RAPD 와 핵 리보솜 DNA ITS2 염기서열에 의한 한국산 전나무속 식물의 유전적 변이 및 계통유전학적 분석

We analyzed the genetic variation and phylogenetic relationship among fourteen taxa of Genus Abies distributed in Korea using random amplified polymorphic DNA (RAPD) and internal transcribed spacer (ITS) 2 sequences of the nuclear ribosomal DNA (rDNA). RAPDs were used for identification of selected taxa by amplification using single 10-mer primers of arbitrary sequence. Fourteen taxa were clearly classified with 10 arbitrary primers, which generated 102 useful polymorphic bands that were evaluated according to the presence or absence of the fragments of equal size. the genetic distance between A. koreana for. chlorocarpa and A, koreana for. rubrocarpa was 0.127, showing the closest among any other pairs. By comparing the base sequences in the ITS2 region of the nuclear rDNA, genetic relationship was assesed among selected taxa. It showed twenty six sites of paint mutation (base substitution) in 243 base pairs (bp) of ITS2 region. The G+C contents of ITS2 region were ranged from 58.3% to 61.7%. The value of nucleotide divergence among selected taxa was ranged from 0.4% to 9.8%. These results showed that RAPD and ITS2 sequence analysis was a useful tool for elucidating phylogenetic relationship and selection of specific marker in Abies species.
Ribosomal DNA 의 ITS 염기서열에 의한 자생 왕벚나무와 재배 왕벚나무의 유연관계

It has long been disputed whether Prunus yedoensis was originated from Mt. Halla in Cheju-do or Cheju-do originated taxon is the same as the cultivated one from Japan. By comparing the base sequences in the ITS 1, ITS 2 and 5.8s coding region of the nuclear ribosomal DNA, genetic relationship was assesed between P. yedoensis Matsumura-Native and P. yedoensis Matsumura-Cultivar. There showed three sites of point mutation (base substitution) and seven insertions or deletions in 236 bp of ITS 1 but no differences in 164 bp of 5.8s coding region. On the other hand, it was shown that four sites of point mutation (base substitution) and seven insertions or deletions in 233 bp of ITS 2. The content of (G+C) in ITS 1 region was 59.4% in natives and 60.2% in cultivar and in ITS 2, 64.0% in natives and 64.3% in cultivar, respectively. But the 5.8s coding region was identical as 56.3% in each taxon. The idea that P. yedoensis Matsumura-Native would be different from the P. yedoensis Matsumura-Cultivar would be strongly substantiated from the similar results obtained in this studies with those of previous studies.

An experimental study for a partial discharge of a pipe bushing and bare/insulated bus bar set has been carried out to understand the fundamentals of the partial discharges occurred in the bushing set, which, however had not been published in the open articles. As a result, the details of partial discharges occurred and their transitions and propagations on the dielectrics were understood and analyzed experimentally and theoretically. The i-v characteristics of the tested bushing set showed very different from the conventional nonuniform field ones. The total current was decreased with the increased source voltage, which is increased conventionally. The ozone monitoring method has been proposed as the partial discharge checking means, which, however, was one of the very convenient and easy methods to check the occurrence of the partial discharge at the working place. The influences of atmosphere temperature(10∼40℃) arid relative humidity(20∼95%) around the bushing set were also investigated, and, it is found that they were not effected dominantly to the leakage current and the taking place partial discharge in the bushing set.
