Tailoring Biomaterials for Cancer Immunotherapy

Despite the great promise of immune checkpoint blockade (ICB) therapy for cancer treatment, the currently available options for ICB treatment pose major clinical challenges, including the risk of severe systemic auto- immune responses. Here, we developed a novel localized delivery platform, immuno-bioglue (imuGlue), which is inspired by the intrinsic underwater adhesion properties of marine mussels and can allow the optimal retention of anti-PD-L1 drugs at tumor sites and the on-demand release of drugs in response to the tumor microenvironment. Using a triple-negative breast cancer and melanoma models, we found that imuGlue could signifycantly enhance anti-tumor efficacy by eliciting a robust T cell-mediated immune response while reducing systemic toxicity by preventing the rapid diffusion of anti-PD-L1 drugs into the systemic circulation and other tissues. It was also demonstrated that imuGlue could be successfully utilized for combination therapy with other immunomodulatory drugs to enhance the anti-tumor efficacy of ICB-based immunotherapy, demonstrating its versatility as a new treatment option for cancer immunotherapy.

Purpose: The purpose of this study was to evaluate the results of the treatment of Schatzker type V and VI tibial plateau fractures using a midline longitudinal incision and dual-plate fixation. Materials and Methods: Ten patients with Schatzker type V and VI tibial plateau fractures treated with a midline longitudinal incision and dual plating were analyzed. The patients were followed for a minimum of one year. Clinical outcomes were evaluated using range of motion, visual analogue scale (VAS) and Knee Society Score. Radiological outcomes were evaluated using the bony union time, medial proximal tibial angle (MPTA) and posterior proximal tibial angle (PPTA). Results: The mean VAS score was 2.2 points, and the mean Knee Society function score was 85 points at the final follow-up. The mean flexion contracture was 2.5o and the mean further flexion was 125o. It took an average of 4 months until bony union occurred. The mean MPTA and PPTA were 90.5o and 4.4o, respectively. There was one case of delayed wound healing, but no other complications were observed. Conclusions: The treatment of Schatzker type V and VI tibial plateau fractures with a midline longitudinal incision and dua -plate fixation resulted in satisfactory clinical and radiological outcomes. This can be an option when treating Schatzker type V and VI tibial plateau fractures.

BACKGROUND: Left atrial (LA) strain is a novel parameter of LA function. However, its reference value has not been established, and the determining factors for LA strain remain elusive. We aimed to present LA strain with reservoir, conduit, and contractile components and associated parameters in a large-sized group of healthy individuals. METHODS: The present study was from a prospective multicenter registry in South Korea. Subjects who had no history of cardiovascular disease with adequate images were eligible for inclusion. LA reservoir, conduit, and contractile strains (LASRES, LASCD and LASCT, respectively) were measured. Left ventricular global longitudinal strain (LV GLS) and early and late diastolic strain rates (DSRe and DSRa, respectively) were also evaluated. RESULTS: Among a total of 324 subjects (mean age: 49 ± 16 years, 167 females), the mean LASRES, LASCD, and LASCT values were 35.9% ± 10.6%, 21.9% ± 9.3%, and 13.9% ± 3.6%, respectively. Mean LV GLS was -20.4% ± 2.2%, and mean DSRe and DSRa were 1.6 ± 0.4 s-1 and 0.8 ± 0.3 s-1, respectively. With aging, LASRES and LASCD showed significant decreases. Factors showing independent associations with LASRES were age (B = -0.425, p < 0.001), DSRe (B = 4.706, p = 0.001), and LV GLS (B = -1.081, p < 0.001). Age (B = -0.319, p < 0.001), DSRe (B = 4.140, p = 0.002), DSRa (B = -3.409, p = 0.018), and LV GLS (B = -0.783, p < 0.001) showed associations with LASCD. With LASCT, only DSRa showed a correlation (R = 0.277, p < 0.001). CONCLUSIONS: We presented LA strain in a large-sized group of healthy subjects. Age is a significant determinant of LA function. Associations of LA strain with diastolic strain rates and LV GLS reflect cardiac mechanics.

Porous silicon with a complex network of nanopores is utilized for photoelectrochemical energy conversion. A novel electroless Pt deposition onto porous silicon is investigated in the context of photoelectrochemical hydrogen generation. The electroless Pt deposition is shown to improve the characteristics of the PS photoelectrode toward photoelectrochemical $H^+$ reduction, though excessive Pt deposition leads to decrease of photocurrent. Furthermore, it is found that a thin layer (< 10 ${\mu}m$) of porous silicon can serve as anti-reflection layer for the underlying Si substrate, improving photocurrent by reducing photon reflection at the Si/liquid interface. However, as the thickness of the porous silicon increases, the surface recombination on the dramatically increased interface area of the porous silicon begins to dominate, diminishing the photocurrent.

A self-sensing circuit for piezo inkjet has been designed in order to monitor the operating condition during printing. In order to verify the circuit, both ink droplet images from strobe LED and vibration signals from the laser vibrometer were measured and compared with self-sensing signal. Experimental results show that selfsensing signal was effective in detecting the pressure wave change due to the bubble trapped in inkjet printhead.

Neutron science and technology, the utilization of neutron beams for a wide variety of scientific and engineering research ranging from materials and life science to industrial applications, has been one of the key elements of modem science and technology. Currently, the neutron science and technology in Korea is in rapid growth with the operation of the 30 MW High-flux Advanced Neutron Application Reactor (HANARO) at the Korea Atomic Energy Research Institute, which is one of the most powerful nuclear research reactors in the world. Furthermore, a state of the art HANARO cold neutron research facility, which will open a new era for the neutron science and technology in Korea, is expected to become available in 2010. In this paper, the progress of neutron science and technology in Korea is reviewed and its unprecedented new opportunities and challenges in coming years are presented.

Background: Despite the reported survival benefit of transarterial chemoembolization/embolization (TACE/TAE) in patients with spontaneously ruptured hepatocellular carcinoma (HCC), clinical outcome of surgical resection compared with TACE/TAE in these patients has not been determined. Methods: A consecutive 54 patients with ruptured HCC at our institution between August 2003 and February 2012 were enrolled. We compared clinical outcomes after surgical resection, TACE/TAE, or supportive care in these patients. Results: The median age was 54 years (range, 30-87 years). The most common cause of HCC was hepatitis B virus infection (66.7%). Median tumor size was 8.5 cm (range, 0.9-25.5 cm), and 4 (7.4%) had HCCs within Milan criteria. Of the 54 patients, 6 (11.1%) patients underwent surgical resection, 25 (46.3%) TACE/TAE, and 23 (42.6%) supportive care. Age, gender, and presence of liver cirrhosis showed no difference among three groups (p>0.05). Patients with surgical resection, TACE/TAE, and supportive care showed significantly difference in Child-Turcotte-Pugh class A (50%, 4%, and 8.7%, respectively; p<0.01) and median tumor size (6.1cm, 8.0cm, and 10cm, respectively; p=0.02), and tumor number (66.7%, 20%, and 8.7%, respectively; p=0.01). The 1-, 3-, and 6- months cumulative survival rates were higher in patients with surgical resection (60%, 60%, and 60%) and TACE/TAE (44%, 28%, and 20%) than supportive care (21.7%, 4.4%, and 0%), respectively (each p<0.01). The 1-, 3-, and 6-months cumulative survival rates tended to be higher in patients with surgical resection than TACE/TAE (p=0.14). Conclusions: Post-treatment outcomes after surgical resection and TACE/TAE were better than supportive care in patients with spontaneously ruptured HCC, and surgical resection might provide better survival benefit than TACE/TAE in these patients. However, reserved liver function should be considered during treatment decision making in such patients.
