      • 참죽나무(Cedrela sinensis Juss)의 培養細胞로 부터 體細胞胚 形成과 人工種子 生産

        참죽나무의 배양세포를 물리 및 화학적인 복합방법으로 동조화를 거쳐 균일한 체세포배형성을 시도하였던바 85%의 높은 향상율을 보였다. 이에 다시 몇가지 아미노산, ABA 및 저온처리를 하여 얻은 정상적인 체세포배를 인공종자로 만들었다. 아미노산 중에서 aspartic acid 및 asparagine의 처리는 정상체세포배형성에 효과적이었는데 인공종자발아에는 별로 성과를 나타내지 않았다. ABA 및 저온처리로 정상체세포배형성율을 향상시킬 수 있었으나 인공종자의 발아에는 저온처리를 한 경우가 더욱 효과적이었다. 그러므로 간단하게 저온처리를 하여 경화된 건실한 체세포배 및 인공종자를 얻을 수 있고 이로 부터 클론식물의 대량생산이 가능할 것으로 사료된다. Somatic embryogenesis in cell suspension cultures of Cedrela sinensis Juss. was synchronized by physical and chemical treatments. Using culture medium containing aspartic acid and asparagine of 5mM each, the quality of somatic embryo from synchronized cell clumps was enhanced, but it was not effective for artificial seed germination. Somatic embryos were singly encapsulated with alginic acid to be used as an artificial seed. To improve the germination rate of artificial seeds, somatic embryos were hardened by ABA and low temperature(4℃) treatments before encapsulation. Low temperature treatment was more effective than ABA treatment.

        김종직 문인 그룹 형성 무대로서의 '서울'

        Terms like 'Yeongnam Sarim(-pa) faction' or 'Group of Kim Jong Jik's students' have suggested for a long time the existence of a group of literary figures whose origin was the Yeongnam region and who were joining each other to form a political presence and most importantly act as a political faction, with Kim Jong Jik at the center. But this kind of notion regarding the nature of this faction shouId be reexamined, as there are aspects displayed by this group rhar have not been accounted for. The politics during King Seongjong's reign was not inducive to (or allowing) political parties, and the Sam-sa ofFices which served as a political stage for Kim's students were actually serving at the time as the intellectuals' most active conduit to high ranking governmental seats. Also, among Kim's students there were a lot of people who were not from the Yeongnam region, and many of them were from renowned houses whose influences even matched the existing high ranking officials' powerful houses. Roughly half of Kim's students were from the capital region. Around 51% of them had ancestors who passed the Mun-gwa national examination. 31% of them had ancestors who had been named meritorious vassals 13% of them even had marital relationships with the Royal family. Also, many of them came to study under the guidance of Kim not because they were living in the same region(Yeongnam), but because they had the chance of visiting him through certain opportunities such as Kim being assigned to several regions they were living in. In that regard, it would be inappropriate to suggest Kim's group was strongly based upon certain local Sajok communicies Kim's srudenrs had been consistently entering the central government or the political arena in general. Before l482(13th year of Seongjong's reign), when Kim was assigned to local regions and was away from the capital, already 18 of them passed the Mun-gwa examination. Seongjong was welcoming the advent of such literary figures, and with their skill and talent, they were assigned to important posts like those in the Hongmun- gwan office, or the post of Daegan or Official Historiographer, which usually required the highest level of morality and integrity from candidates. Even the ones who did not past the examination continued to acquaint themselves with others in Seonggyun-gwan, as we can see from the appeals submitted to the government by Lee Shim Weon & Nam Hyo On, or the Sohak community(Sohak-gyae), and the gathering of the 'Wise Seven at the Bamboo forest'. And when Kim returned to a central post, he was still highly respected by Seongjong, was ordered to supervise the revision of 『Dongguk Yeoji Seungram』, and his students were also establishing themselves as formidable presence inside the governrnent. In Seonggyun-gwan, junior figures like Lee Mok and Nam Gon were deepening their relationships with others, and Nam Hyo On & Kim Gwaeng Pil were emerging as distinctive figures. But in general, they were still not raising (officially) a voice as a unified force in political terms. Kim himself, even with all his fame, was also criticised for not being professional in managing certain official affairs, and many of his students were impeached for either official or private mistakes and misdeeds. Also his students, while still highly respecting Kim, from time to time confronted him for not being the ideal politician they imagined and hoped for. During the Muo-year massacre of scholars, some of them even publicly denied their relationship with Kim or other students. Therefore, we should be careful in defining the nature of Kim's group. Kim's group was not a political faction composed of figures from local regions who came to the capital to represent the inrerests of local Sajok figures. It was rather a group of literary figures who passed the national examination with their exceptional talents and therefore were continuing to grow in political terms with the support of King Seongjong, and other people who shared the values and teachings of Neo-Confudanism. All these people happened to have trained themselves under the guidance of Kim Jong Jik, who was displaying extraordinary scholarship and integrity and wished to train as many people as he couId. And in regional terms, Kim's group was formed upon the basis of the Seoul capital, which served as a stage for their political growth and creation of their own consensus.

      • 의대생의 스트레스에 영향을 미치는 요인 분석

        Background : Generally, it is well known that medical students have lots of stress because they have many classes of which the courses are much different from those of non-medical students. So, this study was designed for the purpose of evaluating the influencing factors on medical students' stress. Method : This study was carried out with the data of 69 students in premedical course and 66 students in medical course, who answered the survey by Kwandong Medical College, Department of Preventive Medicine and Public Health in June, 1999, The independent variables are the general characteristics, the characteristics related with study, the state of health, the activity for free time. The amount of stress is measured by Korean-translated BEPSI (Brief Encounter Psychosocial Instrument). Result : The multiple recession and logistic analysis show that the grade influence the significant difference of the stress level of the students in premedical course. The entrance age, average selfstudy time, hobby and religion influence the significant difference of the stress level of the students in medical course. The grade, avercage self-studying time and smoking habits influence the signifiant difference of the stress level of the medical students. Conclusion : This result will be used for the management of the stress level of the medical students' that influences their study.

        갑상선 수술의 해부학적 지표로서의 Zuckerkandl 결절의 중요성

        Purpose: Zuckerkandl's tubercle (ZT) of the thyroid gland is a well-documented anatomical structure. This study evaluated the anatomical relationship of the ZT in terms of the recurrent laryngeal nerve (RLN) and the superior parathyroid gland (SP). Methods: The study included 325 patients (ten patients with benign tumors and 315 patients with malignancies) who underwent thyroid surgery between February and June 2007. Tubercles were classified according to size: Grade 0 (unrecognizable), Grade I (≤ 5 mm), Grade II (6∼10 mm) and Grade III (>10 mm). The incidence and size of the ZT and its positional relationship to the RLN and SP were investigated during thyroid surgery. Results: ZTs were identified in most patients (right thyroid 89.3%, left thyroid 85.6%). The percentageof tubercles according to grade and location was as follows: Grade 0, right thyroid 10.7% and left thyroid 14.4%; Grade I, right thyroid 7.9% and left thyroid 11.1%; Grade II, right thyroid 43.5% and left thyroid 38.5%; Grade III, right thyroid 37.9% and left thyroid 35.9%. The most common RLN course was in a groove between the ZT and the main body of the thyroid. Most of the SPs are situated cranial to the ZTs and were located at the 1 or 2 o'clock position (96.1%) in the left thyroid and at the 10 or 11 o'clock position (95.2%) in the right thyroid. A greater distance between the ZT and the SP was seen with a decreasing size of the ZT. Conclusion: The ZT was identified during most thyroidectomies, and there was a constant relationship between the ZT and either the RLN or SP. Therefore, identification of the ZT and an understanding of the relationship between the ZT and either the RLN or SP are essential for the performance of safe thyroid surgery.       • 온도, 광, 이산화탄소에 따른 참외 광합성 변화

        본 연구는 참외의 생리활성 및 생산량 예측을 위한 환경 요인에 따른 참외의 광합성 동화량을 분석하고자 실시하였다. 측정 잎은 생장점으로부터 5마디 아래의 완전 전개되고 건전한 잎을 선정하여 측정하였으며 측정 환경은 온도 20°C에서 40°C까지 5°C 간격으로 5지점, 이산화탄소는 0 ~ 1,600ppm 범위에서 200ppm간격으로 9지점, 챔버 내 광량은 400, 1,200μmol·m<SUP>-2</SUP>·s<SUP>-1</SUP> 2지점을 측정하였다. 측정은 청명일 오전 10:00 ~ 12:00, 13:00~15:00시 사이에 실시하였으며 각 3회 반복한 측정값이 평균을 사용하였다. 광의 세기가 1,200μmol·m<SUP>-2</SUP>·s<SUP>-1</SUP> 일 때, 광합성동화량은 20°C에서 최젓값(21.37μmol·m<SUP>-2</SUP>·s<SUP>-1</SUP>), 35°C에서 최댓값(45.31μmol·m<SUP>-2</SUP>·s<SUP>-1</SUP>)을 보였다. 최대 광합성동화량은 20, 25, 30, 35°C 순으로 증가하다가 40°C에서 감소하였으며, 이 때 최댓값과 최솟값의 차이는 △23.93μmol·m<SUP>-2</SUP>·s<SUP>-1</SUP>였다. 이산화탄소 포화점은 20, 25°C에서는 900~1,000ppm사이, 30°C 이상 일 때는 700 ~ 800ppm사이로 나타났다. 이에 반하여 챔버 내부의 광이 400μmol·m-2·s-1 일 때는 40°C에서 최젓값(16.32μmol·m<SUP>-2</SUP>·s<SUP>-1</SUP>), 30°C에서 최댓값(21.49μmol·m<SUP>-2</SUP>·s<SUP>-1</SUP>)을 보였으며, 이 때 최댓값과 최솟값의 차이는 △5.16μmol·m<SUP>-2</SUP>·s<SUP>-1</SUP>였다. 최대 광합성동화량은 500 ~ 600ppm 사이로 관측되었으며, 이후 CO₂농도 증가에 따른 감소는 보이지 않았다. 참외는 30 ~ 35°C 사이에서 광합성 최댓값을 보이며, 광이 약하고 온도가 높을 경우 식물의 광합성 동화가 저해되나 충분한 광 조건이 형성되면 광합성이 증가하는 것을 확인하였다.

      • Immunocytochemical staining of p16<sup>ink4a</sup> protein as an adjunct test in equivocal liquid-based cytology

        <P>For cervical cancer screening, HPV-DNA test is expensive and is not easily available in all clinical situations. Thus, we investigated the role of p16<SUP>ink4a</SUP> immunostaining as another adjunct test to diagnose cervical neoplasia in equivocal liquid based cytology. Eighty-seven patients were randomly selected for this study (3 patients with normal, 84 patients with abnormal including 24 ASCUS, 30 LSIL, and 30 HSIL). We performed p16<SUP>ink4a</SUP> immunostaining on ThinPrep® slide and on each case from the corresponding cervical biopsy tissues. High-risk HPV-DNA testing was also performed on all the subjects. We found that the immunoreactivity of p16<SUP>ink4a</SUP> is strongly correlated with the grade of cytologic and histologic diagnoses as well as with Hybrid Capture 2. In comparing the p16<SUP>ink4a</SUP> immunostaining with the Hybrid Capture 2 for accuracy of the diagnosis of CIN II/III or a higher-grade disease in the case of ASCUS/LSIL on ThinPrep®, no significant differences were observed. Our data implies that p16<SUP>ink4a</SUP> immunocytochemical staining in liquid-based cytology specimens might be used as a good adjunct test to predict cervical histology in equivocal ThinPrep® tests.         Magnetic Pd–Fe3O4 Heterodimer Nanocrystals as Recoverable Catalysts for Ligand-Free Hiyama Cross-Coupling Reactions

        Ligand-free Hiyama cross-coupling reaction was achieved through the use of Pd–Fe3O4 heterodimeric nanocrystals (1 mol% in Pd) as recyclable catalysts. The nanocrystal catalysts exhibited good activities accommodating a variety of substrates including aryl bromides and iodides with substituents of varying electronic and steric properties. Furthermore, the nanocrystal catalyst could be conveniently recovered with the aid of an external magnet and recycled five times without the loss of catalytic activity to a considerable degree.

        Protective Effects of KH-204 in the Bladder of Androgen-Deprived Rats

        Purpose: We investigated the protective effects of the herbal formulation KH-204 in the bladder of androgen-deprived rats. Materials and Methods: Male rats aged eight weeks were randomly divided into four groups, containing eight rats each: sham operation only (normal control group), androgen-deprived only (androgen-deprived control group), and androgen-deprived followed by treatment with 200 mg/kg or 400 mg/kg of KH-204. After 0.5 mg/kg of leuprorelin was subcutaneously injected in the androgen-deprived groups, the oral administration of either distilled water in the two control groups or KH-204 in the treatment group was continued for four weeks. Serum testosterone levels, RhoGEF levels, nitric oxide (NO)-cyclic guanosine monophosphate (cGMP)-related parameters, oxidative stress, and histologic changes were evaluated after treatment. Results: Treatment with the herbal formulation KH-204 (1) increased serum testosterone levels; (2) restored the expression of RhoGEFs, endothelial NO synthase, and neuronal NO synthase; (3) increased the expression of superoxide dismutase; and (4) decreased bladder fibrosis. Conclusions: Our results suggest that the positive effects of KH-204 on the urinary bladder may be attributed to its antioxidant effects or to an elevation in NO-cGMP activity.

      • Antioxidant and Antifibrotic Effect of a Herbal Formulation <i>In Vitro</i> and in the Experimental Andropause via Nrf2/HO-1 Signaling Pathway

        <P>The Korean herbal formulation Ojayeonjonghwan is used for improving late-onset hypogonadism (LOH) symptoms such as erectile dysfunction (ED). A previous research suggested that a modified Ojayeonjonghwan (KH-204) could be used as an alternative to the treatment for ED. The pharmacological effects were examined in different conditions, including in vitro and in vivo. We measured the survival rate of TM3 Leydig cells under the oxidative stress condition. The s.c. injection of leuprorelin was used to induce androgen deprivation. We measured serum testosterone levels, oxidative stress, and apoptosis. The results of the treatment by KH-204 (1) preserved TM3 cells from oxidative stress by improving the expression of nuclear factor erythroid 2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1); (2) lowered the expression of transforming growth factor-beta (TGF-<I>β</I>) 1/SMAD; (3) increased the average of serum testosterone in androgen-deprived male rats; (4) kept the activation of spermatogenesis; (5) upgraded the contents of 8-hydroxy-20-deoxyguanosine (8-OHdG) and degraded the contents of superoxide dismutase (SOD); and (6) reduced apoptosis. We studied that KH-204 improved testicular dysfunction in LOH. It is likely, at least in part, to degrade oxidative stress through the Nrf2/HO-1 pathway. These findings may offer credible evidences for the use of new alternative therapies to treat LOH.</P>
