        Screening nephroprotective compounds from cortex Moutan by mesangial cell extraction and UPLC

        A method for screening nephroprotective compoundsin cortex Moutan, a common traditional Chinesemedicine (TCM) in treating diabetic nephropathy with renalmesangial cell extraction and ultra performance liquidchromatography technique was described in this paper. Wehypothesize that the compounds which bind to cell membranesunder pathological conditions may be the bioactivecompounds in TCMs. Mesangial cells were cultured inmedium containing 5 mM (physiological, NG) or 30 mM(pathological, HG) glucose for 48 h and then incubated withcortex Moutan extract. After the unbound substances werewashed off, the cell membrane-bound compounds weredissociated and concentrated by an SPE column. By comparingthe chromatograms of NG and HG cultured-cellextractions and cortex Moutan extract, three compoundsbound to both NG and HG-cultured mesangial cells wereidentified as paeoniflorin, pentagalloylglucose (PGG) andpaeonol. In vitro studies showed that paeoniflorin, PGG andpaeonol reduced the activity of nicotinamide-adenine dinucleotidephosphate oxidase (NADPH) activity, anddecreased the level of reactive oxygen species (ROS),transforming growth factor-b1 (TGF-b1) and fibronectin inhigh glucose cultured mesangial cells. The results indicatethat paeonol, paeoniflorin and PGG may be the nephroprotectivecompounds from cortex Moutan. This study isexpected to provide a more reliable and effective method forscreening bioactive compounds from the complex TCM systems.

        Fault analysis of CNC equipment based on DEMATEL/ISM/ANP

        To make a reasonable reliability improvement strategy for CNC equipment, it is necessary to clearly understand the fault relation characteristics and the weak links of reliability. While the fault complexity of CNC equipment makes the fault relation characteristics unclear, it is very difficult to determine the weak link of reliability. Therefore, a fault analysis method for CNC equipment based on decision-making and trial evaluation laboratory (DEMATEL)/ interpretive structure modeling (ISM)/analytic network process (ANP) is proposed in this study to describe the relationship between CNC equipment faults accurately and identify reliability weaknesses. First, the fault mechanism was analyzed by the fault phenomenon, and the fault factors of the CNC equipment were identified in view of the system. Second, DEMATEL/ISM/ ANP method was adopted to integrate the interaction direction and intensity between the failure factors, hierarchical structure of the factors, and their relative importance, and a hierarchical network structure model for CNC equipment fault factors was built. Finally, according to the analysis results of the aforementioned model, the fault factors of the CNC equipment were divided into drive, link, dependence, and independent fault factors, and the identification of the weak link of reliability was completed. Results show that the proposed method can quantify the fault factors of the CNC equipment and clearly identify the main direction of its reliability improvement.

        Chondrogenic Differentiation and Three Dimensional Chondrogenesis of Human Adipose-Derived Stem Cells Induced by Engineered Cartilage-Derived Conditional Media

        Due to lack of optimal inductive protocols, how to effectively improve chondrogenesis of adipose-derived stem cells (ASCs) is still a great challenge. Our previous studies demonstrated that the culture media derived from chondrocyte-scaffold constructs (conditional media) contained various soluble chondrogenic factors and were effective for directing chondrogenic differentiation of bone marrow stem cells. Nevertheless, it remains unclear whether the conditional media can induce ASCs towards chondrogenic differentiation, especially for three-dimensional (3D) cartilage formation in a preshaped scaffold. In this study, it demonstrated that the conditional media derived from chondrocyte-scaffold constructs could promote ASCs to differentiate into chondrocyte-like cells, with similar expression of type II collagen to those induced by chondrogenic growth factors. Moreover, the expression level of chondrocyte-specific genes, such as SOX9, type II collagen, and COMP, was even higher in conditional medium group (CM) than that in optimized chondrogenic growth factor group (GF), indicating that the conditional media can serve as an effective inducer for chondrogenic differentiation of ASCs. Most importantly, the conditional media could also induce ASC-scaffold constructs to form 3D cartilage-like tissue with typical lacunae structures and positive expression of cartilage specific matrices, even higher contents of GAG and type II collagen were achieved in CM group compared to GF group. The current study establishes a simple, but stable, efficient, and economical method for directing 3D cartilage formation of ASCs, a strategy that may be more closely applicable for repairing cartilage defects.

        According to increasing sharply use of smartphone in the recent year, domestic banks are showing a lot of attention to service quality and customer satisfaction of smartphone banking for the success of the business. This paper is to find out and analyze the service quality factors of customer satisfaction as the critical success factors of smartphone banking. The result of this research shows that efficiency, fulfillment, and privacy have a significant positive influence on customer satisfaction. On the other hand system availability and empathy don’t have a significant positive influence on customer satisfaction. Customer satisfaction has a significant positive influence on loyalty. These research findings suggest the clue for making smartphone banking successful.

        Antimicrobial and Cytotoxic Activity of Endophytic Fungi from Lagopsis supina

        In this study, five endophytic fungi belonging to the Aspergillus and Alternaria genera were isolated from Lagopsis supina. The antimicrobial activity of all fungal fermented extracts against Staphylococcus and Fusarium graminearum was tested using the cup-plate method. Among them, Aspergillus ochraceus XZC-1 showed the best activity and was subsequently selected for large-scale fermentation and bioactivity-directed separation of the secondary metabolites. Four compounds, including 2- methoxy-6-methyl-1,4-benzoquinone (1), 3,5-dihydroxytoluene (2), oleic acid (3), and penicillic acid (4) were discovered. Here, compounds 1 and 4 displayed anti-fungal activity against F. graminearum, F. oxysporum, F. moniliforme, F. stratum, Botrytis cinerea, Magnaporthe oryzae, and Verticillium dahliae with diverse MIC values (128–512 μg/ml), which were close to that of the positive control antifungal, actidione (64–128 μg/ml). Additionally, compounds 1 and 4 also exhibited moderate antibacterial activity against S. aureus, Listeria monocytogenes, Escherichia coli, and Salmonella enterica, with low MIC values (8–64 μg/ml). Moreover, compounds 1 and 4 displayed selective cytotoxicity against cancer cell lines as compared with the normal fibroblast cells. Therefore, this study proposes that the endophytic fungi from L. supina can potentially produce bioactive molecules to be used as lead compounds in drugs or agricultural antibiotics.

        Investigation of the dynamic bending moment transmitted to the forging manipulator due to press motion

        The bending moment transmitted to the forging manipulator due to press motion during metal forming process is investigated. The dynamic model of the forging manipulator system is established, including the manipulator and the workpiece by using Lagrange equation. The system is modeled as an Euler-Bernoulli beam with spring-mass at the sliding end, which experiences a transient vibration due to displacement excitation. The Winkler foundation model is used to simulate the rotational constraint of the forging dies on the workpiece because of the surface contact. The numerical results are compared with the LS-DYNA simulations, and a good prediction on the bending moment could be obtained with the comparatively simple proposed model. The results indicate that the press position where the forging dies work along the workpiece plays a crucial role for the dynamic bending moment. To further investigate the dynamic effects due to the press motion, the influence of clamp mass of the manipulator is also examined.

        Wind Power Interval Prediction Based on Improved PSO and BP Neural Network

        As is known to all that the output of wind power generation has a character of randomness and volatility because of the influence of natural environment conditions. At present, the research of wind power prediction mainly focuses on point forecasting, which can hardly describe its uncertainty, leading to the fact that its application in practice is low. In this paper, a wind power range prediction model based on the multiple output property of BP neural network is built, and the optimization criterion considering the information of predicted intervals is proposed. Then, improved Particle Swarm Optimization (PSO) algorithm is used to optimize the model. The simulation results of a practical example show that the proposed wind power range prediction model can effectively forecast the output power interval, and provide power grid dispatcher with decision.

        Energy Landscape Paving with Local Search for Global Optimization of the BLN Off-lattice Model

        The optimization problem for finding the global minimum energy structure is one of the mainproblems of protein structure prediction and is known to be an NP-hard problem in computationalmolecular biology. The low-energy conformational search problem in the hydrophobic-hydrophilicneutral(BLN) off-lattice model is studied. We convert the problem into an unconstrained optimizationproblem by introducing the penalty function. By putting forward a new updating mechanismof the histogram function in the energy landscape paving (ELP) method and incorporating heuristicconformation update strategies into the ELP method, we obtain an improved ELP (IELP) method. Subsequently, by combining the IELP method with the local search (LS) based on the gradient descentmethod, we propose a hybrid algorithm, denoted by IELP-LS, for the conformational searchof the off-lattice BLN model. Simulation results indicate that IELP-LS can find lower-energy statesthan other methods in the literature, showing that the proposed method is an effective tool forglobal optimization in the BLN off-lattice protein model.

        Prediabetes Progression and Regression on Objectively- Measured Physical Function: A Prospective Cohort Study

        Background: Prediabetes leads to declines in physical function in older adults, but the impact of prediabetes progression or regression on physical function is unknown. This study assessed this longitudinal association, with physical function objectivelymeasured by grip strength, walking speed, and standing balance, based on the Health and Retirement Study enrolling United States adults aged >50 years.Methods: Participants with prediabetes were followed-up for 4-year to ascertain prediabetes status alteration (maintained, regressed, or progressed), and another 4-year to assess their impacts on physical function. Weak grip strength was defined as <26 kg for men and <16 kg for women, slow walking speed was as <0.8 m/sec, and poor standing balance was as an uncompleted fulltandem standing testing. Logistic and linear regression analyses were performed.Results: Of the included 1,511 participants with prediabetes, 700 maintained as prediabetes, 306 progressed to diabetes, and 505 regressed to normoglycemia over 4 years. Grip strength and walking speed were declined from baseline during the 4-year followup, regardless of prediabetes status alteration. Compared with prediabetes maintenance, prediabetes progression increased the odds of developing weak grip strength by 89% (95% confidence interval [CI], 0.04 to 2.44) and exhibited larger declines in grip strength by 0.85 kg (95% CI, –1.65 to –0.04). However, prediabetes progression was not related to impairments in walking speed or standing balance. Prediabetes regression also did not affect any measures of physical function.Conclusion: Prediabetes progression accelerates grip strength decline in aging population, while prediabetes regression may not prevent physical function decline due to aging.

        Bending static and stress investigation of a microplate of piezoelectric/piezomagnetic material subjected to combined multifield loading. Shear deformable as well as thickness stretched model is used for derivation of the kinematic relations. Multi field governing equations are derived analytically through principle of virtual work. the results are analytically obtained analytically including magnetic/electric potentials, displacement and stress components with variation in multifield loading parameters.
