저온·알칼리성 Protease를 생산하는 Pseudomonas sp. RP-222의 분리 및 조효소의 특성

저온에서 높은 활성을 나타내는 알칼리성 protease를 생산하기 위하여 여러가지 시료로부터 집적배양에 의해 저온성 세균을 분리하였다. 분리된 세균은 저온·저알칼리성 Pseudomonas sp.인 것으로 판명되었으며, 효소생산을 위한 균생육의 최적 pH는 10.0, 온도 20℃에서 4일간 배양하였을 때였다. 이 효소활성의 최적 pH 및 온도는 각각 pH 10.5 및 40℃였으며, pH 및 열안정성은 각각 pH 7.0∼13.0, 온도 50℃ 이하의 범위에서 비교적 안정하였다. 또한 이 효소는 PMSF와 EDTA에 의해서 저해되므로 활성부위에 serine기와 금속이온이 관여하는 것으로 추정되며 계면활성제의 존재하에서도 안정하였다. 효소의 활성은 0.05% Na-α-olefin sulfonate 첨가에 의해 약간 증가되었다. In order to produce alkaline protease, psychrotrophic bacterium which have high enzyme activity at low temperature, was isolated by using enrichment culture from various samples and identified as genus alkalopsychrotropic Pseudomonas sp. RP-222. The optimal culture conditions for enzyme production were pH-10.0, temperature-20℃ and culture time-4 days. The optimum pH and temperature for the enzyme activity were pH 10.5 and 40℃, respectively and the enzyme was relatively stable at pH 7.0∼13.0 and below 50℃. The emzyme was inhibited by ethylenediaminetetraacetate and phenylmethylsulfonylfluoride, indicating that the enzyme was a serine metalloenzyme, but considerably stable in the presence of surface active agents. Activity of the enzyme was increased by the addition of 0.05% Na-α-olefin sulfonate.

목적: 본 연구는 개별 작업치료와 통합된 집단회상프로그램이 장기요양시설 노인의 심리적 건강과 인지능력에 미치는 효과를 알아보고자 하였다, 연구방법: 대상자는 장기시설 노인 중 18명을 각 9명씩 무작위로 실험군과 대조군에 배정하였다. 실험군은 8주간 주 1회 30분씩 개인 작업치료를 실시하고, 집단회상프로그램은 주 2회씩 총16회(30~40)를 실시하였다. 사전·사후검사는 심리적 건강상태(우울, 삶의 질)를 알아보기 위해 한국형 노인우울검사(KGDS)와 삶의 질 척도(SF-8)를 사용하였고, 인지기능은 한국판 간이정신상태 판별검사(MMSE-K)와 신경행동학적 인지상태검사(NCSE)의 기억 항목을 사용하였다. 분석방법은 실험군과 대조군 각각의 중재 전, 후의 차이를 알아보기 위해 월콕슨 부호순위검정을 사용하였고, 두 군 간의 중재 후 변화량의 차이를 알아보기 위해 Mann-Whitney 검정을 사용하였다, 연구결과: 중재 전, 후 비교에서 실험군은 심리적 건강상태인 우울과 삶의 질에서 통계적으로 유의한 차이를 보여 향상되었으나, 인지기능은 통계적으로 유의한 차이가 없었다. 대조군은 우울과 삶의 질, 인지기능에 통계학적 차이를 보이지 않았다. 두 군 간의 중재 전, 후 변화량의 차이비교에서는 한국 판간이 정신상태 판별검사(MMSE-K)변화량에서만 실험군이 대조군에 비해 변화량이 통계학적으로 유의한 차이를 보였다, 결론: 개별 작업치료와 통합된 집단회상 프로그램은 장기시설 노인들에게 심리적 안정과 인지기능 변화에 긍정적인 효과가 있었다. 따라서 장기요양시설에서 개별 작업치료와 통합된 집단회상프로그램을 작업치료의 효과적인 방법으로 제안하고자 한다. Purpose: This study aims to investigate the effect of individual occupational therapy and the integrated group reminiscence counseling on psychological health and cognitive function of the elderly at long-term care facilities, Methods :Among the elderly at long-term care facilities, each eight people were randomly selected as experimental and control groups, respectively. Individual occupational therapy and group remi?niscence counseling were given to experimental group for 30minutes once a week and 30-40 minutes twice a week (total 16 times) for 8 weeks, respectively. Pre-and post-tests were per?formed using KGDS and SF-8 for psychological health(depression and quality of life), MMSE-K and memory item of NCSE for cognitive function, respectively. Wilcoxon sign?ed-ranks tests was used to examine the difference before and after the intervention in two groups, respectively. Mann-Whitney test was used to examine the difference in changes be?tween two groups after the intervention, Results: Experimental group sh wed statistically significant difference in depression and quality of life which were psychological health after intervention, but control group showed no statistically significant difference. Changes of MMSE-K after the intervention between two groups showed statistically significant difference, Conclusion: Individual occupational therapy and group reminiscence counseling gave the positive impact on psychological health and changes in cognitive function of the elderly at care facilities. Therefore, I would like to propose the development of individual occupational therapy and in?tegrated group reminiscence counseling as an effective method of occupational therapy.

The electrochemical properties of the AB₂-type (Zr-Ti-V-Ni-Cr-Co-Mn) metal hydride electrodes prepared by ball milling with AB<sub>5</sub>-type {(LM)Ni<sub>3.6</sub>Al<sub>0.4</sub>Co<sub>0.7</sub>Mn<sub>0.3</sub>}(LM : Lanthanum-rich mischmetal) alloy powder as a surface activator were investigated. By ball milling with AB<sub>5</sub> type alloy powder, the activation of AB₂ type metal hydride electrode was accelerated resulting in an increase of discharge capacity from 35% to 85% of the maximum capacity at the first cycle. As the amount of surface activator increased the activation rate increased, whereas the discharge capacity increased with 10wt% and decreased with 20wt% addition of the surface activator. When the amount of the surface activator was kept constant as 10wt%, the discharge capacity and the activation rate increased with ball milling time up to 20 hours. However beyond 20 hours of ball milling time, they decreased drastically due to the nano-crystallization or amorphorzation of the alloy powder.
호알칼리성 Bacillus sp.F204와 Bacillus sp.K17의 원형질체 융합

To develop cellulase and xylanase-producing strain by protoplast fusion, alkalophilic Bacillus sp. F204 and K17 were treated with NTG(N-methyl-N'-nitro-N-nitrosoguanidine) and isolated antibiotics resistant strains of S20 (Km', Cm') and G70(Str'). The frequency of protoplast formation was about 95% when cells of mid-log phase were treated with 200㎍/㎖ lysozyme at 37℃ for 30-45 minutes. Under addition of 0.4-0.5M sodium succinate, 0.5% casamino acid, 1.5% polyvinylpyrrolidone, 25mM MgCl_2 and 50mM CaCl_2 to the regeneration medium, the regeneration frequency of Bacillus sp. F204 and K17 was 24.9% and 26.2%, respectively. the fusion frequency was 6.6×10^-6 in the presence of 30% polyethylene glycol 6000 containing 50mM Ca^++ at 45℃ for 5 minutes. Cellulase complex and xylanase activities of fusant were compared with parental strains.

Dong Cheon is one of the severely polluted streams by domestic sewage and industrial wastewater. In order to evaluate the effect of domestic sewage and sediment characteristics on water quality, a study was carried out at downstream of Dong Cheon in Busan. The range and mean values of water quality in Dong Cheon were as follows: DO 0.8-11.0mg/l: SS 3-113mg/1: COD 12.4-40.8mg/1: BOD 12.6-48.1mg/1: NH4-N 0-l.10mg/l. Bottom sediment of Dong cheon mainly consisted of fine sand and silt, and contents of organic meter were very high considering the results of sediment BOD. According to the results of this study, stagnation of high pollutant load of water and rapid oxygen uptake of sediment lead the stream water to anoxic condition and it caused offensive oder in the downstream of Done Cheon during summer.

Sludge retention tests were conducted to crossflow filter media in an-hybrid reactors. Sections of this crossflow filter media were cut to fit a laboratory scale reactor. Five an-hybrid reactors with media volume to reactor volume of 0, 20, 40. 60 and 80% were run under various air flow and hydraulic retention time to estimate sludge retention. The result shows that the concentration of microorganism in effluent was generally higher in the reactor having lower media volume. And 40% media volume of total reactor volume in an-hybrid reactor was evaluated to be sufficient to prevent the wash-out of biological solids and thus to reduce start-up period of the an-hybrid reactor.
