      • The effect of grief counseling base on Swanson Caring Model for early adult students after bereavement

        Abstract: Accident injury is a common life crisis for men and women over 20 years old in early adulthood. The purpose of this study was to explore the effect of grief counseling base on Swanson Caring Model for early adult students after the death of classmate who had been together for five years. For 43 students, using the grief counseling strategy, after two months of counseling, 86% of the students felt that the degree of grief counseling had improved by using the grief counseling questionnaire. Method(s): Based on Swanson"s five steps of understanding, being with, doing for, empowering, and maintaining belief, this study develops grief counseling strategies for junior high school students in early adulthood, including observation, listening, learning, and caring. Secondly, the teacher should accompany and share the experience and feelings of getting along with the dead students. Thirdly, the teacher should endow the students with the ability to produce the video of the dead students" reminiscence, share the responsibility of helping the dead students with the farewell ceremony and provide the company of the parents. Finally, the teacher should continue to trust the dead students in the process of caring, Early adult students who wholeheartedly support their bereaved friends. To provide care and care for the early adult students who lost their friends for one month from February to march in 110, and to evaluate the effect of grief counseling with self-administered grief counseling evaluation questionnaire in April, and to understand the students" mood in the latest week with mood thermometer. Result(s): The questionnaire of grief counseling evaluation was recovered to 86%, and the strategies of counseling were analyzed. The preparation of commemorative video was made by group consultation, individual care of tutors and farewell video production of the deceased students and blessing to angels. The improvement of the measures to the grief of early adult students was 85-90%. After one month after the end of the farewell ceremony, there were still 5-10% of the students in the week of April 7, 110. At the time point of two months after the bereavement of friends, there were still slight difficulties in sleeping, restlessness or depression. Conclusion(s): Based on the Swanson Caring Model, this study developed a grief counseling strategy suitable for early adult students after their bereavement, and analyzed its effectiveness. With the passage of time and the implementation of counseling strategies, the degree of grief of early adult students has improved, but a small number of them still feel slightly negative emotions and distress due to the bereavement of their friends. This study aims to provide a reference for class tutors to help students with special events through the application of Swanson Caring Model.

        Prioritization of Sleep: Discrepancies Between Attitude and Practice

        Objectives: With the promotion of sleep through education, an increasing recognition of the importance of sleep among the general public has been observed. However, whether this increased recognition affects sleep practices is unknown. To provide direction for future sleep education programs, this study aimed to explore the consistency between people’s prioritization of sleep among their daily life activities and their actual practice when sleep conflicts with other activities.Methods: The study was a cross-sectional online survey with 898 adults (male:female=372:526; age=39.74±10.04 years). The participants were asked to complete an online survey that required them to 1) rate how important sleep was; 2) prioritize daily life activities including sleep, work, family activities, leisure activities, social activities, and exercise; and 3) recall the actions they undertook at the most recent instance when sleep conflicted with each of the other activities.Results: Although 70.7% of the participants prioritized sleep by placing it in their top three daily life activities, only 35.7% and 38.1% chose to go to sleep when it conflicted with work and family activities, respectively. Gender and occupational factors also significantly influenced the discrepancy between sleep attitudes and practices.Conclusions: Our study demonstrated an inconsistency between reporting sleep as a priority in daily life activities and actual behavioral practices when sleep conflicted with other activities. The results suggest that sleep education should not only emphasize the importance of sleep but also provide practical solutions for circumstances when sleep conflicts with other activities.

        Chinese Translation and Validation of the Child and Adolescent Sleep Checklist for Parents

        Background and ObjectiveaaThe Child and Adolescent Sleep Checklist (CASC) for parents was originally developed and published in Japanese and has served as a valid and effective tool for the assessment of a parent’s perspective of their adolescent child’s sleep behaviors. To allow for comparisons of such perspectives among parents of different cultures, and with different linguistic backgrounds, reliable and valid measurement tools are necessary. The purpose of this study was to translate and validate a Chinese language version of this questionnaire. MethodsaaInternal consistency, factor structure, and criterion validity data were tested using 497 parent/adolescent dyads in Taiwan. Exploratory factor analysis (EFA) and internal consistency reliability were examined in a subset of the cohort (n = 241), while criterion validity and conduct confirmatory factor analysis (CFA) was examined in a second subset of the cohort (n = 256). Preliminary criterion validity was established through a comparison with a measure of chronotype, Morningness- Eveningness Scale for Children. ResultsaaEFA resulted in four factors, consistent with the original version, with the four factors accounting for 60% of the total variance. The results of CFA presented good fit to the data. The questionnaire demonstrated also acceptable to good internal consistency (α = 0.84–0.89). Preliminary criterion validity was evident as the total score and each of the subscale scores on the questionnaire were significantly higher (worse sleep-related behavior) in evening-types. ConclusionsaaThe Chinese language version of the CASC for parents appears to be a psychometrically sound measure to evaluate sleep behaviors in Chinese-speaking adolescents.

        The Gender-Sensitive Social Risk Factors for Internet Addiction in College Undergraduate Students

        Objective The current study aims to explore precipitating and social risk factors for internet addiction (IA) in university undergraduate students, and to provide evidence for interventions and the early prevention of IA in different genders. Methods Four thousand eight hundred and fifty-eight college sophomores completed an online survey on their internet use-related behaviours and social risk factors. Results We found that more male (8.3%) than female students (5.4%) had moderate and severe IA. The main online activity in the moderate and severe IA groups was online gaming in males and online streaming in females. Roommates engaging in similar internetbased entertainment was a risk factor of IA only for males, while not being in a romantic relationship was a risk factor of IA for females only. Infatuation with the internet before college and adjustment problems for college life were shared risk factors for both genders in the mild and moderate IA groups. Conclusion IA was a common phenomenon in college students with shared and unique precipitating and social risk factors in males and females. The gender-sensitive risk factors for IA warranted earlier and individualized intervention and prevention strategies for IA in this population.

        Single-cell RNA sequencing reveals B cell–related molecular biomarkers for Alzheimer’s disease

        In recent years, biomarkers have been integrated into the diagnostic process and have become increasingly indispensable for obtaining knowledge of the neurodegenerative processes in Alzheimer’s disease (AD). Peripheral blood mononuclear cells (PBMCs) in human blood have been reported to participate in a variety of neurodegenerative activities. Here, a single-cell RNA sequencing analysis of PBMCs from 4 AD patients (2 in the early stage, 2 in the late stage) and 2 normal controls was performed to explore the differential cell subpopulations in PBMCs of AD patients. A significant decrease in B cells was detected in the blood of AD patients. Furthermore, we further examined PBMCs from 43 AD patients and 41 normal subjects by fluorescence activated cell sorting (FACS), and combined with correlation analysis, we found that the reduction in B cells was closely correlated with the patients’ Clinical Dementia Rating (CDR) scores. To confirm the role of B cells in AD progression, functional experiments were performed in early-stage AD mice in which fibrous plaques were beginning to appear; the results demonstrated that B cell depletion in the early stage of AD markedly accelerated and aggravated cognitive dysfunction and augmented the Aβ burden in AD mice. Importantly, the experiments revealed 18 genes that were specifically upregulated and 7 genes that were specifically downregulated in B cells as the disease progressed, and several of these genes exhibited close correlation with AD. These findings identified possible B cell-based AD severity, which are anticipated to be conducive to the clinical identification of AD progression.

        Controlled Release of Clenbuterol from a Hydroxyapatite Carrier for the Treatment of Alzheimer’s Disease

        Background Alzheimer’s disease is a neurodegenerative disorder, and Aβ aggregation is considered to be the central process implicated in its pathogenesis. Current treatments are faced by challenges such as serious side effects and reduced drug bioavailability. In this study, we developed a drug delivery system for intramuscular injection that uses cellular activity to achieve constant and long-term drug release. Methods Synthesized mesoporous hydroxyapatite (SHAP) was prepared via co-precipitation, and hydrophobic surface modification using stearic acid was then used to load clenbuterol by physical absorption, thus creating the drug delivery system. Clenbuterol release was achieved through cellular activity, with macrophage uptake triggering lysosome/endosome disruption, cytoplasmic release, extracellular exocytosis, and subsequent systemic circulation. Results We found that clenbuterol-loaded SHAP enabled sustained release for more than 2 weeks and effectively modulated inflammation, reduced Aβ oligomer-induced toxicity, and prevented Aβ aggregation. Conclusions Our findings suggest that treatment with clenbuterol loaded in this SHAP delivery system could be a promising strategy for treating Alzheimer’s disease.

        Volatilization of Nerolidol in Tea and Tea-soup

        Nerolidol is one of most important content in tea aroma, especially in Oolong tea. This experiment focuses on the impact of time and medium on nerolidol volatilization. The results of nerolidol volatilization with infusion time showed that the peak areas of nerolidol increased from 1 to 3 min, then decreased. The maximum peak area of nerolidol volatilization was 25630.78 at 3 min. Tea polyphenol had strong promotion impact on fragrant compound volatize, caffeine had limited impact on volatilization, while polysaccharide inhibited nerolidol volatilization.

        IL-33/ST2 axis mediates hyperplasia of intrarenal urothelium in obstructive renal injury

        The monolayered intrarenal urothelium covers the renal papilla and ureteropelvic junction (UPJ). In response to increased renal pressure during obstruction or ischemic injuries, intrarenal urothelial cells begin to proliferate and form a multilayered urothelium. Little is known regarding the mechanism and pathophysiological role of urothelium hyperplasia during renal obstruction. In this study, we investigated the expression of interleukin (IL)-33, an IL-1 family cytokine, in kidneys with unilateral ureteral obstruction (UUO)-induced obstructive injury. IL-33 levels in hydronephrotic urine and serum were upregulated 2 days after UUO. The number of ST2-expressing immune cells was increased in the UUO kidney. We found that IL-33 was upregulated in vimentin-positive cells in the cortical and medullar layers and the UPJ stroma. Moreover, IL-33 expression was predominantly induced in multilayered keratin 5- positive urothelial cells in the UPJ. IL-33 was not detected in terminally differentiated superficial umbrella cells expressing uroplakin 3a. In vivo, we confirmed that deficiency of IL33 or its receptor ST2 attenuated UUO-induced hyperplasia of the UPJ urothelium. Deficiency of IL33 attenuated the expression of UUO-induced type 2 inflammatory cytokines and upregulated uroplakins and urothelial differentiation signaling in UPJ tissues. Our results collectively suggest that the IL-33/ST2 axis mediates the activation of innate immune responses and contributes to urothelial hyperplasia by regulating urothelial differentiation in obstructive kidney injury.

        An Aligned Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) Scaffold Fixed with Fibronectin to Enhance the Attachment and Growth of Human Endothelial Progenitor Cells

        Repair and regeneration of vascular tissue is a crucial current research focus in the fields of biomedical engineering and regenerative medicine. Numerous studies revealed that cells are required to grow on an appropriate extracellular matrix to maintain or enhance functionality. In the present study, various surface modification methods were evaluated to fix fibronectin on the surface of a bio-based and aligned poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) film for vascular tissue engineering. After chemical modification, the properties of the fibronectin-fixed PHBV films were examined and compared with the original films, including -NH2 group expression, contact angle, mechanical properties, and fibronectin binding amount. Then, cytotoxicity and biocompatibility were measured by culture with L929 cells and endothelial progenitor cells (EPCs) of the fibronectinfixed PHBV films. In addition, cell morphology, cell growth kinetics, acetylated low-density lipoprotein uptake ability, lectin binding ability and specific gene expressions of cultured EPCs on fibronectin-fixed PHBV films were also analyzed. Taken together, our data demonstrated that the surface of the aligned PHBV films could be successfully modified to immobilize fibronectin. Importantly, EPCs cultured on the fibronectin-fixed PHBV films showed excellent cell biocompatibility, a rapid proliferation rate, an aligned growth direction and correct cell functions. We believed that fibronectin-fixed PHBV films can serve as a potential scaffold for vascular tissue engineering.
