Antioxidative and Hepatoprotective Effects of Magnolol on Acetaminophen-induced Liver Damage in Rats

Acute liver failure (ALF), an often fatal condition characterized by massive hepatocyte necrosis, is frequently caused by drug poisoning, particularly with acetaminophen (N-acetyl-p-aminophenol/APAP). Hepatocyte necrosis is consecutive to glutathione (GSH) depletion and mitochondrial damage caused by reactive oxygen species (ROS) overproduction. Magnolol, one major phenolic constituent of Magnolia officinalis, have been known to exhibit potent antioxidative activity. In this study, the anti-hepatotoxic activity of magnolol on APAP-induced toxicity in the Sprague-Dawley rat liver was examined. After evaluating the changes of several biochemical parameters in serum, the levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) were elevated by APAP (500 mg/kg) intraperitoneal administration (8 and 24 h) and reduced by treatment with magnolol (0.5 h after APAP administration; 0.01, 0.1, and 1 μg/kg). Histological changes around the hepatic central vein, lipid peroxidation (thiobarbituric acid-reactive substance/TBARS), and GSH depletion in liver tissue induced by APAP were also recovered by magnolol treatment. The data show that oxidative stress followed by lipid peroxidation may play a very important role in the pathogenesis of APAP-induced hepatic injury; treatment with lipid-soluble antioxidant, magnolol, exerts anti-hepatotoxic activity. Our study points out the potential interest of magnolol in the treatment of toxic ALF.

Objective: Predicting disease relapse and early intervention could reduce symptom severity. We attempted to identify potential indicators that predict the duration to next admission for an acute affective episode in patients with bipolar I disorder. Methods: We mathematically defined the duration to next psychiatric admission and performed single-variate regressions using historical data of 101 patients with bipolar I disorder to screen for potential variables for further multivariate regressions. Results: Age of onset, total psychiatric admissions, length of lithium use, and carbamazepine use during the psychiatric hospitalization contributed to the next psychiatric admission duration positively. The all-in-one found that hyperlipidemia during the psychiatric hospitalization demonstrated a negative contribution to the duration to next psychiatric admission; the last duration to psychiatric admission, lithium and carbamazepine uses during the psychiatric hospitalization, and heart rate on the discharge day positively contributed to the duration to next admission. Conclusion: We identified essential variables that may predict the duration of bipolar I patients’ next psychiatric admission. The correlation of a faster heartbeat and a normal lipid profile in delaying the next onset highlights the importance of managing these parameters when treating bipolar I disorder.
Effects of Acupuncture at Pai-Hui on the Deficit of Memory

Acupuncture has been introduced as one of the complementary therapies in traditional Chinese medicine. In addition to the analgesic effect, acupuncture also has anti-inflammatory, anti-allergic and immunomodulatory effects. Activation of the Bai-Hui by acupuncture was further shown to correct loss of consciousness, as well as to relieve mental abstraction, sluggish speech and hysteria. In this study, we investigated the attenuating effect of Pal-Hui acupuncture on scopolamine (SCOP)-induced or cycloheximide (CXM)-induced impairment of the passive avoidance response in rats. Our preliminary results indicated that acupuncture at Pal-Hui before or immediately after the training trial could significantly attenuate SCOP-induced impairment in rats. The effects were better than those treated before the right-room retention trial. Interestingly, the identical procedure could also significantly attenuate the CXM-induced impairment, but not those treated before the retention trial. The correction effect of acupuncture on CXM-induced impairment was equivalent to that of piracetam treatment. Acupunctures at Pal-Hui 15,30 or 60 min before the training trial could he all effective to attenuate CXM-induced impairment, and its efficacy paralleled acupuncture duration. Furthermore, we demonstrated the mechanism of the attenuating effects of acupuncture at Pal-Hui on CXM-induced memory consolidation deficits by combined with SCOP or p-chloroamphetamine(PCA). The attenuating effects of acupuncture at Pal-Hui was significantly antagonized by PCA but not SCOP. These results suggest acupuncture at Pal-Hui might mainly affect the memory storage process and had preventive and immediate therapeutic effects on the CXM-induced impairment and its efficacy paralleled acupuncture duration. The preventive effect of acupuncture at Pal-Hui on CXM-induced impairment is significantly reduced by serotonergic 5-HT releaser. However, the detailed mechanism of acupuncture on memory function and the differential mechanism of acupuncture on memory function an analgesia must be further investigated.
A Novel Transparent Microwave Thin Film Coating Technique Applied to Dual‑Band Antennas

In this paper, we propose a novel transparent microwave thin film coating technique and discuss its application in planardual-band antennas (0.9/5.55 GHz). We developed a new process for activating the nano-alignment thin film from high tolow resistivity (from 1.96 to 1.29 × 10−4 Ω cm) and from partial to full transparency (from 55 to 83% transmittance) within150 s. The platform of the activation process comprises a periodic electrode, an optical microscope, and an alternating currentsignal generator. The periodic electrode can effectively rearrange the nano-alignment thin film into an ordered arrangement,which enhances the properties of the thin film in the microwave frequency range. A high-transparency and low-resistivitydual-band antenna is designed and fabricated using the proposed microwave thin film coating technique. The dual-bandantenna has operating bandwidths of 740–960 and 5030–7030 MHz and potential applications in transparent electronicssuch as wearable devices and intelligent cars.

The mitogen-activated protein kinase (MAPK) phosphatase-1 (MKP-1) belongs to the MAPK cascades which are central to cell proliferation and apoptosis. The carcinogenic role of MKP-1 has been reported in many types of cancer but it has rarely been investigated in breast cancer. The present study was designed to evaluate the MKP-1 mRNA expression and its possible regulation by methylation of MKP-1 promoter in the model of several breast cancer cell lines and tissues as well as controls. Our data demonstrate MKP-1mRNA expression significantly decreased in five breast cancer cell lines compared to breast controls (P <0.01). Using the methylation-specific PCR (MSP) analysis,the unmethylated reaction (U) is dominant in both normal cell lines and benign breast tumors (100% vs. 86.2%), whereas the methylated reaction (M) is dominant in both breast cancer cell lines and invasive breast tumors (100% vs. 57.2%). In terms of methylation ratio (M/M+U), methylation level in MKP-1 promoter is significantly higher in the invasive breast tumor tissues (n = 152) than in benign breast tumor tissues (n = 29)(P < 0.0001). Assessing the methylation ratio of the promoter of MKP-1 gene to diagnose the breast malignancy (invasive vs. benign), the area under the receiver-operating characteristic (ROC) curve was 0.809(95% CI: 0.711-0.906, P < 0.001). The best performance for this prediction has a sensitivity of 76.32% and a specificity of 82.76% at the cutoff value of 0.38. Taken together, we firstly demonstrated that the promoter methylation of MKP-1 gene is a potential breast cancer biomarker for breast malignancy.

It has been postulated that grana stacking and destacking are mediated by the alteration of the surface charge density on the thylakoid membrane, which is regulated by the LHCII phosphorylation and dephosphorylation cycle. Insect-induced cecidomyiid galls, transformed from Machilus thunbergii Sieb & Zucc. (Lauraceae) leaves, are totally deficient in the pigment–protein complexes CPI and A1 of PSI and AB1 and AB2 of PSII. Although the galls are deficient in LHCII complex, grana stacking is normal. Our data suggest that the LHCII complex may not be the only factor responsible for grana stacking of thylakoid membranes.

<P>Chemical vapour deposition of two-dimensional materials typically involves the conversion of vapour precursors to solid products in a vapour-solid-solid mode. Here, we report the vapour-liquid-solid growth of monolayer MoS2, yielding highly crystalline ribbons with a width of few tens to thousands of nanometres. This vapour-liquid-solid growth is triggered by the reaction between MoO3 and NaCl, which results in the formation of molten Na-Mo-O droplets. These droplets mediate the growth of MoS2 ribbons in the 'crawling mode' when saturated with sulfur. The locally well-defined orientations of the ribbons reveal the regular horizontal motion of the droplets during growth. Using atomic-resolution scanning transmission electron microscopy and second harmonic generation microscopy, we show that the ribbons are grown homoepitaxially on monolayer MoS2 with predominantly 2H-or 3R-type stacking. Our findings highlight the prospects for the controlled growth of atomically thin nano-structure arrays for nanoelectronic devices and the development of unique mixed-dimensional structures.</P>

Angiotensin II (Ang II) is a very important cardiovascular disease inducer and may cause cardiac pathological hypertrophy and remodeling. We evaluated a Chinese traditional medicine, alpinate oxyphyllae fructus (AOF), for therapeutic efficacy for treating Ang II-induced cardiac hypertrophy. AOF has been used to treat patients with various symptoms accompanying hypertension and cerebrovascular disorders in Korea. We investigated its protective effect against Ang II-induced cytoskeletal change and hypertrophy in H9c2 cells. The results showed that treating cells with Ang II resulted in pathological hypertrophy, such as increased expression of transcription factors NFAT-3/p-NFAT-3, hypertrophic response genes (atrial natriuretic peptide [ANP] and b-type natriuretic peptide [BNP]), and Gαq down-stream effectors (PLCβ3 and calcineurin). Pretreatment with AOF (60–100 μg/mL) led to significantly reduced hypertrophy. We also found that AOF pretreatment significantly suppressed the cardiac remodeling proteins, metalloproteinase (MMP9 and MMP2), and plasminogen activator, induced by Ang II challenge. In conclusion, we provide evidence that AOF protects against Ang II-induced pathological hypertrophy by specifically inhibiting the insulin-like growth factor (IGF) II/IIR-related signaling pathway in H9c2 cells. AOF might be a candidate for cardiac hypertrophy and ventricular remodeling prevention in chronic cardiovascular diseases.
