The random amplified polymorphic DNA analysis of four sea cucumbers, Holothuria pardalis, H. monacaria, Stichopus japonicus and Cucumaria chronhjelmi from Geomun-do was performed. Ten primers of 10-mer arbitrary primer kit were used for PCR. Similarity was calculated by Jaccard’s equation. The band pattern was different in three of ten primers. The similarity between S. japonicus and C. chronhjelmi, H.pardalis and C. chronhjelmi, S. japonicus and H. monacaria, S. japonicus and H. pardalis, H. monacaria and C. chronhjelmi, and H. pardalis and H. monacaria was 0.42, 0.55, 0.63, 0.68 and 0.85, respectively.

To achieve a high viability by minimizing thawing damage, ice crystallization must be reduced in the process of thawing. Thermophysical properties of suspensions of Porphyra seriata, Undaria pinnatifida, and Sargassum fulvellum were measured to determine the optimal thawing conditions in the thawing process for species conservation of sea algae. The range of final thawing temperature was –2.49 ~ –0.50°C at thawing speed 1.25°C/min and 2~3°C at 1.5~2.0°C/min, and thawing was completed at temperatures above 3.0°C, enabling the removal process of cryoprotectant. The optimum thawing speed with the minimum thawing time was 0.75~1.0°C/min, and the latent heat of thawing was Undaria pinnatifida 128.37 J/g, Porphyra seriata 118.53 J/g, and Sargassum fulvellum 94.42 J/g, which can be useful for the design and manufacture of algae cryopreservation system. From the change of enthalpy, entropy and free energy according to each thawing rate, the optimum thawing rate for recrystallization temperature pan and viability improvement is suitable over Porphyra seriata, Sargassum fulvellum, Undaria pinnatifida 0.75°C/min, 1.0°C/min, 1.5°C/min respectively. From the results of the thawing process, the optimal method of freezing-thawing process of seedlings for aquaculture will be set up to increase the viability and contribute to the aquaculture industry by being used for growth in the farm. In addition, the results of this study will contribute to the establishment of gene bank, seed bank, and seed vault for various kinds of sea algae

해역이용협의에 따른 협의서 검토현황과 연안이용형태를 분석한 결과, 공유수면 점 사용은 공작물설치와 해수 취배수, 공유수면 매립은 산업단지조성, 항만 어항개발과 도로건설의 점유율이 높았고, 점 사용은 서해에서 그리고 매립은 남해에서 우세하였다. 지역별로는 전남, 경남지역과 인천 경기지역에서 이용행위가 많았다. 해역이용협의 검토량은 2008년이 전년도에 비해 약 200건 이상 증가하였고, 협의과정에서 준설토 해양투기, 매립, 준설, 해수 취배수, 바다골재채취와 규사채취에 대해 보완요청이 주로 이루어졌다. 연안이용은 대부분 해양환경기준 I등급 또는 II등급의 지역에서 집중되었고, 특히, 법령상 해양 규제지역인 특별관리해역에서는 항만 어항개발과 관련된 매립과 공작물설치, 국립공원에서는 공작물설치와 호안정비가 많았다. 또한, 수산자원보호구역에서는 호안정비, 공작물설치와 해수의 취배수 행위가 많이 이루어진 것으로 평가되었다. 해역이용협의제도의 제반적인 합리성과 신뢰성을 높이는 방법 등 개선방안을 제도운영 및 정책적인 측면과 협의서 작성과 관련한 해양환경영향평가 측면으로 구분해서 정책제언을 제시하였다. The review of statements and coastal development characteristics in South Korea were diagnosed by the consultation on the coastal area utilization in Korea. The occupation and the use of public water were dominated by installation of coastal structure and seawater supply and drainage whereas the reclamation areas were predominantly utilized for developing industrial complex, harbor, fishing port and road. The occupation and the use were dominant in western coast, but reclamation was dominant in southern coast of Korea. The number of utilization cases was particularly greater in Jeonnam, Gyeongnam, and Gyeonggi Province including Incheon. The number of the statements reviews increased by more than 200 cases in 2008 compared with 2007, and most of them were the simple statements. The statements related to ocean disposal of dredged sediment, reclamation, dredging sediment, seawater supply and drainage, and marine sand mining were submitted for review. Coastal utilization was especially active in the regions of seawater quality criteria I and II. In particular, special management sea areas designated under the relevant marine regulations were utilized mostly by reclamation for developing harbor, fishing port and coastal structure. Development activities in national parks comprised primarily structure installation and coastal maintenance. In the fisheries resources protection areas, 40% of the total cases accounted for coastal maintenance, 31% for structure installation, and 16% for seawater supply and drainage. In addition, alternative plans for the improvement in policies and system of marine environmental impact assessment were suggested to enhance the function and confidence of the consultation on the coastal area utilization in Korea.

We evaluated the status and problems of golf course developments in the southern coast of Korea. It`s adjacent waters supports nursery and fishing grounds for commercially-important fisheries species, and various sites are designated and protected as marine protection area(MPA), fisheries reserve, or dean area(blue belt) for producing shellfish. We proposed key assessment items for environmental impact assessment(EIA) and checklists in selecting golf course locations. For the protected areas, we suggest that it is essential to limit golf course establishment while setting a minimal distance from the coast to secure a buffer zone for mitigating the environmental impacts. To efficiently utilize existing regional coastal management plans, it is necessary to diagnose how a golf course development will potentially modify geomorphology and scenery, amplify pollutant loads from non-point sources, and disrupt the functions of coastal ecosystem. Especially, continued monitoring and assesssing input loads of hazardous materials originating from agricultural chemicals should be obligatory. Finally, measures for improving the QA/QC analysis were discussed to enhance reliability of environmental data with respect to golf courses and adjacent coastal waters.

The two-step cooling method have been mostly used for the preservation of algae, with cryoprotectants in freezing medium. However, in order to minimize the freezing damages and to get higher survival rate, effect of ice crystal on algae must be reduced in the process of first-step freezing. As size and distribution of ice crystals depend on cooling rate and recrystallization of ice crystal decrease the survival rate, optimal conditions in first-step freezing of cryopreservation must be established. In this study optimal cooling rate and final freezing temperature of Pyropia seriata, Undaria pinnatifida, Sargasum fulvellum were investigated. The mixed suspension of 10% DMSO and 0.5 M sorbitol in 50% seawater used as cryoprotectant. Exothermic temperature, freezing time, latent heat of freezing, temperature range of freezing process were measured by differential scanning calorimetry. Results obtained revealed the optimal cooling rate and final freezing temperature of P. seriata as 1.25oC/min, −30.5oC, U. pinnatifida, 1.0oC/min, −27.8oC and S. fulvellum, 1.0oC/min, −27.1oC respectively.
