        Genome-wide investigation of a Korean synthetic breed, Woori-Heukdon using the Illumina PorcineSNP60K BeadChip

        Background: Woori-Heukdon (KWH) is a Korean synthetic pig breed generated using Chookjin-Duroc (KCD), Chookjin-Chamdon (KCC), and their crossbreds. Currently, there is a severe lack of studies investigating the Korean breed populations including wild boars (KWB) throughout the genome. Objective: This study was performed to investigate the genetic characteristics of Korean pig populations at the genome-wide level. Methods: Using the SNP dataset derived from genotyped and downloaded datasets using the Illumina PorcineSNP60K BeadChip, we compared the genomes of 532 individuals derived from 23 pig breeds to assess the genetic diversity, inbreeding coefficient, genetic differentiation, and population structure. Results: KWB showed the lowest average expected heterozygosity (HE = 0.1904), while KWH showed the highest genetic diversity (HE = 0.02859) among Korean populations. We verified that the genetic composition of KWH, showing USD of 74.8% and KCC of 25.2% in ADMIXTURE analysis. In population structure analyses, KCC was consistently shown to be separated from other pig populations. In addition, we observed gene flow from Western pigs to a part of Chinese populations. Conclusion: This study showed that Korean native pigs, KCC have genetic differences in comparison with Chinese and Western pigs; despite some historical records and recent genetic studies, we could not find any clear evidence that KCC was significantly influenced by Chinese or Western breeds in this study. We also verified the theoretical genomic composition of KWH at the molecular level in structure analyses. To our knowledge, this is the first genomic study to investigate the genomic characteristics of KWH and KCC.

        위절제술 환자의 표준진료지침 개발 및 적용 효과

        Background : Gastric cancer is the most common malignant tumor in Korea. surgical operation is one of the major treatment modalities for gastric cancer patients. Therefore, gastrectomy is one of the most common procedures in General Surgery. There were variation in length of hospital stay and medical treatment for gastrectomy between three surgeons at Asan Medical Center. Clinical pathways have received considerable attention as a tool for recucing the medical practice variation, increasing the efficiency of care process, and improving the quality of care. The aim of this study was to evaluate the effect of a clinical pathway for gastrectomy in gastric cancer patients. Methods : The clinical pathway for gastrectomy was developed and implemented by a multidisciplinary group in Asan Medical Center. A computerized clinical pathway program was developed and revised after a pilot test. A total of 145 patients underwent gastrectomy by three surgeons at Asan Medical Center. We compared the length of hospital stay, patient satisfaction, and unplanned readmission rate between the pre-pathway group(n=67) and the post-pathway group(n=78). We also investigated the degree of satisfaction among the physicians and nurses who were main end-users of the clinical pathway. Results : The clinical pathway was applied to all target patients. The average length of hospital stay was shortened from 12.7days to 10.6days(p<0.01). The degree of patient satisfaction with the care process changed from 90.3% to 89.2% after the implementation of the clinical pathway, but the difference was of satistically significant(p=0.761). Unplanned readmission rate was 2.9% in the pre-pathway group. More than 90% of physicians and nurses answered that the clinical pathway had been a useful tool in their medical practice. Conclusions : The findings of the study demonstrated that implementation of the clinical pathway for gastrectomy produced substantial reduction in the length of hospital stay while improving the quality of patient outcomes. The computerized clinical pathway program can be used as one of the powerful patient management tools for reducing the practice variations and increasing the efficiency of care process in Korea hospital settings.

      • 연쇄중합효소반응을 이용한 태아성별진단

        For sex determination by the PCR method, oligoprimers to Y-chromosome gene, DYZ1, SRY, and AMGL were synthesized and genomic DNA was extracted from male and female placenta for the control use. DYZ1 represented 154 bp single band to 0.001pg/ml male genomic DNA but did not represent 154 bp band in female genomic DNA, SRY represented 341 bp band to 1 pg/ml male genomic DNA but did not represent 341 bp band in female genomic DNA, and AMGL represented 977 and 788 bp double band to 1 pg/ml male genomic DNA and 977 bp single band to 1 pg/ml female genomic DNA in 2% agarose gel electrophoresis stained with ethidium bromide. DYZ1 was 1,000-fold sensitive than SRY and AMGL. DYZ1 and SRY could not identify the PCR failure from female but AMGL identified. To increase the sensitivity, the dual amplification of AMGL was performed and the sensitivity increased to 1,000-fold. During the dual amplification of female genomic DNA mixed with male genomic DNA, 0.00125pg/l, 1:400 part male genomic DNA contamination represented double bands as male. In 2 cases of 46, XY female, DYZ1 and AMGL amplification represented male band but SRY amplification did not represent male band. It was suggested that SRY gene was deleted in two 46, XY female cases. for fetal sex determination, PCR with DYZ1, SRY, and AMGL was performed in 10 cases of chorionic villi and 15 cases of amnionic cells. By the comparison with karyotyping result, fetal sex determination was achieved successfully in all 23 samples using PCR of SRY and AMGL but false result was detected in 3 cases(13%) using DYZ1. According to our results, it was concluded that DYZ1 was 1,000-fold sensitive than SRY and AMGL but could not be used because of its false results, and AMGL and SRY must be used concomitantly for precise sex determination.

        Sinonasal Undifferentiated Carcinoma (SNUC) is a very rare, highly aggressive malignant tumor of the nasal cavity and paranasal sinuses. SNUC tends to present with advanced-stage disease, often with intracranial invasion. It requires an aggressive multimodality therapy that includes surgical resection. A cure rate of less than 20% is generally reported in the literature, with most patients dying within 1 year of onset of the disease. Three patients diagnosed as SNUC were treated at the Yeungnam University Medical Center between the years 2000 and 2003 were analyzed retrospectively. All patients presented with the disease very advanced. The three cases were given chemotherapy or chemotherapy with radiotherapy. Two patients died of the disease, surviving only 6 and 11 months following treatment, respectively. We did a follow-up on just the one remaining case with incomplete controlled disease for 27 months. The overall prognosis of SNUC is very poor. We consider that more intensive multimodality therapies are recommended for all patients with SNUC.

      • 비닐 조각 섞은 모래의 剪斷强度와 變形 特性

        Every industrialized country has environmental problems due to the tremendous amount of industrial, agricultural and home wastes. Polymeric material wastes such as polyethelene, polypropylene, polyvinyl and polyester are growing rapidly in volume and most of them are not disposed properly. Using the polymer wastes as construction material is expected to be a way to mitigate the wastes disposal problem. This paper performes triaxial tests on sand samples mixed with polyethylene film cuttings measuring the effects of admixture on the shear strength and deformation characteristics of sand. The KS standard sand and two kinds of polyethylene film square(0.5×0.5mm) cuttings(0.1 and 0.2mm thickness) are used for test samples. Mixing rates of PEF 5 and 8% by weight are used. The results show that a proper amount of PEF mixed in sand increases the angle of shear resistance and brings a small cohesion intercept. The ductility of PEF mixed sand becomes larger than that of pure sand. Though the tests performed in this study are not much, it shows the possibility of using polymer wastes as construction material.

        초등학교에서의 군 개념 지도에 관한 연구

        In late 19C, German mathematician Felix Klein declaired "Erlangen program" to reform mathematics education in Germany. The main ideas of "Erlangen program" contain the importance of instructing the concepts of functions and groups in school mathematics. After one century from that time, the importance of concepts of groups revived by Bourbaki in the sense of the algebraic structure which is the most important structure among three structures of mathematics - algebraic structure, ordered structure and topological structure. Since then, many mathematicians and mathematics educators devoted to work with the concepts of group for school mathematics. This movement landed on Korea in 21C, and now, the concepts of groups appeared in element mathematics text as plane rigid motion. In this paper, we state the rigid motions centered the symmetry - an important notion in group theory, then summarize the results obtained from some classroom activities. After that, we discuss the responses of children to concepts of groups.

      • 때죽나무 열매의 脂質成分에 관한 硏究

        때죽나무 열매의 一般成分과 diethyl ether로 抽出한 組脂肪質에 대한 몇가지 物理化學的 特徵들, 脂肪質의 構成 및 脂肪酸의 組成을 分析한 結果를 要約하면 다음과 같다. 1. 一般分析値는 水分 8.52%, 組蛋白質22.5%, 組脂肪質 43.65%, 炭水化物 22.5%, 灰分이 2.83%였다. 2. 油脂의 比重은 0.928∼0.935, 굴절율은 1.4828∼1.4840, 요오드값은 109.2∼110.4, 비누화값은 191∼199, 산값은 1.1∼2.7, 에스테르값은 189∼195, 불검화물은 3.4%로 불검화물은 다른 植物性 油脂보다 높았다. 3. 脂肪質의 構成은 中性脂質이 98.4%,糖脂質 1.1%, 燐脂質은 0.5%였으며 中性脂質에서 esterified sterol, triglyceride, free fatty acid, free sterol, diglyceride, monoglyceride등 6가지 성분을 분리, 확인하였으며 이중에서 triglyceride 含量이 가장 많았다. 4. 脂肪酸의 組成은 palmitic acid와 stearic acid등의 포화지방산이 12.3%정도였으며 불포화지방산인 oleic acid가 41.5%, linoleic acid 42.3%, linolenic acid 3%가 含有되어 있었다. The lipid components, fatty acid compositions, physical and chemical characteristics of crude oil in Styrax japonica seed were determined and proximate compositions of it were also analyzed. The results were summerized as follows: 1. Proximate components of the seed were shown to be 8.52% moisture, 22.5% crude protein, 43.65% crude fat, 22.5% carbohy drates, 2.83% ash. 2. Physico-chemical characteristics of the seed oil were determined as follows:specific gravity 0.928∼0.935, refractive index 1.4828∼1.4840, iodine value 109.2∼110.4, saponification value 191∼199, acid value 1.1∼2.7, ester value 189∼195, unsaponifiable content 3.4%. The unsaponifiable content of the lipid is more than that of other vegetable oil. 3. Lipid components were 98.4% neutral lipid, 1.1% glycolipid, 0.5% phospholipid. The neutral lipids were fractionated and identified as six components:esterified sterols, triglycerides free fatty acids, free sterols, diglycerides, monoglycerides, and triglycerides is the most content of those. 4. Fatty acid compositions were quantitatively determined by GLC to give 9.7% palmitic acid, 2.6% stearic acid, 41.5% oleic acid, 42.3% linoleic acid, 3% linolenic acid and arachidonic acid were presented in small quantities.

      • N,N-Dimethylaniline의 아조커플링 반응에 關한 硏究

        The coupling reaction of N, N-dimethylaniline with several monosubstituted benzenediazonium salts has been studied kinetically in aqueous buffered solution. The activation evergies measured in the coupling reaction of N, N-dimethylaniliine with diazotized p-nitroaniline, m-nitroaniline, sulfanilic acid, m-anisidine,p-anisidine, p-bromoaniline, p-chloroaniline, m-toluidine, p-toluidine are 8.46, 8.05, 9.20, 12.32, 15.91, 10.86, 11.54, 13.80, 14.35, 15.47Kcal/mole, respectively. The coupling reactionis more favorable in the presence of an electron-withdrawing group in active components than in the presence of electron-donating group. It has been observed that the coupling reaction with diazotized p-nitroaniline proceeds 1000 times more rapidly than that with the benzenediazonium salt, which is coupled 40 times faster than diazotized p-anisidine. In our experiments, the formation of triazene, the diazoamino compound has not been observed, and the experimental result that the ionic strength of the medium didn't give any influence on the coupling reaction rate leads to the confirmation of the neutral free amine being one of the reasonable reactive species in the coupling reaction. A linear free energy relationship has been essentially observed except for three para substituted active components, for which the extended Hammett plot treated by Yukawa-Tsuno permits a good log k/k??=ρ(σ+ rㆍ????) linear relationship when considering the mesomeric effect. The calculated reaction constant of these azocoupling reaction is ρ=3.92, and the reaction constant of resonance contribution is calculated as r=0.32 at 20 ℃.
