        Dynamic Characteristics of Intersection Structure Consisting of Twin Shield Tunnels and a Transverse Passage

        The intersection structure, composed of twin shield tunnels and a transverse passage, is ubiquitous in the metros around the world. However, the dynamic characteristics of this kind of intersection structure have not been studied a lot yet. By carrying out the corresponding model tests and numerical simulations, the dynamic characteristics of this kind of intersection structure are studied in this paper, which is very helpful for the determination of the ground vibration from this kind of underground structure. The results show that the vibration of the transverse passage is more severe than the main tunnel, manifested that the frequency response function (FRF) of the transverse passage has greater magnitudes than that of the main tunnel at the locations with the same distance from the vibration source. The peak frequencies of the acceleration responses at different locations of the intersection structure under the moving train load are all located in the frequency band 44.9 − 56.1 Hz where the corresponding 1/3 octave band spectra of FRF have the largest values in the considered frequency band 31.5 − 250 Hz, indicating that the natural frequencies of the intersection structure in the frequency band 44.9~56.1 Hz have significant influences on its acceleration responses below 250 Hz induced by the moving train.

        Analysis on the Dynamic Responses of an Overlapped Circular Shield Tunnel under the Different Vibration Loads

        At present, there are many studies on the dynamic response of overlapped tunnels, but most of them are through numerical simulation. Very few has been conducted by the experimental model research and frequency domain analysis. Combined with the model test and the numerical simulation, the dynamic response of the tunnel lining structure under the action of different train speeds and different tunnel clear distances are investigated, which can remedy the deficiencies in the relevant fields and verify the accuracy of numerical simulation results. The results show that as the train speed increases, the amplitude of the energy spectra of the vibration loads decrease significantly. The tunnel response at the sidewalls is smaller than that at the tunnel crown and at the invert in the lower tunnel. As the net distance increases, the amplitudes of the acceleration frequency spectrum and the energy spectra of the lower tunnel decrease, but with a diminishing rate. The dynamic coefficients of circumferential of the upper tunnel under a train load are larger than those in the longitudinal direction in the invert.

        Investigation of the Probability of a Safe Evacuation to Succeed in Subway Fire Emergencies Based on Bayesian Theory

        Emergency evacuation in subway fires is one of the hot issues in public safety. Based on Bayesian theory, a dynamic evacuation risk analysis model considering both psychological and behavioral responses of evacuees was proposed in this contribution. A subway fire scene was modeled using the Event Tree Analysis (ETA) according to the questionnaire survey at Nanjing Xinjiekou station. In this fire scenario, three control events (the fire alarm, the exhaust system and the evacuation route) were related to fire evacuation control and two control events (active escape and passive escape), which were specifically defined, were related to personnel characteristics. Subsequently, the probability of the final state of each scene was calculated by Monte Carlo simulation. Combined with the statistics of accidents and safe evacuation of subway fire cases from 1976 to 2013, the posterior probability distribution of safety evacuation was obtained after real-time dynamic updating of its precursor probability through Bayesian theory. Results show that the posterior probability of safety evacuation to succeed is around 0.8. This indicates that there is still a probability of 0.2 resulting in injury or fatalities in subway accidents even though most passengers are safely evacuated. It also suggests that utilization of 0.2 as the safety threshold would be an appropriate choice.

        A Data-driven Approach to Estimate the Probability of Pedestrian Flow Congestion at Transportation Bottlenecks

        In public gathering places, pedestrian flow congestion may happen in transportation bottlenecks. Limitations exist in the conventional judgment of congestion by considering the crowd density or the walking speed merely. In this paper, a data-driven mathematical approach based on Kernel Density Estimation (KDE) to analyze the probability of pedestrian flow congestion is proposed, which comprehensively considers the walking speed, the crowd density and the flow rate when pedestrians walk towards possible bottlenecks. With the case study of Nanjing metro station during rush hours, the advantages of non-parametric KDE compared with traditional parametric normal distribution estimation are analyzed and the optimal bandwidth for KDE is also discussed. The case study shows that the proposed method can obtain a more reliable quantitative assessment of congestion risk, as it overcomes the limitation of parametric estimation that relies on experience, and also avoids biased assessment of congestion that merely concerns single parameter of pedestrian flow. Finally, an assessment framework for dynamic congestion risk at bottlenecks is suggested. With this framework, the change of congestion situations of a monitored region can be mastered through data-driven approach, and thus the transformation of normal flowing to congestion of the crowd can be quantified through probabilistic analysis.

        Mediation analysis of leisure activities on the association between cognitive function and mortality: longitudinal study of 42,942 Chinese adults 65 years and older

        OBJECTIVES: Previous studies have established associations of cognitive function and leisure activities with mortality. This study aimed to evaluate whether leisure activities causally mediate these associations. METHODS: This longitudinal study included 42,246 participants aged over 65 years from the Chinese Longitudinal Healthy Longevity Survey. The Mini-Mental State Examination and a self-reported scale were used to measure cognitive status and leisure activities, respectively. We examined the associations of cognitive function and leisure activities with mortality using Cox proportional hazards models. Causal mediation analysis was used to assess whether leisure activities mediated the association between cognitive function and mortality. RESULTS: Cognitive function and leisure activities were inversely associated with mortality. Leisure activities accounted for 28.3% (95% confidence interval [CI], 25.6 to 31.1) of the total effect (TE) of cognitive function and mortality. A higher mediated proportion (PM) was observed for physical leisure activities (PM, 20.1%; 95% CI, 18.0 to 22.3) than for social leisure activities (PM, 17.7%; 95% CI, 15.7 to 19.7). The mediating effect was higher among participants at younger ages (PM, 41.5%; 95% CI, 21.3 to 65.4), those with higher education levels (PM, 30.5%; 95% CI, 25.3 to 36.2), and residents of rural China (PM, 42.5%; 95% CI, 25.4 to 62.5). CONCLUSIONS: Cognitive function was associated with inverse mortality. Leisure activities significantly mediated this association. Participation in leisure activities at the early stages of mild cognitive impairment could reduce the risk of mortality, which has a major impact on interventional strategies for healthy aging.

        The antibacterial activity of E. coli bacteriophage lysin lysep3 is enhanced by fusing the Bacillus amyloliquefaciens bacteriophage endolysin binding domain D8 to the C-terminal region

        Bacteriophage endolysin is one of the most promising antibioticsubstitutes, but in Gram-negative bacteria, the outermembrane prevents the lysin from hydrolyzing peptidoglycansand blocks the development of lysin applications. Theprime strategy for new antibiotic substitutes is allowing lysinto access the peptidoglycan from outside of the bacteria byreformation of the lysin. In this study, the novel Escherichiacoli (E. coli) phage lyase lysep3, which lacks outside-in catalyticability, was fused with the N-terminal region of theBacillus amyloliquefaciens lysin including its cell wall bindingdomain D8 through the best manner of protein fusionbased on the predicted tertiary structure of lysep3-D8 to obtainan engineered lysin that can lyse bacteria from the outside. Our results showed that lysep3-D8 could lyse both Gramnegativeand Gram-positive bacteria, whereas lysep3 and D8have no impact on bacterial growth. The MIC of lysep3-D8on E. coli CVCC1418 is 60 μg/ml; lysep3-D8 can inhibit thegrowth of bacteria up to 12 h at this concentration. The bactericidalspectrum of lysep3-D8 is broad, as it can lyse of allof 14 E. coli strains, 3 P. aeruginosa strains, 1 Acinetobacterbaumannii strain, and 1 Streptococcus strain. Lysep3-D8 hassufficient bactericidal effects on the 14 E. coli strains testedat the concentration of 100 μg/ml. The cell wall binding domainof the engineered lysin can destroy the integrity of theouter membrane of bacteria, thus allowing the catalytic domainto reach its target, peptidoglycan, to lyse the bacteria. Lysep3-D8 can be used as a preservative in fodder to benefitthe health of animals. The method we used here proved to bea successful exploration of the reformation of phage lysin.
