Stem Cell Therapy Status in Veterinary Medicine

The stem cell therapy in veterinary medicine continues to grow both experimentally and clinically. Application of adipose-derived mesenchymal stem cell (MSC), umbilical cord blood-derived MSC, bone marrow-derived MSC and skeletal muscle-derived MSC for treatment of various diseases are currently in use for several species. However, the differential efficacies of various approaches are still being investigated. In stem cell therapy, common disease animal models include mouse, rat, rabbit, swine, canine, caprine, equine and dolphin are used. Whereas for clinical models, canine, equine are dolphin are used. We need attention and participation from the researchers of tissue engineering and regenerative medicine, and now it is necessary to develop mutual understanding and cooperation between veterinary and human medicine fields. This review is focused on some of the recent status of stem cell therapy in the veterinary medicine.

A Shihtzu (6-year-old, intact female) was referred to Konkuk University Teaching Animal Hospital for cystic calculus and hydronephrosis of right kiney. The dog was suspected to have ureteral rupture and peritonitis. Radiographic findings included cyctic calculi and right kidney enlargement. On ultrasonographic examination hyperechoic mass with severe acoustic shadowing was located in the right proximal ureter which was dilated. Hydronephrosis of the right kidney, right ureteral caculus and cytic calculus were confirmed by radiography and ultrasonography. Cystotomy for removal of calculus in the bladder and ureterotomy for removal of calculus in the right ureter were performed. Excretory urography (EU) performed one month after surgery revealed that the right ureter was homogeneously opacified and decreased to 3 to 6 mm in diameter. Surgical removal of ureteroliths was appropriate treatment for the ureteral and cystic calculi. The result suggests that ureterotomy is effective treament for the dilation and calculi of ureter.

In this paper an experimental study is presented to investigate the effect of a step edge and a stationary droplet on the dynamic behavior of impacting droplet on a wall. The main parameters are the distance from the edge and the center-to-center distance between two droplets. Photographic images are presented to show coalescence dynamics, shape evolution and contact line movement. The emphasis is on presenting the spreading length of droplet for the step edge and two coalescing droplets along their original centers. It is clarified that the droplet exhibits much different dynamic behavior depending on the location of the step edge. The momentum of impacting droplet was better transferred to the stationary droplet as the center-to-center distance between two droplets was reduced, which results in more spreading of coalescing droplet.

A diaphragm is a musculotendinous partition that separates abdominal and thoracic organs and assists in ventilation. Three dogs were presented after being hit by a car, and one dog was presented with the history of respiratory problems including intermittent dyspnea and coughing. Survey thoracic radiography, positive contrast celiography and sonography revealed traumatic diaphragmatic hernia in three dogs and congenital peritoneopericardial diaphragmatic hernia in one dog. In three dogs including a dog with congenital peritoneopericardial diaphragmatic hernia, herniorrhaphy was performed. Among the three dogs underwent surgery, two dogs with traumatic diaphragmatic hernia recovered uneventfully. However the dog with congenital peritoneopericardial diaphragmatic hernia died from pulmonary edema at 2 days after surgery. Positive-contrast ceilography and ultrasonography provides to investigate the integrity of the diaphragm. Chronic diaphragmatic hernia and concurrent injuries are considered as prognostic factors.
Ununited Anconeal Process in a Labrador Retriever Dog

A 6 month-old male Labrador retriever was presented for intermittent lameness on the left forelimb after exercise. The dog was suffering progressive lameness that had started two months before. On mediolateral radiographic view of the left elbow, proximal margin of the anconeal process was irregular. A lucent, indistinct line separating the anconeal process from the ulna was observed, when the elbow joint was flexed. The lateral approach to the elbow joint was used and the ununited anconeal process was removed. The limb was bandaged after surgery for 7 days to support soft tissue and exercises were restricted. The dog showed normal limb function 3 weeks after surgery. On a telephone conversation with the owner 18 months after surgery, the dog was reported to maintain normal function of the limb.

A 3-year-old castrated male Siberian husky was presented for evaluation after being hit by a car. On physical examination, the dog showed open-mouth, displacement of mandible and hypersalivation with blood ting. The base of tongue was transected almost 80% on the bias from right dorsal side to the left ventral side. Radiography demonstrated separation of mandible symphysis, and fracture of right condyle and vertical ramus. After debridement of the necrotic tissue, tongue apposition with simple interrupted suture was performed. Mandibular symphysis, condyle and mandibular vertical ramus fractures were fixed using pin, cerclage wire, T-plate, and K-wires. The mouth was irrigated daily using chlorhexidine after surgery. The sutures that were loose here or untied at tongue were re-sutured under sedation. The transected tongue was healed and recovered its normal movement after 6 weeks.
Belt Loop and Circumcostal Gastropexy Techniques of Canine Gastric Dilatation-Volvulus: 4 Cases

Four dogs were presented with the history of progressive abdominal distension and regurgitation. Survey abdominal radiographs revealed gastric dilatation-volvulus (GDV) with a distended, gas-filled stomach and double bubbles. The mean time from onset of clinical signs to presentation to a clinic was 3.25 hours. In three dogs, orogastric tubes were inserted and their stomachs were decompressed. However, we failed to insert the tube in the remaining one dog. Among these four dogs, gastrotomy was performed in two dogs to remove the gastric contents and to decompress the stomach additionally during surgery. The dogs with GDV were treated with belt-loop gastropexy (n=3) or circumcostal gastropexy (n=1) to prevent recurrence. Necrosis of gastric or splenic tissues was not observed during surgical intervention. All four dogs recovered uneventfully, and no recurrence was found in long term follow-up during 1~3 years.
Ventral Fixation of Atlantoaxial Joint under Fluoroscopic Guidance Using Screws in a Chihuahua Dog

A 5-year-old Chihuahua dog was evaluated for a tetraparesis. The dog was presented with a non-ambulatorytetraparesis and neck pain. Radiography demonstrated an atlantoaxial (AA) subluxation and increased distance betweenthe dorsal arch of the C1 and the dorsal spinous process of the C2. The AA joint was fixed with ventral transarticularfixation using two screws under fluoroscopic guidance. Neck brace was applied during 3 weeks post-operation. After4 weeks, the dog was fully ambulatory with improved neurological function. Postoperative radiographs confirmedreduction of the luxation and no migration of screws. The dog showed complete resolution of clinical signs withoutenabled adequate stabilization of the AA joints using ventral transarticular screw fixation which improved neurologicoutcome of the patient.

In the development of novel tissue engineering and regenerative medicine techniques in humans, preclinical studies using laboratory animal models are required to examine the pathogenic mechanisms and the potential treatment of diseases for the clinics and an indispensable step for determination of their safety and efficacy. Regrettably, the etiological pathology of diseases of spontaneous diseases may not be exactly the same despite the similarities in signs and symptoms observed in artificial laboratory animal models. Therefore, the artificial laboratory animal model is not fully suitable for studies on specific target diseases. However, the animals, especially companion animal, that have spontaneous and frequently occurring diseases due to genetic and environmental factors mostly likely mimic the naïve pathology of diseases in humans. Consequently, these species can be better subjects of preclinical studies and veterinary clinical trials in order to possibly gauge the effectiveness of the therapeutic strategies in the clinical stage. Hence, research on the spontaneous and frequently diagnosed diseases of animals predisposed by the type of breeding, age, and gender that are applicable in tissue engineering and regenerative medicine could be an important data for future advancements. In this study, we conducted a survey on the spontaneous and common diseases of canine and feline species to provide a foundation on the effectiveness and practical application of clinical studies. Therefore, this review can contribute significant development in tissue engineering and regenerative medicine in the future.
