For the accelerating fermentation of high salt anchovy using halophilic proteolytic microorganisms, various halotolerant or halophilic protease-producing strains were isolated from 13 types of high salt fermented fish pastes. Among 13 halotolerant and 4 halophilic isolates which produced proteases on PS agar medium containing a different NaCl concentration from 0% to 20%, one halotolerant and protease-producing strain, CK-14 was identified as a member of Bacillus sp., and another halophilic and protease-producing strain , LF-7, a member of Vibrio sp. through fatty acid composition analysis. The NaCl range for growth and protease production of Bacillus sp. CK-14 and Vibrio sp. LF-7 in PS broth was extended from 0% to 12.5%, and from 5% to 20%, with an optimal concentration of 5% and 7.5%, respectively. The optimal temperature and pH for cell growth and protease production of Bacillus sp. CK-14 and Vibrio sp. LF-7 were 30℃ and 35℃, pH range of 8.0 to 8.5 and 6.5 to 9.5, respectively. The maximal protease activity of Bacillus sp. CK-14 was 3200 unit after 6% incubation, and that of Vibrio sp. LF-7 was 3980 unit after 120hr incubation under optimal culture conditions.
길항세균 Pseudomonas cepacia(P713 & P41)의 근권정착력 및 수박덩굴쪼김병 (Fusarium oxysporum f.sp.niveum)에 대한 생물적방제효과

In order to find out effect of the antagonistic bacterium for Fusarium wilt of watermelon, the two strains of Pseudomonas cepacia were tested. Inhibition zone formed by the antagonist was not only confirmed with a dual culture but also examined conidial germination and mycelial growth of the fungus by using culture filterate. Root colonization of the two antagonistic Pseudomonas cepacia (P7l3 & P4l) were successfully confirmed on the cucurbit rhizospheres through DLF(double layered filterpaper method) and Ahmad and Baker methods, and there was no difference of root colonization between the two isolates of the antagonist. With regard to competition between the antagonists and pathogen, the two bacteria were not challanged by the casual fungus. Furthermore, numerous antagonistic bactera were observed at root tip, terminal root and leteral root through scanning electron microscope. Biocontrol effect of 40% to 100% forFusarium wilt of watermelon were confirmed by percent of diseased plants under the pots and polyethylene film house experiments. : Pseudomonas cepacia, root colonization, Fusarium oxysporum f. sp. Recent experimental studies and clinical cases have suggested that grafting tissue from the adrenal medulla into the brain may alleviate the symptoms of Parkinson's disease. Autologous adrenal medullary grafts into the striatum was performed stereotactically in two patients with Parkinson's disease. Both patients were bedridden preoperatively, but 4 months after the surgery both became ambulatory and one was even capable of returning to his job. Even though the number of cases was small⑵ and the follow-up period was short, this study shows that adrenal medullary transplantation into the striatum may have some beneficial effects in patient with Parkinson's disease. Further experimental and clinical trials are necessary to see whether this procedure will be a recommendable surgical treatment for patients suffering from Parkinson's disease.
Pseudomonas sp.β-1,4-Glucosidase유전자의 발현

The Pseudomonas β-glucosidase gene was subcloned in a IPTG-induced E. coli expression vector PKK 223-3 containing the tac promoter. Recombinant plasmid inserted β-glucosidase gene in EcoRI site of pKK 223-3 was constructed and named pKKB 10. The E. coli JM109 carrying pKKB10 was grown under antibiotic selection to mid-log phase. Each culture was divided and IPTG was added to one portion. The concertration of inducer necessary for maxium induction of β-glucosidase activity was 6mM isopropyl-β-D-thiogalactoside(IPTG) and the addition of IPTG had no inhibitiory effect on cell growth. A 8-fold increase in the level of β-glucosidase activity expressed by pKKB10 was detected 5 hours after 6mM IPTG induction. The Pseudomonas β-glucosidase gene was also subcloned in a temperature-regulated E. coli expression vector PAS1 containing the leftward promoter pL of bacteriophage lambda. Recombinant plasmid inserted β-glucosidase gene in BamHI linker of PAS1 was constructed and named PASG20. The level of gene expression in E. coli N 4830-1 habouring PASG20 was controlled by thermal inactivation of the heat-sensitive lambda c1857 repressor. Under optimal condition, the recombinant β-glucosidase gene was expressed to a level of 6.2-fold.
단어출현빈도분석과 AHP를 통한 기술 연구 동향과 기대 효과 분석: 바이오 에너지 분리막을 중심으로

[연구목적] 본 연구는 과학 기술 연구 동향과 기술 도입 기대 효과를 함께 분석하는 방안을 바이오 에너지 분리막 분야에 적용하여 연구개발 전략 제시에 도움이 되는 연구 방안을 제시하고자 하였다. [연구방법] 본 연구는 분리막 기술 연구 방향과 기대효과를 분석하기 위해 각각에 대해 두 가지 연구방법을 이용하였다. 우선 텍스트 마이닝을 통해 단어출현 빈도분석으로 선행연구의 연구 방향을 확인하였고, AHP를 통해 기대효과를 알아보았다. [연구결과] 분석결과 분리막 기술의 연구 방향은 주로 분리막 관련 다양한 공정과 공정개선을 통한 성능 향상에 있는 것으로 확인되었다. 분리막 기술을 통한 기대 효과는 바이오 에너지 공정에서 발생하는 환경 문제 해결이 주된 것으로 나타났다. [연구의 시사점] 분리막 관련 연구는 분리막 성능에 중점을 두고 진행되고 있으며 분리막을 실제 공정에 도입하는 측에서 기대하는 부분은 분리막 도입으로 인한 효과와 분리성능 향상에 의한 효과에 중점을 두고 있음을 확인하였다. 이는 분리막 기술 연구가 실제 니즈를 잘 반영하여 이루어지고 있음을 보여준다. 이처럼 특정 기술의 연구 방향과 도입에 따른 기대효과를 함께 분석할 수 있으며, 이는 연구개발 전략 제시에 도움이 될 수 있음을 확인하였다. [Purpose] Purpose of this study is to propose a research methodology that is helpful in presenting the R&D strategy by applying analysis on research trends and expected effect of technology introduction together in the field of bioenergy membranes. [Methodology] For this purpose, we used two research methodologies. Word frequency analysis of text mining was used to analyze the research trends of previous studies, and the expected effect was investigated through AHP. [Findings] This study found that the research trends of membrane is mainly to improve performance through various processes and expected effect focus on environmental problem solving arising from bioenergy process. [Implications] It was confirmed that main research trends of membrane is performance of membrane and expected effect is performance improvement by introduction of membrane in bioenergy processes. This shows that research on membrane is well reflected in actual needs. This methodology can be helpful in presenting the consumer-oriented R&D strategy.
Epidermal Growth Factor가 人體纖維芽細胞 增殖에 미치는 影響

The effects of epidermal growth factor(EGF) on the cell proliferation were estimated in normal human fibroblast and established adenovirus type 12-simian murine sarcoma virus 40 hybrid virus-transformed human fibroblast. In the presence of culture medium containing 1.0ng per ㎖ epidermal growth factor had more effect on the cell proliferation of transformed cell lines than normal cell lines. The data suggest that epidermal growth factor stimulates cell proliferation in additive manner of virus-transformed human fibroblast.
