify a novel role of DRG2 in G2/M progression.
Expression of Keratin 10 in Rat Organ Surface Primo-vascular Tissues

The primo-vascular system is described as the anatomical structure corresponding to acupuncture meridians and has been identified in several tissues in the body, but its detailed anatomy and physiology are not well understood. Recently, the presence of keratin 10 (Krt10) in primo-vascular tissue was reported, but this finding has not yet been confirmed. In this study, we compared Krt10 expression in primo-vascular tissues located on the surface of rat abdominal organs with Krt10 expression on blood and lymphatic vessels. Krt10 protein (approximately 56.5 kDa) was evaluated by western blot analysis and immunohistochemistry. Krt10 (IR) in the primo-node was visualized as patchy spots around each cell or as a follicle-like structure containing a group of cells. Krt10 IR was also identified in vascular and lymphatic tissues, but its distribution was diffuse over the extracellular matrix of the vessels. Thus Krt10protein was expressed in all three tissues tested, but the expression pattern of Krt10 in primo-vascular tissue differed from those of blood and lymphatic vascular tissues, suggesting that structural and the regulatory roles of Krt10 in primo-vascular system are different from those in blood and lymphatic vessels.
DRG2 Deficiency Causes Impaired Microtubule Dynamics in HeLa Cells

The developmentally regulated GTP binding protein 2 (DRG2) is involved in the control of cell growth and differentiation. Here, we demonstrate that DRG2 regulates microtubule dynamics in HeLa cells. Analysis of live imaging of the plus-ends of microtubules with EB1-EGFP showed that DRG2 deficiency (shDRG2) significantly reduced the growth rate of HeLa cells. Depletion of DRG2 increased ‘slow and long-lived’ subpopulations, but decreased ‘fast and short-lived’ subpopulations of microtubules. Microtubule polymerization inhibitor exhibited a reduced response in shDRG2 cells. Using immunoprecipitation, we show that DRG2 interacts with tau, which regulates microtubule polymerization. Collectively, these data demonstrate that DRG2 may aid in affecting microtubule dynamics in HeLa cells.

인터넷 사용이 Daily Activity 중에서 큰 비중을 차지함에 따라 인터넷은 광고 매체로서의 큰 영향력을 가지게 되었다. 적은 비용으로 효과가 큰 인터넷 마케팅의 툴로는 메일링, 온라인 배너, 검색 광고, 블로그, 웹싸이트, 소셜 미디어, UCC 등이 있으며, 이는 전세계적으로 공통된 추세로 나타난다. 이런 환경하에서 다수의 해외 법인을 가지고 있는 글로벌 기업은 디지털 마케팅 수행 방법에 대한 관심을 갖게 되었다. 하지만 디지털 마케팅의 필요성에 대한 연구는 많이 이루어졌으나, 실제 적용할 수 있는 추진 방법에 대한 연구는 많지 않다. 따라서 본 연구는 글로벌 기업에 적합한 디지털 마케팅을 정의하고, 이행 전략 수립 및 방법론을 제시한다. 또한 실제 현장 적용 가능하도록 사례를 통해 글로벌 디지털 마케팅 상향 표준화를 실현시키는 데 기여하고자 한다. Internet growth has expanded beyond developed markets and now represents a significant portion of media consumption around the world. This growth has led to the creation of a new set of cost efficient but highly effective marketing tools (e.g. emails, banner ads, search engine marketing, blogs, social medias, UCC, etc.) that multinational companies can leverage to execute their marketing strategies and campaigns. Although there have been studies focused on the changing landscape in the age of digital marketing, few have provided a tangible methodology that allows for prompt execution of company strate-gies in this dynamic environment. This study has set out to contribute to the global standardization of digital marketing while helping multinational companies define, develop, and execute successful international marketing strategies.
Infrared spectroscopy characterization of normal and lung cancer cells originated from epithelium

The vibrational spectral differences of normal and lung cancer cells were studied for the development of effective cancer cell screening by means of attenuated total reflection infrared spectroscopy. The phosphate monoester symmetric stretching νs(PO32-) band intensity at ∼970 cm-1 and the phosphodiester symmetric stretching νs(PO2-) band intensity at ∼1,085 cm-1 in nucleic acids and phospholipids appeared to be significantly strengthened in lung cancer cells with respect to the other vibrational bands compared to normal cells. This finding suggests that more extensive phosphorylation occur in cancer cells. These results demonstrate that lung cancer cells may be prescreened using infrared spectroscopy tools.
