목적 : Schedule induced polydipsia(SIP) 흰쥐 모델은 강박장애 연구를 위한 중요한 동물 모델중의 하나로 제안되어 왔다. 이전 연구에서 선택적 세로토닌 재흡수 억제제인 fluoxetine이 SIP 흰쥐 모델에서 음수량을 감소시켰다는 보고가 있다. 최근에는 vasopressin이 강박장애의 병인과 관련이 있다고 한다. 따라서 본 연구에서는 SIP 흰쥐 모델을 이용하여 강박장애의 치료시 사용되는 fluoxetine이 vasopressin 발현에 어떠한 영향을 미치는 지를 알아보고자 하였다.방법 : SIP 흰쥐 모델은 동일한 배에서 출생한 수컷 Spraque-Dawley 흰쥐를 사용하여 제작하였다. 각각의 사육상자에는 1정당 90mg 무게인 사료(pellet)를 자동급이장치(automatic dispenser)에서 60초당 1정씩 공급되게끔 고정된 시간 간격(fixed time 60 seconds; 이하 FT-60s)으로 하루 150분간씩 총4주간에 걸쳐서 먹이를 공급하였다. 4주후에 SIP 모델 흰쥐에게 fluoxetine(5 mg/kg)을 3주간 복강내 주사하였다.결과 : Fluoxetine을 처치한 SIP 흰쥐군이 생리식염수를 처치한 SIP 흰쥐군에 비하여 음수량과 시상하부의 실방핵 부위에서 vasopressin의 면역반응성이 뚜렷하게 감소하였다. Objective : The purpose of the present study was to examine whether fluoxetine affects vasopressin expression in SIP rats. Methods : To obtain SIP rats, male Sprague-Dawley rats were exposed to a fixed-time feeding schedule; the pellet dispenser automatically dispensed one pellet (90 mg) at each fixed-time 60 s, for 150 min training sessions conducted daily for 4 weeks. After the 4-week training session, SIP rats were treated with fluoxetine (5 mg/kg) for 3 weeks. Immunohistochemistry was carried out to determine the alteration of vasopressin expression. Results : SIP rats treated with fluoxetine showed a significant reduction in volume of water intake and in vasopressin immunoreactivity in the hypothalamic paraventricular nucleus compared to SIP rats with saline treatment. Conclusion : These results suggest that fluoxetine may affect SIP by altering the expression of vasopressin.

Objectives : Alteration of hippocampus was demonstrated in the maternal social separation(MSS) pups, separated from dams on postnatal day(pnd) 14 and placed alone. Therefore, to understand the molecular events involved in the MSS, we have initiated a search for gene profiles that are up or down-regulated in the hippocampus of MSS pups. Methods : Analysis of cDNA microarray was performed by using total RNA extracted from the hippocampus of control and MSS pups on pnd 17. Also, passive-avoidance test was demonstrated on pnd 35. Results : Up-regulation of Nedd4a was observed in the hippocampus of MSS pups. Also, MSS rats showed less elongation of latency in passive avoidance test. Conclusion : We suggest that environmental effects of MSS may be altered the neural and/or glial differentiation and synapse formation-related genes which may lead cognitive alterations in MSS rats.
Inhibitory Effects of an Aqueous Extract of Gynura procumbens on Human Mesangial Cell Proliferation

Gynura procumbens (Lour.) Merr. has been used in some parts of Southeast Asia as a folk medicine to treat kidney diseases, diabetes mellitus, and hyperlipidemia. The present work was undertaken to prove the mechanisms of G. procumbens in the management of glomerular diseases. We investigated the effect of an aqueous extract of G. procumbens on cell proliferation, DNA synthesis, and the expressions of TGF-β1, PDGF-BB, CDK1, CDK2, and CDK4 in fetal bovine serum-activated human mesangial cells (MCs). The G. procumbens extract inhibited proliferation, DNA synthesis, expressions of PDGF-BB, CDK1, and CDK2 mRNA, and expression of TGF-β1 protein in MCs. The inhibitory effect of G. procumbens on MC proliferation may be mediated by suppression of PDGF-BB and TGF-β1 expressions and the modulation of CDK1 and CDK2 expression. Therefore, G. procumbens shows promise as an adjunct therapy in preventing progressive renal diseases.
