황산마그네슘이 인간제대정맥내피세포의 VEGF와 Caspase-3에 미치는 영향

목적 : 황산마그네슘이 혈관 내피 세포의 caspase-3와 VEGF의 농도에 어떠한 영향을 미치는지를 알아보기 위하여 본 연구를 하였다. 방법 : 임신 말기에 계획된 제왕절개를 시행한 태아의 제대로부터 정맥내피세포(HUVECs)를 추출하여 TNF-α로 처리한 군과 처리하지 않은 두 군으로 나누어 황산마그네슘을 각 농도 별(0, 4, 8, 12 mM)로 가하여 배양한 후 배양 상청액으로 VEGF에 대한 ELISA assay를, caspase-3에 대하여 colorimeter assay를 시행하여 그 농도를 측정하였다. 결과 : HUVECs에서의 VEGF의 발현 농도는 TNF-α로 처리한 경우가 처리하지 않은 경우에 비하여 유의적으로 증가하였다. TNF-α를 가한 경우는 황산마그네슘의 농도에 따른 VEGF의 농도에 유의적인 차이가 없으나 TNF-α를 가하지 않은 경우 황산마그네슘의 농도가 4 mM 및 8 mM인 경우에 가장 낮은 농도로 발현되었다. caspase-3의 농도는 TNF-α와 황산마그네슘의 농도에 따른 유의적인 차이가 없었다. 결론 : HUVECs에서의 VEGF의 발현은 TNF-α에 의하여 증가되나 황산마그네슘의 농도 변화에 따른 영향은 없고, TNF-α에 의한 자극이 없는 경우는 황산마그네슘의 치료적 농도에서 그 발현은 감소되는 반면 caspase-3의 발현은 TNF-α와 황산마그네슘의 영향을 받지 않는 것 같다. Objectives : To determine the effects of magnesium sulfate on Caspase-3 and Vascular endothelial growth factor (VEGF) of human umbilical vein endothelial cells (HUVECs) under presence or absence of Tumor necrosis factor (TNF-α). Methods : HUVECs were isolated from normal term umbilical cords and cultured in several physiolo-gically relevant concentrations of magnesium sulfate with or without exposure of TNF-α. The concentrations of VEGF and caspase-3 were estimated by colorimetric assay and ELISA assay, respectively. Results : The concentration of VEGF in HUVECs significantly increased in the presence of TNF-α compared with in the absence of TNF-α. However, the concentration of VEGF did not show significant difference in several concentrations of magnesium sulfate concentrations with addition of TNF-α and it showed the lowest concentration under 4 mM and 8 mM of magnesium sulfate concentration without addition of TNF-α. The concentration of caspase-3 in HUVECs did not show statistically significant difference with the addition of TNF-α and magnesium sulfate. Conclusion: TNF-α induce HUVECs to stimulate the VEGF, and magnesium sulfate might not have the effects on the expression of VEGF with addition of TNF-α. However, the concentration for treatment of magnesium sulfate inhibits the expression of VEGF without addition of TNF-α. Magnesium sulfate might not have an effect of the expression of caspase-3 with or without addition of TNF-α.
Nonobstetric surgery during pregnancy and perioperative fetal monitoring

we present here a 2 cases of nonobstetric surgery during pregnancy and intraoperative fetal monitoring. Case report: A 34-year-old Korean nullipara woman, with her 31 weeks 3days of normal pregnancy with no disease history, was referred to our outpatient clinic for recent frequent seizure and cognitive disorder. Solid tumor involving left thalamus and left lateral ventricle was detected on her brain MRI. She underwent small craniectomy with tumor biopsy and bilateral V-P shunt. Perioperatively, cardiotocography (CTG) was done to monitor fetal status. CTG showed a lack of beat-to-beat variability with normal fetal heart rate during the whole operation procedure. We observed the pattern and postoperative CTG showed recovery of variability. Pathological examination revealed a brain glioblastoma grade IV and at gestational age 32weeks and 4days cesarean section was done with necessity of maternal early radiation therapy. Although the premature baby had mild neonatal hyperbilirubinemia, her general status is tolerable till now. 2nd case, A 39-year-old Korean nullipara woman, with 30weeks pregnancy with no underlying disease, came to emergency center for severe low abdominal pain. Flat abdomen x-ray was suspicious of intestinal volvulus and emergency laparotomy was done. Post operative diagnosis was sigmoid colon volvulus and operation title was Hartman operation. During the operation, we checked fetal heart rate with ultrasound probe instead of CTG because it was abdominal operation. Postoperative recovery was uneventful and full term delivery was done with gestational age of 39weeks 3 days. The mother and baby were discharged with no complication. Conclusion:Sometimes there are clinical situations in which urgent surgery in pregnancy is necessary. Intraoperative fetal heart rate (FHR) monitoring of the infant during non-obstetric surgery is controversial and we must interprete intraoperative CTG correctly and not take a mistake of doing unnecessary cesarean section
Multi-residue analysis of 63 pesticides in livestock products using GC-MS/MS

The aim of this research is to develop a rapid and easy multi-residue analytical method for livestock products (beef, pork, chicken, milk, egg and fat) using gas chromatography-tandem mass spectrometry (GC-MS/MS). To determine multi-residue in livestock products, samples were prepared using QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) based method. Residual pesticides were extracted with acetonitrile followed by addition of anhydrous magnesium sulfate (MgSO₄) and sodium chloride (NaCl). Then, the extracts were cleaned up using MgSO₄, octadecyl (C18) and primary secondary amine (PSA). The method was validated by evaluating the following parameters: selectivity, linearity, accuracy, precision, limit of detection (LOD) and limit of quantification (LOQ). The matrix-matched calibration curves were constructed using 8 levels (2.5-100 ug/ml) and coefficient of determination (r²) was above 0.98. Average recoveries spiked at three levels (0.01, 0.02 and 0.1 mg/kg) were most in the range of 70~120% and standard deviation values were less than 32%, which is under the Codex guideline (CODEX CAC/GL 40). Therefore, the developed method is suitable and could be usefully used the multi-residue analysis in livestock products.

The widespread use of sulfonamides can result in the residue of sulfonamides in the foods of animal origin that are the major concerns of consumers and regulatory bodies due to their adverse reaction such as the development of antibiotic resistance. A rapid and efficient multi‑residue analytical method was developed to screen and confirm 31 sulfonamides in livestock samples in a single run, using ultra‑high‑performance liquid chromatography com bined with comprehensive mass spectrometric approaches. In this study, a novel sample preparation procedure was used, based on a modified QuEChERS method (Quick, Easy, Cheap, Effective, Rugged, and Safe). The linearity, sensitivity, accuracy, and precision of the method were validated according to the Codex guidelines. The response of the detector was linear for each target compound over a wide concentration range, with a correlation coeffi cient (r2) greater than 0.98. The limit of detection (LOD) and limit of quantification (LOQ) ranged from 0.3 to 5 ng g−1 and from 1 to 19 ng g−1 and the average recoveries (%) for three laboratories ranged from 85 to 109% with a CV (n = 5) below 22%. The applicability of this screening method was verified using real livestock samples. The proposed analyti cal method achieves identification and quantification of target sulfonamides at trace levels in a short analysis time. None of the samples contained residues that exceeded the maximum residue limit (MRL).

The corpus callosum consists of white fibers connecting the cerebral hemispheres. Agenesis of the corpus callosum is an uncommon congenital anomaly which is easily diagnosed in the postnatal period by ultrasound and computed tomographic scan or MRI, but its prenatal sonographic diagnosis is difficult because of fetal head positioning and limiting trans-axial scans. We experienced a case of agenesis of the corpus callusum with chromosomal anomaly. The prenatal sonographic findings are ventricular abnormalities that demonstrated dilatation of lateral ventricles and disproportionate enlargement of the occipital horns, which were suggestive findings for the corpus callosal agenesis. We could confim the diagnosis of the corpus callosal agenesis with chromosome anomaly by postnatal MRI and chromosome analysis.

To determine multi-residue in livestock products, residual pesticides were extracted with acetonitrile followed by addition of anhydrous magnesium sulfate (MgSO<sub>4</sub>) and sodium chloride (NaCl). Then, the extracts were cleaned up using MgSO<sub>4</sub>, octadecyl (C<sub>18</sub>) and primary secondary amine (PSA). To verify the applicability of the developed method, selectivity, linearity, accuracy, precision, limit of detection (LOD) and limit of quantification (LOQ) was evaluated. The coefficient of determination (R<sup>2</sup>) was above 0.98 within the concentration range from 2.5 to 100 μg/L. The average recoveries were most in the range of 70∼120% and standard deviation values were less than 32%, which is satisfied the Codex guideline (CODEX CAC/GL 40). These results suggest that the developed method can be applied to the real market livestock products successfully. Therefore, the developed method is suitable and could be included in the Korean Food Code for enabling the safety management of pesticides.
