萬金湯 및 加味萬金湯이 배양 척수후근신경절 세포에 미치는 영향

The purpose of this study is to examine the toxic effects caused by xanthine oxidase/hypoxanthine (XO/HX) and the effects of herbal extracts such as Mangeum-tang (萬金湯: MGT) and Gamimangeum-tang (加味萬金湯: GMGT) on the treatment of the toxic effects. The results of these experiments were XO/HX, an oxygen radical-generating system, decreased the survival rates of the cultured cells on XTT assay, the amount of DNA syntheses, and the amount of neurofilaments, and increased c-fos positive cells. MGT and GMGT have the efficacy of increasing the survival rates of the cultured cells by increasing the amount of neurofilaments and DNA synthesis and decreasing the c-fos positive cells damaged by XO/HX. From the above results, it is suggested that MGT and GMGT have marked efficacy as a treatment for the damages caused by the XO/HX-mediated oxidative stress. And MGT and GMGT are thought to have certain pharmacological effects.

Tritiated thymidine autoradiography has been applied to the duodenal progenitor population (crypts of Lieberkuhn) in one day postnatal male chick to determine 1) the cell cycle of the progenitor cell pool, 2) the characteristics of DNA synthetic index, and the 3) mitotic index. Available amounts of colchicine were injected into the animals before sacrifice to arrest the dividing cell nucleus at metaphase. The results of this investigation are as follows: 1) The generation time was 14 hours, 2) DNA synthetic index was 23.3%, and 3) The mitotic index was 1.8%. The cause of shortening of DNA synthetic phase and the lengthening of post mitotic phase were discussed.

Authors have experienced 104 cases of the intracranial aneurysms with subarachnoid hemorrhage, who were proved to have aneurysm by cerebral angiography for past 8 years. Aneurysm surgery under controlled hypotension has shown far better result than that of surgery under hypothermia, carotid ligation or conservative treatment. Of the 104 cases, the site distribution of aneurysms and the result of various treatment are analyzed. Followings are the results. 1. Anterior communicating aneurysm was 44 per cent of the cases and it was most common site of the intracranial aneurysm. 19 per cent of aneurysms located on internal carotid artery at the posterior communicating artery, and middle cerebral artery aneurysm was 16%. The site distribution of aneurysms in Korean has shown marked difference from the reports of other countries. 2. Of 40 cases, who received conservative therpy, 15 cases (37.5%) were expired, and most of them were dead due to rebleeding from aneurysms. 3. The direct attack of aneurysms through intracranial approach was performed under moderate hypothermia till 1970, and controlled hypotension was introduced thereafter. In 49 cases of intracranial aneurysm surgery, 7 (14.2%) were expired following surgery. 4. Mortality of the carotid ligation for 15 cases of the intracranial aneurysms was 40 per cent. Cerebral ischemia or rebleeding was cause of death. 5. Intracranial aneurysm surgery under hypotension was carried out in 16 cases, and one case (6.2%) was expired. Induced hypotension and careful identification of aneurysm and neighbouring structures by magnifying operating glasses promoted operative result in aneurysm surgery.
Regulatory Effect of Inflammatory Reaction by Anglicae Dahuricae Radix

Anglicae dahuricae Radix (ADR), the dried roots of Angelica Dahurica Benth et Hook (Umbelliferae), is a traditional herbal medicine used to treat colds, headache, skin diseases such as acne and possess analgesic, antipyretic and drainage effects. In the present study, the author evaluated the effect of ADR on regulation of inflammatory reaction. ADR reduced the ear-swelling responses derived from compound 48/80 in dose-dependent manner significantly. ADR inhibited the PMA plus A23187-induced productions of IL (Interleukin)-8, IL-1β, granulocyte macrophage-colony stimulating factor (GM-CSF) and tumor necrosis factor (TNF)-α from human mast cells (HMC)-1. In addition, ADR blocked PMA plus A23187-induced ERK1/2 phosphorylation. Ⅰ suggest that ADR regulates inflammatory reaction through inhibition of inflammatory cytokines such as IL-8, IL-1β and GM-CSF.
백강잠 전탕액이 Hydrogen Peroxide에 의해 손상된 배양 척수운동신경세포에 미치는 효과

To evaluate the effect of Batryticatus Bombycis(BB) water extract on cultured mouse spinal motor neuron which was inhibited by hydrogen peroxide (H_2O_2)-induced oxygen radicals, MTT assay, NR assay, TBARS assay and SRB assay were carried out after the cultured mouse spinal motor neuron were preincubated with various concentrations of BB water extract for 3 hours prior to exposure of H_2O_2. H_2O_2, a oxygen radical, decreased the survival rate of the cultured mouse spinal motor neuron cells on NR assay and MTT assay. BB water extract have efficacy of decreasing a amount of lipid peroxidation increased by H_2O_2 in mouse spinal motor neuron and increasing total protein synthesis activity decreased by H_2O_2 in mouse spinal motor neuron. From the above results, It is concluded that BB has marked efficacy as a treatment for the damages caused in the H_2O_2-mediated oxidative process.

Objectives & Methods:To evaluate the effect of Bujabanha-tang water extract on cultured mouse spinal sensory neurons which was inhibited by hydrogen peroxide (H2O2)-induced cytotoxicity, MTT assay, NR assay and neurofilament enzymeimmunoassay(EIA), LDH activity assay were carried out after the cultured mouse spinal sensory neurons were pre-incubated with various concentrations of Bujabanha-tang water extract for 3 hours prior to exposure of hydrogen peroxide.Results:H2O2, an oxygen radical, decreased the survival rate of the cultured mouse spinal sensory neuron cells on NR assay and MTT assay.Bujabanha-tang water extract has efficacy of increasing neurofilament decreased by hydrogen peroxide and decreasing LDH activity increased by hydrogen peroxide in cultured mouse spinal sensory neurons.Conclusions:From the above results, it is suggested that Bujabanha-tang has marked efficacy as a treatment for the damages caused in the H2O2-mediated oxidative process.
