        New Permeable Structure for Controlling Debris Flows in the Wenjiagou Gully

        Aimed to control debris flows more effectively, we attempt to develop a new structure called a “sloping roof grill dam”, which consists of guidance walls, separation grills and lateral deposit areas. A series of flume tests were performed to verify the function of this new structure, focusing on the trapping efficiency (β ), deceleration efficiency (μ) and coarse-fine sediment separation extents (η) under different conditions of structure and debris flows. Experimental results show that both the trapping and deceleration efficiency increase with a decrease in permeability, whereas the separation extent shown the opposite effect. Multiple regression analysis indicates that β and η are linearly dependent on the permeability, deceleration efficiency and magnitudes of debris flow. In addition, an increase in beam gap can lead to an increase in the impact area downstream for the debris flow after the control of the structure. Finally, a two-level installation of the structure is tested to determine whether there is an enhancement of the debris control, which indicated that although the trapping effect of the two-level structure is considerably greater than that of a single-level structure (2.11 times at maximum), the separation extent of the two-level structure is not distinct and may be even lower.

      • Inhibitory Effects of Syk Transfection on Lung Cancer Cell Invasion

        Objective: Spleen tyrosine kinase (Syk) is closely related to tumor invasion and metastasis, and has been shown to have potential inhibitory effects in tumors. In this study, we constructed a eukaryotic expression vector for Syk and analyzed its effects on invasive ability of the A549 non-small cell lung cancer cell line in vitro. Methods: A fragment of Syk was obtained by RT-PCR from human lung cancer cells and cloned into the expression vector pLNCXSyk. After restriction endonuclease digestion, PCR and DNA sequencing confirmation, the recombinant Syk expression plasmid was transfected into A549 human lung cancer cells using lipofectamine protocols. After selection, the cells stably expressed Syk. Detection of Syk expression of the cells by RT-PCR, and invasive ability were examined. Results: The eukaryotic expression plamid pLNCXSyk was constructed and expressed stably in the A549 human lung cancer cells. The RT-PCR results showed that Syk mRNA expression was upregulated significantly (P<0.05). Lower invasion through a basal membrane were apparent after transfection (P<0.05). Conclusions: A eukaryotic expression plasmid to cause Syk expression in lung cancer cells can obviously inhibit their invasive ability in vitro.

        Inverse Analysis of Inconel 718 Laser-Assisted Milling to Achieve Machined Surface Roughness

        This manuscript proposes an inverse analysis method for the machined surface roughness in laser-assisted milling on Inconel 718. The method solves the forward problem considering the tool profile and the elastic recovery of machined surface and applies the variance-based recursive method to guide the updating mechanism of process parameters to match the measurements. Subsequently, the inverse analysis identifies four process parameters of feed per tooth, tool tip radius, minimum cutting thickness, and tool tip angle, and finds the optimal solution for target performance, the surface roughness. The measurements are collected under the single beam coaxial laser-assisted milling spindle. The proposed modified Kalman filter algorithm introduces the gain coefficient G when updating the process parameters to improve robustness and accuracy. The inverse analysis is conducted on all measurements, and the average error of target performance is 0.460% when the laser is on and 0.394% when the laser is off. The average difference of process parameters is less than 5%, and the selection process is done in 50 loops within a minute. Therefore, the proposed inverse analysis model is robust, adaptive to different initial guesses and measurements, highly accurate, and saves computation time.

        Dielectric properties of Mg0.95Ni0.05TiO3 ceramic modified by Nd0.5Na0.5TiO3 at microwave frequencies

        The effect of Nd0.5Na0.5TiO3 addition on the microwave dielectric properties of Mg0.95Ni0.05TiO3 ceramics was investigated. The Nd0.5Na0.5TiO3 content plays a significant role in improving the dielectric properties. Microwave dielectric measurements show an increase in the dielectric constant (εr) and temperature coefficient of the resonant frequency (τf) and a decrease in the quality factor (Q × f value) with increasing Nd0.5Na0.5TiO3 content. By properly adjusting the Nd0.5Na0.5TiO3 content, a zero τf value can also be obtained, which make Mg0.95Ni0.05TiO3 a promising material in wireless systems. At 1300 ℃, the 0.81Mg0.95Ni0.05TiO3–0.19Nd0.5Na0.5TiO3 ceramics possess excellent microwave dielectric properties: a dielectric constant (εr) of 25.61, a Q × f value of 69,100 GHz, and a τf value of -6 ppm/℃. The effect of Nd0.5Na0.5TiO3 addition on the microwave dielectric properties of Mg0.95Ni0.05TiO3 ceramics was investigated. The Nd0.5Na0.5TiO3 content plays a significant role in improving the dielectric properties.         Transcriptional Analysis of 10 Selected Genes in a Model of Penicillin G Induced Persistence of Chlamydophila psittaci in HeLa Cells

        Chlamydophila psittaci is an important intracellular pathogen. Persistent infection is an important state of the host-parasite interaction in this chlamydial infection, which plays a significant role in spreading the organism within animal populations and in causing chronic chlamydiosis and serious sequelae. In this study, a C. psittaci persistent infection cell model was induced by penicillin G, and real-time quantitative PCR was used to study the transcriptional levels of 10 C. psittaci genes (dnaA, dnaK, ftsW, ftsY, grpE, rpsD, incC, omcB, CPSIT_0846, and CPSIT_0042) in acute and penicillin-G-induced persistent infection cultures. Compared with the acute cultures, the penicillin-G-treated cultures showed a reduced chlamydial inclusion size and a significantly decreased number of elementary body particles. Additionally, some enlarged aberrant reticulate body particles were present in the penicillin- G-treated cultures but not the acute ones. The expression levels of genes encoding products for cell division (FtsW, FtsY) and outer membrane protein E encoding gene (CPSIT_0042) were downregulated (p < 0.05) from 6 h post-infection onward in the persistent infection cultures. Also from 6 h post-infection, the expression levels of DnaA, DnaK, IncC, RpsD, GrpE, and CPSIT_0846 were upregulated (p < 0.05); however, the expression level of OmcB in the persistent infection was almost the same as that in the acute infection (p > 0.05). These results provide new insight regarding molecular activities that accompany persistence of C. psittaci, which may play important roles in the pathogenesis of C. psittaci infection.

        Optical Characters Measurement and Simulation of 2D Cross Grating Microstructures of Butterfly Wings

        In this paper, the reflectance of the butterfly wings are measured by using the microspectrophotometer as the optical character with the reflectance on several scales on the wings. Reflectance spectra from different wing scale zones with the same color are dramatically different. And it suggests that the tiny structural difference has the great influence on the reflectance property. In addition, the microstructures of the butterfly wings can be simplified to a 2D crossing grating. By simulating a two-dimensional model using rigorous coupled-wave analysis technique, the optical properties of the butterfly wings were investigated. The simulation results depend strongly on the structural parameters and refractive index. The research will reveal the mechanisms of the structural color of 2D crossing grating butterfly wings.
