6-(3.4-Dichlorophenyl)amino-7-chloro-5.8-quinolinedione (RCK50) was tested for antifungal activities in mice systemically infected with Candida albicans. The therapeutic potential of RCK50 was also assessed in comparison with ketoconazole. RCK50 had ED_50 0.22±0.01 ㎎/㎏. Ketoconazole as a positive control had ED_50 6.00±1.70 ㎎/㎏. Intraperitoneally administered RCK50 at the ED_50 for 7 days and 14 days reduced Candida albicans colony count in the kidneys and liver. And administered RCK50 at the ED_50 for 14 days improved survival rates. The genotoxicities of RCK50 had been evaluated. RCK50 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. RCK50 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK50 has no genotoxic potential under these experimental conditions. Acute oral toxicity studies of RCK50 were carried out in ICR mice of both sexes. RCK50 did not show acute oral toxicities and LD_50 values were over 2.850 ㎎/㎏ in ICR mice.

Hepatocellular carcinoma(HCC)is one of common causes of cancer-related death in Korea where the majority of HCC patients were Hepaitc B virus(HBV)carriers and have cirrhosis. Transarterial chemoembolization(TACE)is commonly applied to the treatment of multinodular HCC in Korea and careful selection of candidate is important for the risk of various side effects. Besides common side effects as fever, nausea, abdominal pain and elevation of liver enzyme, TACE may predispose to hepatic failure, ischemic cholecystitis, pulmonary embolism, cerebral embolism and pneumonitis. In previous studies, some cases of pulmonary and cerebral embolism cases were reported but lipiodol pneumonitis after TACE was rarely reported. A 65-year-old woman with a multinodular HCC associated with HBV infection, was treated with TACE. Seven days after the procedure, nonspecific respiratory symptoms such as dyspnea and dry cough developed. Chest X-ray and chest computed tomography showed diffuse ground glass opacities in whole lung fields, suggestive of lipiodol pneumonitis. After several days of supportive care with steroid administration, radiologic abnormalities and subjective symptoms were much improved, considered that the disease was compatible with lipiodol pneumonitis.

Objective: The aim of this study was to propose a prediction model based on machine learning algorithms for spontaneous preterm delivery (sPTD) prediction in singleton pregnancies using the cervical elastographic markers obtained at midtrimester. Methods: Multicenter prospective study was performed between June 2018 to December 2020 by the Korean Research Group of Cervical Elastography (grant ID: HI18C1696). Women with singleton pregnancies who underwent ultrasound between 18+0 and 24+0 weeks gestation were eligible for analysis. Cervical elastography were performed using E-cervixTM (WS80A and HERA, Samsung Medison). sPTD was defined as delivery <37 weeks gestation due to either spontaneous preterm labor or rupture of membranes. The final dataset included data from 1,415 patients and this dataset was split into training and validation datasets. Three machine learning classification algorithms (Lasso regression, random forest and XGBoost) were used to build sPTD prediction models. Model performance in predicting sPTD from elastographic data and clinical factors was assessed using validation dataset. Results: Among 1,415 patients, 75 (5.3%) had sPTD. The area under the receiver operating characteristic curve (AUC) results were as follows: for Lasso regression- 0.742, for the random forest- 0.769 and for the XGboost 0.776. In subgroup analysis, the XGBoost model had a better predictive performance in both nulliparity and multiparity group (AUC 0.736 and 0.741, respectively). Results of variable importance for XGBoost showed that indication for prior preterm birth, maternal weight, body mass index, cervix length at examination, hardness ratio (HR) 10 and 30, and elasticity contrast index (ECI) were significant contributors of predicting sPTD. Conclusion: The prediction model using midtrimester cervical elastographic parameters had a potential value in predicting spontaneous preterm birth in single pregnancies.

The detection of implanted stem cells in vivo almost always depends on postmortem histology. In this study, we sought to demonstrate the feasibility of adenovirus-mediated transient expression of the firefly luciferase(Fluc) reporter gene and longitudinal monitoring of implanted embryonic stem(ES) cells in rat corpus cavernosum by optical bioluminescence. Mouse ES cells(ES-E14TG2a) were transiently transfected with Ad-CMVFluc( MOI=100) and implanted into the corpus cavernosum of male Sprague-Dawley rats that were derived into the following groups: sham operated controls and injection of 5 different doses of ES cells(1×103, 1×104, 1×105, 1×106 and 5×106). Cell survival was assessed histologically and by bioluminescence imaging every other day after cellular implantation. All injected rats with cell numbers over 1×104 revealed the bioluminescence in the genital area until terminating the experiment(for 2 weeks), and histology showed increased smooth muscle contents compared to control rats. The study demonstrated a feasibility of adenovirus-mediated transduction of an imaging reporter gene applied for optical imaging of ES cells. The location, magnitude and survival of ES cells in rat corpus cavernosum can be monitored noninvasively with bioluminescence imaging system.
