새로운 LacZ Gene Fusion 및 Gene Cartridge Vector의 개발에 관한 연구

In this study, a new lacZ gene vector, plasmid PGEM3-lacZ was constucted, which could be used for the study of the gene regulation and the function of proteins. Plasmid pMC1871 was digested with restriction endonuclease Pst I and Sma I to obtain a DNA fragment with the lacZ-gene(3091 bp). Recombinant plasmid pGEM3-lacZ was constructed by inserting the lacZ gene DNA(Pst I-Sma I cut DNA; 3091 bp) of pMC1871 into the Pst I and Hinc Ⅱ sites of plasmid pGEM3. After then, Recombinant plasmid pGEM3-lacZ DNA was transformated into E. coli HB 101. pGEM3-lacZ DNA/HB 101 was screened on the NZY plate with ampicillin and X-gal(5-Bromo-4-Chloro-3-Indolyl-β-D-Galactoside) by selecting blue colonies. The restriction map of pGEM.3-1acZ DNA was constructed by the following restriction enzymes :Bgl I, Xbal I, Bam HI, EcoR I, Hind Ⅲ, Kpn I, Pst I, Sma Ⅰ, Sal Ⅰ. From the restriction map, the lacZ DNA was confirmed to be present at the expected position in the plasmid pGEM3-lacZ. pGEM3-lacZ/HB101 gives about 10 times more lacZ DNA than pMC 1871/HB101.

본고는 2009년 3월부터 진행해 온 한문 고전 『번역용례사전』 구축을 위한 연구의 중간성과를 소개하고 향후의 연구 방향을 점검하기 위한 것이다. 여기서는 조선 선조시기에 이루어진 교정청본 『論語諺解』와 율곡 이이가 언해한 『論語栗谷先生諺解』를 대상으로 그동안 연구에서 추출된 의미 있는 결과들을 소개하였다. 우리는 이 번역용례사전이 한문 원문에 문법적으로 접근함으로써 그 구조를 보다 쉽게 파악할 수 있게 하고, 조선시대 사대부들의 경전 해석과 그에 사용된 번역어를 통해 초학자의 학습과 전문 번역사의 번역어 구사에 도움이 되고, 특히 허사의 다양한 쓰임을 종합적으로 살필 수 있으며, 조선 사대부의 경학적 성과를 파악해 볼 수 있을 것이라는 기대를 갖는다. 그러나 두 종의 『논어언해』 모두가 자유로운 번역어를 구사하기보다는 가능한 원문을 훼손하지 않는 원천 언어 지향적인 번역을 했기 때문에 우리말 번역어가 상대적으로 빈약하며, 번역되지 않은 원문 그대로의 단어들이 많다는 점은 하나의 한계가 될 수 있을 것이다. 이 점은 보다 많은 자료를 종합하여 다양한 사례들을 통해 어느 정도 극복될 수 있을 것이며, 원문의 한자어를 그대로 쓰는 경우라도 두 책이 표현 방식을 달리하고 있는 점은 전문 번역사의 번역어 구사에 유연성을 제공할 수 있을 것으로 보인다. This paper aims to present the interim report about the study on the construction of 《Translation Example-based Dictionary》for Chinese classics which has been worked on since March of 2009 and to examine the research direction for the future. In this paper, the significant results drawn from the research up to now are reported on the edited Gyojeongcheong version of 《The Annotation of The Confucian Analects》, which was done during the era of King Seonjo of the Joseon Dynasty, and 《The Yulgok's Annotation of The Confucian Analects》annotated by Yulgok Yi Yi. We expect that this translation example-based dictionary with grammatical approach to original Chinese text allows easier comprehension of the structure, helps beginners to learn and professional translators to command translation words, enables us to comprehensively examine the various applications of function words, and lets us understand the achievements of noblemen of the Joseon Dynasty in the area of the study of Confucian classics. However, as both the two kinds of the annotation of the Confucian Analects pursued the original language oriented translation that destroys the original text as little as possible rather than commands liberated translation words, the Korean translation words are relatively scarce, and the fact that there are many original words which were not translated can be one of limitations. This will be overcome to a certain degree through various examples after putting more data together, and it seems even when the original Chinese characters were used as it were, the aspect that the two books used different ways of expression can provide the flexibility in the commanding of translation words for professional translators.

‘Tamnara’ was originated from the cross between ‘Campbell Early’ (Vitis labruscana) and ‘Himrod Seedless’ (Vitis sp.) in grape breeding program for disease resistance and cold hardness by NHRI in 1981. ‘Tamnara’ had a budburst on 22 April, flowering on 4 June, and ripening on 7 September, similar to ‘Campbell Early’ at Suwon. The berry weight was 7.5 g, 2 g bigger than that of ‘Campbell Early’. It had a total soluble solids (TSS) of 17.2%, about 3% TSS higher than ‘Campbell Early’, and low acidity (0.39%). The clusters were cone-shaped (conical) and had circular, juicy, and black-skin colored berries with abundant blooms. Clusters with moderately dense berry setting required no cluster thinning and resulted in low incidence of fruit cracking. ‘Tamnara’ vines had good cold hardness, resistance to downy mildew and anthracnose. It is recommended that ‘Tamnara’ be planted as replacement for ‘Campbell Early’, the leading table grape in Korea.

‘Heukgoosul’ (4X), resulted from the cross between ‘Golden Muscat’ (2X, Vitis sp.) and ‘Pione’ (4X, Vitis sp.) in grape breeding program by NHRI in 1988, was preliminarily selected in 1995. After regional adaptability tests under the name of ‘Wonkyo RA-07’ at seven sites during 1996∼2000, it was finally selected in 2000. Although most tetraploid cultivars had a berry shattering problem, ‘Heukgoosul’ showed a low incidence of berry shattering, resulting in a good berry set compared to ‘Kyoho’. ‘Heukgoosul’ had a budburst on 21 April, flowering on 3 June, and fruit maturity 15 September (almost same as ‘Kyoho’ at Suwon), and it was considered a midseason cultivar. The mean berry weight was 14.4 g, about 2.6 g bigger than ‘Kyoho’, and mean soluble solids were 18.4Bx (similar to ‘Kyoho’). The skin color was black with abundant bloom and the flesh firmness was soft and juicy. It was recommended that it be pruned to 6 to 12-node canes, 2 to 4 nodes shorter than ‘Kyoho’, because it is not as vigorous. ‘Heukgoosul’ may be a substitute for ‘Kyoho’, due to its large sized berries with high quality and several characteristics for facilitating vineyard management such as good cold hardness, good disease tolerance.
