      • Effect of High CO<sub>2</sub> Pretreatment and MA Packaging on the Change of Sensory Quality, Physiological and Biochemical Characteristics of Green Asparagus (Asparagus officinalis L.) during Postharvest Storage

        Asparagus is vulnerable to thrips that is the major insect pest of asparagus. The effect of high carbon dioxide pretreatment (60%CO2, v/v) for killing thrips on asparagus sensory quality, physiological and biochemical changes during postharvest storage was investigated. After 60% of CO<sub>2</sub> pretreatments for 24h(CO<sub>2</sub>24h) at ambient temperature or for 48h(48h) or 72h(72h) at 4°C, the asparagus were stored in MA package (10,000 cc/m²ㆍdayㆍatm oxygen transmission rate film) and non-MA package (perforated film) at 4°C for 30 and 25 days. In all treatments, MA package obtained 5 days longer shelf life than non-MA package. Irrespective of the duration, CO<sub>2</sub>24h was worst in non-MA package but 72h in MA package with highest fresh weight loss rate. The O<sub>2</sub> and CO<sub>2</sub> content was similar in all packages, nearly 19-21% and 6-11% respectively. The ethylene content was little affected in CO<sub>2</sub>24h, but lower at 4°C. In MA packages, the concentration of ethylene was nearly 1ppm whereas was 2ppm in non-MA package in the end. The best visual quality was obtained in 48h MA packages which kept the marketability that determined by the score≥3, and the worst was the control(air) 24h in non-MA packages. The off-odor was similar with the visual quality, and lower temperature inhibited the odor, it indicates that the temperature and duration of CO<sub>2</sub> pretreatment affect asparagus sensory quality during the postharvest storage. It is worth noted that control 60h has the lowest firmness in tip and stem of spear in MA packages, and 60% CO<sub>2</sub> increased the firmness. The CO<sub>2</sub> pretreatments retarded hue angle decrease which related to duration in tip but not in stem. CO<sub>2</sub> pretreatments were obviously reduced the soluble solids content at 4°C and ambient temperature and the lowest was CO<sub>2</sub>24h and the highest was 48h. The electrolyte leakage was less in CO<sub>2</sub> pretreatments in MA packages. In conclusion, the effect of high CO<sub>2</sub> pretreatments on asparagus was depended on the duration and temperature, 48h in MA package was better in keeping the sensory quality physiological and biochemical characteristics.

        포장 방법에 따른 아스파라거스의 저온저장 중 품질변화

        저장 중 생체중 감소율은 MAP 처리구들은 0.5% 이하의 낮은 수치를 보였으며, 무포장 대조구의 경우 저장 종료일 3%에 육박하였으나, 아스파라거스 최대 생체중 감소 허용량보다는 낮았다. 포장 내 이산화탄소 농도는 동일한 필름을 사용하였으나 포장 단위에 따라 크게 차이를 보였는데, 소포장 MA 처리구는 3% 내외를 유지하였고 6kg 박스 MAP 처리구는 최대 12%까지 증가하였다. 포장 내 에틸렌 농도는 MA저장한 100g과 6kg 포장 모두 저장 직후 3일까지 급속히 증가하였다가 이 후 감소하여 저장 종료일까지 5μL·L<SUP>-1</SUP> 수준을 유지하였다. 저장 중 패널테스트로 조사한 외관상 품질은 저장 15일째부터 처리에 따른 차이가 나타나 종료일에 6kg 박스 MA처리구가 가장 양호하였고, 소포장 MA 처리구가 가장 낮았다. 이취는 소포장 MA가 가장 높았으며, 6kg박스 대조구가 가장 낮았다. 경도의 경우 처리에 따른 차이에 통계적 유의성은 나타나지 않았는데, 화두는 소포장 처리구, 줄기는 6kg 박스 MA처리구가 가장 높았다. 저장 종료일의 전해질 용출량은 소포장 MA 처리구가 가장 낮았으며 6kg 박스 대조구가 가장 높았다. 화두와 줄기의 Hue angle은 모두 저장 기간이 길어짐에 따라 감소하는 경향을 보였으나, 처리구간의 차이가 나타나지 않았다. 저장 10일째 총 세균수는 소포장 MA 처리구가 가장 낮았으나, 종료일인 20일째에는 모든 처리구에서 유사하게 나타났다. 대장균수는 10일째 모든 처리구에서 검출되지 않았으나, 20일째에는 6kg 박스 MA 처리구가 가장 낮았으며 곰팡이는 저장기간 모든 처리구에서 관찰되지 않았다. Effect of 6kg large unit with the carton box (20% open ratio) and MA box (10,000cc·m<SUP>-2</SUP>·day<SUP>-1</SUP>·atm<SUP>-1</SUP> oxygen transmission rates modified atmosphere package), and the 100g small unit with MA film on asparagus sensory quality were evaluated. The CO₂ concentration depended largely on the packing unit and maintained at around 3% in small MA packages, whereas in the MA box increased to 12%. Ethylene concentration rapidly increased until after 3 days of storage in MA packages and then decreased to maintain 5μL·L<SUP>-1</SUP>. Unrelated to the unit size, the lower weight loss was obtained in MA packages. A significant difference in visual quality was shown since the 15th day, the best and worst were the MA box and small MA package on the finish day. Off-odor was the highest in small MA packages and the lowest in the carton box (< 3.0). Although there was no significant difference in firmness among all treatments, the packages showed the highest firmness in tips and stems, respectively. The sugar content and hue angle decreased during storage, but there was no statistical difference in all treatments. EL was lowest and highest in small MA package and carton box, respectively. On the 10th day, the total aerobic bacteria was lowest in small MA packages, but no significant difference on the 20th day. E. coli was not found in all treatments on the 10th day, while it was the lowest in the MA box on the 20th day. The mold and yeast were not observed during the whole storage. Based on the above results, the carton box packaged with 10,000cc OTR film was more effective in maintaining the quality of green asparagus with the suitable CO₂ concentration for asparagus cold storage.

      • The Heuristic Algorithm of Wavelet Image Denoising Based on Rough Set

        In this paper, we propose a novel approach to explore image denoising for patch based image process. The importance measurement model of Rough Entropy and the importance reduction method of wavelet coefficients are given. This paper combines the rough set theory with the denoising method of wavelet threshold, regarding the high-frequency information blocks in the transform domain as similar ones, and adopting importance Reduction Methods to contract the coefficients. The simulation results show that this method is effective.

        Genome wide methylation analysis to uncover genes related to recurrent pregnancy loss

        Background Recurrent pregnancy loss (RPL) refers to two or more consecutive spontaneous abortion before 24 weeks of gestation, representing 1% of couples of childbearing age. Epigenetic factors including dysregulation of DNA methylation of some genes may play a role in RPL. Objective To identify RPL related genes modulated by DNA methylation expressed in decidua and blood. Methods Three decidua samples each from RPL patients and normal controls were recruited to perform genome-wide bisulfte sequencing (GWBS) and transcriptome sequencing. Based on the above results, 22.52 kb of diferential methylation regions (DMRs) from 17 genes were verifed by bisulfte sequencing PCR at specifc region (Hi-MethylSeq) in another 15 decidua (7RPL vs. 8 Controls) and 13 blood (5RPL vs. 8 Controls) samples. Results 23 genes showed signifcantly diferential cytosine methylation status and distinct expression level between PRL patients and healthy controls synergistically. Three signaling pathways were found to be shared between genes with both hypomethylated diferential methylation regions (DMR) and upregulated diferential gene expression (DGE). The results from Hi-MethylSeq showed that the hypermethylation of SGK1 in both blood and decidua samples in RPL patients, which was consistent to its lower expression in endometrium reported earlier. SGK3 and CREB5 also showed modulated methylation level in RPL decidua. Conclusion Our fnding supported that aberrant methylation of SGK1 and CREB5 could be a cause of the dysregulation of these gens in the endometrium, which is one of cause of reproductive failure. The function of SGK3 in reproduction system deserves further investigation.

        Investigating Impacts of CoronaVac Vaccination in Males on In Vitro Fertilization: A Propensity Score Matched Cohort Study

        Purpose: This study aimed to evaluate the influences of SARS-CoV-2 vaccination (CoronaVac) on male fertility and investigate the impact of a history of the CoronaVac vaccination in males on gamete and embryo development and in vitro fertilization (IVF) outcomes. Materials and Methods: A prospective cohort study enrolled couples undergoing IVF cycles between June and August 2021 at Reproductive Medicine Center, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology in China. According to the history of SARS-CoV-2 vaccination in males, the participants were divided into the vaccination group and the non-vaccination group. A self-controlled study of semen analyses for males before and after CoronaVac vaccination was conducted. Baseline characteristics were matched using propensity score matching. Participants were categorized into the unexposed group (non-vaccination) and exposed group (vaccination), and the population was 271 for each. Semen parameters and IVF outcomes were the main outcomes. Results: Generally, no statistically significant differences were exhibited between the matched cohorts regarding embryo developmental parameters, including fertilization rate, cleavage rate, high-quality embryo rate, blastocyst formation rate, and available blastocyst rate, as well as clinical outcomes, such as implantation rate, biochemical pregnancy rate, and clinical pregnancy rate. Moreover, males after vaccination seemed to have fluctuating semen parameters including increased semen volume, lower motility, and decreased normal forms of sperm, while the motile sperm counts were similar. In addition, all semen parameters were above the lower reference limits. Conclusions: Our findings suggested that CoronaVac vaccinations in males may not have adverse effects on patient performance or the gamete and embryonic development potential during assisted reproductive technology (ART) treatments.

      • Multi-target Tracking Algorithm Based on TLD under Dynamic Background

        The tracking of multi-target under dynamic background is a challenging issue in computer vision. We improve TLD algorithm by introducing multi-threading mechanism to expand the number of the tracking targets. Thus the excellent framework of TLD for accurate long-time single target tracking is kept, and the tracking of multiple moving targets is realized at the same time. The time cost of tracking also reduces effectively by this algorithm to meet the real-time demand in many dynamic scenes.

        Identification and differential expression of microRNAs in Madin–Darby canine kidney cells with high and low tumorigenicities

        Background: Madin-Darby canine kidney (MDCK) cells are widely used for vaccine production, however, the safety of MDCK cells needs to be considered seriously because of high tumorigenicity. Micro RNAs (miRNAs) that are involved in the tumorigenicity of MDCK cells have been never been reported. Objective: To reveal the role of miRNA in the tumorigenic phenotype of MDCK cell line. Methods: The miRNA expression profiles of two monoclonal MDCK cells (M09CL and M35CL) with low tumorigenicity and one MDCK cell line (M73P) with high tumorigenicity were characterized and investigated by using small RNA-seq technology. Results: A total of 5 known miRNAs and 5 novel miRNAs were highly expressed in M73P. In addition, 4 known miRNAs and 4 novel miRNAs were highly expressed in M09CL and M35CL. The target genes of the differentially expressed miRNAs were significantly enriched in several biological processes, and the majority of these genes were involved in pathways in cancer and the MAPK signaling pathway. Through interaction analysis, 4 up-regulated miRNAs (cfa-miR-452, cfa-miR-8826, cfa-miR-224, and cfa-miR-2387) and their crucial target genes related to the tumor regulation network were identified. Results indicated these 4 miRNAs might play crucial roles in the tumorigenesis of MDCK cells. Conclusion: Our findings, which were based on the functional prediction of miRNAs and target genes, suggested that miRNAs might influence the tumorigenicity of MDCK cells by regulating target genes. Moreover, the results provided important data for understanding the miRNA-mediated regulatory networks that control the tumorigenicities of MDCK cells.
