        Effects of Microbiota on the Treatment of Obesity with the Natural Product Celastrol in Rats

        Background: Obesity has become one of the most serious issues threatening the health of humankind, and we conducted this study to examine whether and how celastrol protects against obesity. Methods: We fed male Sprague-Dawley rats a high-fat diet and administered celastrol to obese rats for 3 weeks. By recording body weight (BW) and other measures, we identified the effective dose of celastrol for obesity treatment. Feces were collected to perform 16S rRNA sequencing, and hypothalami were extracted for transcriptome sequencing. We then treated leptin knockout rats with celastrol and explored the changes in energy metabolism. Male Institute of Cancer Research (ICR) mice were used to test the acute toxicity of celastrol. Results: We observed that celastrol reduced BW and promoted energy expenditure at a dose of 500 μg/kg BW but that food intake was not changed after administration. The diversity of the gut microbiota was improved, with an increased ratio of Bacteroidetes to Firmicutes, and the gut microbiota played an important role in the anti-obesity effects of celastrol. Hypothalamic transcriptome analysis showed a significant enrichment of the leptin signaling pathway, and we found that celastrol significantly enhanced energy expenditure, which was mediated by the leptin signaling pathway. Acute lethal toxicity of celastrol was not observed at doses ranging from 0 to 62.5 mg/kg BW. Conclusion: Our study revealed that celastrol decreased the BW of obese rats by enhancing energy expenditure but not by suppressing food intake and that this effect was mediated by the improvement of the gut microbiota and the activation of the hypothalamic leptin signaling pathway.

        체육계열 대학생의 전공 선택 동기가 전공 만족도 및 취업준비행동에 미치는 영향

        목적 본 연구는 체육계열 대학생을 대상으로 전공선택동기가 전공만족도 및 취업준비행동에 미치는 영향을 파악하여 취업준비행동 향상을 위한 실증적 기초자료를 제공하기 위해 시행되었다. 방법 이를 위하여 경기도 용인 소재의 Y대학에 재학 중인 체육계열 대학생에게 설문지에 자기평가기입법으로 응답하여 최종 133부의 설문지를 분석처리 하였다. 자료처리는 Windows Program SPSS Statistics Version 22.0으로 p<.05로 설정하여 빈도 분석, t-test, 상관관계분석과 다중회귀분석을 실시하였다. 결과 체대생의 성별에 따른 전공선택동기의 차이 검증 결과, 외재적 동기(p=.016)에서 유의한 차이가 나타났으며, 전공만족도의 차이 검증 결과, 유의한 차이가 나타나지 않았으며, 취업준비행동의 차이 검증 결과, 비공식적 준비(p=.037)에서 유의한 차이가 나타났다. 전공선택동기가 전공만족도에 미치는 영향 분석 결과, 내재적 동기는 수업만족과 사회인식만족에 유의한 영향을 미치며, 외재적 동기는 수업만족, 관계만족, 사회인식만족에 유의한 영항을 미치는 것으로 나타났다. 전공선택동기가 취업준비행동에 미치는 영향을 검증한 결과, 외재적 동기요인이 비공식 준비행동과 적극적 준비행동에 유의한 영향을 미치며, 내재적 동기요인이 공식적 준비행동에 유의한 영향을 미치는 것으로 나타났다. 전공선택동기의 내재적 동기와 외재적 동기는 전공만족도와 취업준비행동에 정(+)적 영향을 미치고 있음을 검증하였다. 결론 전공만족도 향상을 통해 취업준비행동을 향상시키는 것은 만족스러운 대학생활 및 취업에 있어 매우 중요하다고 할 수 있다. Objectives This study was conducted to provide empirical basic data for improving the employment preparation behavior of physical education undergraduate students. Methods The subject of this study is a physical education undergraduate student enrolled in Y University. For the period from November 15 to December 20, 2019, the final 133 questionnaires that were answered by the self-evaluation method were analyzed with Windows Program SPSS Statistics Version 22.0. Results Differences in major selection motives according to gender were found in extrinsic motivation (p=.016), and differences in job preparation behavior were found in informal preparation (p=.037). Intrinsic motivation has a significant effect on class satisfaction and social cognition satisfaction, and extrinsic motivation has a significant effect on class satisfaction, relationship satisfaction, and social cognition satisfaction. Extrinsic motivational factors have a significant effect on informal preparation behaviors and active preparation behaviors, and intrinsic motivational factors have a significant effect on formal preparation behaviors. Conclusions Improving employment preparation behavior through improvement of major satisfaction is very important and can be done for satisfactory university life and employment.

      • Construction Design and Theoretical Research on Cloud-based Intelligent Platform for Physical Fitness Test and Health Examination Data of College Students

        PURPOSE : To build a cloud intelligent analysis platform for physical measurement data and physical examination data for college students, so as to enhance the physical health of college students and realize the deep integration of sports, education and medical treatment. METHODS : On this basis, a cloud-based intelligent early warning and screening analysis module for the physical fitness test level and the prevalence rate in college students can be established. Then, a cloud-based intelligent construction module for non-medical health intervention means and scientific physical education (PE) curriculum should be built, aiming to strengthen the corresponding physical fitness test indexes and improve weak health examination indexes, and further mitigate and prevent relevant diseases. RESULTS : Through the Internet of things technology, we can realize the co-platform link of physical examination instruments and physical examination instruments, process and analyze data in the same system, and realize the source combination of sports and medical treatment. CONCLUSIONS : The establishment of cloud intelligent college student physical examination and physical examination platform is the need of The Times, and is also one of the effective measures to realize Outline of the Healthy China 2030 Plan (the Outline).

        What are the Key Location Factors for McDonald's, KFC and Starbucks stores in China?

        Since Chinese retail industry foreign investment mainly distributed in urban areas, there were significant differences in number of shops in different cities. Previous researches on FDI mainly focused on national and regional level, which could not explain this phenomenon. So, it is imperative to conduct researches to determine which factors attracted more foreign investment in retail industry. Based on distributed data of Starbucks, KFC and McDonald shops in 296 cities in China mainland, this paper explores their location determining factors. Apart from traditional market size and economic agglomeration effect, the number of university students in each city is also a key driving factor. Although the number of university students is a negative variable in distribution of Starbucks shops, it is a positive variable in distribution of KFC shops. In addition, results of this research show that location of Starbucks and KFC are not affected by the open policy system, which is different from location of McDonald's. The implication for this paper is explained from two perspectives. Firstly, theoretically, this paper promotes the discussion on FDI location determinants from city level, and provides significant reference for establishing location model of foreign investment in China on macro-level. Secondly, from practical point of view, this paper offers some suggestions on attracting foreign investment for city managers, and provides a few advices on location for other retail brands and potential investors on the three brands.

        Microphysical Structures of an Extreme Rainfall Event Over the Coastal Metropolitan City of Guangzhou, China: Observation Analysis with Polarimetric Radar

        A record-breaking nocturnal rainfall event (543 mm in 16-h) under weak synoptic forcing occurred in the metropolitan city of Guangzhou, China, during 6–7 May 2017. The evolution and microphysical structures of this torrential rainfall event are investigated using S-band polarimetric radar datasets. The torrential rainfall concentrated in two cores: one over Huadu District (HD) in which the storms were initiated between urban areas and mountains at mid-night, and the other over Huangpu and Zengcheng District (ZC) which was characterized by locally triggered storms merging with the storms from HD. The two heavy precipitation regions show some similarities, including strong reflectivity factor for horizontal polarizations (ZH) magnitude, low centroid cumulonimbus structures, and column shape of differential reflectivity (ZDR). But obvious differences can also be viewed between them. Compared to HD, ZC has higher precipitation intensity, longer precipitation duration, and larger accumulated rainfall. Besides, ZC also has a relatively lower ZDR value of ~ 0.2 dB and a higher specific differential phase (KDP) of approximately ~ 0.35° km− 1, which indicates the larger population of medium-sized rain droplet and higher water content in ZC. The radar-retrieved drop size distributions (DSDs) (i.e., mass-weighted diameter, logarithmic normalized intercept, and liquid water content) show that small size particles and high particle number concentration are more obvious in the storm over ZC. Combined with the retrieved DSDs, the merger process brings more medium-sized raindrops to ZC, and increases the possibility of raindrop growth via the accretion of cloud water by rain, which leads to enhancement of precipitation. In addition, strong KDP may be a good indicator of intensity for extreme precipitation.

        The Combined Tensile and Torsional Behavior of Irregular Fibers

        Most fibers are irregular, and they are often subjected to combined loading conditions during processing and enduse. In this paper polyester and wool fibers under the combined tensile and torsional loads have been studied for the first time using the finite element method (FEM). The dimensional irregularities of these fibers are simulated with sine waves of different magnitude and frequency. The breaking load and breaking extension of the fibers at different twist or torsion levels are then calculated from the finite element model. The results indicate that twist and level of fiber irregularity have a major impact on the mechanical properties of the fiber and the effect of the frequency of irregularity is relatively small.

        MRU-Net: A remote sensing image segmentation network for enhanced edge contour Detection

        Remote sensing image segmentation plays an important role in realizing intelligent city construction. The current mainstream segmentation networks effectively improve the segmentation effect of remote sensing images by deeply mining the rich texture and semantic features of images. But there are still some problems such as rough results of small target region segmentation and poor edge contour segmentation. To overcome these three challenges, we propose an improved semantic segmentation model, referred to as MRU-Net, which adopts the U-Net architecture as its backbone. Firstly, the convolutional layer is replaced by BasicBlock structure in U-Net network to extract features, then the activation function is replaced to reduce the computational load of model in the network. Secondly, a hybrid multi-scale recognition module is added in the encoder to improve the accuracy of image segmentation of small targets and edge parts. Finally, test on Massachusetts Buildings Dataset and WHU Dataset the experimental results show that compared with the original network the ACC、mIoU and F1 value are improved, and the imposed network shows good robustness and portability in different datasets.

        Self-Tuning Position Control for the Linear Long-Stroke, Compound Switched Reluctance Conveyance Machine

        This paper proposes a long-stroke linear switched reluctance machine (LSRM) with a primary and a secondary translator for industrial conveyance applications. The secondary one can translate according to the primary one so that linear compound motions can be achieved. Considering the fact that either one translator imposes a time-variant, nonlinear disturbance onto the other, the selftuning position controllers are implemented for the proposed linear compound machine and experimental results demonstrate that the absolute steady-state error values can fall into 0.03 mm and 0.05 mm for the secondary and primary translator, respectively. A composite absolute precision of less than 0.6 mm can be achieved under the proposed control strategy.

        Understanding soybean (Glycine max) domestication and improvement at a genetic level is important to inform future efforts to further improve a crop that provides the world's main source of oilseed. We detect 230 selective sweeps and 162 selected copy number variants by analysis of 302 resequenced wild, landrace and improved soybean accessions at >11× depth. A genome-wide association study using these new sequences reveals associations between 10 selected regions and 9 domestication or improvement traits, and identifies 13 previously uncharacterized loci for agronomic traits including oil content, plant height and pubescence form. Combined with previous quantitative trait loci (QTL) information, we find that, of the 230 selected regions, 96 correlate with reported oil QTLs and 21 contain fatty acid biosynthesis genes. Moreover, we observe that some traits and loci are associated with geographical regions, which shows that soybean populations are structured geographically. This study provides resources for genomics-enabled improvements in soybean breeding.

        Intracellular Localization and Sustained Prodrug Cell Killing Activity of TAT-HSVTK Fusion Protein in Hepatocelullar Carcinoma Cells

        Gene therapy with nonviral vectors using the suicide gene/prodrug activating system of herpes simplex virus type-1 thymidine kinase (HSV1-TK)/ganciclovir (GCV) is inefficient in killing malignant tumor cells due to two major factors: (a) an unsatisfactory bystander effect; (b) short-lived expression of the protein. To study the capacity of the protein transduction domain (PTD) of HIV-1 TAT protein to enhance HSV1-TK/GCV cancer gene therapy, we constructed three fusion proteins TAT-TK, TK-TAT and TK. TATTK retained as much enzyme activity as TK, whereas that of TK-TAT was much lower. TAT-TK can enter HepG2 cells and much of it is translocated to the nucleus. The transduced HepG2 cells are killed by exogenously added GCV and have bystander effects on untransduced HepG2 cells. Most importantly, the introduced recombinant protein is stable and remains functional for several days at least, probably because nuclear localization protects it from the cytoplasmic degradation machinery and provides access to the nuclear transcription machinery. Our results indicate that TAT fusion proteins traffic intercellularly and have enhanced stability and prodrug cell killing activity. We conclude that TAT has potential for enhancing enzyme prodrug treatment of liver cancers.
