        Bioassay-guided Isolation of Novel Compound from Paeonia suffruticosa Andrews Roots as an IL-$1{\beta}$ Inhibitor

        The inhibition of Interleukin-1beta (IL-$1{\beta}$) is of substantial interest for the treatment of rheumatoid arthritis. Using an in vitro assay with RAW 264.7 cells, oxo-acetic acid 2-ethoxy-4-(3-hydroxy-2-oxopropyl) phenyl ester (1) was isolated from the roots of Paeonia suffruticosa Andrews as an inhibitor of IL-$1{\beta}$ with an $IC_{50}$ value of 56 ${\mu}M$. Compound 1 is a novel phenylesteric compound from P. suffruticosa Andrews. Compound 1 was shown to inhibit the production of pro-inflammatory cytokines in RAW 264.7 cells. Thus, a possible new action of novel compound is provided explaining the anti-rheumatoid arthritic properties of P. suffruticosa Andrews.

        Anti-inflammatory effects of β-sitosterol-β-D-glucoside from Trachelospermum jasminoides (Apocynaceae) in lipopolysaccharide-stimulated RAW 264.7 murine macrophages.

        <P>Trachelospermum jasminoides (Apocynaceae) has pharmacological effects that include anti-inflammatory, anti-bacterial and anti-viral activities, which have been observed from various studies. Of these pharmacological effects, the anti-inflammatory capacity of compounds from T. jasminoides is not yet known exactly. In this study, we investigated the compound that can be used for the suppression of lipopolysacchaide (LPS) stimulated inflammatory responses in macrophages among the five isolated compounds. β-sitosterol-β-D-glucoside (1) was found to reduce nitric oxide (NO) production from LPS-induced RAW 264.7 cells the most. In addition, compound 1 strongly inhibited the interleukin 6 (IL-6) activities of stimulated macrophages. Treatment of RAW 264.7 cells with compound 1 reduced secretion of inflammatory elements including tumour necrosis factor - alpha (TNF-α) and interleukin 1 beta (IL-1β). Thus, compound 1 may be a useful candidate for the development of new drugs to treat endotoxemia and inflammation accompanied by the overproduction of NO.</P>

        Metaplastic Meningioma Overspreading the Cerebral Convexity

        Meningioma is relatively common, benign, and extra-axial tumor accounting for about 20% of primary brain and spinal cord tumors. The World Health Organization (WHO) classified these tumors into Grade I (benign), Grade II (atypical), and Grade III (anaplastic) meningioma. Grade I meningioma which is slowly growing tumor and have some rare subtypes. Among them, metaplastic subtype is defined as a tumor containing focal or widespread mesenchymal components including osseous, cartilaginous, lipomatous, myxoid or xanthomatous tissue, singly or in combinations. We report a rare metaplastic meningioma overspreading nearly whole cerebral convexity from main extra-axial tumor bulk in the parietal lobe.

        Phenolic Compounds from Pueraria lobata Protect PC12 Cells against Aβ-induced Toxicity

        Bioassay-guided fractionation of the EtOAc-soluble extract of Pueraria lobata based on the inhibition of Aβ-induced toxicity in PC12 cells resulted in the isolation of four known active compounds, genistein (8), biochanin A (9), sissotrin (10), and puerol B (11). Of these, genistein (8) and biochanin A (9) exhibited potent neuroprotective effects with ED50 values of 33.7 and 27.8 μM, respectively. In addition, a new coumestan, 2-(α,α-dimethylallyl)coumestrol (1) was isolated and characterized, but proved to be inactive, as were additional seven known compounds. The structure of new compound 1 was determined using spectroscopic techniques.

        중견기업 디지털전환에 대한 탐색적 연구

        본 연구는 국내 중견기업의 디지털전환 현황을 살펴보고 디지털전환 촉진 방안의 객관적인 근거자료로 활용하고자 함이 목적이다. 국내 중견기업 200개 기업에 설문지를 구성, 배포하여 150개 기업에서 응답을 받았다. 주요 결과를 살펴보면, 설문 응답기업 150개 기업 중에서 79개 기업(52.6)%이 디지털전환을 추진하고 있고 디지털전환 추진분야는 업무프로세스 및 설비 자동화, 기존 설비 확장 등(53.2%), 제조공정 빅데이터/AI 기반 스마트화(43.0%) 순으로 나타났다. 국내 중견기업의 디지털전환을 촉진하기 위해 필요한 지원분야는 중견기업 맞춤형 금융지원 확대(56.0%)가 가장 높게 나타났고 구성원들의 디지털전환 역량을 강화하기 위한 필요한 조직 차원의 지원분야는 기술개발 역량교육(46.7%)이 가장 높게 나타났다. 본 연구는 설문결과를 토대로 국내 중견기업의 디지털전환 촉진 방안의 정책적 제언을 제시함으로써 향후 국내 중견기업이 디지털전환을 가속화하고 글로벌 시장경쟁력을 강화하는데 기여할 것으로 사료된다. The purpose of this study is to examine the current status of digital transformation of domestic mid-sized companies and to use it as an objective basis for digital transformation promotion plans. A questionnaire was formed and distributed to 200 domestic midsized companies and received responses from 150 companies. Looking at the main results, 79 companies (52.6)% of the 150 companies responding to the survey are pursuing digital transformation, and the areas for digital transformation are business process and facility automation, expansion of existing facilities, etc. (53.2%), manufacturing process big data/ AI-based smartization (43.0%)appeared in the order. The area of support required to promote digital transformation of domestic mid-sized companies was the highest in the expansion of financial support tailored to mid-sized companies (56.0%). 46.7%) was the highest. This study is expected to contribute to accelerating digital transformation and strengthening global market competitiveness by presenting policy suggestions for promoting digital transformation of domestic mid-sized companies based on the results of the survey.

        중소기업의 사회적 책임활동이 조직문화유형에 따른 조직몰입에 미치는 영향에 관한 연구

        본 연구의 목적은 중소기업의 사회적 책임활동이 조직몰입에 미치는 영향을 알아보고 나아가 조직문화 유형에 따라서 중소기업의 사회적 책임활동이 조직몰입에 미치는 영향을 검증하기 위한 것이다. Quinn과 Rohrbaugh(1983)의 4가지 조직문화유형을 토대로 조사하였다. 연구의 가설을 다음과 같다. 첫째, 중소기업의 사회적 책임활동은 조직몰입에 정(+)의 영향을 미칠 것이다. 둘째, 중소기업의 사회적 책임활동이 조직몰입에 미치는 정(+)의 영향은 조직문화 유형(합의문화, 발전문화, 위계문화, 합리문화)에 따라 달라질 것이다. 본 연구를 위한 연구방법은 현재 중소기업에 재직 중인 중소기업연수원 교육생 300명을 대상으로 설문조사를 실시하여 284부를 실증분석에 사용하였다. 연구 결과는 다음과 같다. 첫째, 중소기업의 사회적 책임활동이 조직몰입에 정(+)의 영향을 미칠 것이다 라는 가설은 채택되었다. 둘째, 합리문화를 제외한 합의·발전·위계의 조직문화는 중소기업의 사회적 책임활동이 조직몰입에 미치는 정(+)의 영향을 강화시킨다는 가설이 채택되었다. 본 연구는 중소기업에 종사하고 있는 구성원들의 CSR 활동에 대한 인식이 조직몰입에 정(+)의 영향을 미치는데 조직문화 유형에 따라서 그 영향력이 강화된다는 결론을 도출함으로써 조직몰입을 향상시키기 위해서는 CSR 활동에 따른 구성원들의 인식 제고 뿐만 아니라 조직문화의 형성 또한 중요하다는 시사점을 제공한다. The study aims to understand how the corporate social responsibility(CSR) of small and medium enterprises influences on the organizational commitment, and furthermore, how the organizational culture types act as elements in the situation. The research is based on the four types of organization culture by Quinn & Rohrbaugh(1983). Hypothesis is established for such purpose, and the study model is set up by collecting opinions from leading researches and professionals, as follows. Firstly, CSR of small and medium enterprises will influence on the organizational commitment. Secondly, CSR of small and medium enterprises will have a significant effect on the organizational commitment in the environment of consensus group culture. Thirdly, CSR of small and medium enterprises will have a significant effect on the organizational commitment in the environment of developmental group culture. Fourthly, CSR of small and medium enterprises will have a significant effect on the organizational commitment in the environment of hierarchy group culture. Lastly, CSR of small and medium enterprises will have a significant effect on the organizational commitment in the environment of rationality group culture. For the research, a survey was carried out subjecting 300 trainees in the training center for small and medium enterprises, who are currently in service for small and medium enterprises, and 284 questionnaires were analyzed. For the analysis, reliability analysis, primary factors analysis, multiple regression analysis and T-test were carried out, and the result is as follows. Firstly, the hypothesis that CSR of small and medium enterprises will influence on the organizational commitment was proved to be true. Secondly, the hypothesis that CSR of small and medium enterprises will have a significant effect on the organizational commitment in the environment of consensus group culture was proved to be true. Thirdly, the hypothesis that CSR of small and medium enterprises will have a significant effect on the organizational commitment in the environment of developmental group culture was proved to be true. Fourthly, the hypothesis that CSR of small and medium enterprises will have a significant effect on the organizational commitment in the environment of hierarchy group culture was proved to be true. And lastly, CSR of small and medium enterprises will have a significant effect on the organizational commitment in the environment of rationality group culture was not proved to be true that it does not have a significant effect. In conclusion, it becomes clear that CSR of small and medium enterprises does influence on the organizational commitment, and especially, consensus group culture is highly influential than others. The company environment with the consensus group culture that emphasizes a sense of ownership, a sense of responsibility has a great effect on the organizational commitment and the participation of the members. On the other hand, the rationality group culture that is task-oriented and focuses on the ability regarding performance and the actual results does not have a significant influence. It shows that, for CSR of small and medium enterprises to have a significant effect on the organizational commitment, the managements need to consider whether their organization culture appeals positive influences to the members.

        Establishment of a non-invasive ischemic hearing loss animal model using the rose bengal dye and 532 nm low-level light energy (photo-thrombosis) in Republic of Korea: animal study

        Background and Objectives: This research was designed to establish an animal model of cochlear ischemia using non-invasive photochemical reaction.Materials and Methods: Rose bengal (RB) was administered to male Sprague-Dawley rats at 6 weeks of age through an intraperitoneal (IP) or intravenous (IV) injection. At five minutes after the RB injection, a 532 nm diode laser was used to irradiate the rats for 5, 10, and 15 minutes using a transcanal approach. The results of the auditory brainstem response at one, three, and seven days after the procedure were compared. On the 10th day, the cochleae were obtained to prepare tissue sections for pathohistological examination.Results: For all the nine rats in the RB IP-injection group, hearing loss was not observed in any of the nine different conditions. For rats in the RB IV-injected group, hearing loss was not observed when 532 nm light was used for irradiation at 150 mW or 175 mW for 5 or 10 minutes. However, for rats irradiated for 15 minutes, hearing loss of more than 70 dB was observed at all frequencies one day later. Hearing loss of more than 50 dB persisted until 10 days later. Further pathohistological examinations of these rats revealed the destruction of the normal structure of the organ of Corti and stria vascularis.Conclusion: The results of the present study demonstrate that the IV injection of RB with transcanal green light irradiation can induce cochlear ischemia in rats. This method could be useful for developing a model of cochlear ischemia.

        율무근의 식물화학적 성분 연구 및 Tyrosinase 저해 활성

        본 연구는 천연 미백소재 탐색을 위하여 율무(C. lachryma-jobivar. ma-yuen) 부산물 추출물의 tyrosinase 저해활성을 검증하였으며, 그 중에서 뿌리 추출물은 159.3 μg/mL의 IC50 값을 나타내었다. 율무 뿌리 추출물의 순차적 용매 분획에 대한 활성 검증 후 가장 활성이 우수한 EtOAc 분획물에 대하여 DiaionHP-20 column chromatography, MPLC및 preparative HPLC를 수행해 율무근으로부터 여섯 개의 화합물을 정제하였고, 이 물질들의 구조 분석은 LC-MS와 NMR 데이터 해석을 바탕으로 진행하여 각각 (+)-icariol A<sub>2</sub> (1), zhepiresionol (2), 4-hydroxybenzaldehyde(3), trans-ρ-coumaric acid (4), N-(2-hydroxy-4-methoxyphenyl)-2-hydroxyacetamide (5), coixol (6)로 구조를 규명하였다. 화합물 1은 이 식물에서 처음으로 분리되었으며, 화합물 5는 자연계로부터 처음으로 분리된 화합물로 확인하였다. 이들 화합물 중 4-hydroxybenzaldehyde (3), trans-ρ-coumaric acid (4)와 coixol (6)은 순서대로 707.4, 6.5, 62.4 μM의 IC<sub>50</sub>값을 나타내어 대조군으로 사용한 arbutin (IC<sub>50</sub> =618.7 μM)과 비교시 유사하거나 우수한 활성을 확인하였다. 이로 미루어 보아, 율무근 추출물과 활성물질의 tyrosinase 저해활성을 확인할 수 있었으며, 율무근 추출물의 미백소재로서의 적용 가능성을 확인할 수 있었다. In the course of screening tyrosinase inhibitory activity, EtOAc-soluble fraction of Coix lachryma-jobi var. mayuen Stapf. (Gramineae) roots showed significant inhibition. Further fractionation of the EtOAc-soluble fraction resulted in six compounds, which were identified as (+)-icariol A<sub>2</sub> (1), zhepiresionol (2), 4-hydroxybenzaldehyde (3), trans-ρ-coumaric acid (4), N-(2- hydroxy-4-methoxyphenyl)-2-hydroxyacetamide (5), and coixol (6). The chemical structures of these compounds were identified on the basis of spectroscopic methods (MS, 1D and 2D NMR) and comparison with literature values. Compound 1 was first reported from this plant. Also, this is the first time that the isolation of compound 5 has been reported from nature source. Among the isolated compounds, compounds 4 and 6 showed enzyme inhibitory activity, with IC<sub>50</sub> values of 6.5 and 62.4 μM, respectively, in comparison with these of positive control, arbutin.
