        Design of a variable valve hydraulic lift system for diesel engine

        A continuous variable valve lift (VVL) mechanism for a diesel engine is proposed in this research. The hydraulic mechanism, whichconsists of a driving plunger, a driven plunger, a hydraulic cylinder, and a hydraulic oil tank, is the key part of the VVL mechanism. Simulation is conducted to study the relationship between maximum valve lift and rotation angle of driving plunger. Calculation resultsindicate that the maximum valve lift decreases with increasing rotation angle. A prototype was manufactured and successfully tested in asingle-cylinder diesel engine. The experiments, which were conducted at different rotation angles of the driving plunger, validate theaccuracy of the calculation results. The difference between experimental valve lift curve and cam lift curve is considered the result of oilleakage. Rotating the driving plunger is an effective method to regulate valve lift. Working stability of the VVL mechanism is validatedthrough a multi-cycle operation. Experimental results indicate that the VVL mechanism is effective and reliable for realizing continuousVVL in engines.

        Effectiveness of Ionic Polymer Soil Stabilizers on Warm Frozen Soil

        Warm frozen soil has characteristics such as low shear strength and high compressibility. To improve its mechanical strength, the ionic polymer soil stabilizers (IPSS): named XRH stabilizer and acid cation exchange resin (CER) were adopted for reinforcing the warm frozen soil from the Beiluhe region of the Qinghai–Tibetan Plateau. To evaluate the effects of IPSS on warm frozen soil and analyze the mechanisms responsible, measurements were made of the physical, chemical and mechanical properties of warm frozen soil, both untreated and following treatment with IPSS. The addition of IPSS to the soil resulted in a significant decrease in the plasticity index, freezing temperature, and cation–exchange capacity of soil. The warm frozen soil were reinforced with XRH and CER, the cohesion increased by 87.9% and 43.1%, respectively, and the compressibility decreased by 44.5% and 41.1%, respectively. The trends of mechanical strength and unfrozen water content of reinforced warm frozen soil were the same, with extreme values being observed in response to treatment with an IPSS content of about 1% (optimal proportion). In addition, the curing mechanism of IPSS on warm frozen soil was analyzed. Overall, applying IPSS to the soil was an effective and feasible measure for reinforcing a warm frozen soil foundation.

        RhoGDI2 induced malignant phenotypes of pancreatic cancer cells via regulating Snail expression

        Background: Rho GDP dissociation inhibitor 2 (RhoGDI2) has been shown to contribute to the aggressive phenotypes of human cancers, such as tumor metastasis and chemoresistance. Objective: This study aimed to assess the effects of RhoGDI2 on tumor progression and chemoresistance in pancreatic cancer cells. Methods: The expression of RhoGDI2 in pancreatic cancer cells was detected by Western blot analysis. Gain-of-function and loss-of-function approaches were done to examine the malignant phenotypes of the RhoGDI2-expressing or RhoGDI2-depleting cells. The correlation between RhoGDI2 and Snail was also analyzed. Results: Differential expression of RhoGDI2 protein in pancreatic cancer cell lines was identified. Gain-of-function and loss-of-function experiments showed that RhoGDI2 induced the malignant phenotypes of pancreatic cancer cells, including proliferation, migration, invasion, and gemcitabine (GEM) chemoresistance. The upregulation of RhoGDI2 stimulated the expression of Snail, resulting in the altered expression of epithelial marker E-cadherin and mesenchymal marker Vimentin, which were characteristics of the tumorigenic activity of epithelial-mesenchymal transition. The expression of RhoGDI2 and Snail was upregulated in clinical tumor samples, and higher expression of RhoGDI2 or Snail was significantly associated with poor patient survival in pancreatic ductal adenocarcinoma (PDAC). Conclusion: The findings indicated that RhoGDI2 promoted GEM resistance and tumor progression in pancreatic cancer and that RhoGDI2 might be a potential therapeutic target in patients with PDAC.

        Neoadjuvant Chemotherapy-Guided Bladder-Sparing Treatment for Muscle-Invasive Bladder Cancer: Results of a Pilot Phase II Study

        Purpose Reduced quality of life after cystectomy has made bladder preservation a popular research topic for muscle-invasive bladder cancer (MIBC). Previous research has indicated significant tumor downstaging after neoadjuvant chemotherapy (NAC). However, maximal transurethral resection of bladder tumor (TURBT) was performed before NAC to define the pathology, impacting the real evaluation of NAC. This research aimed to assess real NAC efficacy without interference from TURBT and apply combined modality therapies guided by NAC efficacy. Materials and Methods Patients with cT2-4aN0M0 MIBC were confirmed by cystoscopic biopsy and imaging. NAC efficacy was assessed by imaging, urine cytology, and cystoscopy with multidisciplinary team discussion. Definite responders (≤ T1) underwent TURBT plus concurrent chemoradiotherapy. Incomplete responders underwent radical cystectomy or partial cystectomy if feasible. The primary endpoint was the bladder preservation rate. Results Fifty-nine patients were enrolled, and the median age was 63 years. Patients with cT3-4 accounted for 75%. The median number of NAC cycles was three. Definite responders were 52.5%. The complete response (CR) was 10.2%, and 59.3% of patients received bladder-sparing treatments. With a median follow-up of 44.6 months, the 3-year overall survival (OS) was 72.8%. Three-year OS and relapse-free survival were 88.4% and 60.0% in the bladder-sparing group but only 74.3% and 37.5% in the cystectomy group. The evaluations of preserved bladder function were satisfactory. Conclusion After stratifying MIBC patients by NAC efficacy, definite responders achieved a satisfactory bladder-sparing rate, prognosis, and bladder function. The CR rate reflected the real NAC efficacy for MIBC. This therapy is worth verifying through multicenter research.
