      • High Expression of Bcl-2 Protein Predicts Favorable Outcome in Non-small Cell Lung Cancer: Evidence from a Systematic Review and Meta-analysis

        Background: The prognostic value of Bcl-2 protein expression in non-small cell lung cancer (NSCLC) is under debate. We therefore systematically reviewed the evidence for Bcl-2 protein effects on NSCLC survival to elucidate this issue. Materials and Methods: An electronic search in Pubmed and Embase complemented by manual searches in article references were conducted to identify eligible studies to evaluate the association between Bcl-2 protein expression and overall survival (OS) as well as disease free survival (DFS) of NSCLC patients. Combined hazard ratios (HRs) with corresponding 95% confidence intervals (95%CIs) were pooled using the random-effects model. Results: A total of 50 trials (including 52 cohorts) encompassing 7,765 patients were pooled in the meta-analysis regarding Bcl-2 expression and OS of NSCLC patients. High expression of Bcl-2 protein had a favorable impact (HR=0.76, 95%CI=0.67-0.86). In the group of Bcl-2 expression and DFS, 11 studies including 2,634 patients were included. The synthesized result indicated high expression of Bcl-2 protein might predict good DFS (HR=0.85, 95%CI=0.75-0.95). Conclusions: Our present meta-analysis demonstrated favorable prognostic values of Bcl-2 expression in patients with NSCLC. Further prospective trails are welcomed to validate the utility of assessing Bcl-2 in NSCLC patient management.

        Physiological character and molecular mapping of leaf-color mutant wyv1 in rice (Oryza sativa L.)

        The seed of an excellent indica restorer line Jinhui10 (Oryza sativa L. ssp. indica) was treated by ethyl methanesulfonate (EMS); a leaf-color mutant displaying distinct phenotype throughout development grown in paddy field was identified from the progeny. The mutant leaf showed white-yellow at seedling stage and then turned to yellow-green at tillering stage, after that, virescent color appeared until to maturity. The mutant was thus temporarily designed as wyv1. The chlorophyll contents decreased significantly and the changing was consistent with the chlorotic level of wyv1 leaves. Chlorophyll fluorescence kinetic parameters measured at the seedling stage showed that co-efficiency of photochemical quenching (qP),actual photosystem II efficiency (ΦPS II), electron transport rate (ETR) and initial chlorophyll fluorescence level (Fo), net photosynthetic rate (Pn) and maximum photochemical efficiency (Fv / Fm) significantly decreased in severe chlorotic leaf of the mutant compared with that of wild type. However,no significant differences were observed for Pn and Fv/Fm between virescent leaf and normal green leaf. Genetic analysis suggested that the mutant phenotype was controlled by a single recessive nuclear gene which was finally mapped between SSR marker Y7 and Y6 on rice chromosome 3 based on F2 population of Xinong1A / wyv1. Genetic distances were 0.06 cM and 0.03 cM respectively, and the physical distance was 84kb according to the sequence of indica rice 9311. The results must facilitate map-based cloning and functional analysis of WYV1 gene.

      • Galectin-9 Acts as a Prognostic Factor with Antimetastatic Potential in Hepatocellular Carcinoma

        Considerable research has been conducted concerning galectin-9 and carcinomas, but little information is available about any relation with the hepatocellular carcinoma. In this study, we employed a small interfering RNA (siRNA) targeting galectin-9 to down-regulate the expression in HepG2 cells. As a result, after galectin-9 expression was reduced, cell aggregation was suppressed, while other behaviour such as the proliferation, adhesion and invasion to ECM, cell-endothelial adhesion and transendothelial invasion of the cells were markedly enhanced. When tumors of 200 patients with hepatocellular carcinoma were tested for galectin-9 expression by immunohistochemistry, binding levels demonstrated intimate correlations with the histopathologic grade, lymph node metastasis, vascular invasion and intrahepatic metastasis (P<0.05). Moreover, survival analysis indicated that patients with galectin-9 expression had much longer survival time than those with negative lesions, and the Log-rank test indicated that this difference was statistical significant (P<0.0001). The Cox proportional hazards model suggested that negative galectin-9 expression in hepatocellular carcinoma represented a significant risk factor for patient survival. We propose that galectin-9 might be a new prognostic factor with antimetastatic potential in patients with hepatocellular carcinoma.

        Airfoil optimization based on multi-objective bayesian

        This paper proposes a novel aerodynamic optimization framework for airfoils, which utilizes OpenFOAM, an open-source computational fluid dynamics software, and a Bayesian network to achieve efficient optimization of airfoil aerodynamic performance. Aerodynamic analysis of the NACA 4-digit airfoil was performed by adopting the Spalart-Allmaras turbulence model to solve the Reynolds-averaged Navier–Stokes equations. With the use of this framework, the optimal lift-to-drag ratio can be found by using a small number of objective evaluations. The optimal angle of attack and aerodynamic shape can be obtained under different thicknesses. Finally, after various aerodynamic objectives are arranged and combined, the Pareto fronts were obtained by the multi-objective Bayesian algorithm. Compared with the original NACA four-digit airfoil, the lift-to-drag ratio of the airfoil after single-objective optimization is greatly improved as the thickness increases, and the airfoil after multi-objective optimization achieves different Pareto sets according to different sailing phases.

        Induction of Apoptosis in Human Hepatoma SMMC-7721 Cells using 2',4'-Dihydroxy-6'-methoxy-3',5'-dimethylchalcone, a Chalcone from Buds of Cleistocalyx operculatus

        The molecular mechanism to induce apoptosis in human hepatoma SMMC-7721 cells was investigated using 2',4'-Dihydroxy-6'-methoxy-3',5'-dimethylchalcone (DMC) isolated from the buds of Cleistocalyx operculatus. Typical morphological changes in apoptotic body formation after DMC treatment were observed using acridine orange (AO)/ethidium bromide (EB) staining. After 48 h of incubation with 0, 6.25, 12.5, and 25 μM of DMC, the percentages of annexin V-FITC-positive/PI negative cells were 1.51, 9.33, 33.84, and 61.32%, respectively. Apoptosis induced by DMC was blocked by z-VAD-fmk, a caspase inhibitor. Analysis of fluorescence intensities of rhodamine 123 in cells showed that DMC induced a loss of mitochondrial membrane potential (ΔΨm). Western blot results showed that DMC decreased the Bcl-2/Bax ratio,suggesting that the Bcl-2 family is involved in control of apoptosis. All of these signal transduction pathways are involved in initiating apoptosis. DMC is a potential anticancer agent that deserves further exploration.

        New Delay-range-dependent Exponential Estimates for Singular Systems with Time-varying Delay

        This paper considers the problem of exponential stability for continuous-time singular systems with in-terval time-varying delay. By defining a novel Lyapunov-Krasovskii function and giving a tighter up-per bound of its derivative, a new delay-range-dependent exponential admissibility criterion, which not only guarantees the regularity, absence of impulses and exponential stability of the system but also gives the estimates of decay rate and decay coefficient, is established in terms of linear matrix inequality (LMI). The resulting criterion has advantages over the result previously reported by Haidar et al. [17] in that it involves fewer matrix variables but has less conservatism, which is established theoretically. Examples are provided to demonstrate the advantage of the proposed criterion.

        Gene Mapping and Expression Analysis of a Novel Mutant reproduce organs absent (roa) in Rice

        Mutant plays an important role in function analysis in plant. A rice flower mutant reproduce organs absent (roa), showing a stable inheritance during several years of study, was identified in rice (Oryza sativa L. ssp. Indica) cultivar Jinhui10 treated with EMS and used in this study. This mutant showed following: elongated palea and pedicle; absence of inner three whorls of floral organs; multi-whorls glume like organs inside the lemma/palea; spikelet meristem like organ upon the pedicle. These phenotypes suggested that ROA is a key gene in rice spikelet development. Genetic analysis confirmed that the mutant traits were controlled by a single recessive nuclear gene. By gene mapping, ROA was restricted between two SSR markers RM221 and RM1342 on the chromosome 2. It concluded that ROA was a novel gene involving in flower development in rice. Besides, the mutation of ROA influenced the transcription level of floral homeotic genes; the expression of floral homeotic genes decreased in roa panicle compared with wild-type, and it suggested that ROA affected flower development by influencing the expression of floral homeotic genes.

      • Clinical and Prognostic Significance of SOX11 in Breast Cancer

        Recently, the transcription factor SOX11 has gained extensive attention as a diagnostic marker in a series of cancers. However, to date, the possible roles of SOX11 in breast cancer has not been investigated. In this study, immunohistochemical staining for SOX11 was performed for 116 cases of breast cancer. Nuclear SOX11 was observed in 42 (36.2%) and cytoplasmic SOX11 in 52 (44.8%) of breast cancer samples. Moreover, high expression of cytoplasmic and nuclear SOX11 was associated with clinicopathological factors, including earlier tumor grade, absence of lymph node metastasis and smaller tumor size. Kaplan-Meier survival curves demonstrated high nuclear SOX11 expression to be associated with more prolonged overall survival than those with low expression and it could be an independent predictor of survival for breast cancer patients. It is worthwhile to note that cytoplasmic SOX11 was not correlated with prognosis of breast cancer patients. These data suggest the possibility that nuclear SOX11 could be as a potential target for breast cancer therapy.
