Effects of climatic factors and picking time on availability of seeds of Chamaecyparis obtusa
손석규,김찬수,황석인,정진현,최완용 한국산림과학회 2008 한국산림과학회지 Vol.97 No.2
Seed production and germination could be influenced by some factors. The picking time andclimate factors are regarded as the elements to obtain sound seeds. We have observed the seed productivityand germination of seeds from trees of Chamaecyparis obtusa selected in a clonal seed orchard. Dependingon picking time, various shapes from liquid material, something jellied to the fully matured one weregerminal aparatuses appeared from the all seeds which had been picked from after at the end of August. The highest germination rate of about 30% was observed from the seeds picked from 20th of Septemberand 10th of October. Seed production was about two times higher in 2005 than in 2006 and the averagegermination rate was also higher in 2005. We have also analyzed the effects of climatic factors about twoconsecutive years on seed productivity. Among the climatic factors, monthly sum of temperature and ofChamaecyparis obtusa seeds.
Inverse PCR 기법을 이용한 양황철 DNA 의 Regulatory Region 의 탐색
손석규,현정오 ( Suk Gyu Son,Jung Oh Hyun ) 한국산림과학회 1998 한국산림과학회지 Vol.87 No.3
This research was conducted to identify plant regulatory regions by gene tagging method. A promoterless GUS coding sequence was introduced to Populus nigra × P. maximowiczii via Agrobacterium strains(LBA4404/EHA101), and putative transgenic poplars were selected by culturing on medium containing G418(60㎎/ℓ) and by GUS assay. Among them one positive plant was to amplify the native sequences flanking to the introduced GUS gene in plant genome by inverse PCR method and from this 730 by DNA product was obtained. After subcloning and sequencing, it has 88% homology to the Eucalyptus gunnii CAD(cinnamyl alcohol dehydrogenase) gene. The GUS gene fused with the putative promoter reinserted into poplar leaves by particle bombardment method to test the funtional promoter activity. Upon staining with X-gluc, many blue sprots appeared on the leaf segments bombarded by the chimeric gene 2-3 days, thus the isolated DNA fragment contain some possible coding region as well as a putative regulatory sequences of poplar CAD gene.
The Time for Collecting of Cryptomeria japonica Seeds
손석규,김찬수,강영재,강병옥,김효정,김창수 한국자원식물학회 2009 한국자원식물학회지 Vol.22 No.6
The time of seed collection is regarded as one of major concerns to obtain sound seeds. The physical and germinal aspects of Cryptomeria japonica D. Don (Taxodiaceae) seeds were analyzed to determine the optimum harvesting time in Korea. Cones were picked every 10 days from the 30th of July to the 30th of October in both 2005 and 2006. Seeds were collected from picked cones. Seed size and weight were not significant in two consecutive years. The 1,000-seed weight was 3.3 g for cones picked at the 18th of August and 5.3 g for cones picked at the 30th of September. The size of seeds was increased as the time of collection from the 18th of August to the 30th of September: from 19.3 mm to 21.3 mm in length and from 15.8 mm to 18.5 mm in width. Average germination rates in 2005 was 18.3% and 19.6% in 2006. The highest germination rate was 34.3% from seeds collected at the 30th of September in 2005. In 2006, the highest germination rate was 31.7% for seeds collected at the same date as the 2005 seeds. After the end of September, germination rate was decreased in both years. The results implied that the best cone picking time for Korean C. japonica seeds is around the end of September.
노각나무 성숙 접합자 배로부터 부정아 유도에 미치는 BA 및 NAA 효과
손석규,조윤진,문흥규 한국자원식물학회 2004 한국자원식물학회지 Vol.17 No.3
Zygotic embryos of Stewartia koreana Nakai were cultured to determine the effects both of BA and NAA on adventitious shoot induction. Multiple shoots (about 11 per explant) were formed when the embryos were treated with 1.0 mg/L BA alone. On the other hand, NAA appeared to inhibit shoot induction when treated with BA. Adventitious shoots looked differ in form and color by the combinations of BA and NAA treated. When both BA and NAA were present, the shoots became short and developed dark color. The highest rooting was observed at 0.5 mg/L NAA. The results could be useful for the establishment of in vitro regeneration system for Stewartia koreana Nakai.
