      • Mechanistic Analysis of Taxol-induced Multidrug Resistance in an Ovarian Cancer Cell Line

        Objectives: To establish a taxol-resistant cell line of human ovarian carcinoma (A2780/Taxol) and investigate its biological features. Methods: The drug-resistant cell line (A2780/Taxol) was established by continuous stepwise selection with increasing concentrations of Taxol. Cell morphology was assessed by microscopy and growth curves were generated with in vitro and in vivo tumor xenograft models. With rhodamine123 (Rh123) assays, cell cycle distribution and the apoptotic rate were analyzed by flow cytometry (FCM). Drug resistance-related and signal associated proteins, including P-gp, MRPs, caveolin-1, PKC-${\alpha}$, Akt, ERK1/2, were detected by Western blotting. Results: A2780/Taxol cells were established with stable resistance to taxol. The drug resistance index (RI) was 430.7. Cross-resistance to other drugs was also shown, but there was no significant change to radioresistance. Compared with parental cells, A2780/Taxol cells were significantly heteromorphous, with a significant delay in population doubling time and reduced uptake of Rh123 (p<0.01). In vivo, tumor take by A2780 cells was 80%, and tumor volume increased gradually. In contrast, with A2780/Taxol cells in xenograft models there was no tumor development. FCM analysis revealed that A2780/Taxol cells had a higher percentage of G0/G1 and lower S phase, but no changes of G2 phase and the apoptosis rate. Expression of P-gp, MRP1, MRP2, BCRP, LRP, caveolin-1, PKC-${\alpha}$, Phospho-ERK1/2 and Phospho-JNK protein was significantly up-regulated, while Akt and p38 MARK protein expression was not changed in A2780/Taxol cells. Conclusion: The A2780/Taxol cell line is an ideal model to investigate the mechanism of muti-drug resistance related to overexpression of drug-resistance associated proteins and activation of the PKC-${\alpha}/ERK$ (JNK) signaling pathway.

        Physiological responses of yellow‑horn seedlings to high temperatures under drought condition

        Drought and hot stresses are the primary limiting factors for the growth and seed production of yellow-horn (Xanthoceras sorbifolia Bunge), especially in Xinjiang Province of China. A better understanding of its physiological and biochemical characteristics under drought–hot stress is requisite for its efficient cultivation in the arid and semi-arid areas. In this study, the physiological and biochemical responses of 3-month-old yellow-horn seedlings were evaluated after 7 days of treatments of five temperatures (25 °C/20 °C, 30 °C/25 °C, 35 °C/25 °C, 40 °C/25 °C and 45 °C/25 °C) under two watering conditions [adequate water supply (HW): 80 ± 5% FC (field capacity); low water supply (LW): 40 ± 5% FC] by a pot experiment. The increased temperature (above to 35 °C) markedly aggravated the damage on biomass accumulation and membrane integrity of yellow-horn seedlings under the LW. The accumulation of soluble sugars and proteins exhibited a growing trend as the temperature increased from 25 to 40 °C, which declined when the temperature was higher than 40 °C except for the soluble protein accumulation under HW. In addition, captured responsive characteristics of the gas-exchange parameters and chlorophyll fluorescence have indicated that the combinations of high temperature (above to 35 °C) and LW induced significant decrease in photosynthetic activities of yellow-horn seedlings. All these results showed that the hot stress significantly aggravated the drought damage on yellow-horn plant growth, especially when the temperature increased above 35 °C under drought stress.

        Ginsenoside compound K inhibits nuclear factor-kappa B by targeting Annexin A2

        Background: Ginsenoside compound K(C-K), a major metabolite of ginsenoside, exhibits anticancer activity in various cancer cells and animal models. A cell signaling study has shown that C-K inhibited nuclear factor-kappa B ($NF-{\kappa}B$) pathway in human astroglial cells and liver cancer cells. However, the molecular targets of C-K and the initiating events were not elucidated. Methods: Interaction between C-K and Annexin A2 was determined by molecular docking and thermal shift assay. HepG2 cells were treated with C-K, followed by a luciferase reporter assay for $NF-{\kappa}B$, immunofluorescence imaging for the subcellular localization of Annexin A2 and $NF-{\kappa}B$ p50 subunit, coimmunoprecipitation of Annexin A2 and $NF-{\kappa}B$ p50 subunit, and both cell viability assay and plate clone formation assay to determine the cell viability. Results: Both molecular docking and thermal shift assay positively confirmed the interaction between Annexin A2 and C-K. This interaction prevented the interaction between Annexin A2 and $NF-{\kappa}B$ p50 subunit and their nuclear colocalization, which attenuated the activation of $NF-{\kappa}B$ and the expression of its downstream genes, followed by the activation of caspase 9 and 3. In addition, the overexpression of Annexin A2-K320A, a C-K binding-deficient mutant of Annexin A2, rendered cells to resist C-K treatment, indicating that C-K exerts its cytotoxic activity mainly by targeting Annexin A2. Conclusion: This study for the first time revealed a cellular target of C-K and the molecular mechanism for its anticancer activity.

        Effect of temperature and spring-mass systems on modal properties of Timoshenko concrete beam

        An exact solution for the title problem was obtained in closed-form fashion considering general boundary conditions. The expressions of moment, shear and shear coefficient (or shear factor) of cross section under the effect of arbitrary temperature distribution were first derived. In view of these relationships, the differential equations of Timoshenko beam under the effect of temperature were obtained and solved. Second, the characteristic equations of Timoshenko beam carrying several spring-mass systems under the effect of temperature were derived based on the continuity and force equilibrium conditions at attaching points. Then, the correctness of proposed method was demonstrated by a Timoshenko laboratory beam and several finite element models. Finally, the influence law of different temperature distribution modes and parameters of spring-mass system on the modal characteristics of Timoshenko beam had been studied, respectively.

        Time-varying H∞ Control for Discrete-time Switched Systems with Admissible Edge-dependent Average Dwell Time

        The problem of H∞ control for discrete-time switched systems is investigated via admissible edgedependent average dwell time (AED-ADT) method in this paper. By virtue of a convex combination of positive definite matrices, a novel multiple piecewise convex Lyapunov function (MPCLF) is designed, which can relax the restricted conditions of Lyapunov functions at switching points and interval interior points. Based on the MPCLF approach, the time-varying H∞ state feedback controllers, guaranteeing that the corresponding closed-loop system is globally uniformly exponentially stable (GUES) with a prescribed H∞ performance, are established for the considered switched system. Finally, three numerical examples are provided to illustrate the effectiveness of the proposed approaches.

      • XPD Lys751Gln and Asp312Asn Polymorphisms and Gastric Cancer Susceptibility: A Meta-analysis of Case-control Studies

        Background: Published data regarding the association between xeroderma pigmentosum group D (XPD) Lys751Gln and Asp312Asn polymorphisms and gastric cancer susceptibility havew been inconclusive. This meta-analysis was therefore performed toobtain a more precise estimation of any relationship. Materials and Methods: A comprehensive literature search was conducted to identify all case-control studies of Lys751Gln and Asp312Asn polymorphisms and susceptibility to gastric cancer. Summary odds ratios (ORs) and its 95% confidence intervals (95% CIs) were calculated using a random-effects model with the software STATA (version10.0). Results: A total of 12 case-control studies including 3,147 cases and 4,736 controls were included. Overall, no significant associations were found in some models (for Lys751Gln: Lys/Gln vs Lys/Lys: OR=1.144, 95% CI=0.851-1.541, Gln/Gln vs Lys/Lys: OR=1.215, 95% CI = 0.740-1.955, dominant model: OR=1.137, 95% CI=0.818-1.582; recessive model: OR=1.123, 95% CI=0.765-1.650; for Asp312Asn: Asp/Asn vs Asp/Asp: OR=1.180, 95% CI=0.646-2.154, dominant model: OR=1.380, 95% CI = 0.812-2.346), but significantly elevated susceptibility was found for Asp312Asn polymorphism in some models (Asn/Asn vs Asp/Asp: OR=2.045, 95% CI=1.254-3.335, recessive model: OR=1.805, 95% CI =1.219-2.672), for the additive model, the XPD Lys751Gln and Asp312Asn polymorphisms were not significantly associated with gastric cancer susceptibility. In stratified analyses, significantly elevated susceptibility was found for some models in the Chinese population. Conclusion: This meta-analysis suggested the XPD Asp312Asn polymorphism might be a potential biomarker of gastric cancer susceptibility in overall population, while both XPD Lys751Gln and Asp312Asn polymorphisms might be risk factors of gastric cancer susceptibility in Chinese.

      • Microstructure and cutting performance of CrTiAlN coating for high-speed dry milling

        <P>Using a closed field unbalanced magnetron sputtering system, the cemented carbide end mills were coated with a CrTiAIN hard coating, which consisted of a Cr adhesive layer, a CrN interlayer and a CrTiAIN top layer. The microstructure and mechanical properties of the coating were investigated by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), micro indentation and scratch test. The cutting performance of the coated end mills were conducted by high-speed dry milling hardened steel (P20, HRC 45). The results indicates that the coating is composed of (Cr, Ti, Al)N columnar grains with nanolayers. The coating exhibits good adhesion to cemented carbide substrate and high microhardness of around 30 GPa. The coated end mills show significant improvement on tool life and much lower cutting force as compared to the uncoated ones. And the related mechanisms were discussed.</P>
