      • The Effects of Fermented <i> Laminaria japonica</i> on Short-Term Working Memory and Physical Fitness in the Elderly

        <P>Considering the neuroprotective and antioxidant potential of fermented<I> Laminaria japonica</I> A. (FST), the purpose of the present study is to establish whether FST may be considered a viable, efficacious supplement that can be administered in later life to offset neurodegenerative conditions associated with aging. Forty senior subjects participated in a randomized, double-blind, and placebo-controlled study. Two groups were formed, one FST group (n = 32, 72.35 ± 5.54 yrs) and one placebo (CON) (n = 28, 74.57 ± 5.69 yrs), which received 1.5 g/day of FST for 6 weeks. Subjects were asked to abstain from any regular exercise. In order to analyze short-term memory, a variety of neuropsychological tests were implemented. Body composition, physical fitness evaluations, antioxidant function, and inflammatory markers were also included in the analyses pre- and posttest. We demonstrated that FST significantly improved neuropsychological test scores, including higher scores in the K-MMSE, numerical memory test, Raven test, and iconic memory, compared to the CON group. Shorter test trial times in the 6-minute walk test were observed in the FST group (<I>p</I><0.001 and<I> p</I><0.05, trials 1 and 2, respectively). FST also significantly increased antioxidant activity of GPx, GSR, and SOD, reduced the production of TBARS, and lowered 8-oxoDG levels. The present study highlights the potential widespread application of FST in protecting against the degenerative effects of aging on short-term memory and physical function. Neuropsychological evaluation indicates that FST may provide a protective mechanism against cognitive impairment associated with dementia. Neuromuscular integrity and physical function are typically compromised in aging and dementia patients; thus, whether by stimulation of muscle-related growth factors or an increase in serum BDNF, FST supplementation may act to preserve physical function in the elderly. The bioactive constituents of FST such as GABA and fucoidan acting to provide improvements in antioxidant activity following FST supplementation may protect against progressive degeneration purportedly caused by reactive oxygen species.</P>

        GABA-enriched fermented Laminaria japonica improves cognitive impairment and neuroplasticity in scopolamine- and ethanol-induced dementia model mice

        BACKGROUND/OBJECTIVES: Fermented Laminaria japonica (FL), a type sea tangle used as a functional food ingredient, has been reported to possess cognitive improving properties that may aid in the treatment of common neurodegenerative disorders, such as dementia. MATERIALS/METHODS: We examined the effects of FL on scopolamine (Sco)- and ethanol (EtOH)-induced hippocampus-dependent memory impairment, using the Passive avoidance (PA) and Morris water maze (MWM) tests. To examine the underlying mechanisms associated with neuroprotective effects, we analyzed acetylcholine (ACh) and acetylcholinesterase (AChE) activity, brain tissue expression of muscarinic acetylcholine receptor (mAChR), cAMP response element binding protein (CREB) and extracellular signal-regulated kinases 1/2 (ERK1/2), and immunohistochemical analysis, in the hippocampus of mice, compared to current drug therapy intervention. Biochemical blood analysis was carried out to determine the effects of FL on alanine transaminase (ALT), aspartate transaminase (AST), and triglyceride (TG) and total cholesterol (TC) levels. 7 groups (n = 10) consisted of a control (CON), 3 Sco-induced dementia and 3 EtOH-induced dementia groups, with both dementia group types containing an untreated group (Sco and EtOH); a positive control, orally administered donepezil (Dpz) (4㎎/㎏) (Sco + Dpz and EtOH + Dpz); and an FL (50 ㎎/㎏) treatment group (Sco + FL50 and EtOH + FL50), orally administered over the 4-week experimental period. RESULTS: FL50 significantly reduced EtOH-induced increase in AST and ALT levels. FL50 treatment reduced EtOH-impaired step-through latency time in the PA test, and Sco- and EtOH-induced dementia escape latency times in the MWM test. Moreover, anticholinergic effects of Sco and EtOH on the brain were reversed by FL50, through the attenuation of AChE activity and elevation of ACh concentration. FL50 elevated ERK1/2 protein expression and increased p-CREB (ser133) in hippocampus brain tissue, according to Western blot and immunohistochemistry analysis, respectively. CONCLUSION: Overall, these results suggest that FL may be considered an efficacious intervention for Sco- and EtOH-induced dementia, in terms of reversing cognitive impairment and neuroplastic dysfunction.

        Changes in Muscle Biomarkers Following Swimming Exercise in Streptozotocin-induced Rats

        [PURPOSE] The purpose of this study was to assess the effect of swimming exercise in diabetic atrophy by monitoring changes in biomarkers associated with musculoskeletal growth and abnormalities. [METHODS] Ninety-two male Sprague-Dawley rats were randomly divided into four groups: control plus exercise group (CO+Ex), control and no exercise group (CO+No Ex), diabetes plus exercise group (DM+Ex) and diabetes and no exercise group (DM+No Ex). Swimming exercise was implemented for 60 min/day, 5 day/week for a total of eight weeks. Muscle strength and atrophy biomarkers as well as muscle morphology were compared at the first, second, fourth and eighth week of experimentation. [RESULTS] ACE levels were significantly higher in the DM groups (p<.05), however swimming exercise did not significantly affect ACE levels. Irisin levels were significantly higher in the DM and DM+Ex groups compared to the CON, in week one (p<.05), and greater in the DM+Ex group at week two compared to CON+Ex (p<.05), but swimming did not significantly affect irisin levels over the eight-week period in DM rats. In terms of muscle damage, Myl3 expression was unchanged in the CON groups, but significantly higher in the DM+Ex group compared to the CON+Ex group (p<.05). AST was found to be in higher levels in the DM+No Ex group, compared to CON and CON+Ex (p<.05); and DM+Ex group, compared to the CON+Ex group at weeks one, four and eight (p<.05). Tibialis anterior muscle fibers were thicker in the DM+Ex group compared to DM+No Ex, but only at week four (p<.05). Otherwise, no other notable differences were recorded in muscle morphology. [CONCLUSIONS] These results suggest that swimming exercise alone may not be sufficient to induce muscle strength and/or protective effects against muscle atrophy associated with diabetes mellitus. However, this mode of exercise may be considered safe by not compounding the deteriorative effects of this disease.

        Effects of γ-aminobutyric acid-enriched fermented sea tangle (Laminaria japonica) on brain derived neurotrophic factor-related muscle growth and lipolysis in middle aged women

        This study evaluated the effects of γ-aminobutyric acid (GABA)-enriched fermented sea tangle (GFST), as a functional food, on brain derived neurotrophic factor (BDNF)-related muscle growth and lipolysis, in a sarcopenic obesity high-risk group. Twenty-one middle-aged women (53-63 y) participated in this randomized, double-blind, placebo controlled study. Participants ingested either 1,000 mg of GFST (n = 10) or a sucrose placebo (CON) (n = 11) everyday, for 8 weeks. Subjects were asked to abstain from any regular exercise. Fasting venous blood samples, body composition and muscular strength were measured before and after supplementation period. Collectively, we demonstrated that GFST significantly decreased total fat mass and triglyceride in body composition, as well as significantly increasing serum BDNF (p < 0.001), angiotensin converting enzyme (p < 0.001), human growth hormone and insulin-like growth factor-1 levels (p < 0.05 and p < 0.05, respectively) accompanied by increased total lean mass (p < 0.01). Furthermore, the reported improvements in total work, knee extension and flexion at 60° s-1 (p < 0.05), and peak torque normalized to body weight of knee flexion at 60° s-1 (p < 0.05), support an ergogenic effect of GABA associated with increased growth factor levels. The use of GFST, as a functional food ingredient, to elicit anti-obesity effects and stimulate the release of muscle-related growth factors with increasing serum BDNF levels may provide a protective intervention for age-related degeneration such as sarcopenic obesity.

        Fermented Laminaria japonica improves working memory and antioxidant defense mechanism in healthy adults: a randomized, double-blind, and placebo-controlled clinical study

        A randomized, double-blind, and placebo-controlled clinical study was used to determine the cognitive functions related to working memory (WM) and antioxidant properties of fermented Laminaria japonica (FLJ) on healthy volunteers. Eighty participants were divided into a placebo group (n = 40) and FLJ group (n = 40) that received FLJ (1.5 g/day) for 6 weeks. Memory-related blood indices (brain-derived neurotrophic factor, BDNF; angiotensin-converting enzyme; human growth hormone, HGH; insulin-like growth factor-1, IGF-1) and antioxidant function-related indices (catalase, CAT; malondialdehyde, MDA; 8-oxo-2’-deoxyguanosine, 8-oxo-dG; thiobarbituric acid reactive substances, TBARS) were determined before and after the trial. In addition, standardized cognitive tests were conducted using the Cambridge Neuropsychological Test Automated Batteries. Furthermore, the Korean Wechsler Adult Intelligence Scale (K-WAIS)-IV, and the Korean version of the Montreal Cognitive Assessment (Mo- CA-K) were used to assess the pre and post intake changes on WM-related properties. According to the results, FLJ significantly increased the level of CAT, BDNF, HGH, and IGF-1. FLJ reduced the level of TBARS, MDA, and 8-oxo-dG in serum. Furthermore, FLJ improved physical activities related to cognitive functions such as K-WAIS-IV, MoCA-K, Paired Associates Learning, and Spatial Working Memory compared to the placebo group. Our results suggest that FLJ is a potential candidate to develop functional materials reflecting its capability to induce antioxidant mechanisms together with WM-related indices.

        Clinical and Imaging Findings of Neonatal Seizures Presenting as Diffuse Cerebral White Matter Abnormality on Diffusion-Weighted Imaging without any Structural or Metabolic Etiology

        Purpose Some patients with neonatal seizures show diffuse, symmetric diffusion-restricted lesions in the cerebral white matter. The aim of this study was to describe clinical and imaging findings of patients with neonatal seizures who had diffuse, symmetric diffusion-restricted lesions without any structural or metabolic etiology. Materials and Methods A total of 56 neonates aged less than 1 week underwent brain magnetic resonance imaging (MRI) for evaluation of seizures from November 2008 to February 2017. After excluding 43 patients, 13 patients showed diffuse white matter abnormality on diffusion- weighted imaging. Initial and follow-up clinical and MRI findings were analyzed retrospectively. Results All 13 patients were born at full term. Among the ten patients who underwent a stool test for viruses, six were positive for rotavirus and one for astrovirus. MRI revealed diffuse, symmetric diffusion- restricted lesions distributed along the cerebral white matter, thalami, and midbrain variably. Conclusion Diffuse, symmetric diffusion-restricted lesions involving the cerebral white matter can be seen in patients with neonatal seizures without any structural or metabolic etiology. Rotavirus is commonly but not exclusively detected in these patients. Nevertheless, viral infection-associated encephalopathy should be considered for patients with characteristic clinical and MRI findings.
