      • Methylated Alteration of SHP1 Complements Mutation of JAK2 Tyrosine Kinase in Patients with Myeloproliferative Neoplasm

        SHP1 negatively regulates the Janus kinase 2/signal transducer and activator of transcription (JAK2/STAT) signaling pathway, which is constitutively activated in myeloproliferative neoplasms (MPNs) and leukemia. Promoter hypermethylation resulting in epigenetic inactivation of SHP1 has been reported in myelomas, leukemias and other cancers. However, whether SHP1 hypermethylation occurs in MPNs, especially in Chinese patients, has remained unclear. Here, we report that aberrant hypermethylation of SHP1 was observed in several leukemic cell lines and bone marrow mononuclear cells from MPN patients. About 51 of 118 (43.2%) MPN patients including 23 of 50 (46%) polycythaemia vera patients, 20 of 50 (40%) essential thrombocythaemia and 8 of 18 (44.4%) idiopathic myelofibrosis showed hypermethylation by methylation-specific polymerase chain reaction. However, SHP1 methylation was not measured in 20 healthy volunteers. Hypermethylation of SHP1 was found in MPN patients with both positive (34/81, 42%) and negative (17/37, 45.9%) JAK2V617F mutation. The levels of SHP1 mRNA were significantly lower in hypermethylated samples than unmethylated samples, suggesting SHP1 may be epigenetically inactivated in MPN patients. Furthermore, treatment with 5-aza-2'-deoxycytidine (AZA) in K562 cells showing hypermethylation of SHP1 led to progressive demethylation of SHP1, with consequently increased reexpression of SHP1. Meanwhile, phosphorylated JAK2 and STAT3 were progressively reduced. Finally, AZA increased the expression of SHP1 in primary MPN cells with hypermethylation of SHP1. Therefore, our data suggest that epigenetic inactivation of SHP1 contributes to the constitutive activation of JAK2/STAT signaling. Restoration of SHP1 expression by AZA may contribute to clinical treatment for MPN patients.

        Facile Fabrication of Electrospun Silica Nanofibrous Membrane with Hydrophobic, Oleophilic and Breathable Performances

        We report a simple and versatile method to prepare hydrophobic composite SiO2 membrane. The electrospun SiO2 membrane was selected due to its good flexibility and thermal stability. The hydrophobic SiO2 membrane was succussfully prepared by simply evaporating a thin polydimethylsiloxane (PDMS) layer on the fiber surface. The characterization results show that the PDMS layer is too thin to be observed. The PDMS coating has no influence on the porous structure of the fibrous membrane, but imparts the good hydrophobicity and oleophilicity to the SiO2 nanofibers. As demonstration, the hydrophobic SiO2-PDMS membrane displays good oil absorption performance from the oil/water mixture, as well as filtration membrane for oil/water separation. Additionally, due to the proper pore size and hydrophobic surface, the SiO2- PDMS membrane shows good waterproof performance and breathability at the same time.

        Purification and Characterization of A Novel Skatole-degrading Protease from Lactobacillus brevis 1.12

        Skatole, a substance with an unpleasant flavor,is released from the cecum and colon of entire male pigs. In this study, a novel skatole-degrading protease from the fermentation broth of Lactobacillus brevis 1.12 was extracted and purified through (NH4)2SO4 precipitation and chromatography with DEAE-Sephadex A-25 and Sephadex G-75. The purified protease was found to be homogeneous in terms of SDS-PAGE and had a molecular mass estimate of 57.5 kDa. This skatole-degrading protease had an optimal pH of 5.0 and optimal temperature of 35oC. The enzyme was only inhibited greatly at a 10 mmol 1/L concentration of K+. The results suggest that the novel skatole-degrading protease was purified. The biochemical characteristics of this protease can provide useful information for its potential use in industrial applications.

        Distinctive Slow β Relaxation and Its Impact on Mechanical Property of LaCe Based Bulk Metallic Glasses

        The slow β relaxation which occurring at temperatures far below glass transition temperature, typically playing importantroles in the plastic deformation behavior of metallic glasses. Compared with polymer glasses, most of the metallic glasses donot exhibit obvious β relaxation based on dynamic mechanical analysis. In the current work, prominent β relaxation behaviorsof a series (LaxCe100−x) Al10Co25(x = 50, 60, 70, 80) bulk metallic glasses (BMGs) at low temperature are observed usingdynamic mechanical analysis. Compared with other BMGs, the LaCe based BMGs show a pronounced β relaxation peak andrelative low peak temperature, the activation energy of the β relaxation based on Arrhenius equation are calculated around79.3 kJ/mol to 86.4 kJ/mol, which obey an empirical rule that Eβ ≈ (26 ± 4) RTg, with the coefficient of 23. Experimentalresults from room temperature compression tests for the LaCe based BMGs indicate that low temperature activated β relaxationbehaviors facilitate the good plasticity of the LaCe based BMGs.

        Variable Research on Engineering Characteristics of Soybean Urease Reinforced Sand

        In this study, based on soybean urease and standard sand, the unconfined compressive strength test and permeability test of four variables of urease concentration, urea concentration, calcium chloride concentration and sodium alginate content under different combinations of values were carried out, and the range analysis and variance analysis of test data were carried out. On this basis, the coupling model of unconfined compressive strength and permeability coefficient of soybean urease reinforced sand was established based on four test variables. It was found that the engineering characteristics of soybean urease reinforced sand were significantly affected by the combination of variable values. The maximum value of unconfined compressive strength under the test combination condition was 6.5 times of the minimum value, and the minimum value of permeability coefficient was only 1/4 of the maximum value. The primary and secondary order of influence variables of unconfined compressive strength was: urea concentration > calcium chloride concentration > urease concentration > sodium alginate content, and the primary and secondary order of influence variables of permeability coefficient was: urea concentration > calcium chloride concentration > sodium alginate content > urease concentration. The influence of urea concentration on unconfined compressive strength and permeability coefficient was obvious, and the influence of the other three variables was not obvious. In addition, it was confirmed that the established coupling model of unconfined compressive strength and permeability coefficient of soybean urease reinforced sand had high correlation coefficient and significant linear regression, which can be used for subsequent research and engineering application.

        Clinical Impact of Drug Adherence of Tyrosine Kinase Inhibitors in Children with Ph-Positive Acute Lymphoblastic Leukemia

        Purpose This study aimed to explore the impact of ABL1–tyrosine kinase inhibitors (TKIs) adherence on the survival of chromosome-positive (Ph+) acute lymphoblastic leukemia (ALL) children and clarify the potential predictors of patients’ prognosis from TKIs intake practices.Materials and Methods Ninety newly diagnosed Ph+ ALL patients who received TKIs were enrolled. We collected the baseline characteristics and adverse events in all children; moreover, TKIs adherence was measured by an eight-item Morisky medication adherence scale (MMAS-8). Progression-free survival (PFS) and overall survival (OS) analysis were performed, and risk factors for PFS and OS were evaluated.Results Among all patients, 69 cases were regarded as adherers, while 21 were non-adherers. The median duration of TKIs interruption was significantly prolonged in the non-adherence group than in the adherence group (13 [0-101] vs. 56 [11-128], p < 0.001). Additionally, dose reduction occurred in 55.2% of non-adherers versus 23.0% of adherers (p=0.002). The PFS and OS in adherers were significantly higher versus non-adherers (p=0.020 and p=0.039). MMAS-8 score was an independent risk factor for PFS (p=0.010) and OS (p=0.031). Among non-adherers, the median OS was only 23.1% (4.2%-42%) in patients aged ≤ 10 years versus 54.4% (38.8%-70%) in adolescents. Most of the patients who experienced TKIs non-adherence suffered pancytopenia.Conclusion TKIs adherence during treatment significantly influenced the survival of pediatric Ph+ ALL patients, and non-adherers with age ≤ 10 years were more vulnerable to TKIs disruption. The cumulative TKIs dose should be especially emphasized to patients with age ≤ 10 years, which may result in an inferior achievement of relevant treatment milestones.

        Molecular detection and genetic diversity of bovine papillomavirus in dairy cows in Xinjiang, China

        Background: Bovine papillomatosis is a type of proliferative tumor disease of skin and mucosae caused by bovine papillomavirus (BPV). As a transboundary and emerging disease in cattle, it poses a potential threat to the dairy industry. Objectives: The aim of this study is to detect and clarify the genetic diversity of BPV circulating in dairy cows in Xinjiang, China. Methods: 122 papilloma skin lesions from 8 intensive dairy farms located in different regions of Xinjiang, China were detected by polymerase chain reaction. The genetic evolution relationships of various types of BPVs were analyzed by examining this phylogenetic tree. Results: Ten genotypes of BPV (BPV1, BPV2, BPV3, BPV6, BPV7, BPV8, BPV10, BPV11, BPV13, and BPV14) were detected and identified in dairy cows. These were the first reported detections of BPV13 and BPV14 in Xinjiang, Mixed infections were detected, and there were geographical differences in the distribution of the BPV genotypes. Notably, the BPV infection rate among young cattle (< 1-year-old) developed from the same supply of frozen sperm was higher than that of the other young cows naturally raised under the same environmental conditions. Conclusions: Genotyping based on the L1 gene of BPV showed that BPVs circulating in Xinjiang China displayed substantial genetic diversity. This study provided valuable data at the molecular epidemiology level, which is conducive to developing deep insights into the genetic diversity and pathogenic characteristics of BPVs in dairy cows.

        The Dynamic Mechanical Properties for Recycled Aggregate Concrete under Tensile-Compressive States

        The experiments under biaxial tensile-compressive (T-C) complex stress were carried out for two kinds of substitution percentages of recycled coarse aggregate (RCA) on biaxial proportional loading (α = σ1:σ3 =1:0, 0: -1, 0.05: -1, 0.1: -1, 0.25: -1, 0.5: -1, 1: -1 and-1: -1) and four loading deformation rates of 10−5/s, 10−4/s , 10−3/s, and 10−2/s. The research was completed in the static-dynamic true triaxial machine. The experimental phenomena indicate the tensile failure appears in these specimens of recycled aggregate concrete (RAC) under biaxial T-C stress states. The dynamic mechanical behavior for RAC under biaxial T-C loadings was affected by the strain rates, stress ratios and its substitution percentages of RCA. Accompanied by the increase of strain rates, its strength of RAC is improved. As the tensile stress increases, its compressive strength decreases. The T-C strengths for RAC under biaxial T-C states are less than its corresponding uniaxial T-C strengths at a certain identical strain rates, respectively. Considering the effect of loading rates, a new tensile-compressive failure criterion is established under complex stress states.

      • Lack of Effects of HER-2/neu on Prognosis in Colorectal Cancer: a Meta-analysis

        Background: The prognostic value of human epidermal growth factor receptor-2 (HER-2/neu) for survival of patients with colorectal cancer (CRC) is still ambiguous. We therefore performed a meta-analysis to evaluate its prognostic significance. Materials and Methods: We searched the MEDLINE and EMBASE databases for published literature investigating associations between HER-2/neu status and overall survival of patients with CRC. A meta-analysis was performed using a DerSimonian-Laird model and publication bias was investigated by Begg's and Egger's tests. Subgroup analysis was also conducted according to the study design type, study quality score, cut-off value for HER-2/neu overexpression, publication region, patient number and publication year. Results: A total of 17 eligible studies involving 2,347 patients were identified for this meta-analysis. The combined hazard ratio (HR) was 1.31 (95% confidence interval (CI): 0.96-1.79), suggesting that HER-2/neu overexpression was not significantly associated with overall survival of patients with CRC. However, subgroup analysis revealed that HER-2/neu overexpression had an unfavorable impact on survival when the analysis was restricted to subgroups of study quality score ${\leq}5 $(HR=1.56, 95%CI: 1.17-2.10), Asian patients (HR=1.74, 95%CI: 1.22-2.49), patient number ${\leq}106$ (HR=1.57, 95%CI: 1.01-2.44), publication year before 2003 (HR=1.59, 95%CI: 1.02-2.49), and prospectively designed study (HR=3.62, 95%CI: 1.42-9.24). The effect disappeared in subgroups of study quality scores > 5 (HR=0.69, 95%CI: 0.33-1.44), non Asian patients (HR=1.14, 95%CI: 0.77-1.70), patients' number > 106 (HR=1.07, 95%CI: 0.67-1.72), publication year after 2003 (HR=1.13, 95%CI: 0.76-1.69), and retrospectively designed study (HR=1.22, 95%CI: 0.89-1.67). Conclusions: Our meta-analysis suggests that HER-2/neu overexpression might not be a significantly prognostic indicator for patients with CRC. Further studies are required to confirm these results.

        Experimental and Numerical Investigation on the Dynamic Responses of the Remaining Structure under Impact Loading with Column Being Removed

        The dynamic response and stability of building structures under accidental loads such as earthquake, explosion, and impact have been focused by experts. The significant influence will be induced once the damaged structure begins to collapse. In this study, a reinforced concrete frame structure model was designed and tested to study the dynamic response law of the structure under the condition of the failure of the middle column of the model structure, the remaining structure successively failed the load-bearing column and continued to be subjected to impact load and uniform load. The analytical method was used to calculate the ultimate uniform load subjecting to the structure under the failure of four load-bearing columns, because its value was less than the load in this test, the structure collapsed. The finite element calculation model was established, and compared the simulation results to the test results to verify its rationality. Then, the influence of the impact energy and the structural stores on the dynamic response of the structure were analyzed. The results showed that the peak value of the dynamic response of the damaged structure did not increase strictly with the increase of the number of failure columns, which may be related to the sequence of failure columns.
