        Apoptosis of Human Burkitt’s Lymphoma Cells Induced by 2-N,NDiethylaminocarbonyloxymethyl-1-diphenylmethyl-4-(3,4,5-trimethoxybenzoyl) piperazine Hydrochloride (PMS-1077)

        Piperazine is one of the heterocycles which are associated with diverse pharmacological activities. 2-N,N-Diethylaminocarbonyloxymethyl-1-diphenylmethyl-4-(3,4,5-trimethoxybenzoyl) piperazine hydrochloride (PMS-1077) is a trisubstituted piperazine which contains a trimethoxybenzene ring and a benzhydrylpiperazine fragment, both of which can induce cell proliferation regression by different mechanisms. We have therefore examined the effects of PMS-1077 on Human Burkitt’s lymphoma cells (Raji). The viability of Raji cells was determined by MTT assay and also assessed by trypan blue dye exclusion method. The results demonstrate that PMS-1077 can suppress the proliferation of Raji cells in a dose- and timedependent manner, while inhibit colony formation ability of Raji cells merely in a dose-dependent manner in vitro. Meanwhile, morphological changes were observed using fluorescence microscope. Flow cytometric analysis through PI stains showed that PMS-1077 blocked the growth of Raji cells in the G0/G1 period, and induced apoptosis of Raji cells after 48 h of incubation. Cell apoptosis induced by PMS-1077 was further confirmed by staining with Annexin-V FITC and PI. Preliminary study by molecular docking suggests that PMS-1077 may inhibit tubulin polymerization. More experiments are in progress in our laboratory to reveal the mode of action of PMS-1077 in the induction of apoptosis of Raji cells.

        A putative plastidial adenine nucleotide transporter, BRITTLE1-3, plays an essential role in regulating chloroplast development in rice (Oryza sativa L.)

        Differentiation from proplastids into chloroplasts isa light- and energy-dependent process. How this process isregulated is still poorly understood at the molecular level. We herein report a new putative plastidial adenine nucleotidetransporter, BRITTLE1-3 (referred to as OsBT1-3), encoded bythe rice (Oryza sativa) White Stripe Leaf 2 (WSL2) gene. Loss of OsBT1-3 function results in defective chloroplastbiogenesis, severely reduced photosynthetic efficiency, andfinally a white stripe leaf phenotype in the first four leaves. The expression levels of genes related to chlorophyllbiosynthesis and photosynthesis are drastically reduced,accompanied with over accumulation of reactive oxygenspecies (ROS) in the wsl2 mutant. OsBT1-3 is targeted tothe chloroplasts and it expresses in almost all tissues inplants, especially in young leaves. OsBT1-3 consists of 419amino acids and exhibits features of all mitochondrialcarrier proteins, including a typical transmembrane-spanningdomain and a highly conserved sequence motif designatedas the ‘mitochondrial energy transfer signatures’. Phylogeneticanalysis shows that OsBT1-3 is a putative plastidialadenine nucleotide transporter and is most closely relatedto ZmBT1-2. Together, these observations suggest that thenew putative adenine nucleotide transporter, OsBT1-3,plays an essential role in regulating chloroplast biogenesisand maintenance of ROS homeostasis during rice seedlingde-etiolation.

        Green-revertible Chlorina 1 (grc1) is Required for the Biosynthesis of Chlorophyll and the Early Development of Chloroplasts in Rice

        The nuclear genes involved in chloroplast developmentand chlorophyll biosynthesis must be investigated tounderstand their functions in plant growth and development. In this study, we isolated and identified a unique leaf-colormutant of rice with a green-yellow phenotype before thefour-leaf stage and named the mutation green-revertiblechlorina 1 (grc1). The mutants had significantly lower plantheight, number of tillers, and panicle length and headedsignificantly earlier than the wild type. The levels ofchlorophylls, carotenoids, and chlorophyll precursors werealso lower. The mutation in grc1 affected chloroplastultrastructure, particularly thylakoid development. Geneticanalysis indicated that the green-yellow phenotype wascontrolled by a single recessive gene. We mapped the grc1gene to a 32.4-kb region on the long arm of chromosome 6. Through map-based cloning, we identified a 45-bp insertionin the genomic region of LOC_Os06g40080, which encodeda heme oxygenase. Expression of LOC_Os06g40080 wassignificantly down-regulated in the grc1 mutant. Subcellularlocalization showed that this heme oxygenase was localizedin the chloroplast. In summary, we isolated and identified thegene for grc1, which plays an important role in chlorophyllbiosynthesis and chloroplast development in rice.

      • HCV, Acute, LT : A Disparate Subset of Double Negative T cells Contributes to the Outcome of Murine Fulminant Viral Hepatitis via Effector Molecule Fibrinogen-like Protein 2

        Aims: The underlying pathogenesis of fulminant viral hepatitis (FVH) has not been fully elucidated. As a subset of regulatory T cells, CD3+CD4-CD8- double negative (DN) T cells can suppress T cell responses. In this study, we present new insights into the immune mediated mechanisms involved in FVH caused by murine hepatitis virus strain 3 (MHV-3). Methods: The phenotype and cytokines of DN T cells were detected by flow cytometric analysis. The levels of mfgl2 were measured by real-time PCR and western-blot. The function of mfgl2 was measured by PCA. Results: After MHV-3 infection, the proportions of DN T cells increased significantly in BALB/cJ mice, and splenic DN T cells expressing high levels of CD69 were recruited by MHV-3 infected hepatocytes to the liver. Serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and total bilirubin (TBil) increased, accompanied by massive hepatocyte necrosis. These DN T cells were predominantly consisted of a TCRαβ+ subset expressing high levels of CD44, and did not produce cytokine except IL-2. Adoptive transfer of this subset of DN T cells to the MHV-3 infected mice resulted in an increase of murine fibrinogen-like protein 2 (mfgl2) expression in association with massive fibrin deposition in the liver. Following MHV-3 infection, membrane mfgl2 expression and functional procoagulant activity (PCA) increased remarkably in the DN T cells. Introduction of a recombinant adenovirus which encoded a microRNA specifically targeting mfgl2 gene (Ad-mfgl2-miRNA) in vivo significantly inhibited the hepatic expression of mfgl2, increased mice survival. However, under this condition, adoptive transfer of the DN T cells accelerated the disease progression and reversed the benefit from mfgl2 gene silence, led to a 100% death. Conclutions: Our results demonstrated that DN T cell-derived mfgl2 may serve as an important effector molecule contributing to the pathogenesis of MHV-3-induced FVH.

      • Contribution of Macrophage Migration Inhibitory Factor -173G/C Gene Polymorphism to the Risk of Cancer in Chinese Population

        Background: Macrophage migration inhibitory factor (MIF) -173G/C (rs755622) gene polymorphism has been associated with cancer risk. Previous studies have revealed that MIF -173G/C gene polymorphism may increase cancer in the Chinese population, while results of individual published studies remain inconsistent and inconclusive.We performed this meta-analysis to derive a more precise estimation of the relationship. Materials and Methods: We conducted a search on PubMed, Embase, MEDLINE, Cochrane Library, Chinese National Knowledge Infrastructure (CNKI), Wanfang, Weipu on Dec 31, 2014.Odds ratio (OR) and 95% confidence interval (95% CI) were used to assess the association. A total of eight studies including 2,186 cases and 2,285 controls were involved in this meta-analysis. Results: The pooled results indicated the significant association between MIF -173G/C polymorphism and the risk of cancer for Chinese population (CC + CG vs GG: OR=1.14, 95%CI=1.02-127, pheterogeneity<0.01; P=0.023; CC vs CG+GG: OR=1.12, 95%CI=1.02-1.23, pheterogeneity<001; P=0.017;CC vs GG: OR=1.18, 95%CI=1.04-1.33, pheterogeneity<001; P=0.008; CG vs GG:OR=1.03, 95%CI=0.91-1.15, pheterogeneity<001; P=0.656; C vs G:OR=1.24, 95%CI=1.14-1.25, pheterogeneity<001; P<001). Subgroup analysis showed that in patients with "solid tumors", heterogeneity was very large (OR=0.94,95%CI=0.83-1.06,pheterogeneity=0.044; p=0.297). Within "non-solid tumors", the association became even stronger (OR=6.62, 95 % CI=4.32-10.14, pheterogeneity<0.001; p<0.001). Conclusions: This study suggested that MIF -173G/C gene polymorphism may increase increase cancer in the Chinese population.Furthermore, more larger sample and representative population-based casees and well-matched controls are needed to validate our results.

      • Peptidoglycans Promotes Human Leukemic THP-1 Cell Apoptosis and Differentiation

        The innate immune system coordinates the inflammatory response to pathogens. To do so, its cells must discriminate self from non-self utilizing receptors that identify molecules synthesized exclusively by microbes. Toll-like receptors have a crucial role in the detection of microbial infection in mammals and insects. In mammals, they have evolved to recognize conserved products unique to microbial metabolism. These include lipopolysaccharide (LPS), lipotechoic acids, and peptidoglycans (PGN). We show here that TLRs, including TLR2, are expressed on the THP-1 human leukemia cell line. Activation of TLR2 signaling in THP-1 by PGN induces the synthesis of various soluble factors and proteins including interleukin-$1{\beta}$, interleukin-8 and TNF-${\alpha}$ and apoptosis of THP-1 with PGN dose and time dependence. Moreover, in this study we show that PGN induces apoptosis of THP-1 cells in a TNF-${\alpha}$-dependent manner. These findings indicate that TLR2 signaling results in a cascade leading to tumor apoptosis and differentiation, which may suggest new clinical prospects using TLR2 agonists as cytotoxic agents in certain cancers.

        Optical Characters Measurement and Simulation of 2D Cross Grating Microstructures of Butterfly Wings

        In this paper, the reflectance of the butterfly wings are measured by using the microspectrophotometer as the optical character with the reflectance on several scales on the wings. Reflectance spectra from different wing scale zones with the same color are dramatically different. And it suggests that the tiny structural difference has the great influence on the reflectance property. In addition, the microstructures of the butterfly wings can be simplified to a 2D crossing grating. By simulating a two-dimensional model using rigorous coupled-wave analysis technique, the optical properties of the butterfly wings were investigated. The simulation results depend strongly on the structural parameters and refractive index. The research will reveal the mechanisms of the structural color of 2D crossing grating butterfly wings.

      • Comparison of Primary Breast Cancer Size by Mammography and Sonography

        Purpose: To compare tumor size by mammography and sonography and align with pathological results in primary breast cancer cases. Materials and Methods: We retrospectively reviewed 95 primary breast cancer patients who underwent mammography and sonography from January 2011 to June 2012. The largest tumor diameter was chosen as sizing reference for each imaging modality. The measurements of mammography and sonography were considered concordant if they were within the measurement of pathological results ${\pm}0.5cm$. Pearson's correlation coefficient was calculated for imaging results. Results: The range of the maximum diameter was 0.6cm-10.5cm and mean value was $3.81{\pm}2.04cm$ by pathological results, 0.7cm-12.4 cm and $3.99{\pm}2.19cm$ by mammography, and 0.9cm-11.0cm and $3.63{\pm}2.01cm$ by sonography, respectively. Sonography (R: 0.754), underestimated tumor size, but had a better correlation with pathological tumor size compared to mammography (R: 0.676), which overestimated tumor size. Conclusions: Sonography is superior to mammography in assessment of primary breast cancer.

        Common Model EMI Prediction in Motor Drive System for Electric Vehicle Application

        Common mode (CM) conducted interference are predicted and compared with experiments in a motor drive system of Electric vehicles in this study. The prediction model considers each part as an equivalent circuit model which is represented by lumped parameters and proposes the parameter extraction method. For the modeling of the inverter, a concentrated and equivalent method is used to process synthetically the CM interference source and the stray capacitance. For the parameter extraction in the power line model, a computation method that combines analytical method and finite element method is used. The modeling of the motor is based on measured date of the impedance and vector fitting technique. It is shown that the parasitic currents and interference voltage in the system can be simulated in the different parts of the prediction model in the conducted frequency range (150 kHz-30 MHz). Experiments have successfully confirmed that the approach is effective.
