        RNA m5C modification upregulates E2F1 expression in a manner dependent on YBX1 phase separation and promotes tumor progression in ovarian cancer

        5-Methylcytosine (m5C) is a common RNA modification that modulates gene expression at the posttranscriptional level, but the crosstalk between m5C RNA modification and biomolecule condensation, as well as transcription factor-mediated transcriptional regulation, in ovarian cancer, is poorly understood. In this study, we revealed that the RNA methyltransferase NSUN2 facilitates mRNA m5C modification and forms a positive feedback regulatory loop with the transcription factor E2F1 in ovarian cancer. Specifically, NSUN2 promotes m5C modification of E2F1 mRNA and increases its stability, and E2F1 binds to the NSUN2 promoter, subsequently reciprocally activating NSUN2 transcription. The RNA binding protein YBX1 functions as the m5C reader and is involved in NSUN2-mediated E2F1 regulation. m5C modification promotes YBX1 phase separation, which upregulates E2F1 expression. In ovarian cancer, NSUN2 and YBX1 are amplified and upregulated, and higher expression of NSUN2 and YBX1 predicts a worse prognosis for ovarian cancer patients. Moreover, E2F1 transcriptionally regulates the expression of the oncogenes MYBL2 and RAD54L, driving ovarian cancer progression. Thus, our study delineates a NSUN2-E2F1-NSUN2 loop regulated by m5C modification in a manner dependent on YBX1 phase separation, and this previously unidentified pathway could be a promising target for ovarian cancer treatment.

        FPGA‑based real‑time simulation of LCC‑HVDC systems with C‑NAM method

        The real-time simulation of a line-commutated converter-high voltage direct current (LCC-HVDC) system based on a field programmable gate array (FPGA) is proposed to meet the increasingly complex dynamic characteristics and real-time simulation requirements of HVDCs. In addition, a real-time simulation platform based on a CPU + FPGA is established forsimulation verification. First, a simulation model of the 12-pulse rectifier of an LCC-HVDC is established using the compact nodal analysis method (C-NAM). When compared with the traditional node analysis method, C-NAM reduces the number of multiplication executions in a single simulation step, and the degree of program serialization is signifi cantly reduced,which greatly improves the real-time simulation speed of a FPGA. Then, this simulation model is programmed in a FPGA, and the optimization algorithm further shortens the simulation step size. Finally, a 12-pulse LCC-HVDC is compared and verified with a 1 μs simulation step on a FPGA real-time simulation platform, and a simulation analysis verifies the accuracy of the model. This method can improve the simulation scale of an LCC-HVDC system, and enhance the versatility of the LCC-HVDC real-time simulator based on a FPGA.

        A FEEDBACK-DRIVEN BUBBLE G24.136+00.436: A POSSIBLE SITE OF TRIGGERED STAR FORMATION

        <P>We present a multi-wavelength study of the IR bubble G24.136+00.436. The J = 1-0 observations of (CO)-C-12, (CO)-C-13, and (CO)-O-18 were carried out with the Purple Mountain Observatory 13.7 m telescope. Molecular gas with a velocity of 94.8 km s(-1) is found prominently in the southeast of the bubble, shaped as a shell with a total mass of similar to 2 x 10(4) M-circle dot. It was likely assembled during the expansion of the bubble. The expanding shell consists of six dense cores, whose dense (a few of 10(3) cm(-3)) and massive (a few of 10(3) M-circle dot) characteristics coupled with the broad linewidths (>2.5 km s(-1)) suggest that they are promising sites for forming high-mass stars or clusters. This could be further consolidated by the detection of compact H II regions in Cores A and E. We tentatively identified and classified 63 candidate young stellar objects (YSOs) based on the Spitzer and UKIDSS data. They are found to be dominantly distributed in regions with strong molecular gas emission, indicative of active star formation, especially in the shell. The H II region inside the bubble is mainly ionized by a similar to O8V star(s), of the dynamical age of similar to 1.6 Myr. The enhanced number of candidate YSOs and secondary star formation in the shell as well as the timescales involved, indicate a possible scenario for triggering star formation, signified by the 'collect and collapse' process.</P>

        Prescribed-time Stabilization with Time-varying Output Constraints for Switched Nonlinear Systems

        This paper studies the problem of the prescribed-time stabilization for a class of switched nonlinear systems with time-varying output constraints. Firstly, a new coordinate design of the output enables the constraints to be incorporated into the original system. Secondly, a fractional term will be played an essential role in achieving prescribed-time stabilization. Then, based on the method of Barrier Lyapunov function (BLF) and backstepping, a recursive algorithm is developed to construct a controller to achieve the prescribed-time stabilization with timevarying output constraints. Finally, two examples are provided to show the effectiveness of the proposed results.

        Correlation of UGT2B7 Polymorphism with Cardiotoxicity in Breast Cancer Patients Undergoing Epirubicin/Cyclophosphamide-Docetaxel Adjuvant Chemotherapy

        Purpose: The present study aimed to investigate correlations between uridine glucuronosyltransferase 2B7 (UGT2B7) -161 singlenucleotide polymorphism C to T (C>T) and the occurrence of cardiotoxicity in Chinese breast cancer (BC) patients undergoingepirubicin/cyclophosphamide-docetaxel (EC-D) adjuvant chemotherapy. Materials and Methods: 427 BC patients who had underwent surgery were consecutively enrolled in this prospective cohortstudy. All patients were scheduled to receive EC-D adjuvant chemotherapy regimen, and they were divided into UGT2B7 -161 CC(n=141), UGT2B7 -161 CT (n=196), and UGT2B7 -161 TT (n=90) groups according to their genotypes. Polymerase chain reactionwas performed for determination of UGT2B7 -161 genotypes. Cardiotoxicity was defined as an absolute decline in left ventricularejection fraction (LVEF) of at least 10% points from baseline to a value less than 53%, heart failure, acute coronary artery syndrome,or fatal arrhythmia. Results: LVEF values were lower at cycle (C) 4, C8, 3 months after chemotherapy (M3), M6, M9, and M12 compared to C0 (allp<0.001), in BC patients undergoing EC-D adjuvant chemotherapy. Cardiotoxicity was recorded for 4.2% of the overall populationand was lowest in the UGT2B7 -161 TT group (1.1%), compared to UGT2B7 -161 CT (3.1%) and UGT2B7 -161 CC (7.8%) group(p=0.026). Multivariate logistic regression revealed that UGT2B7 -161 T allele could independently predict a low occurrence ofcardiotoxicity in BC patients undergoing EC-D adjuvant chemotherapy (p=0.004). Conclusion: A UGT2B7 -161 T allele serves as a potential biomarker for predicting a low occurrence of cardiotoxicity in BC patientsundergoing EC-D adjuvant chemotherapy.

        Impacts of peat on nitrogen conservation and fungal community composition dynamics during food waste composting

        Peat, as a heterogeneous mixture of decaying plant debris and microbial residues, has been widely used in many fields. However, little research focused on the impact of peat addition on food waste composting. To fill this gap, a composting experiment of food waste mixed with five varying percent peat 0, 5, 10, 15, and 20% (w/w, dry weight) was designed to investigate the effect of different dosages of peat on nitrogen conservation, physiochemical parameters, and fungal community dynamics during composting. The results showed that adding peat elevated the peak temperature of composting, lowered final pH, reduced ammonia emissions and increased the final total nitrogen content. Compared to control, adding 5, 10, 15, and 20% peat decreased ammonia emissions by 1.91, 10.79, 23.73, and 18.26%, respectively, during 42 days of composting. Moreover, peat addition increased fungal community diversity especially during maturation phase. The most two abundant phyla were Basidiomycota and Ascomycota in all treatments throughout the composting process. At the end of composting, in treatments with adding 10 and 15% peat, the richest fungi were Scedosporium spp. and Coprinopsis spp., respectively. Simultaneously, canonical correlation analyses showed that pH, moisture content, and seed germination index had significant association with fungal community composition. The study also showed that fungal community and nitrogen conservation had no direct obvious relation during composting. Overall, the results suggest that the addition of peat could efficiently enhance nitrogen conservation through reduction of ammonia emissions and 15% peat addition is the optimal formula for food waste composting.

        Optimization and application of multiple tuned mass dampers in the vibration control of pedestrian bridges

        An effective design approach for Multiple Tuned Mass Dampers (MTMDs) in pedestrian bridges was proposed by utilizing the transfer function to obtain each TMD’s optimum stiffness and damping. A systematic simulation of pedestrian excitations was described. The motion equation of a typical MTMD system attached to a Multi-degree-of-freedom (MDOF) system was presented, and the transfer function from the input pedestrian excitations to the output acceleration responses was defined. By solving the minimum norm of the transfer function, the parameters of the MTMD which resulted in the minimum overall responses can be obtained. Two applications of lightly damped pedestrian bridges attached with MTMD showed that MTMDs designed through this method can significantly reduce the structural responses when subjected to pedestrian excitations, and the vibration control effects were better than the MTMD when it was considered as being composed of equal number and mass ratios of TMDs designed by classical Den Hartog method.

        Optimal Blood Suppression Inversion Time Based on Breathing Rates and Heart Rates to Improve Renal Artery Visibility in Spatial Labeling with Multiple Inversion Pulses: A Preliminary Study

        Objective: To determine whether an optimal blood suppression inversion time (BSP TI) can boost arterial visibility and whether the optimal BSP TI is related to breathing rate (BR) and heart rate (HR) for hypertension subjects in spatial labeling with multiple inversion pulses (SLEEK). Materials and Methods: This prospective study included 10 volunteers and 93 consecutive hypertension patients who had undergone SLEEK at 1.5T MRI system. Firstly, suitable BSP TIs for displaying clearly renal artery were determined in 10 volunteers. Secondly, non-contrast enhanced magnetic resonance angiography with the suitable BSP TIs were performed on those hypertension patients. Then, renal artery was evaluated and an optimal BSP TI to increase arterial visibility was determined for each patient. Patients’ BRs and HRs were recorded and their relationships with the optimal BSP TI were analyzed. Results: The optimal BSP TI was negatively correlated with BR (r1 = -0.536, P1 < 0.001; and r2 = -0.535, P2 < 0.001) and HR (r1 = -0.432, P1 = 0.001; and r2 = -0.419, P2 = 0.001) for 2 readers (κ = 0.93). For improving renal arterial visibility, BSP TI = 800 ms could be applied as the optimal BSP TI when the 95% confidence interval were 17–19/min (BR1) and 74–82 bpm (HR1) for reader#1 and 17–19/min (BR2) and 74–83 bpm (HR2) for reader#2; BSP TI = 1100 ms while 14–15/min (BR1, 2) and 71–76 bpm (HR1, 2) for both readers; and BSP TI = 1400 ms when 13–16/min (BR1) and 63–68 bpm (HR1) for reader#1 and 14–15/min (BR2) and 64–70 bpm (HR2) for reader#2. Conclusion: In SLEEK, BSP TI is affected by patients’ BRs and HRs. Adopting the optimal BSP TI based on BR and HR can improve the renal arterial visibility and consequently the working efficiency.
