        산업계관점 대학평가에 대한 인식 및 개선방안 연구

        The purpose of this study is to analyze the perceptions of college officials about the evaluation of university education on the perspective of industry, to make policy proposals for improvement by analyzing the preparing process of the departments that have received the evaluation of the best universities, In order to achieve the purpose of the study, we conducted literature review, survey studies, and in-depth interviews. The survey was conducted by professors related to this system, and finally the results of 51 responses were used for analysis. The contents of the survey were composed of the recognition, appropriateness, the utilization of the results, the proliferation and improvement plan of the evaluation of university education on the perspective of industry. In-depth interviews conducted 1:1 interviews with 5 professors. The evaluation of university education on the perspective of industry is generally perceived to be a bridge between industry and university education, but there is room for improvement in evaluation criteria, operating methods, and promotion strategies to public.

        Salmonella typhimurium에서 SPI2의 ssaK와 ssaJ의 발현조절

        Salmonella typhimurium에서 병독성 발현, 특히 숙주세포내로의 침입에 중요한 역할을 하는 유전자들의 집단(gene cluster)인 Salmonella Pathogenicity Island 2(SPI2)의 발현에 대한 다양한 환경요인들의 영향을 조사하였다. 이를 위해 SPI2의 주요 유전자인 ssaJ와 ssaK의 promoter를 포함하는 regulatory region을 promoterless lac operon과 융합시켜 reporter를 제조하였다. 그리고 산소농도, 삼투압, pH, 탄소원 결핍 및 glycerol 참가 등 여러 환경요인들의 변화가 이 reporter 유전자의 발현에 미치는 효과를 조사한 결과 저산소, 낮은 삼투압, 약 알칼리 등이 ssaJ와 ssaK의 발현을 증가시켰으며 위의 세조건이 함께 존재할 때 그 발현이 가장 크게 증가하는 것으로 나타났다. 그러나 탄소원 결핍이나 glycerol 첨가는 이 두 유전자들의 발현에 영향을 주지 않았다. 또한 이상과 같은 환경인자들의 효과는 S. typhimurium의 세 가지 야생형인 LT2, UK1과 SL1344 모두에서 동일한 양상을 보였다. 다른 한편 SPI1의 transcriptional activator를 암호화하는 조절유전자인 hilA의 돌연변이는 ssaJ와 ssaK의 발현에 영향을 미치지 않음도 밝혀냈다. 따라서 이상의 결과는 SPI1과 SPI2가 서로 별개의 조절계에 의해 그 발현이 조절됨을 보여준다. Salmonella Pathgenicity Island 2 plays an important role in Salmonella pathogenicity, especially invasion into host cell. We have investigated the effect of various environmental factors, such as oxygen level, osmolarity, pH, carbon starvation and glycerol addition on the expression of SPI2. For this research, we constructed the reporter plasmids, in which the promoter-less lac operons are fused with the regulatory regions (including promoter) of ssaJ and ssaK, major genes in SPI2. The study using the reporters showed that low oxygen, low osmolarity, or weak alkali conditions increased the expression levels of ssaJ and ssaK and when these three conditions exist simultaneously, the expression levels of ssaJ and ssaK are the highest. However carbon starvation and glycerol addition did not affect the expression of ssaJ and ssaK. These environmental effects on the expression levels of ssaJ and ssaK are the same in three Salmonella typhimurium wild types, LT2, UK1, and SL1344. In addition, we confirmed that the mutation in hilA, a regulatory gene encoding a transcriptional activator of SPI1, had no effect on the expression of ssaJ and ssaK. Thus, these results strongly suggest that the expressions of SPI2 and SPI1 are regulated by different control systems.

        전문대학 관광영어 관련 학과의 현장실습 개선방안에 관한 연구

        Tourism and English-related departments in junior colleges have been providing students internships for the enlargement of job opportunities in the future and for the cultivation of experts with both practical skills in the tourism field and fluent foreign language proficiency. The purpose of the study is to provide suggestions for the improvement of the student internship system of tourism and english-related departments in junior colleges. For this, literature review and a questionnaire were conducted. Based on the result of the study, the following suggestions could be included in the students` internship system. First, the internship system should be flexible in when it starts and how long it will last according to the needs of students and industry. Second, the internship system might be based on a consortium among industries to improve the students` satisfaction in the field training. Third, the program could be enlarged by connecting industry-college programs with the governments` financial support. Fourth, the program could be a multi-step approach toward industry for the students to get familiar with the industry.

        전문대학 관광외국어 전공자의 학사학위 취득 방안에 관한 연구

        Currently, tourism-related departments in junior college are attempting to establish a B.A. degree program for the cultivation of experts with both fluent foreign language proficiency and practical skills, in the tourism field. The purpose of the study is to provide suggestions for an establishment of a B.A. degree in tourism management in junior college through combining Credit Bank System courses with Advanced Major courses. For this, a questionnaire was administered. A 'Flexible, Selective, Practical Model' and the following were suggested: First, environmently friendly education rather than theory-focused, eclectic education system combining Credit Bank System courses with Advanced Major courses, and a flexible adjustment system of semester and class hours; Second, strategies for making the courses known to the applicants should be developed such as a homecoming day, development of publicity materials like brochures, leaflets, etc; Third, licences related with tourism, like the Overseas Tour Escort Licence, need to be included as national licences to be accepted as credits to reduce the students' education period.

        일본 돗토리현(鳥取縣)의 지역관광에 관한 연구 -돗토리, 구라요시, 요나고, 사카이미나토시를 중심으로-

        The purpose of this study is to examine characteristics of geographical tourism resources in Tottori prefecture, Japan. Especially, this study aims to discover geographical landscapes, urban tourism changes and major regional marketing strategies for successful tourism development based on the literature review and field survey. Tottori prefecture is located in the Chugoku region and is the least populous prefecture in Japan. Four cities are located in Tottori prefecture: Tottori, Kurayoshi, Yonago and Sakaiminato city. Tottori city is the capital of this prefecture and is home to the Tottori Sand Dunes, Japan`s only large dune system. Kurayoshi city is located in the central part, and Yonago and Sakaiminato city are located in the west of Tottori prefecture. Local governments in tottori are trying to strengthen their local competitiveness through regional tourism focused marketing strategies. The strategies are as follows; First, the Improvements of accessibility degree and demands for foreign travelers visiting Tottori prefecture could be increased by using of tranport facilities such as Kitaro airport(e.g. Asiana Airline servicing between Incheon and Yonago) and Sakaiminato seaport(e.g. DBS Cruises Ferry between Donghae and Sakaiminato). Second, the establishments of indoor museum and electric illumination winter event in the Tottori Sand Dunes could be strengthen physical geographical Images of Tottori region. Third,New attractiveness of Tottori prefecture could be created by local place-making through utilizing animation contents such as and ``Ge Ge Ge no Kitaro`` and ``Detective Conan``. Fourth, owing to tourists` visiting areasmaking use of the filming locations of Korean TV drama, the interests of tourists could be promoted by city marketing plans.

        Salmonella enterica serovar Typhimurium에서 Type III 분비장치의 표적단백질들의 분비신호 확인 및 Type III 분비장치를 이용한 Secretion Vector의 개발

        소장의 상피세포내로 세균 세포가 들어가는 과정(invasion)은 Salmonella의 감염에서 중요한 단계이다. invasion은 Salmonella type III 분비장치에 의해 분비되는 단백질들에 의해 유도된다. type III 분비단백질들은 특이하게, 일반적인 분비단백질들이 가지는 N-말단 분비 신호 펩타이드를 가지고 있지 않는 것을 알려져 있다. Yersinia에서의 최근 연구에서 type III 분비장치에 의해 인지되는 분비신호는 분비 단백질을 암호화하는 mRNA의 5'말단부의가 형성하는 2차 구조에 있을 것이라는 보고가 있다. 본 연구에서는 Salmonella type III 분비장치의 분비신호를 조사하기 위해 type III 분비단백질중 하나인 sopE를 택하여 ompR과의 translational fusion을 만들었다. translational fusion을 위해 사용된 sopE DNA절편은 프로모터와 시작 콘돈으로부터 10, 15 코돈을 포함하는 절편이다. Immunoblot으로 확인한 결과, OmpR을 포함하는 fusion 단백질이 형질전환 Salmonella 세포로부터 분비되었다. 이러한 결과는 Salmonella의 type III 분비신호가 분비단백질을 암호화하는 mRNA의 5'말단에 위치할 가능성을 제시하고 있다. 또한, 이러한 분비신호를 활용하여 유용한 외래 단백질을 세균 세포 내에서 효과적으로 생산, 분비할 수 있는 secretion vector의 원형을 개발하였다. Invasion process of bacterial cell into intestinal epithelium is important in Salmonella infection. The invasion is induced by the proteins secreted by type III secretion appratus of Salmonella. It has been known that the proteins do not have N-terminal signal peptide existing in general secreted proteins. Recent studies on Yersinia reported that secretion signal of type III appratus may lie on 5'end secondary structure of mRNA of secreted protein. In this study, we constructed translational fusion of ompR and sopE, encoding type III secretion protein of Salmonella, and observed secretion of the fusion protein for investigating the secretion signal of Salmonella type III appratus. The sopE DNA fragments of the translational fusion contain the region of promoter and from start code to tenth or to fifth code. These translational fusions indicate that type III secretion signal of Salmonella is located on 5'end of mRNA encoding secreted protein. We constructed prototype of secretion vector using this signal to produce useful foreign protein.
