Effects of B(a)P on preimplantation mouse embryos in vitro were studied. Preimplantation mouse embryos were exposed to a concentration of 0.3, 1, 3 and 10 $\mu$M B(a)P for 72 hrs. The toxicological effects of B(a)P were evaluated by morphological observation of embryos up to the blastocyst stage, and by measuring DNA, RNA and protein synthesis by radioactive precursor incorporation. At 1 $\mu$M B(a)P did not affect preimplantation development but interfered with hatching and ICM formation. Suppressing effect of ICM formation was dose dependent. At the eight cell stage, the developmental rate was decreased at above 3 $\mu$M of B(a)P. At the blastocyst stage, attachment and trophoblast outgrowth were diminished at the 10 $\mu$M of B(a)P and ICM formation was decreased at 1 $\mu$M of B(a)P. Inner cell number of blastocyst was decreased dose dependently. So, number of ICM was one of the most sensitive and toxicological end point. The RNA incorporation rate of 0.1 $\mu ^3$H-uridine was dosedependent and the protein incroporation of 0.5 $\mu Ci ^{35}$S-methionine showed a significant decrease after 48 hrs. But the DNA incorporation rate of methyl-$^3$H thymidine was not affected. Our results suggested that B(a)P did not affect the DNA replication but transcription was inhibited by dose dependent manner. There delay of development during the blastocyst stage was mainly due to the inhibition of RNA synthesis followed by protein synthesis.
Captafol과 Ethanol이 혈액상의 생화학적인 변화에 미치는 영향

Captafol (1H-isoindole-1.3 (2H)-dione, 3a,4,7,7a-tetrahydro-2-1.1.2.2-tetrachloroethylthio), is widely used as broad spectrum fungicide in agriculture and industrial application. N-sulfenylphthalimide fungicides are relatively low toxic to human and animal by oral administration. However it was reported that the fungicides have carcinogenic and mutagenic potentials, in subacute and chronic toxicity experiments. As mentioned the above, there are many experimental reports on the toxicity of captan and folpet, the toxicological researches on captafol are few, therefore in this experiment we examined the subacute toxicity of captafol which is widely used as fungicide in biochemical parameters in serum. The experimental results were summerized as follows: 1) In captafol administered group, the body weight was significantly decreased but the biochemical parameters such as AST, ALT, LDH, BUN, TG in serum were remarkably elevated in comparison to that of control group. 2) Captafol plus ethanol administered group, serum AST, ALT, BUN were elevated in comparison to that of captafol treated group.
방사선 조사 인삼이 랫드의 태자와 신생자의 발달 및 모체기능에 미치는 영향에 관한 연구

Korean ginseng products have been fumigated with ethylene oxide(EO) for sterilization and prolongation of storage periods. However, there had been controversies indicating that consumption of EO treated foods might cause harmful effects in human. In Korea, the use EO gas for sterilization of food was banned in 1991. Since then, irradiation technique has been developed as an alternative. This study was carried out to evaluate the safety of irradiated ginseng on peri- and postnatal developmental toxicity in rats. Either EO gas fumigated or gamma-irradiated ginseng was administered to pregnant Wistar rats by oral gavage from gestational day 16 to postnatal day 21. The amount of irradiation used in this study was 5, 10 and 30kGy, respectively. There were no treatment related changes of dams in deaths, clinical signs, and parturition. No treatment related changes in food consumption, body/organ weight and lactation of dams were observed. Also, no F1 fetuses in external abnormality, physical development, reflex/sensory functions and behavioral development were found. The results of this study showed that gamma-irradiated ginseng, up to 30 kGy, has no adverse effects on the peri- and postnatal development of rats.

Either EO gas fumigated or Gamma-Irradiated ginseng was administered to pregnant Wistar rats by oral gavage from gestational day 7 to 17. The amount of irradiation used in this study was 5, 10 and 30 kGy, respectively. There were no treatment related changes of dams in deaths, clinical signs, food consumption and body/organ weight. No treatment related changes in implantation ratio, litter size, sex ratio and body/oragan weight of fetuses were observed. Also, no F1 fetuses with external, visceral, head and skeletal malformation were observed. The results of this study showed that Gamma-Irradiatedg ginseng, up to 30 kGy, has no adverse effects on fetal development of rats.
