      • Comparison of Serum Tumor Associated Material (TAM) with Conventional Biomarkers in Cancer Patients

        Objective: To compare expression level of serum tumor associated materials (TAM) with several conventional serum tumor biomarkers, eg., carcinoembryonic antigen (CEA), carbohydrate antigen19-9 (CA19-9), carbohydrate antigen 15-3 (CA15-3), alpha-fetoprotein(AFP), in selected solid tumors. Methods: Patients diagnosed histologically or cytologically with liver, breast, esophageal, gastric, colorectal or pancreatic cancers were enrolled into this study. After diagnosis, the level of TAM was determined by chemical colorimetry, and levels of conventional tumor markers was measured by chemiluminescence methods. Results: A total of 560 patients were enrolled into this study. No statistically significant difference was detected in TAM and the above mentioned tumor biomarkers in terms of their positivity and negativity ( P>0. 05). Conclusions: Detection of TAM in liver, breast, esophageal, gastric, colorectal, and pancreatic cancer patients demonstrates a good accordance with CEA, CA199, CA153, and AFP, thus suggesting that further study is warranted to verify whether TAM could be a surrogate for these conventional biomarkers.

        Analysis of the Baroreceptor and Vestibular Receptor Inputs in the Rostral Ventrolateral Medulla following Hypotension in Conscious Rats

        Input signals originating from baroreceptors and vestibular receptors are integrated in the rostralventrolateral medulla (RVLM) to maintain blood pressure during postural movement. The contributionof baroreceptors and vestibular receptors in the maintenance of blood pressure following hypotensionwere quantitatively analyzed by measuring phosphorylated extracellular regulated protein kinase(pERK) expression and glutamate release in the RVLM. The expression of pERK and glutamate releasein the RVLM were measured in conscious rats that had undergone bilateral labyrinthectomy (BL)and/or sinoaortic denervation (SAD) following hypotension induced by a sodium nitroprusside (SNP)infusion. The expression of pERK was significantly increased in the RVLM in the control groupfollowing SNP infusion, and expression peaked 10 min after SNP infusion. The number of pERKpositive neurons increased following SNP infusion in BL, SAD, and BL+SAD groups, although theincrease was smaller than seen in the control group. The SAD group showed a relatively higherreduction in pERK expression when compared with the BL group. The level of glutamate release wassignificantly increased in the RVLM in control, BL, SAD groups following SNP infusion, and thispeaked 10 min after SNP infusion. The SAD group showed a relatively higher reduction in glutamaterelease when compared with the BL group. These results suggest that the baroreceptors are morepowerful in pERK expression and glutamate release in the RVLM following hypotension than thevestibular receptors, but the vestibular receptors still have an important role in the RVLM.

        Genome-wide identification and analysis of long noncoding RNAs in longissimus muscle tissue from Kazakh cattle and Xinjiang brown cattle

        Objective: In recent years, long noncoding RNAs (lncRNAs) have been identified in many species, and some of them have been shown to play important roles in muscle development and myogenesis. However, the differences in lncRNAs between Kazakh cattle and Xinjiang brown cattle remain undefined; therefore, we aimed to confirm whether lncRNAs are differentially expressed in the longissimus dorsi between these two types of cattle and whether differentially expressed lncRNAs regulate muscle differentiation. Methods: We used RNA-seq technology to identify lncRNAs in longissimus muscles from these cattle. The expression of lncRNAs were analyzed using StringTie (1.3.1) in terms of the fragments per kilobase of transcript per million mapped reads values of the encoding genes. The differential expression of the transcripts in the two samples were analyzed using the DESeq R software package. The resulting false discovery rate was controlled by the Benjamini and Hochberg's approach. KOBAS software was utilized to measure the expression of different genes in Kyoto encyclopedia of genes and genomes pathways. We randomly selected eight lncRNA genes and validated them by quantitative reverse transcription polymerase chain reaction (RT-qPCR). Results: We found that 182 lncRNA transcripts, including 102 upregulated and 80 downregulated transcripts, were differentially expressed between Kazakh cattle and Xinjiang brown cattle. The results of RT-qPCR were consistent with the sequencing results. Enrichment analysis and functional annotation of the target genes revealed that the differentially expressed lncRNAs were associated with the mitogen-activated protein kinase, Ras, and phosphatidylinositol 3-kinase (PI3k)/Akt signaling pathways. We also constructed a lncRNA/mRNA coexpression network for the PI3k/Akt signaling pathway. Conclusion: Our study provides insights into cattle muscle-associated lncRNAs and will contribute to a more thorough understanding of the molecular mechanism underlying muscle growth and development in cattle.

        Nuclide assignment for an isolated band observed in the 10B + 114Cd reaction

        Three experiments based on the nuclear reactions 14N + 110Pd, 10B + 114Cd and 11B + 114Cd were performed. Coincidences between γ-rays emitted from these reactions were measured with the same detection array which consisted of 14 HPGe detectors. A new band structure was identified using an offline analysis of coincidence data. Because of the isomeric nature of the bandhead, the new band cannot be connected to any known state in any residual nuclei of the used reactions. To assign it to a specific nucleus, we evaluated the experimental population intensities of various known bands in known residual nuclei and investigated the entrance channel effects in the three different experiments. The population intensity patterns of the known bands were observed to exhibit regular systematics. Based on such experimental systematics, we suggest that the most reasonable nuclide assignment for the new band is the 120I nucleus.

        Additive Role of the Vestibular End Organ and Baroreceptors on the Regulation of Blood Pressure in Rats

        Contribution of the vestibular end organ to regulation of arterial pressure was quantitatively compared with the role of baroreceptors in terms of baroreflex sensitivity and c-Fos protein expression in the rostral ventrolateral medulla (RVLM). Baroreflex sensitivity and c-Fos protein expression in the RVLM were measured in conscious rats that had undergone bilateral labyrinthectomy (BL) and/or baroreceptor unloading. BL attenuated baroreflex sensitivity during intravenous infusion of sodium nitroprusside (SNP), but did not significantly affect the sensitivity following infusion of phenylephrine (PE). Baroreflex sensitivity became positive following sinoaortic denervation (SAD) during infusion of PE and attenuated sensitivity during infusion of SNP. Baroreflex sensitivity also became positive following double ablation (BL+SAD) during infusion of PE, and attenuated sensitivity during infusion of SNP. c-Fos protein expression increased significantly in the RVLM in the sham group after SNP administration. However, the BL, SAD, and SAD+BL groups showed significant decreases in c-Fos protein expression compared with that in the sham group. The SAD group showed more reduced c-Fos protein expression than that in the BL group, and the SAD+BL group showed less expression than that in the SAD group. These results suggest that the vestibular system cooperates with baroreceptors to maintain arterial pressure during hypotension but that baroreceptors regulate arterial pressure during both hypotension and hypertension. Additionally, afferent signals for maintaining blood pressure from the vestibular end organs and the baroreceptors may be integrated in the RVLM.

        An effective route for the fabrication of multi-walled carbon nanotubes-reinforced ROMP-based nanocomposites by solution casting technique

        <P><B>Abstract</B></P> <P>This work reports the solvent-soluble multi-walled carbon nanotubes (MWCNTs), which can be used for the fabrication of ring-opening metathesis polymerization (ROMP)-based nanocomposites by solution casting technique. As the first and most important step of the solution casting technique, the excellent dispersibility of the MWCNTs in different solvents is achieved by the functionalization of the nanotube surface with norbornene oligomers. The norbornene-functionalized MWCNTs (nMWCNTs) have outstanding dispersion stability in water, tetrahydrofuran (THF), acetone, and ethanol, especially the maximum nanotube concentration of 3.9mg/mL in THF. The incorporation of nMWCNTs into poly(5-ethylidene-2-norbornene) (poly(ENB)) by solution casting technique results in significant improvements in the mechanical properties over the neat poly(ENB) and the pristine MWCNT-reinforced poly(ENB) nanocomposites. The route developed here not only avoids the dramatic increase of the viscosity occurring in the bulk polymerization but also provides the feasibility of high loadings of MWCNT reinforcements, consequently broadening the potential applications of ROMP-based nanocomposites.</P>

        Effect of synthesis conditions on the particle size and morphology of YAG powder

        Yttrium aluminum garnet (YAG) is currently a widely applied structural and functional material. To prepare highperformance YAG ceramics we prepared pure YAG powder, after calcining the precursor at 1100 ºC, by co-precipitation with ammonia as the precipitant and aluminum nitrate as well as yttrium nitrate as raw materials. The preparation conditions for the YAG precursor were a pH of 9, a titration rate of 10 ml/ minute and a reaction time of 60 minutes. The mean paricle size of the YAG powder was 11ìm. The mean particle size for the YAG powder increased with an increase in the pH and at a higher pH the YAG particles were more irregular in shape. The mean particle size and particle size distribution range of the YAG powder increased with an increase in the titration rate. For the YAG powder with a higher mean particle size, the particle size distribution range was wider but the size of most YAG particles was still small and the YAG particles were nearly spherical.

        Numerical Investigation of the Segmental Lining Performance for a Shield Tunnel

        The performance of segmental linings in China's Yellow River Crossing Tunnel is studied as a case study, and a complete three-dimensional numerical model is developed for the segmental joint to perform a bending test and reproduce the joint rotational performance. Besides, an improved three-dimensional solid-spring model together with a straightforward two-dimensional beam-spring model are presented for the segmental lining to take the influences of the joint into account. The bending test shows that the rotation stiffness of joints presents complex and nonlinear characteristics. The segmental lining simulation reveals that the segmental joint has little impact on the axial forces of the lining. However, the bending rigidity of the segmental lining corresponding to the joint location is weakened, which leads to discontinuous stress distribution, decreased bending moments and increased deformations. According to the comparison, the proposed three-dimensional solid-spring model excluding contact has the advantage of mesh generation, and it is capable of reproducing the three-dimensional segmental lining effects. The proposed two-dimensional beam-spring model combined with the calculation methods of all the spring stiffnesses is an effective and further simplified method for the segmental lining modeling, which incorporates the influences of the joint and staggered format.

        A novel panel of serum miR-21/miR-155/miR-365 as a potential diagnostic biomarker for breast cancer

        Purpose: Insufficient sensitivity and specificity prevent the use of most existing biomarkers for early detection of breast cancer. Recently, it was reported that serum microRNAs (miRNAs) may be potential biomarkers in many cancer diseases. In this study, we investigated whether serum levels of 5 miRNAs including miR-21, miR-125b, miR-145, miR-155, and miR-365 could discriminate breast cancer patients and healthy controls. Methods: Serum levels of miRNAs were measured by using quantitative real-time polymerase chain reaction in 99 breast cancer patients and 21 healthy controls. The abundance change of serum miRNAs were also evaluated following surgical resection in 20 breast cancer patients. Receiver operating characteristic (ROC) curve analysis was performed to assess the sensitivity and specificity of miRNAs as diagnostic biomarkers. Results: Serum levels of miR-21 and miR-155 was significantly higher, while miR-365 was significantly lower in breast cancer as compared with healthy controls. The serum levels of miR-21 and miR-155 significantly decreased following surgical resection. Additionally, the serum level of miR-155 at stages I and II was significantly higher compared to stage III. The serum miR-145 level was remarkably higher in progesterone receptor (PR)-positive patients than PR-negative. The positivity of miR-21, miR-155, and miR-365 was high compared to CA 153 and CEA in breast cancer. ROC curve analyses of a combination of miR-21, miR-155, and miR-365 yielded much higher area under curve and enhanced sensitivity and specificity in comparison to each miRNA alone. Conclusion: The combination of serum miR-21/miR-155/miR-365 may potentially serve as a sensitive and specific biomarker that enables differentiation of breast cancer from healthy controls.
