        Development of SNP Markers to Distinguish Various Watermelon Traits and Validation Using Fluidigm Genotyping Assay

        Watermelon [Citrullus lanatus (Thunb.) Matsum and Nakai] is one of the economically most important fruit crops of the Cucurbitaceae family. Among different watermelon traits, disease resistance and fruit quality are the important traits for growers and consumers. The single nucleotide polymorphism (SNP) markers similar to those traits can potentially and cost-effectively distinguish the genetic variations among these traits. Consequently, we developed 33 SNP makers linked to different watermelon traits associated with fruit quality and disease resistance, and validated in the genetic resources of watermelon and F1 breeding lines using ‘Fluidigm SNP Genotyping’ assay. Most of the SNP markers distinguished the alleles into three different types such as reference allele, alternative allele and heterozygous from watermelon genotypes for various traits. The SNP markers ‘ZymFL-T81P’ (ZYMVresistance), ‘FON1-U161’ and ‘FON1-S075’ (Fusarium wilt-resistance), ‘Pmr21-Cla831’ (PM-resistance), and ‘ClGBS-J168’ and ‘GBS-GC230’ (GSB-resistance) can successfully differentiate resistant (R), susceptible (S) and heterozygous watermelon genotypes. Similarly, the SNP marker associated with sugar content, citrulline content, arginine content, rind hardness, flesh firmness, fruit shape, rind strip pattern of watermelon fruit and seed coat colour can successfully distinguished the watermelon genetic resources and F1 breeding lines as reference allele (A) type, alternative allele (B) type and heterozygous (H). These SNP markers could be utilized for marker assisted selection as well as screening of a large number of watermelon germplasm for fruit quality and disease resistance. However, further validation like artificial inoculation of pathogens for the traits related to disease resistance is required in watermelon crops.

        SYNTHESIS OF THE KNOWLEDGE MODEL FOR FAULT DIAGNOSIS USING QUALITATIVE SIMULATION

        Fault diagnosis requires the knowledge models which describe the behavior of the chemical processes. However it is cumbersome and time consuming work to generate these models. Also the knowledge acquisition required for generation is difficult. The objective of this study is to examine whether the knowledge aquisition for fault diagnosis knowledge models, especially bottom-up models, is possible through qualitative simulation. Qualitative simulation is based on the study of modeling, representation of feedback control and state transition using the QSIM algorithm. Also in this study, qualitative simulation is applied to a buffer tank level control system using the simulation strategy of this paper. The results of the simulation show several behaviors of the processes and the usability for the generation of a knowledge model. However when several different results of simulation are generated, some are spurious solutions which result from insufficient information.

        The Clinical Results of Conservative Treatment of Frozen Shoulder Using Continuous Passive Motion

        Background: The purpose of this study is to administer conservative treatment in 30 patients diagnosed with idiopathic frozen shoulder, following the suggested frozen shoulder rehabilitation program and to assess the clinical outcome using a prospective study. Methods: Thirty patients diagnosed with idiopathic frozen shoulder, treated with steroid hormone injection on the articular joint with an intra-articular steroid (triamcinolone 40 mg+lidocaine 4 ml) injection and started on stepwise shoulder extension exercise were chosen. The subjects were divided into two groups of 15 people each with one group undergoing rehabilitation with continuous passive motion (CPM) and the other group without it. Follow-ups were done before rehabilitation and at 4-week intervals with the 24th being the final week. At every follow-up, passive range of motion (ROM) was measured and surveys on pain and clinical score were administered. Results: In the last follow-up, both groups showed statistically significant improvements in all evaluation criteria. However, no statistical difference in all values of the ROM and Constant score evaluation criteria was observed between the groups. Only in the last follow-up, group 1 had a visual analog scale (VAS) score of 2.4 ± 2.1 points, which was lower, with statistical significance, than the VAS score of group 2, which was 4.4 ± 3.1 points (p<0.001). Conclusions: Study using CPM in treatment of frozen shoulder has been inadequate, meaning that there is still room for improvement and need for more study on setting a more specific protocol and guidelines for this procedure.

        스틸보강재가 부착된 프로파일 및 고강도 모르타르를 이용한 균열손상 콘크리트관의 보강효과

        본 논문에서는 내하력이 저감된 노후화 된 콘크리트 하수관거의 보강공법의 타당성을 검증하기 위하여 정적재하 시험을 실시하였다. 내경 0.8 m, 1.0 m, 1.5 m인 원형관과 폭 2.0m, 2.5 m인 상자형관을 대상으로 하였으며, 신관을 대상으로 정적 재하시험을 통하여 관 내부에 충분한 균열을 발생시킨 후 보강공법을 적용 한 후 다시 정적재하시험을 실시하였다. 본 연구에서 적용 된 보강공법은 균열관 내부에 스틸보강재가 추가된 프로파일과 고강도 모르타르를 이용하여 화학적 열화에 대한 저항성과 단면 내력을 증진 시킨 것이다. 보강공법 적용 후 단면 내력은 균열발생 이전 신관에 비해 원형관의 경우 1.66~3.50배, 상자형관의 경우 1.66~3.10배 정도 증가하였다. 또한 ABAQUS 6.6을 이용하여 보강공법 적용 전ㆍ후 콘크리트 원형관과 상자형관의 비선형 해석을 수행하였다. C3D8R 솔리드 요소를 사용하였고, concrete damage plasticity option을 적용하였다. 원형관과 상자형관에 배근된 철근의 영향성을 해석상 반영하기 위하여 합성 재료식을 유도하여 적용하였다. In this study, in order to verify the reinforcement effects of the cracked concrete tubes and culverts, static load test was conducted. After the load carrying capacity of the original concrete tubes (nominal diameter 0.8 m, 1.0 m, 1.5 m) and box culverts (inner width 2.0 m. 2.5 m) was reduced by the cracking test, the cracked concrete specimens were strengthened by installing profile with steel stiffener and high strength mortar. And then, the maximum load tests were conducted the renewal concrete tubes and box culverts. According to the method application, the load carrying capacity increased 1.66~3.50 times than it of the original tubes before applying the method. In case of the original box culverts, the load carrying capacity increased 1.66~3.10 times than the case before installing profile and high strength mortar. Also non-linear analysis was carried out by using the commercial FEM program of ABAQUS 6.6. Solid (C3D8R) elements and concrete damage plasticity option was applied to the analysis. For reflecting confined reinforcing bars in the analysis, the composite material properties were used.

        Infliximab, which is indicated for refractory rheumatoid arthritis (RA), ankylosing spondylitis (AS) and Crohn’s disease, has cutaneous adverse events including skin rash, urticaria, pruritus, and lupus-like eruption. Psoriasis induced by infliximab is very rare. In Korea, it is infrequently reported in Crohn’s disease or RA and never reported in AS. We encountered a case of psoriasis induced by infliximab treatment for AS, and report here on this case along with a review of the relevant literature.

        과학교육연구소에서는 학생들의 창의적 능력을 길러줄 수 있는 모델을 제시하고, 창의적 문제의 모델을 제시해 주어야 겠다는 입장에서 충청남도의 초·중고등학교 학생들을 대상으로 「제1회 충청남도 초·중고생 창의력 경연대회」를 실시하였다. 대회 실시 결과를 분석해 본 결과 저학년보다는 고학년이, 초등학생보다는 고등학생으로 올라갈 수록 창의적인 아이디어를 내는 양이나 질에 있어서 뒤 떨어지는 것으로 분석되었으며, 상급학교로 올라갈수록 창의적 아이디어를 내지 못하였고, 발표력 또한 떨어지는 것으로 나타났다. 그리고 본 대회에서 출제되었던 창의적인 문제와 같은 유형의 프로젝트형 창의성 문제를 각급 학교의 교실에서도 방과후 활동프로그램으로 보급될 수 있도록 사법대학 차원에서 프로그램을 개발할 필요가 있다. 특히 학생들은 선생님한테서만 배우는 것이 아니라 학생들 스스로가 각기 서로의 스승이 되고 지도자가 되어 상호 유기적으로 배우고 가르침을 준다고 볼 때 본 대회에서 실시하고 있는 장기문제와 같이 학생들이 협동해서 문제를 풀어 나가는 공동연구(group study)를 수행하여 문제를 해결한다면 학생중심의 교육활동을 할 수 있는 학교 교육 현장으로 변화시키는데 결정적인 역할을 할 것으로 기대된다. The Institute of Science Education has hosted The First Creativity Competition for Elementary and High School Students in the Chung Nam Province with the aim to enhance the creative ability of the students. Based on the evaluation from the competition, results showed that as the students get older, creativity and presentation ability decreases. Therefore, it is recommended that problems which require creative thinking, such as those presented at the competition, need to be introduced into the educational curriculum from an early age through methods such as after school activities. Toward this end, the School of Education need to train student teachers who will become the teachers to the future generations. Students learn not only from teachers but also from each other. The First Creativity Competition can be evaluated as having contributed toward enhancing student centered education, group study, and introducing a model toward creativity education where students cooperate with each other to reach a common end.

        Expression Characteristics of LSH Genes in Brassica Suggest their Applicability for Modification of Leaf Morphology and the Use of their Promoter for Transgenesis

        The functions of DUF640/ALOG (Arabidopsis LSH1 and Oryza G1) domain proteins, which are found in most land plants, have not been well characterized, but some of these proteins regulate inflorescence architecture in rice and specify organ boundaries in Arabidopsis. Arabidopsis DUF640-domain genes are initially identified as LIGHT-SENSITIVE HYPOCOTYLS (LSH) genes. Chinese cabbage leaves have large, white midribs and photosynthetic leaf blades (or lamina). A DUF640 domain gene of Brassica rapa, BrLSH2, is specifically expressed in the midrib of Chinese cabbage. Arabidopsis and rice possess ten LSH family genes, but B. rapa has 24 LSH genes, which can be categorized into two or four groups based on sequence identity. Here, we examined the expression patterns of the LSHs in various Brassica species and analyzed the promoter sequence of the BrLHS2 gene. The transcript levels of most LSH genes were very high in the midrib but low in the leaf blade. These genes were evenly expressed throughout the petiole region of Korean cabbage and highly expressed in the leaf base region near the stem and in the border area in B. oleracea. In addition, BrLSHs were expressed in both bundle and mesophyll cells of the midrib. These expression patterns suggest the possible use of these genes to generate leafy vegetables with altered leaf morphology. The BrLSH2 promoter, which contains auxin- and cytokinin-responsive elements as well as leaf development-related elements, may confer midrib-specific expression, suggesting that this promoter may be useful for the production of midrib-targeted transgenic Chinese cabbage.
