        김치유래 젖산균의 균체지방산 분석을 이용한 분류학적 연구

        표준균주를 포함한 230여개의 김치유래 젖산균에 대한 균체지방산(FAMEs)을 분석하였다. FAMEs profiles는 Euclidian Distance 17.5에 의해 7개의 Major Cluster와 1개의 Single Cluster로 나뉘어졌다. 이중 A, B, C 및 Cluster는 Leuconostoc속으로 분석되어졌고, F는 Lactobacillus속으로 분석되어졌다. 그리고 E와 G cluster는 두개의 Genus가 혼재되어 나타났으며 보충적인 연구가 필요하다. 앞으로 김치유래 젖산균의 균체지방산 분석결과를 기반으로 한 데이타베이스에 95가지 탄소원을 이용하는 수치분류학적 접근방법 및 Pyrolysis Mass Spectrometry 등의 화학적 분석 방법과 분자친화적 연구를 통한 종합적 분류정보 체계가 갖추어지면 젖산균의 신속, 정확한 동정 및 연구에 활발히 이용되어질 것이다. Two hundreds and thirty lactic acid bacteria, mostly isolated from Kimchi, including type strains were sued for analysis of cellular fatty acids. The 230 test strains were recoverd in 7 major and 1 single clusters defined a Euclidian distance of 17.5. These aggregate taxa were equivalent to the genus Leuconostoc (aggregate group A, B, C and D), and the genera Leuconostoc and Lactobacillus (aggregate group G). It is concluded as evident that FAMEs (Fatty Acid Methyl Esters) profile of cell can be used as a criterion in classification of lactic acid bacterial from kimchi. Additional comparative taxonomic studies need to be carried out on well chosen representative strains to determine the most appropriate methods of value.

        ABSTRACTAn Influence of Travel Agency's Culture on Organizational Effectiveness Sun-Hee Lee·Yong-Keun Lee The Effects of corporate culture on the organizational effectiveness and/or corporate competitiveness have been widely recognized and discussed among both the incumbent managerial and the academicians of economics and sociology in recent years. The purpose of this study is to review the functions of the corporate culture and to examine the factors of the corporate culture among the travel agencies.The present study starts with a conceptual review of the corporate culture. The characteristics of corporate culture is shaped usually with the two major elements; the styles of management and decision-making of the top managerial and the subculture of the employees. Each of the four components of Corporate culture is operationalized as follows; ① tough-guy, macho culture, ② work hard, play hard culture, ③ bet-your-company culture, ④ process culture. These four components of corporate culture become the independent variables while the recognition of environment becomes the dependent variables in the analytical model proposed in this paper. The recognition of environment such as danger and feed-back speed is divided into pre-IMF and post-IMF.Some results of the findings are as follows: First, before IMF, as the danger is recognized low and feed-back speed fast, the corporate culture is shown as work hard, play hard culture. Second, after IMF, as the danger is recognized high and feed-back speed fast, the corporate culture is shown as tough-guy, macho culture.

        추황배(Pyrus pyrifolia Nakai cv. Chuhwangbae) 과피로부터 1종의 Sterol과 3종의 배당체 화합물의 단리·동정

        본 연구에서는 배의 유용성 증명을 위한 일환으로 배의 화학성분을 분자수준에서 밝히고자 하였다. 이에 배 과피 MeOH 추출물을 용매분획하여 얻은 EtOAc-산성 분획과 EtOAc-중성분획을 대상으로 Sephadex LH-20, silica gel, 그리고 ODS colmn chromatography와 HPLC를 이용하여 정제 및 단리하였다. 그 결과, EtOAc-산성 분획과 EtOAc-중성 분획으로부터 각각 2종씩의 화합물을 단리하였다. 단리된 화합물 1-4는 1H- 및 13C-NMR 분석을 통하여 각각 (S)-(+)-2-cis-abscisic acid O-β-D-glucopyranosyl ester (화합물 1), 1-[4-O-β-D-glucopyranosyl]phenyl ethanone(piceoside, 화합물 2), β-sitosterol (화합물 3), 그리고 β-sitosteryl 3-O-β-D-glucopyranoside (화합물 4)로 동정되었다. 단리된 3종의 배당체 화합물(화합물 1, 2, 4)들은 본 연구에 의해 배로부터 처음 동정되었으며, 화합물 3은 추황배로부터 처음 동정되었다. 본 연구결과가 배 함유 성분연구는 물론 배의 기능성 해명 연구에도 추후 중요한 기초자료로 활용되길 기대한다. We isolated and identified antioxidants from acidic and neutral ethyl acetate fractions of the peel of pear (Pyrus pyrifolia N. cv. Chuhwangbae). We isolated 4 compounds from the methanol extract, by using 3 different types of column chromatography (Sephadex LH-20, silica gel, and octadecylsilane) and preparative HPLC. We identified the isolated compounds as (S)-(+)-2-cis-abscisic acid O-β-D-glucopyranosyl ester (compound 1), 1-[4-O-β-D-glucopyranosyl]phenyl ethanone (picroside, compound 2), β-sitosterol (compound 3), and β-sitosteryl 3-O-β-D-glucopyranoside (compound 4) by nuclear magnetic resonance analysis. We are the first to report the identification of compounds 1, 2, and 4 from pear.

        치과용 인산칼슘계 충전 및 재생재료 개발(Ⅰ)

        Because calcium phosphate materials including hydroxyapatite are bioactive and exhibit good biocompatibility, these compositions are actively tried as the hard substituting materials. At the beginning of the development of the calcium phosphate materials, these materials were used as sintered ceramics. After then, plastic calcium phosphate cements were developed to make the shape of the material directly at the site of application. Reports on the dental calcium phosphate materials can be assorted into some categories; development of root apex sealing material or canal sealer, development of pulp capping material or cavity liner, or the reinforcement through addition of resin materials. Studies on the biocompatibility and drug delivery system may be other categories. The final purpose of this study is the development of dentine substituting materials based on calcium phosphate, which is highly biocompatible with the tooth pulp and additionally can stimulate or accelerate the dentine formation. In this preliminary study, PMVE-Ma (polymethy1 viny1 ether-maleic anhydride), HPMC (hydroxypropy1 methy1 cellulose), acrylic resin and acrylic-maleic copolymer was added into the PBS (phosphate buffered solution) to optimize the mechanical and chemical properties of calcium phosphate cement. From this study, AC-1 (4:1 mixture of cement powder and resin powder mixed chemical cure acrylic resin solution) showed the highest diametral tensile strength after storage in 100 % relative humidity for 24 hours, and AM-3, AM-2 (4:1 mixture of cement powder and resin powder mixed with 40 or 30 % acrylic-maleic copolymer) followed. The ranking of compressive strength from the highest in the same condition was AC-1, AM-2 and HP-4 (cement powder mixed with 15 % PMVE-Ma in DW). HP-4 showed the highest diametral tensile strength after storage in normal saline for 24 hours, and AC-1 and AC-2 (9:1 mixture of cement powder and resin powder mixed chemical cure acrylic resin solution) followed. The ranking of compressive strength from the highest in this condition was AC-1, HP-4 and HP-6 (cement powder mixed with the 1:1 mixture of 2 % HPMC in DW and 15 % PMVE in DW). From the point of the mechanical properties, AC-1, HP-4, AM-3 and AM-2 seemed proper for dentine filling material. Addition of resin materials did not influence on the speed and ratio of hydroxyapatite formation. From this study, appropriate compositions based on calcium phosphate for dentine filling materials can be found.

        바륨 페라이트를 함유한 알지네이트 자성 복합 입자의 제조

        Magnetic polymer composite materials with high coercivity have potential applications in medical diagnostic technologies, magnetic drug delivery, and hyperthermic cancer treatment. The purpose of this study was to prepare the magnetic composite particles by encapsulating barium ferrite powders with alginate and to investigate their physical and biological properties. 0.4 g of sodium alginate was dissolved in 20 ml of distilled water and different amount (0.4~4 g) of barium ferrite powder was dispersed in alginate solution using an ultrasonication method. The resulting barium ferrite-alginate slurry was added dropwise to a CaCl2 solution to form magnetic Ca-alginate composite beads. They were separated by filtration and washed several times with water and ethanol. The final product was then completely dried at 40℃ under vacuum to obtain brownish black powders (0.1~1 mm in size) with a high magnetic response when submitted to a small magnet. Average size of magnetic composite particles was dependent on the amount of barium ferrite and the viscosity of slurry . The morphological and dimensional analyses of magnetic composite particles were performed by optical microscopy and scanning electron microscopy. Magnetic polymer composite particle was found to maintain the high coercivity. The saturation magnetization value for magnetic Ca-alginate composites increased with the increase of the barium ferrite/alginate ratio. The amount of self-heating induced by hysteresis under an alternating magnetic field was measured as a function of barium ferrite/alginate ratio in distilled water. According to the results, the encapsulated magnetic composite is expected to be useful for hyperthermia and chemotherapy remarkably.

        Background : Mucins possess the unique function of protecting and lubricating the epithelial surface and other important functions such as call growth, direct implication in the fetal development, the epithelial renewal and differentiation, the epithelial integrity, carcinogenesis, immune regulation, cellular adhesion and metastasis. Purpose : This study was done to provide the significance of alteration of MUC1 and MUC2 expression in colorectal adenocarcinoma, A series of 131 colorectal adenocarcinomas including 11 mucinous carcinomas were screened immunohistochemically for their expression of MUCI and MUC2, Materials and mehtods : Of 131 carcinomas, 76 (58,5%) were MUCI positive and 91 (68, 9%) were MUC2 positive, In normal colonic goblet cells, MUCl was not expressed but MUC2 was expressed in cytoplasm, Conclusion There were up-regulation of MUCI and down-regulation of MUC2 in colorectal carcinomas, The frequency of MUC2 positivity according to differentiation was statistically reliable.         광도 및 광원 차이가 치과용 광중합 콤포짓트레진의 중합깊이에 주는 영향

        Inadequately polymerized resin has greater water sorption and solubility, possibly manifested clinically with early color instability and degradation. The purpose of this study was to determine the influence of total light energy (light intensity X irradiation time) of halogen and plasma arc curing units on the polymerization deth of five universal resin composites by measuring the Vickers hardness number (VHN/0.1, 10) at the surface and 0.5, 1.0, 1.5, 2.0, 2.5and 3.0㎜ under the surface. The total light energy for the halogen light curing unit (VIP, Bisco, U.S.A.) was 4,000, 8,000, 16,000 mJ/㎠, and VHN was measured immediately after curing and after 24-hours storage in 37℃ distilled water. Polymerization behavior of plasma arc curing unit (Flipo, Serial No. P03G02221, Lokki, France) was also determined, and the irradiation time was 3 and 9 seconds. From this experiment, the following results were obtained. 1.VHN immediately after light curing was 33.2∼123.3, and showed a great variation depending on the brand of the materials. The curing depth was different depending on the brand of the materials by the variables of light source, total light energy and light intensity. 2.VHN after 24 hours was 36.3∼115.1, and in almost all cases, these values increased in comparison to those immediately after light curing. However, when the total light energy was absolutely insfficient for curing, these values decreased. 3.When the same total light energy was irradiated with different light intensity, the differences in curing depth and VHN were depended on the brand of the materials. 4.In some brand of the materials, the curing depth and VHN were similar regardless of the light source (halogen or plasma arc) when the total light energy was the same.

        배양조건에 따른 치수세포 RPC-C2A의 특성 : 배양용기 및 BMP 처리 Culture Ware and BMP Treatment

        Odontoblasts are highly differentiated cells aligned at the periphery of the dental pulp. Pulp tissue responds to injury by generating reparative dentine elaborrated by odontoblast-like cells that differentiated from pulp cells. In vitro, dental pulp cells cultured under certain conditions are able to express typical markers of cell differentiation; however, no culture sysem can simulates clinical pulp response. The purpose of this study was to evaluate the effect of culture ware and BMP on the seeding. growth sne differentiation of pulp derived cell line, RPC-C2A. Large pore-sized PET membrane (0.4 ㎛, Cell Culture Insert, BD Science, USA), rhBMP-4 (R & D Systems, USA) and polystyrene modified by vacuum-gas plasma treated culture surface (Primaria, BD Science, USA) were tested. RPC-C2A cells are maxillary incisor pulp cells from male Wistar rats (Kasugai el al., 1988). Cells were cultured on a traditional polystyrene culture ware (PS), transparent PET membrane (MI), Insert + rhBMP-4 (20 ng/ml, MB), and Primaria culture ware (PM) up to 21 days. Culture media was 10% FBS added DMEM with 10 mM β-glycerophosphate, 10^-8 M dexamethasone, 50 ㎍/ml ascorbic acid and 50 mM sodium fluoride. Formation of multilayered cell layer and mineral nodule was determined with a phase contrast microscope and EDXA. There were no differences in cell attachment and growth rate among four culture conditions up to three-days culture. Multilayered cells were observed after six-days culture, and mineral nodules were observed in MB condition after seven-days culture. After nine-days culture, mineral nodules were observed in PS, and new nucleation of mineral nodules was prominest in MI and MB conditions. Exfoliation of cells from the dish was observed in PS after 13-days culture, and vaeuoles were formed in MI after 19-days culture. After 21-days culture, cell death was accompanied by increase of mineral nodules in MB. Growth and development of pulp cells were influenced by the culture ware and BMP. In MB condition, the duration for the formation of multilayered cells and mineral nodule was shortened compared to other conditions.

        치주조직 재생을 위한 칼슘 포스페이트 글라스가 MC3T3-E1 세포의 증식, 분화 및 석회화에 미치는 영향

        The purpose of this work was to exploring the influence of the prepared calcium phosphate glass on the proliferation, differentiation, and mineralization of the culture system of the pre-osteoblastic MC3T3-E1 cells. Polystytene was employed as a control in this experiment. Cells were cultured in a differentiated medium onto the calcium phosphate glass as well as polystyrene, and the number of cells and the protein contents were determined. To examine osteoblast differentiation, the alkaline phosphatase activity was measured. Mineralization was evaluated by staining the calcium deposit with Alizarin red. Culture onto the calcium phosphate glass exhibited no significant difference in cell proliferation compared with control group. The alkaline phosphatase activity in the experimental group, however. was enhanced In the calcium phosphate glass significantly at 10-18 days than that of the control group (p<0.05). Enhancement of bone-like tissue formation by the addition of the calcium phosphate glass was observed since 7 days. The results of the present study indicate that the prepared calcium phosphate glass promotes osteogenesis by increasing collagen synthesis and calcification of the extracellular matrix. It maybe due to greater calcium concentration in the culture medium released from the calcium phosphate glass.
