        Correlation between Intravoxel Incoherent Motion Magnetic Resonance Imaging Derived Metrics and Serum Soluble CD40 Ligand Level in an Embolic Canine Stroke Model

        Objective: To determine the relationship between intravoxel incoherent motion (IVIM) imaging derived quantitative metrics and serum soluble CD40 ligand (sCD40L) level in an embolic canine stroke model. Materials and Methods: A middle cerebral artery occlusion model was established in 24 beagle dogs. Experimental dogs were divided into low- and high-sCD40L group according to serum sCD40L level at 4.5 hours after establishing the model. IVIM imaging was scanned at 4.5 hours after model establishment using 10 b values ranging from 0 to 900 s/mm2. Quantitative metrics diffusion coefficient (D), pseudodiffusion coefficient (D*), and perfusion fraction (f) of ischemic lesions were calculated. Quantitative metrics of ischemic lesions were normalized by contralateral hemisphere using the following formula: normalized D = Dstroke / Dcontralateral. Differences in IVIM metrics between the low- and high-sCD40L groups were compared using t test. Pearson’s correlation analyses were performed to determine the relationship between IVIM metrics and serum sCD40L level. Results: The high-sCD40L group showed significantly lower f and normalized f values than the low-sCD40L group (f, p < 0.001; normalized f, p < 0.001). There was no significant difference in D*, normalized D*, D, or normalized D value between the two groups (All p > 0.05). Both f and normalized f values were negatively correlated with serum sCD40L level (f, r = -0.789, p < 0.001; normalized f, r = -0.823, p < 0.001). However, serum sCD40L level had no significant correlation with D*, normalized D*, D, or normalized D (All p > 0.05). Conclusion: The f value derived from IVIM imaging was negatively correlated with serum sCD40L level. f value might serve as a potential imaging biomarker to assess the formation of microvascular thrombosis in hyperacute period of ischemic stroke.

        Numerical and Experimental Study on Strain Rate Effect of Ordinary Concrete under Low Strain Rate

        The strain rate effect of ordinary concrete is researched through the particle discrete element method. A random construction method of three-dimensional aggregate is used, and the numerical sample consistent with the experimental specimen is constructed using particle flow code. Moreover, the simulation method that can properly reflect the strain rate effect with the parallel bond model is proposed. The meso-parameters are calibrated by triaxial compression test. Based on the proposed simulation method and calibrated meso-parameters, the numerical tests of direct tension, uniaxial compression, and cyclic loading under strain rates from 10−5 s−1 to 10−1 s−1 are carried out. The results show that the strain rate effect of concrete can be simulated with particle flow code by assuming that the micromechanical properties of materials vary with the strain rate, and the strength and failure characteristics of numerical samples under different strain rates are described well by the proposed method. In addition, the different mechanical responses of the samples to the strain rate in the compression test and the tensile test are obtained, and the changes of mechanical parameters and damage degree with strain rates in the cyclic loading test are also successfully simulated. This study can provide a feasible numerical method for the follow-up research of dynamic mechanical behavior of concrete and offer theoretical guidance for the stability assessment of concrete engineering.

        Cyclic Loading Tests on Composite Joints with Flush End Plate Connections

        The tests on composite joint specimens with flush end plate connections subjected to cyclic loads have been conducted inTongji University. One bare steel joint specimen has also been tested for comparison. Various instrumentations have been usedto measure: beam strains, column strains, rebar strains, connection deformations and deflections of specimens. The testobservations are reported in detail in this paper. The main results are as folows: (1) the moment-rotation relationships of thenability; (2) the composite joint specimens show large strength resistance and god ductility, and all the rotations of conectionare greater than 0.03rad as required by the FEMA-97; (3) similar failure modes have been identified from the observation ofthe composite specimens, which are found to be concentrated around the joint zone; and (4) the slipage between the concreteslab and steel beam is very small, which shows that betwen the concrete slab and steel beam the ful composition can be

      • Over-Expression of Beclin-1 Facilitates Acquired Resistance to Histone Deacetylase Inhibitor-Induced Apoptosis

        Apoptotic cell death plays a predominant role in histone deacetylase (HDAC) inhibitor-induced cytotoxicity. Nuclear morphological changes and activation of apoptotic executors are involved in CTS203-induced cell death. However, emerging issues of HDAC inhibitor-resistance have been observed in patients. Herein, MCF-7 cells were continuously exposed to CTS203 until the derived cells could proliferate normally in its presence. The newly obtained CTS203-resistant cells were nominated as MCF-7/203R. Compared to MCF-7 original cells, the MCF-7/203R cells were less sensitive to CTS203-induced apoptosis, with a minimal 6-fold higher $IC_{50}$ value. In contrast, the expression of Beclin-1 was dramatically up-regulated, positively correlated to the acquisition of CTS203-resistance. Our results revealed the participation of autophagy in acquired HDAC inhibitor-resistance and further identified Beclin-1 as a promising target for anti-drug resistance.

        Characterization on the Beam Profile for Defect Detection of Unidirectional CFRP Composites Using NDE Techniques

        Nondestructive evaluation (NDE) technology has garnered interest as an inspection tool in various industries. In this study, ultrasonic technology was applied to the defect monitoring of carbon fiber reinforced plastic (CFRP) composite materials. By using ultrasonic technology, major ultrasonic reflection signals were observed in sequential order for defect evaluation in unidirectional composites. As ultrasonic waves propagate through the unidirectional CFRP composite, mode conversion, attenuation, and scattering results in a relative decrease in the peak-to-peak amplitude of the reflected ultrasonic signals, indicating the presence of defects. In the pitch-catch mode, three to four ultrasonic reflection signals were observed, and by implementing the beam profile of the reflected signals, we confirmed that the signal-to-noise ratio can be utilized as a key parameter for NDE by providing an effective scanning range of ultrasonic amplitudes.

        CD64 Expression Is Increased in Patients with Severe Acute Pancreatitis: Clinical Significance

        Background/Aims: Upregulated CD64 expression on neutrophils is the most useful marker for acute bacterial infections and systemic inflammation. However, it is unknown whether CD64 is involved in the pathogenesis of acute pancreatitis (AP). This study was designed to determine whether CD64 is implicated in severe acute pancreatitis (SAP), and thus, is a suitable marker for SAP. Methods: SAP was induced in rats with an intraperitoneal injection of L-arginine. CD64 expression in the rat pancreas was determined by quantitative real-time polymerase chain reaction (qRT-PCR) and immunohistochemistry. Additionally, the CD64 mRNA expression in peripheral blood leukocytes from 21 patients with mild acute pancreatitis (MAP) and 10 patients with SAP was investigated at the time of admission and during remission by qRT-PCR. Results: CD64 mRNA and protein expression in the pancreas was significantly higher in rats with SAP, compared to the controls. The CD64 expression was higher in the patients with SAP than in the patients with MAP. During remission, CD64 mRNA decreased in both the MAP and SAP patients. The area under the curve of CD64 expression for the detection of SAP was superior to both the Ranson and the Acute Physiology and Chronic Health Evaluation II scores. Conclusions: The CD64 level was significantly increased in correlation with the disease severity in SAP and may act as a useful marker for predicting the development of SAP.         Partial dehulling increases the energy content and nutrient digestibility of barley in growing pigs

        Objective: The hull attached to the barley kernel can be mechanically removed thus reducing the fiber content of the barley. This experiment was carried out to evaluate the effects of partial dehulling on the nutrient digestibility as well as the digestible energy (DE) and metabolizable energy (ME) content of barley in pigs. Methods: Two hulled barley samples (high fiber barley [HF] and low fiber barley [LF]) with either high or low fiber contents were obtained from the Hubei and Jiangsu Provinces of China. A portion of the two barleys was mechanically dehulled (dehulled high fiber barley [DHF] and dehulled low fiber barley [DLF]). Thirty barrows (initial BW = 31.5±3.2 kg) were assigned to one of five diets in a randomized complete block design. The five diets consisted of 96.9% corn, HF, LF, DHF, or DLF supplemented with 3.1% minerals and vitamins. Each diet was fed to six barrows housed in individual metabolism crates for a 10-d acclimation period followed by a 5-day total but separate collection of feces and urine. Results: The daily loss of gross energy (GE) in feces was lower (p<0.01) for pigs fed DHF than for those fed HF. The daily N intake and fecal N loss were lowest (p<0.01) for pigs fed the corn diet. The DE and ME as well as the apparent total tract digestibility (ATTD) of dry matter, GE, organic matter, neutral detergent fiber (NDF) and acid detergent fiber (ADF) of DHF or DLF were higher (p<0.01) than the values in HF and LF, respectively while the values except the ATTD of NDF and ADF in DLF were higher (p<0.01) than the values in DHF and were comparable to corn. Conclusion: The DE and ME contents as well as the ATTD of nutrients in both DHF and DLF barley were improved compared with the HF and LF barley. Moreover, the nutritive value of DLF barley was comparable to the yellow-dent corn used in the study.

      • Potential Study Perspectives on Mechanisms and Correlations Between Adiposity and Malignancy

        Adiposity is a well-recognized risk factor of type 2 diabetes and cardiovascular disease, and recently there is increasing evidence that excess body weight is an avoidable cause of cancer, including gastrointestinal, endometrial, esophageal adenocarcinoma, colorectal, postmenopausal breast, prostate, and renal malignancies. The mechanisms whereby adiposity is associated with tumor development remains not well understood. There are some most studied hypothesized mechanisms such as, high levels of insulin and free levels of insulin-like growth factors, sex hormones, adipocytokines, and inflammatory cytokines, adiposity-induced hypoxia, and so on. The potential mechanisms and conclusions in adiposity associated with increased risk for developing malignancy, and the underlying cellular and molecular mechanisms will be studied very well in the near future.

      • Gene Polymorphism of XRCC1 Arg399Gln and Cervical Carcinoma Susceptibility in Asians: A Meta-analysis Based on 1,759 Cases and 2,497 Controls

        Many epidemiological studies in Asian populations have investigated associations between the Arg399Gln gene polymorphism of X-ray repair cross complementing gene 1 (XRCC1) and risk of cervical carcinoma, but no conclusions have been available because of controversial results. Therefore a meta-analysis was conducted for clarification. Relevant studies were identified by searching the Pubmed, Embase, the Web of Science, Cochrane Collaboration's database, Chinese National Knowledge Infrastructure (CNKI), Wanfang database and China Biological Medicinse (CBM) until September, 2012. A total of eight studies were included in the present meta-analysis, which described 1,759 cervical carcinoma cases and 2,497 controls. Odds ratios (ORs) and corresponding 95% confidence intervals (95%CIs) as effect size were calculated by fixed-effect or random-effect models. The overall results indicated that the XRCC1-399G/A polymorphism was marginally associated with cervical carcinoma in Asians: OR (95%CI): 1.16 (1.07, 1.26) in the G/A vs G/G inheritance model, 1.24 (0.87, 1.76)in A/A vs G/G inheritance model, 1.13 (1.01, 1.27) in the dominant inheritance model and 1.18 (0.94, 1.47) in the recessive inheritance model. Subgroup analyses on sample size showed no significant correlation in the small-sample size group but the large-sample size group was consistent with the outcomes of overall meta-analysis. In the subgroup analysis by regions, we only found significant association under the G/A vs G/G inheritance model in the Chinese population. For the non-Chinese populations, no correlation was detected in any genetic inheritance model. In the Asian populations, XRCC1-399G/A gene polymorphism was implied to be associated with cervical carcinoma.

        Single Nucleotide Polymorphisms of NLRP12 Gene and Association with Non-specific Digestive Disorder in Rabbit

        The NLRP12 (NLR family, pyrin domain containing 12) serves as a suppressor factor in the inflammatory response and protects the host against inflammation-induced damage. In the present study, we aimed to study the polymorphisms of NLRP12 gene and its association with susceptibility to non-specific digestive disorder (NSDD) in rabbits. We re-sequenced the entire coding region of the rabbit NLRP12 gene and detected a total of 19 SNPs containing 14 synonymous and five non-synonymous variations. Among them, the coding SNP (c.1682A>G), which would carry a potential functional implication, was subsequently subjected to genotyping for case-control association study (272 cases and 267 controls). The results revealed that allele A was significantly protective against NSDD with an odds ratio value of 0.884 (95% confidence interval, 0.788 to 0.993; p = 0.038). We also experimentally induced NSDD in growing rabbits by feeding a fibre-deficient diet and subsequently investigated NLRP12 mRNA expression. The mRNA expression of NLRP12 in healthy status was significantly higher than that in severe NSDD (p = 0.0016). The highest expression was observed in individuals carrying the protective genotype AA (p = 0.0108). These results suggested that NLRP12 was significantly associated with the NSDD in rabbits. However, the precise molecular mechanism of NLRP12 involving in the development of rabbit NSDD requires further research.
