Streptozotocin (STZ) is a naturally occurring nitrosoamide used extensively to produce diabetes in experimental animals. Our previous study has demonstrated that i.v. administration of streptozotocin induces significant red blood cell hemolmysis in rats. Since it has been reported that the highest concentration of STZ is found in the liver, the effect of STZ in freshly isolated rat hepatocytes has been investigated. STZ treatment (10mM) did not cause significant loss of viability throughout 4 hour incubation, while high dose of STZ-induced hyperglycemia in vivo did not affect its hepatotoxicity. To investigate the mechanism of the toxicity, intracellular total glutathione level was determined. Treatment with 10mM STZ which was not toxic to hepatocytes led to complete depletion of intracellular glutathione level within 1 hour incubation. These results suggest that STZ-induced hepatotoxicity may be independent on the intracellular glutathione depletion.

Background: The Purpose of this study was to evaluate the value of Spencer technique on the range of motion (ROM), Pain, function in patients with shoulder adhesive capsulitis. Methods: subjects consisted of 30patients who were diagnosed shoulder adhesive capsulitis. All subjects are randomly assigned to 2groups: Spencer technique (ST) group (n=15), self assistive ROM exercise(S-A ROM E) group (n=15). The subjects performed an intervention program 30 minuets per day and was repeated 3 times a week for 4 weeks a total of 12 times. ROM of flexion, abduction, external rotation, internal rotation were measured using a goniometer. The visual analog scale (VAS), Shoulder pain and disability index (SPADI) were used to measure pain, functional ability. Results: In the intergroup comparisons after the intervention, ROM of flexion, abduction, internal rotation, VAS, SPADI were significantly different(p<.05). Spencer technique was more effective for improving ROM, pain, functional ability than self assistive ROM exercise. Conclusions: Our study suggest that considering Spencer technique for the patient with shoulder adhesive capsulitis. Further studies on Spencer technique are needed in the future.

Safety pharmacology studies are conducted to investigated the potential undesirable pharmacodynamic effects of a substance on physiological functions in relation to exposure in the therapeutic range and above. In the International Conference on Harmonisation (ICH), the guideline “S7A : Safety Pharmacology Studies for Human Pharmaceuticals” has been developed and reached Step 5 of the ICH process in 2001. Now the Korea Food and Drug Administration (KFDA) are going to transfer “The Guideline for General Pharmacology” into “The Guideline for Safety Pharmacology”. Safety pharmacology studies should be performed in compliance with Good Laboratory Practice (GLP). Thus, the present paper reviews the Japanese GLP guidelines for pharmaceuticals to help the conduct and inspection of safety pharmacology studies in compliance with GLP. We also reviewed the ICH guidelines “S7B revised : The Nonclinical Evaluation of the Potential for Delayed Ventricular Repolarization (QT Interval Prolongation) by Human Pharmaceuticals” and “E14 : The Clinical Evaluation of QT/QTc Interval Prolongation and Proarrhythmic Potential for Non-antiarrhythmic Drugs” to apply our drug approval systems.

Demineralized bone particle (DBP), one of the significant natural bioactive materials, has a powerful inducer of new bone growth. It has been recognized that DBP contains many kinds of osteogenic and chondrogenic cytokines as bone morphogenic protein (BMP). BMP acts as local mitogen to stimulate proliferation of mesenchymal stem cell. Bone marrow stromal cell (BMSCs) can be differentiated in culture into osteoblasts, chondrocytes, and myoblasts with controlling of the environment of cell growth. In this study, we developed the DBP loaded poly(L-lactide-co-glycolide) (PLGA) scaffolds for the possibility of the application of the tissue engineered bone. PLGA/DBP scaffolds were prepared by solvent casting/salt leaching method and characterized by porosimeter, scanning electron microscopy, and differential scanning calorimetry. BMSCs were isolated from femur of New Zealand white rabbit and cultured with osteo-medium for osteogenic differentiation. DBP impregnated PLGA scaffolds with BMSCs were implanted into the head and femur of New Zealand white rabbit to observe the effect of DBP on the osteoinduction compared with control scaffolds. Thin sections were cut from paraffin embedded tissues and histological sections were stained hematoxylin & eosin and safranin-O. It can be observed that the porosity was above 94.6% and the pore size was above 69.8 ?m. In vivo study, we could observe that similar to bone tissue region in PLGA/BMSCs and PLGA/BMSCs/DBP groups, but bone tissue region did not occur almost in control scaffolds. From these results, it seems that DBP play an important role for bone induction on BMSCs.
Hypoxanthine과 Ovarian Steroids가 생쥐난자 성숙에 미치는 영향

The influence of hypoxanthine and ovarian steroids on the meiotic maturation process of mouse oocytes was investigated for the qualified application of culture medium in in vitro fertilization(IVF). Mouse oocytes were cultured in hypoxanthine and various ovarian steroids(progesterone, estradiol-17${\beta}$ and testosterone) and their effects on the oocyte maturation had been observed. When mouse oocytes were cultured in the various concentration(1-4mM) of hypoxanthine, meiotic maturation of cumulus cell-enclosed oocytes was inhibited by presence itself, which was a dose-dependent effect in meiotic arrest of mouse oocytes. The presence of progesterone, estradiol-17${\beta}$ and testosterone have made the mouse oocyte mature properly. Meanwhile maturation of cumulus cell-enclosed oocyte was severely inhibited by 3 hoursculture in the media of progesterone supplemented with hypoxanthine. However the continuous presence lasting 24 hours of progesterone even supplemented with hypoxanthine had got rid of the inhibition of oocytes maturation. Not only estradiol-17${\beta}$ supplemented with hypoxanthine but also testosterone supplemented with hypoxanthine exert the severe inhibition of the maturation of cumulus cell-enclosed oocytes for 3-hours culture. However the continuous presence lasting 24 hours of estradiol-17${\beta}$ and testosterone even supplemented with· hypoxanthine had relieved the inhibition of oocytes maturation. These results make us suggest that hypoxanthine inhibits the mouse oocyte maturation, particularly markedly in conjunction with ovarian steroids for short period, which indicated some sort of the synergistic inhibitory retationship between the ovarian steroids and hypoxanthine.

Lactoferrin is a multifunctional protein that is found in milk, neutrophils, and other biological fluids, and its receptors have also been identified in the central nervous system. Recently, it was reported that bovine milk-derived lactoferrin (BLF) produced analgesia via a μ-opioid receptor-mediated response in the spinal cord. However the precise mechanism of this analgesic effect is remains unclear. In Randall-Selitto paw pressure study, each single administration of morphine (10 mg/kg) and BLF (50, 100 and 200 mg/kg) induced analgesia, however, NMDA receptor antagonist MK-801 (0.1, 0.2 and 0.3 mg/kg), inhibited analgesia induced by BLF(100mg/kg). Intracerebroventricular infusion (I.C.V) of N-methyl-D-aspartic acid (NMDA) (0.3 μg/8.0 μl/hr/day), as a NMDA receptor agonist, reversed inhibition of MK-801 (0.3mg/kg) on analgesia induced by BLF (100mg/kg). These results suggest that BLF have analgesic effect, through NMDA receptor activation.
