        Unsteady aerodynamic forces on a vibrating long-span curved roof

        The present paper discusses the characteristics of unsteady aerodynamic forces on long-span curved roofs. A forced vibration test is carried out in a wind tunnel to investigate the effects of wind speed, vibration amplitude, reduced frequency of vibration and rise/span ratio of the roof on the unsteady aerodynamic forces. Because the range of parameters tested in the wind tunnel experiment is limited, a CFD simulation is also made for evaluating the characteristics of unsteady aerodynamic forces on the vibrating roof over a wider range of parameters. Special attention is paid to the effect of reduced frequency of vibration. Based on the results of the wind tunnel experiment and CFD simulation, the influence of the unsteady aerodynamic forces on the dynamic response of a full-scale long-span curved roof is investigated on the basis of the spectral analysis.

        Relationship among porcine lncRNA TCONS_00010987, miR-323, and leptin receptor based on dual luciferase reporter gene assays and expression patterns

        Objective: Considering the physiological and clinical importance of leptin receptor (LEPR) in regulating obesity and the fact that porcine LEPR expression is not known to be controlled by lncRNAs and miRNAs, we aim to characterize this gene as a potential target of SSC-miR-323 and the lncRNA TCONS_00010987. Methods: Bioinformatics analyses revealed that lncRNA TCONS_00010987 and LEPR have SSC-miR-323-binding sites and that LEPR might be a target of lncRNA TCONS_00010987 based on cis prediction. Wild-type and mutant TCONS_00010987-target sequence fragments and wild-type and mutant LEPR 3'-UTR fragments were generated and cloned into pmiRRB-REPORT<sup>TM</sup>-Control vectors to construct respective recombinant plasmids. HEK293T cells were co-transfected with the SSC-miR-323 mimics or a negative control with constructs harboring the corresponding binding sites and relative luciferase activities were determined. Tissue expression patterns of lncRNA TCONS_00010987, SSC-miR-323, and LEPR in Anqing six-end-white (AQ, the obese breed) and Large White (LW, the lean breed) pigs were detected by real-time quantitative polymerase chain reaction; backfat expression of LEPR protein was detected by western blotting. Results: Target gene fragments were successfully cloned, and the four recombinant vectors were constructed. Compared to the negative control, SSC-miR-323 mimics significantly inhibited luciferase activity from the wild-type TCONS_00010987-target sequence and wild-type LEPR-3'-UTR (p<0.01 for both) but not from the mutant TCONS_00010987-target sequence and mutant LEPR-3'-UTR (p>0.05 for both). Backfat expression levels of TCONS_00010987 and LEPR in AQ pigs were significantly higher than those in LW pigs (p<0.01), whereas levels of SSC-miR-323 in AQ pigs were significantly lower than those in LW pigs (p<0.05). LEPR protein levels in the backfat tissues of AQ pigs were markedly higher than those in LW pigs (p<0.01). Conclusion: LEPR is a potential target of SSC-miR-323, and TCONS_00010987 might act as a sponge for SSC-miR-323 to regulate LEPR expression.

        Clinicopathologic and Prognostic Significance of the Zinc Finger of the Cerebellum Family in Invasive Breast Cancer

        Purpose: Five members of the zinc finger of the cerebellum (ZIC) family—ZIC1, ZIC2, ZIC3, ZIC4, and ZIC5—have been shown to be involved in various carcinomas. Here, we aimed to explore the clinicopathologic and prognostic roles of ZIC family members in invasive breast cancer patients using immunohistochemical analysis, western blotting analysis, and real-time quantitative polymerase chain reaction (RT-qPCR). Methods: A total of 241 female invasive breast cancer patients who underwent radical mastectomy between 2009 and 2011 were enrolled. ZIC proteins in 241 pairs of breast tumors and corresponding normal tissues were investigated using immunohistochemistry and the clinicopathologic roles of proteins were analyzed using Pearson’s chi-square test. Kaplan-Meier curves and Cox regression analysis were also used to analyze the prognostic value of the ZIC proteins. In addition, 12 pairs of fresh-frozen breast tumors and matched normal tissues were used in the western blotting analysis and RT-qPCR. Results: Only ZIC1 expression in normal tissues was obviously higher than that in tumors (p<0.001). On multivariate analysis, ZIC1 expression (in overall survival analysis: hazard ratio [HR], 0.405, 95% confidence interval [CI], 0.233–0.702, p=0.001; in disease-free survival analysis: HR, 0.395, 95% CI, 0.234–0.669, p=0.001) was identified as a prognostic indicator of invasive breast cancer. Conclusion: ZIC1, but not the other proteins, was obviously decreased in breast tumors and associated with clinicopathologic factors. Thus, ZIC1 might be a novel indicator to predict the overall and disease-free survival of invasive breast cancer patients.

        Deduction and exploration of the evolution and function of vertebrate GFPT family

        Background: Glutamine-fructose-6-phosphate aminotransferase (GFPT) is a key factor in the hexosamine metabolism pathway. It regulates the downstream factor O-GlcNAc to change cell function and plays an important role in the metabolism and immune process of tissues and organs. However, the evolutionary relationship of GFPT family proteins in vertebrates has not been elucidated. Objective: To deduce and explore the evolution and function of vertebrate GFPT family. Methods: 18 GFPT sequences were obtained from Homo sapiens (H. sapiens), Trachypithecus francoisi (T. francoisi), Mus musculus (M. musculus), Rattus norvegicus (R. norvegicus), Gallus gallus (G. gallus), Zootoca vivipara (Z. vivipara), Xenopus tropicalis (X. tropicalis), Danio rerio (D. rerio), Rhincodon typus (R. typus), Plasmodium relictum from National Center for Biotechnology Information (NCBI). The physical and chemical characteristics and molecular evolution of GFPT family proteins and nucleic acid sequences were analyzed by ClustalX2, Gene Doc, MEGA-X, SMART, Datamonkey, R etc. RESULTS: Based on the neighbor-joining (NJ) phylogenetic tree and evolution fingerprints, GFPT family members of vertebrates can be divided into two groups: the GFPT1 group and the GFPT2 group. Seven positive selection sites were identified by IFEL and integrated methods mixed effects model of evolution (MEME) and fixed effects likelihood (REL). Finally, we predicted 28 phosphorylation sites and 18 ubiquitousness sites in the human GFPT1 sequence, 10 phosphorylation sites, and five ubiquitousness sites in GFPT2. Gene ontology (GO) analyzes the protein molecules and KEGG signaling pathways of vertebrates interacting with GFPT family proteins. Conclusions: Our work confirmed that higher animals GFPT family may have differentiated GFPT1 and GFPT2, which meets their own functional needs. This knowledge answers the question what the origin and evolution of GFPT family in vertebrates and provided the basis for disease treatment and function research of GFPT protein.

        Full-scale testing and modeling of the mechanical behavior of shield TBM tunnel joints

        For shield TBM (Tunnel Boring Machine) tunnel lining, the segment joint is the most critical component for determining the mechanical response of the complete lining ring. To investigate the mechanical behavior of the segment joint in a water conveyance tunnel, which is different from the vehicle tunnel because of the external loads and the high internal water pressure during the tunnel's service life, full-scale joint tests were conducted. The main advantage of the joint tests over previous ones was the definiteness of the loads applied to the joints using a unique testing facility and the acquisition of the mechanical behavior of actual joints. Furthermore, based on the test results and the theoretical analysis, a mechanical model of segment joints has been proposed, which consists of all important influencing factors, including the elastic-plastic behavior of concrete, the pre-tightening force of the bolts and the deformations of all joint components, i.e., concrete blocks, bolts and cast iron panels. Finally, the proposed mechanical model of segment joints has been verified by the aforementioned full-scale joint tests.

        Effect of support on the performance of Mn–Cu oxides for low temperature selective catalytic reduction of NO with NH3

        MnOx–CuOx/TiO2 and MnOx–CuOx/Al2O3 catalysts were prepared by sol–gel method and used for low temperature selective catalytic reduction of NO with NH3. The results showed that MnOx–CuOx/TiO2 had better catalytic activity and SO2 resistance than MnOx–CuOx/Al2O3 in the temperature range of 100–250 ℃. The properties of the catalysts were characterized by using XRD, N2 adsorption, temperature programmed reduction (H2-TPR) and temperature programmed desorption (NH3-TPD). It was found that the support has a great impact on the acidity of catalyst; TiO2 and Al2O3 can promote the formation of Lewis acid sites and Bro¨ nsted acid sites respectively.

        A study on load-sharing structure of multi-stage planetary transmission system

        Unequal load distribution is a crucial factor in decreasing bearing capacity and stability of the planetary transmission system. In thispaper, a dynamical model of two-stage helical planetary gear transmission system is established based on lumped-parameter method andLagrange general function. Nonlinearity of gear tooth backlash and error is taken into account. Four load-sharing structures are proposedto study the load-sharing performance. A method to calculate dynamic sensitivity of load-sharing coefficient to errors is presented thatcan provide a reference to component precision determination in order to make planetary system have a better load distribution. Finally, anumerical method of load-sharing performance is validated by a test. These results provide fundamental basis for multi-stage planetarygear transmission system design.

        Improved Global Maximum Power Point Tracking Method Based on Voltage Interval for PV Array under Partially Shaded Conditions

        The power-voltage (P-V) curve of photovoltaic (PV) arrays connected in parallel to bypass diodes would have several local maximum power points (LMPP) under partial shading conditions (PSC). Conventional maximum power point tracking (MPPT) methods fail to search for the global maximum power point (MPP) because the searched peak point may remain at the LMPP on the P-V curve under PSC. This study proposes an improved MPPT algorithm to ensure that PV arrays operate at global maximum power point (GMPP) under PSC. The proposed algorithm is based on a critical study and a series of observations of PV characteristics under PSC. Results show the regularity of voltage interval between LMPPs. The algorithm has the advantages of rapidly reaching GMPP, maintaining stability, and recovering GMPP quickly when the operating condition changes. Simulation and experimental results demonstrate the feasibility of the proposed algorithm.
