      • TLC/HPTLC에서 측정된 자외/가시부 스펙트럼의 표준화 및 검색

        To improve the identification power of TLC/HPTLC the in situ reflectance spectra obtained directly from plates with commercial scanner are used. The spectrum normalization should be carried out prior to comparing and searching the spectra from library for the identification of compounds. Because the reflectance does not obey the Lambert-Beer's law, there arise some problems in normalization. These problems could be solved to some extent by normalizing the spectra with regression methods. The spectra are manipulated with the regression function of a curve obtained from the correlation plot. When the parabola was used as the manipulating function, the spectra were identified with the accuracy of 97% and this result was better than that of conventionally used the point and area normalization method.

      • 모세관 전기영동법을 이용한 1,2,4-트리메틸벤젠 대사체의 분석

        방향족 탄화수소인 tnmethylbenzene (TMB)은 그 사용량이 늘어갈 뿐 아니라 직업적으로 폭로되는 양도 증가하고 있으므로 생물학적 모니터링 및 흡수, 대사, 배설에 관한 연구가 중요시되고 있다. 일반적으로 TMB는 간의 산화효소에 의해 하나의 메틸기가 산화되고 이것은 glycine과 포합되어 배설되는 것으로알려져 있다. 본 연구에서는 1,2,4-TMB의 대사체를 합성하고, 모세관의 전기영동법으로 분석할 수 있는 방법을 개발하였다. 모세관 전기영동법으로 흰쥐의 뇨 중에서 1,2,4-TMB의 대사체인 3,4, 2,4, 2,5-dimethylbenzoic acid 및 3,4 2,4 2,5-dimethylhippuric acid를 분석하기 위하여 내경 75㎛, 총길이 35cm (검출기까지 29cm)인 용융실리카 모세관을 15℃로 유지하면서 양단에 10kV의 전압을 걸어주고, 전해질로는 15mM β-CD, 3% 2-프로판올을 포함하는 01m 인산완충액 (pH 7)을 사용하였으며, 검출신호는 UV 210nm와 254nm에서 동시에 모니터링하였다. 뇨 시료의 분석 결과 배설된 1,2,4-TMB의 대사체의 상대량은 3,4-이성질체가 56.&%, 2,4-이성질체가 30.5%, 2,5-이성질체가 12.8%였다. 이 방법은 노동자의 뇨 분석에도 적용될 수 있을 것으로 생각된다. The metabolites of 1,2,4-trimethylbenzene (TMB) were synthesized and determined by capillary electrophoresis (CE). The optimum conditions of CE for the separation and determination of 3,4-, 2,4-, 2,5-dimethylbenzoic acid and 3,4-, 2,4-, 2,5-dimethylhippuric acid from the rat wine were as following: the fused silica capillary(75μm i.d. X 36 cm length, 29 cm to detector) was used and kept at 15℃ The applied voltage was 10kV and compounds were detected at UV 210 nm and 254 nm. The running electrolyte was 0.1 M phosphate buffer (pH 7) contalI1ing 15 mM of β -CD and 3% of 2-propanol. The relative amount of the metabolite of 1,2.4-TMB in the rat urine was 56.7% of 3,4-isomer, 30.5% of 2,4-lsomer and 12.8% of 2,5-isomer. This method can be applied to the analysis of TMB-metabolites in human wine.

        패턴분석법에 의한 대황의 비교 연구

        Three species, such as Rheum palmatum L., R. tanguticum Maxim. and R. officinale Baillon are recognized as the source plants of Rhei Rhizoma in Korean Pharmacopeia. However, other herbal sources such as R. undulatum L. and Rumex crispus L. have been often misused as Rhei Rhizoma. A pattern analysis method to discriminate Rhei Rhizoma in Korean Pharmacopeia from other herbal plants using HPLC and TLC chromatograms was developed. The multivariate peak data of the chromatograms of methanol extracts of Rhei Rhizoma were used for hierarchical clustering analysis, principal components analysis and similarity calculation. Besides of the statistic analysis, TLC patterns of samples could be used as criteria of the discrimination. The developed pattern analysis method was specific and could be readily utilized for comprehensive evaluation of Rhei Rhizoma.

        The trace elements in liver, kidney and testicle of rats were analysed by Inductively Coupled Plasma-Mass Spectrometer(ICP-MS). Ten elements(Cr, Ni, Cu, Zn, Mo, Cd, Pd, Bi, Th, U) were determined simultaneously and the detection limit of each elements was lower than ppb level.

      • 크로마토그래프법을 이용한 사삼(沙蔘) 및 유사생약의 패턴분석 비교 연구

        Adenophora Radix is the dried root of Adenophora triphylla var. japonica Hara or the other crude drugs belong to genus Adenophora. However, other crude drug like Codonopsis lanceolata or C. pilosula were often misused as Adenophora radix. A pattern analysis method to evaluate the Adenophora Radix and similar crude drugs were carried out with the methanol and ethylacetate extract by HPLC and TLC. The components were separated by HPLC on ODS column with gradient elution of solvent A (10% acetonitrile) and B (70% acetonitrile). The multivariate peak data of the chromatograms of extracts of samples were used for hierarchical clustering analysis, principal components analysis and similarity calculation. The similarities like Euclidean distance, cosine and correlation for the methanol extract of A. tetraphylla and A. remitiflora showed good correlation with A. triphylla var. japonica, indicating that the similarity could be used as a chemical taxonomy. The genus Adenophora and genus Codonopsis were well classified by the hierarchical clustering method for the cluster analysis. The developed pattern analysis method was specific and could be readily utilized for comprehensive evaluation of Adenophora Radix.

      • HPLC-ECD를 이용한 흰쥐 뇌의 선조체 중 신경전달물질의 동시분석시 최적 조건

        흰쥐 뇌의 선조체로부터 catecholamine, indoleamine 및 대사산물을 동시분석할 수 있는 간편한 방법을 제시하였다. 기기는 ECD를 장착한 HPCL를 사용하였고, 고정상인 C_18 컬럼에서, 이동상은 10mM citric acid, 0.13mM Na_4EDTA, 0.58mM SOS를 함유하는 35mM sodium acetate buffer(pH 3.4) : MeOH=85:15가 최적이었다. 이 때 컬럼온도는 30℃였다. 이 조건으로 dopamine(DA), 3, 4 -dihydroxyphenyl acetic acid(DOPAC), homovanilic acid(HVA), 5-hydroxyindole acetic acid(5-HIAA), serotonin(5-HT) 및 noradrenaline(NA)을 미량까지 분석할 수 있었고, 검출한계는 각 성분에 대해 1회 주입당 2~10pg이었다. 이 방법으로 흰쥐의 주령 및 성별에 따른 선조체내 생체아민과 대사산물의 함량을 비교하였다. 7주령 쥐에서 암컷은 수컷보다 DA와 5-HT의 함량이 유의성 있게 높은 것으로 나타났으며, 주령이 증가함에 따라 암컷에서 DOPAC의 함량이 유의적으로 증가하였다. A simple, efficient and sensitive method was described for the simultaneous determination of catecholamine, indoleamine and related metabolites from the homogenates of the rat brain striatum by HPLC-ECD. The optimum mobile phase on a reverse phase C_18 column was 35mM sodium acetate buffer(included 10mM citric acid, 0.13mM Na_4EDTA, 0.58mM SOS, pH 3·4):MeOH=85:15. The column temperature was 30℃. Dopamine(DA), 3, 4-dihydroxyphenyl acetic acid(DOPAC), homovanilic acid(HVA), 5-hydroxyindole acetic acid(5-HIAA), serotonin(5-HT) and noradrenaline(NA) could be separated and analysed to very small amount. The detection limits of this method were 2~10pg per injection for all components. The effects of age and sex of rat on the contents of the catecholamines and their metabolites in rat brain striatum were studied. The levels of DA and 5-HT contents of the 7 week old female rats were higher than those of the 7 week old male rats. As the age of rat increases, the contents of DOPAC increased significantly.

      • HPLC-ECD를 이용한 흰쥐의 뇌 중 감마아미노부티르산 및 글루탐산의 정량

        A sensitive and efficient assay method was applied to determine the level of glutamic acid (GA) and γ-aminobutyric acid (GABA) in frontal cortex and hippocampus of rat administrated with ethanol and drugs. The compounds were derivatized with ο-phthalaldehyde (OPA) and 2-mercaptoethanol for precolumn analysis. The condition for the simultaneous determination of GA, GABA and beta-aminobutyric acid (BABA) by high performance liquid chromatography with electrochemical detection was reverse phase C_18 column as stationary phase, 0.1M phosphate buffer containing 0.1mM Na_4EDTA: methanol=55:45 (v+v) pH 3.8 as mobile phase and 0.7V electrode voltage. The stability of reaction product of GA, GABA and BABA with OPA could be increased by adding the same volume of polyethylene glycol 400 to reaction mixture. The GABA level in frontal cortex of rat was significantly decreased by the administration of picrotoxin and diazepam, but it was significantly increased by the administration of red ginseng total saponin, N-methyl-D-glucamine and (-)-deprenyl.

First, we are concerned with the explicit distribution function of the partial sum □n when the random variables are independent and has identically uniform distribution, next, we determine the n-fold convolution distribution of □n when the identically distributed condition is not satisfied.
