<P>Summary</P><P>Apicidin is a cyclic tetrapeptide produced by certain isolates of <I>Fusarium semitectum</I> and has been shown to inhibit Apicomplexan histone deacetylase. An apicidin-producing strain (KCTC16676) of the filamentous fungus was mutated using an <I>Agrobacterium tumefaciens</I>-mediated transformation, resulting in 24 apicidin-deficient mutants. Three of the mutants had a T-DNA insertion in a gene that encodes a non-ribosomal peptide synthetase (NRPS). Results of sequence, expression, and gene deletion analyses defined an apicidin biosynthetic gene cluster, and the NRPS gene was named as apicidin synthetase gene 1 (<I>APS1</I>). A 63 kb region surrounding <I>APS1</I> was sequenced and analysis revealed the presence of 19 genes. All of the genes including <I>APS1</I> were individually deleted to determine their roles in apicidin biosynthesis. Chemical analyses of the mutant strains showed that eight genes are required for apicidin production and were used to propose an apicidin biosynthetic pathway. The apicidin analogues apicidin E, apicidin D<SUB>2</SUB> and apicidin B were identified from chemical analysis of the mutants. The cluster gene <I>APS2</I>, a putative transcription factor, was shown to regulate expression of the genes in the cluster and overexpression of <I>APS2</I> increased apicidin production. This study establishes the apicidin biosynthetic pathway and provides new opportunities to improve the production of apicidin and produce new analogues.</P>

Chalcone isomerase (CHI) is a key enzyme ofthe flavonoid pathway that plays crucial roles in plant responsesto various pathogens. Our previous study using comparativetranscriptome analysis of soybean inoculated with or withoutPhytophthora sojae revealed Glyma.20G241500, a homologof CHI1A in Arabidopsis thaliana, to be induced by P.sojaein soybean. In the present study, we isolated the gene fromsoybean cultivar Nannong 10-1, designating it GmCHI1A. GmCHI1A transcripts were detected in soybean roots, stemsand leaves, with the highest levels in roots. Interestingly,GmCHI1A localizes to the cortical endoplasmic reticulum(ER). GmCHI1A was overexpressed in hairy roots to furtheruncover the roles of CHIs in soybean. Compared to emptyvector-expressing hairy roots (EV) infected with P. sojae,hairy roots overexpressing GmCHI1A (OE) exhibited decreasedP. sojae biomass accumulation, shortened the lesion lengthsand reduced zoospores germination. These results demonstratethat GmCHI1A plays positive roles in the response ofsoybean to P. sojae.
A New Triterpenoid from Brucea javanica

A new triterpenoid, bruceajavanin C (1), together with bruceosides A and B (2 and 3), bruceines D and E (4 and 5), yadanziosides A and G (6 and 7), (20R)-O-(3)-α-L-arabinopyranosylpregn- 5-ene-3β,20-diol (8), and α-D-glucopyranoside, (3β, 20R)-3-hydroxypregn-5-en-20-yl (9) were isolated from the aerial parts of Brucea javanica. The structure of 1 was elucidated on the basis of 2D-NMR spectroscopic analysis. In addition, compounds 1, 3, 4, 5, and 6 exhibited mild inhibitory effect on NO production in LPS-stimulated RAW264.7 cells.
SYNTHESIS OF YTTRIA-STABILIZED CUBIC ZIRCONIA NANOCRYSTALS BY ULTRASONIC–MICROWAVE ROUTE

The yttria-stabilized zirconia (YSZ) nanocrystals with uniform size, high purity, and high degree of crystallinity, were prepared by ultrasonic–microwave-assisted method. The structure, optical properties and morphologies of YSZ nanocrystals were characterized by X-ray powder diffraction (XRD), Raman spectroscopy, UV–vis absorption, scanning electron microscope (SEM) and transmission electron microscopy (TEM). The SEM and TEM images of the YSZ nanocrystals indicate that the product is a mono-dispersion structure with an average particle size of about 25 nm.

Purpose Nonylphenol (NP) is an endocrine disruptor found in products such as cleaners, plastics, and detergents. It exerts actions similar to endogenous 17-estradiol (E2) and is reported to influence various cancers. However, its role in colon cancer remains elusive. Materials and Methods Colon cancer cell lines COLO 205 and SW480 were employed in our study. The cells were treated with NP or E2 followed by measurement of apoptosis and proliferation using flow cytometry and MTT assays, respectively. G protein–coupled estrogen receptor 30 (GPR30) expression was visualized using immunofluorescence and Western blot. To investigate the underlying mechanism, the expression levels of GPR30, p-protein kinase A (PKA), c-myc, cyclin D1, and ERK1/2 were analyzed using Western blot. Meanwhile, the GPR30 antagonist G15 was utilized to validate the role of GPR30 in colon cancer progression. Finally, the effect of a GPR30 inhibitor on tumor growth was determined in vivo using tumor xenograft mouse models. Results NP facilitated the proliferation of colon cancer cells and induced apoptosis failure in vitro. Western blot revealed increased GPR30 expression levels in response to NP treatment. Cyclin D1, p-PKA, c-myc, and proliferating cell nuclear antigen, proteins that regulate the cell cycle, were all upregulated by NP, and NP-mediated ERK1/2 activation and subsequent cell proliferation were abrogated by the GPR30 inhibitor G15. Moreover, colon cancer mice that received G15 administration demonstrated impaired tumor growth in vivo. Conclusion Low dose NP promotes the growth of colon tumors through GPR30-mediated activation of ERK1/2 signaling.

Simulations of the severe precipitation event that occurred in the warm sector over southern China on 08 May 2014 are conducted using the Advanced Weather Research and Forecasting (WRF-ARWv3.5.1) model to investigate the roles of microphysical latent heating and surface heat fluxes during the severe precipitation processes. At first, observations from surface rain gauges and groundbased weather radars are used to evaluate the model outputs. Results show that the spatial distribution of 24-h accumulated precipitation is well reproduced, and the temporal and spatial distributions of the simulated radar reflectivity agree well with the observations. Then, several sensitive simulations are performed with the identical model configurations, except for different options in microphysical latent heating and surface heat fluxes. From the results, one of the significant findings is that the latent heating from warm rain microphysical processes heats the atmosphere in the initial phase of the precipitation and thus convective systems start by self-triggering and selforganizing, despite the fact that the environmental conditions are not favorable to the occurrence of precipitation event at the initial phase. In the case of the severe precipitation event over the warm sector, both warm and ice microphysical processes are active with the ice microphysics processes activated almost two hours later. According to the sensitive results, there is a very weak precipitation without heavy rainfall belt when microphysical latent heating is turned off. In terms of this precipitation event, the warm microphysics processes play significant roles on precipitation intensity, while the ice microphysics processes have effects on the spatial distribution of precipitation. Both surface sensible and latent heating have effects on the precipitation intensity and spatial distribution. By comparison, the surface sensible heating has a strong influence on the spatial distribution of precipitation, and the surface latent heating has only a slight impact on the precipitation intensity. The results indicate that microphysical latent heating might be an important factor for severe precipitation forecast in the warm sector over southern China. Surface sensible heating can have considerable influence on the precipitation spatial distribution and should not be neglected in the case of weak large-scale conditions with abundant water vapor in the warm sector.
Characteristics of registered studies for Coronavirus disease 2019 (COVID-19): a systematic review

Background: The World Health Organization characterized the Coronavirus disease 2019 (COVID-19) as a pandemic on March 11th. Many clinical trials on COVID-19 have been registered, and we aim to review the study characteristics and provide guidance for future trials to avoid duplicated effort. Methods: Studies on COVID-19 registered before March 3rd, 2020 on eight registry platforms worldwide were searched and the data of design, participants, interventions, and outcomes were extracted and analyzed. Results: Three hundred and ninety-three studies were identified and 380 (96.7%) were from mainland China, while 3 in Japan, 3 in France, 2 in the US, and 3 were international collaborative studies. Two hundred and sixty-six (67.7%) aimed at therapeutic effect, others were for prevention, diagnosis, prognosis, etc. Two hundred and two studies (51.4%) were randomized controlled trials. Two third of therapeutic studies tested Western medicines including antiviral drugs (17.7%), stem cell and cord blood therapy (10.2%), chloroquine and derivatives (8.3%), 16 (6.0%) on Chinese medicines, and 73 (27.4%) on integrated therapy of Western and Chinese medicines. Thirty-one studies among 266 therapeutic studies (11.7%) used mortality as primary outcome, while the most designed secondary outcomes were symptoms and signs (47.0%). Half of the studies (45.5%) had not started recruiting till March 3rd. Conclusion: Inappropriate outcome setting, delayed recruitment and insufficient numbers of new cases in China implied many studies may fail to complete. Strategies and protocols of the studies with robust and rapid data sharing are warranted for emergency public health events, helping the timely evidence-based decision-making.
