        Diaporthe taoicola and D. siamensis, Two New Records on Citrus sinensis in China

        Two Diaporthe species isolated from fruit of Citrus sinensis in China were characterized based on morphology and multilocus phylogeny of ITS, tef1, and tub2 gene sequences. The phylogeny indicated that the two species match Diaporthe taoicola and D. siamensis. A critical examination of phenotypic characteristics confirmed the phylogenetic results. Diaporthe taoicola was morphologically characterized by producing Alpha conidia with tapering toward both ends. Meanwhile, D. siamensis produced cylindrical or ellipsoidal Alpha conidia with two oil drops. Pathogenicity tests revealed that both species were pathogenic to fruit of C. sinensis. To our knowledge, the two species were firstly reported on Citrus sinensis in China.

        Variable Research on Engineering Characteristics of Soybean Urease Reinforced Sand

        In this study, based on soybean urease and standard sand, the unconfined compressive strength test and permeability test of four variables of urease concentration, urea concentration, calcium chloride concentration and sodium alginate content under different combinations of values were carried out, and the range analysis and variance analysis of test data were carried out. On this basis, the coupling model of unconfined compressive strength and permeability coefficient of soybean urease reinforced sand was established based on four test variables. It was found that the engineering characteristics of soybean urease reinforced sand were significantly affected by the combination of variable values. The maximum value of unconfined compressive strength under the test combination condition was 6.5 times of the minimum value, and the minimum value of permeability coefficient was only 1/4 of the maximum value. The primary and secondary order of influence variables of unconfined compressive strength was: urea concentration > calcium chloride concentration > urease concentration > sodium alginate content, and the primary and secondary order of influence variables of permeability coefficient was: urea concentration > calcium chloride concentration > sodium alginate content > urease concentration. The influence of urea concentration on unconfined compressive strength and permeability coefficient was obvious, and the influence of the other three variables was not obvious. In addition, it was confirmed that the established coupling model of unconfined compressive strength and permeability coefficient of soybean urease reinforced sand had high correlation coefficient and significant linear regression, which can be used for subsequent research and engineering application.

        Eco-Friendly Electrochemical Biosensor based on Sodium Carboxymethyl Cellulose/Reduced Graphene Oxide Composite

        Reduced graphene oxide (GO) and carboxymethyl cellulose sodium (CMC) composite film was prepared by a simple solution blending method. The obtained composite film was used to modify glassy carbon electrode (GCE) to detect Vitamin B6 (VB6) with electrochemical measurements. Morphologies and stucture of the composite were characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD), fourier transform infrared spectroscopy (FTIR) and electron paramagnetic resonance (EPR). The electrochemistry performances of the GCE modified by GO/CMC composite were investigated by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). The results revealed that CMC and GO were able to form a homogeneous mixture, and the modified GCE presented extremely strong sensitivity towards VB6 such as high detection sensitivity, low detection limit, small Ret value, certain stability and excellent reversibility. The good performance might be attributed to high electrical conductivity and large surface-to-volume ratio of GO which could accelerate electron transfer between the surface of GCE and VB6. The test results showed that GO/CMC composite was a novel electrochemical biosensor for the detection of VB6.

        Facile Fabrication of Electrospun Silica Nanofibrous Membrane with Hydrophobic, Oleophilic and Breathable Performances

        We report a simple and versatile method to prepare hydrophobic composite SiO2 membrane. The electrospun SiO2 membrane was selected due to its good flexibility and thermal stability. The hydrophobic SiO2 membrane was succussfully prepared by simply evaporating a thin polydimethylsiloxane (PDMS) layer on the fiber surface. The characterization results show that the PDMS layer is too thin to be observed. The PDMS coating has no influence on the porous structure of the fibrous membrane, but imparts the good hydrophobicity and oleophilicity to the SiO2 nanofibers. As demonstration, the hydrophobic SiO2-PDMS membrane displays good oil absorption performance from the oil/water mixture, as well as filtration membrane for oil/water separation. Additionally, due to the proper pore size and hydrophobic surface, the SiO2- PDMS membrane shows good waterproof performance and breathability at the same time.

        Synthesis and Structural Studies of an Organic Complex and its Association with BSA

        The self-assembly of one novel organic complex based on chlorogenic acid (HCA) and 2,2'-bipyridine (2,2'-bipy) has been synthesized and characterized. The complex achieved by hydrogen-bonding interactions, adopted a 1:1 stoichiometry in a solid state. The proton transfer occurred from the carboxyl oxygen to the aromatic nitrogen atom to form salts CA${\cdot}$(2,2'-Hbipy), the 2,2'-Hbipy molecule individually occupies the pseudo-tetragonum that is formed with CA. In this paper, the interactions of CA${\cdot}$(2,2'-Hbipy) with bovine serum albumin (BSA) were studied by fluorescence spectrometry. For CA${\cdot}$(2,2'-Hbipy), HCA and 2,2'-bipy, the average quenching constants for BSA were $2.4384{\times}10^4$, $4.653{\times}10^3$, and $3.059{\times}10^3\;L{\cdot}mol^{-1}$, respectively. The mechanism for protein fluorescence quenching is apparently governed by a static quenching process. The Stern-Volmer quenching constants and corresponding thermodynamic parameters ${\Delta}$H, ${\Delta}$G and ${\Delta}$S were calculated. The binding constants and the number of binding sites were also investigated. The conformational changes of BSA were observed from synchronous fluorescence spectra.

        Inhibitory Effects of Crocetin On High Glucose-induced Apoptosis in Cultured Human Umbilical Vein Endothelial Cells and Its Mechanism

        Dysfunction of endothelial cell is considered as a major cause of vascular complications in diabetes. Crocetin has been shown to have strong antioxidant activities. In present study, we tested whether crocetin inhibited high glucose-induced apoptosis in cultured human umbilical vein endothelial cells $(HUVEC_s)$ and to explore its possible mechanism. Exposure to high glucose (33 mM) for 72h induced a pronounced increase in apoptosis compared with normal glucose (5.5 mM),as evaluated by cell chromatin staining with Hoechst 33,258 and cell death detection ELISA. High glucose attenuated activation of Akt and endothelial nitric oxide synthase (eNOS). Crocetin (0.1 ${\mu}M$, 1.0 ${\mu}M$) prevented high glucose-induced apoptosis, which correlates with the increase of activation of p-Akt, following the up-regulation of eNOS and NO production. Pretreatment with phosphatidylinositol 3' kinase (PI3K) inhibitor LY294002 or eNOS inhibitor $N^G-nitro-$ arginine methyl ester (LN or L-NAME) inhibited crocetin' effect on p-Akt or eNOS, respectively. For the first time, results of our study suggest that crocetin inhibits high glucose-induced apoptosis, at least partly, via PI3K/Akt/eNOS pathway in $(HUVEC_s)$ and crocetin may exert a beneficial effect in preventing diabetes-associated cardiovascular complications.

        Octadecyl-Modified Graphene as an Adsorbent for Hollow Fiber Liquid Phase Microextraction of Chlorophenols from Honey

        Octadecyl-modified graphene (graphene-C18) was fabricated and used as adsorbent in hollow fiber liquid phase microextraction (HF-LPME) for the first time. The extraction performance of graphene-C18 reinforced HF-LPME was evaluated using chlorophenols as model analytes. The factors affecting the extraction efficiency, such as extraction time, pH of the sample solution, agitation rate, the concentration of graphene-C18 and salt addition were optimized. After the graphene-C18 reinforced HF-LPME of the chlorophenols from honey sample, the analytes were separated and determined by high-performance liquid chromatography. The linearity was observed in the range of 5.0-200.0 ng g−1 for 2-chlorophenol and 3-chlorophenol, and 2.0-200.0 ng g−1 for 2,3-dichlorophenol and 3,4-dichlorophenol, respectively. The limits of detection (S/N = 3) of the method were lower than 1.5 ng g−1. The recoveries of the method were between 88% and 108%. The method is simple, sensitive and has been resoundingly applied to analysis of chlorophenols in honey samples.

        Preparation and Characterization of an Antibody Antagonist That Targets the Porcine Growth Hormone Receptor

        A series of antagonists specifically targeting growth hormone receptors (GHR) in different species, such as humans, rats, bovines, and mice, have been designed; however, there are currently no antagonists that target the porcine growth hormone (GH). Therefore, in this study, we developed and characterized a porcine GHR (pGHR) antibody antagonist (denoted by AN98) via the hybridoma technique. The results from enzyme-linked immunosorbent assay, fluorescence activated cell sorter, indirect immunoinfluscent assay, and competitive receptor binding analysis showed that AN98 could specifically recognize pGHR, and further experiments indicated that AN98 could effectively inhibit pGH-induced signalling in CHO-pGHR cells and porcine hepatocytes. In addition, AN98 also inhibited GH-induced insulin-like growth factor-1 (IGF-1) secretion in porcine hepatocytes. In summary, these findings indicated that AN98, as a pGHR-specific antagonist, has potential applications in pGH-pGHR-related research on domestic pigs.

      • Plasma Folate and Vitamin B <sub>12</sub> Levels in Patients with Hepatocellular Carcinoma

        <P>Folate and vitamin B<SUB>12</SUB> involved in the one-carbon metabolism may play a key role in carcinogenesis and progression of hepatocellular carcinoma (HCC) through influencing DNA integrity. The purpose of this study is to evaluate the association of plasma folate and vitamin B12 levels with HCC in a case-control study on 312 HCC patients and 325 cancer-free controls. Plasma concentrations of folate and vitamin B<SUB>12</SUB> in all the subjects were measured by electrochemiluminescence immunoassay. Meanwhile, the information of HCC patients’ clinical characteristics including tumor-node-metastasis (TNM) stage, tumor size and tumor markers were collected. The patients of HCC had significantly lower folate levels than those of controls; there was no significant difference in the mean of plasma vitamin B<SUB>12</SUB> levels. We also observed an inverse association between the levels of plasma folate and HCC: the adjusted odds ratios (OR) (95% confidence intervals (CI)) of HCC from the highest to lowest quartile of folate were 0.30 (0.15–0.60), 0.33 (0.17–0.65), and 0.19 (0.09–0.38). Compared to the subjects in the lowest quartile of plasma vitamin B<SUB>12</SUB>, only the subjects in the highest quartile of vitamin B<SUB>12</SUB> exhibited a significant positive relationship with HCC, the adjusted OR was 2.01 (95% CI, 1.02–3.98). HCC patients with Stage III and IV or bigger tumor size had lower folate and higher vitamin B<SUB>12</SUB> levels. There was no significant difference in the mean plasma folate levels of the HCC cases in tumor markers status (AFP, CEA and CA19-9 levels), whereas patients with higher CEA or CA19-9 levels retained significantly more plasma vitamin B<SUB>12</SUB> than those with normal-CEA or CA19-9 level. In conclusion, plasma folate and vitamin B<SUB>12</SUB> levels could be associated with HCC, and might be used as predictors of clinical characteristics of HCC patients. However, further prospective studies are essential to confirm the observed results.</P>
