      • 노화가 인체 중간엽 줄기세포로부터 조골세포로의 증식 및 분화에 미치는 영향

        연구배경: 일반적으로 골다공증과 연관된 위험인자로는 연령, 폐경, 약물, 불충분한 칼슘섭취, 만성질환 및 운동부족 등이 있는데, 특히 노화가 진행할수록 골밀도가 감소하는 것은 잘 알려져 있다. 노화와 관련하여 진행되는 골소실은 조골세포 및 전구조골세포의 기능적 결핍에 의한 골형성의 감소가 주요한 요인으로 여겨지고 있다. 그 동안 연령이 조골모 세포의 양과 조골모 세포로부터 성숙조골세포로의 분화 및 증식에 미치는 영향에 대한 일부 보고들이 있었으나 아직 일치된 견해는 없는 형편이다. 방법: 다양한 연령의 사람으로부터 골수를 채취, 중간엽 줄기세포가 포함된 단핵세포를 분리한 후 조골세포로 분화하기 좋은 조건하에서 배양하였다. 대상군은다시 젊은군과 노령군으로 구분하여 다양한 변수를 비교 분석하였다. 일차배양에서는 CFU-F를 계수하여 골수내 중간엽 줄기세포의 수를 추산하였고, 칼슘측정을 통하여 기질의 무기화 정도를 비교하였다. 계대배양후 이차배양에서는 시기별로 알카리성 포스파타제 활성도를 측정하고 오스테오칼신 mRNA의 발현을 관찰하여 젊은군과 노령군 사이의 증식능 차이를 비교하였다. 또한 이차배양 시기별로 MTT 측정을 하여 양군간에 증식능 차이가 있는지 알아보았다. 결과: 1. 일차배양 15일째에 평균 CFU-F의 수는 젊은군에서 유의하게 많았다(젊은군 148.3±28.9, 노령군 54.3±9.1, p=0.02). CFU-F의 평균면적은 젊은군에서 넓은 경향을 보였으나 통계적으로 유의하지는 않았다. 2. 일차배양 17일 경과 후 양군간에 기질 칼슘 침착정도는 유의한 차이를 보이지 않았다(젊은군 103.6±50.6, 노령군: 114.0±56.5, p=NS). 3. 이차배양 10일째에 젊은군에서 알카리성 포스파타제 활성도가 고령군에 비해 유의하게 높았다(젊은군: 935.5±115.0 U/mg, 노령군: 578.4±115.7U/mg,p.0.05). 고령군에서는 시간 경과에 따른 변화가 미약했으며 전반적으로 알카리성 포스파타제의 활성도가 젊은군에 비해 낮았다. 4. 이차배양도중 오스테오칼신 mRNA의 발현은 배양시기별로 젊은군에 비해 고령군에서 더 낮은 경향을 관찰할 수 있었다. 5. 이차배양 10일과 15일에 젊은군에서 노령군보다세포증식이 유의하게 증가된 양상을 보였다(10앓 젊은군 0.73±0.05, 노령군 0.58±0.04, p=0.05, 15일; 젊은군 0.80±0.05, 노령군 0.70±0.03, p=0.05).결론: 이상의 연구에서 저자들은 노령군에서 젊은군보다 골수 내 중간엽줄기세포의 수가 적고, 노령군에서 유래한 전구조골세포의 성숙조골세포로의 증식 및 분화가 젊은군 보다 감소해 있는 것을 관찰할 수 있었다. Background: Osteoblasts originate from osteoprogenitor cells in bone marrow stroma, termed mesenchymal stem cells (MSCs) or bone marrow stromal cells. Each MSC forms colonies (colony forming units-fibroblasts [CFL-Fs]) when cultured ex vivo. There are some reports about the age-related changes of the number and osteogenic potential of osteoprogenitor cells, but any relationship has not been clearly established in humans. In this study, we counted MSCs using CFU-Fs count and examined the proliferative capacity and differentiation potential of osteoprogenitor cells. Finally, we analyzed how these parameters varied with donor age. Methods: Bone marrow was obtained from the iliac crest of young (n=6, 27.2±8.6 years old) and old (n= 10, 57.4k6.7 years old) healthy donors. Mononuclear cells, including MSCs, were isolated and cultured in osteogenic medium. In primary culture, we compared the colony-forming efficiency of MSCs between the two groups and determined the matrix calcification. When primary culture showed near confluence, the cells were subcultured. Alkaline phosphatase activity, osteocalcin expression by RT-PCR and proliferative potential by MTT assay were examined by the time course of secondary culture. Results: At the 15th day of primary culture, the mean number of CFU-Fs was significantly higher in the younger donors (young: 148.3±28.9, old: 54.3±9.1, p=0.02) and the mean size of CFL-Fs was also larger in the younger donors than the older donors. However, matrix calcification was not different between the two groups (young: 103.6±50.6, old: 114.0±56.5, p=NS). In secondary culture, alkaline phosphatase activities were significantly lower in the older donors. The younger donors showed peak alkaline phosphatase activity at day 10, while the older donors didn't showed a remarkable peak (young: 935.5±115.OU/mg, old: 578.4±115.7U/mg, p<0.05). Total cell number as a proliferative index increased progressively during the secondary culture and a significantly greater cell number was noted in the younger donors. Osteocalcin expression was generally upregulated in the younger donors, but this was not statistically significant. Conclusion: Our study shows that the number of osteoprogenitor cells is decreased during aging and that the proliferative capacity and differentiation potential of osteoprogenitor cells seem to be reduced during aging (J Kor SOC Endocrinol 18:296-305, 2003).

        The prevalence of PPNG among pretreated gonorrhea cases isolated at the STD clinic of Choong-Ku Public Health Center in Seoul has been studied and reported annually since 1981. In 1991, 123 strains of N.gonorrhoeae were isolated, among which 58(47.1%) were PPNG. In 1992, 98 starains of N.gonorrhoeae were isolated, among which 51(52.0%) were PPNG. In all, 109(49.3%) strains were found to be PPNG among 221 strains isolated between 1991-1992. The prevalence of PPNG in Seoul showed increased tendency till 1989, thereafter, it has been stationary or slightly decreasing.

        太子蔘의 造血作用에 對한 實驗的 硏究

        In order to investigate the hematopoietic effects of Pseudostellariae Radix various experiment about hematopoiesis were studied. The results were obtained as follows: 1. In the proliferous effect of bone marrow cells, water extracts of Pseudostellariae Radix played an important role on the proliferating bone marrow cells compared with control group. 2. The water extracts of Pseudostellariae Radix activated the bone marrow cells. 3. The water extracts of Pseudostellariae Radix promoted the proliferation of the bone marrow cells into cluster, BFU-E, CFU-GEMM. 4. In the bone marrow cells treated with CTX, the water extracts of Pseudostellariae Radix promoted the SCF, EPO, IL-4, IL-6 gene expression, but gene expression of its receptors, c-kit, EPO-R, TPO, c-mpl, EKLF, GATA-1 were not promoted significantly. According to the above results, water extracts of Pseudostellariae Radix promoted not only the differentiation of the bone marrow cells into CFU-GEMM which mother cell of erythrocytes, thrombocytes, monocytes and macrophages, but also the differentiation of the bone marrow cells into BFU-E. On the other hand, in the gene expression that have a lot of hematopoietic effects in the early differentiation of haematopoietic stem cells by promoting the gene expression of SCF, EPO, IL-4, IL-6 and so forth. Above results also demonstrates that the water extracts of Pseudostellariae Radix have somewhat meaningful effects in hematogenesis but not sufficient to explain its mechanisms about hematogenesis, therefore further deep studies should be accomplished about its mechanisms.

        사람폐암세포주 (PC-14)에서 Cyclosporin A에 의한 Adriamycin 내성의 극복

        Cyclosporin A and verapamil were tested using MTT assay to evalute the modification effect on the resistance to adriamycin in a human lung cancer cell line(PC-14) and its resistant subline(PC-14/A). PC-14/A was derived by the continuous exposure of PC-14 to incremental concentrations of adriamycin. PC-14/A was 2.5 times more resistant to adriamycin in terms of ICso than PC-14. Cyclosporin A alone, at a concentration of 2.5㎍/㎖, inhibited the growth of PC-14 to 68.3%. 2.5㎍/ ㎖ and 5.0㎍/㎖ of cyclosporin A showed an increase in the cytotoxicity of adriamycin (p<0.01) with 5.0㎍/㎖ being greater than 2.5㎍/㎖(p<0.01). Excluding the direct cytotoxic effect, however, cyclosporin A did not increase in the sensitivity of PC-14 to adriamycin but only showed an additional cytotoxic effect with adriamycin. Verapamil (up to 6.0㎍/㎖) did not inhibit the growth of PC-14. 3.0㎍/㎖ of verapamil did not increase the cytotoxic effect of adriamycin. The combination of cyclosporin A and verapamil with adriamycin enhanced the cytotoxicity of adriamycin, but the result was similar to that of cyclosporin A with adriamycin. 5.0㎍/㎖ of cyclosporin A modified the adriamycin resistance of PC-14/A(SR, 3.2). However, 3.0㎍/㎖ of verapamil did not significantly reverse the adriamycin resistance of PC-14/A. The modified effect of the combination of 5.0㎍/㎖ of cyclosporin A and 3.0㎍/㎖ of verapamil was similar to that of 5.0㎍/㎖ of cyclosporin A alone in PC-14/A. These results demonstrate that cyclosporin A has an additional cytotoxic effect with adriamycin in PC-14 and PC-14/A and has overcome the acquired resistance to adriamycin in PC-14/A. They also suggest that cyclospoin A may have the therapeutic potential in the treatment of human lung cancer.

        Metal matrix composites have toughness, machinability and heat conductivity, which can be obtained in metals, as well as hardness, high temperature stability, which can be obtained in ceramics. The purpose of this study is to find the optimum production condition of a compocasting method which is composed of stirring SiC particles inside a matrix which is presented as a solid or liquid state. During the compocasting method. SiC particles are mechanically incorporated into the matrix. From this study, sound AZ91D/SiC_p composites were obtained when stirring the matrix for 60minutes between 583-588℃.

        Rhabdomyosarcoma, which is a very rare and highly malignant disease in urologic field, has three categories-pleomorphic, alveolar, embryonal, and may arise from any part of the body that contains embryonal mesenchyme, whether striated muscle tissue is present or not. The common site in the genitourinary tract are prostate, bladder, and vagina. Herein we experienced a rhabdomyosarcoma of the pelvic cavity and reviewed literature briefly.

        개인선량계 교정을 위한 환산인자 계산

        MCNP 코드를 사용하여 IAEA에서 권고하고 있는 물팬톰과 미국내 선량계 성능시험 프로그램에서 규정하고 있는 PMMA 팬톰내 0.07mm 및 10mm 깊이에서의 환산인자 H(d)/Ka을 계산하였다. 계산은 팬톰의 한면에 수직으로 입사하는 단일에너지 광자의 확장정열빔에 대해 수행하였다. 결과는 팬톰내에서 정의되는 선량당량으로 선량계를 교정할 때 환산인지로 사용될 수 있다. MCNP code was used to calculate conversion factor H(d)/Ka at the depths of 0.07 and 10mm within a water phantom recommended by IAEA and within a PMMA phantom required by the US dosimeter proficiency testing programmes. The calculations were performed for an expanded parrallel beam of monoenergetic photons of perpendicular incidence on one faces of the phantom. The results can be used as conversion factor in calibrating individual dosemeters in terms of the dose equivalent quantities defined directly in the phantom.

        This research aims at surveying the MIA and the menarche ages by using a follow-up survey of the Korean students height of the two schools in which the job of their parents and their living areas are somewhat different, some result can be withdrawn from this research. 1. The annual increase marks the height point at the age of ten and eleven, the average age of the object menarche marks 12.78 and the average height of them is marked by 151.5㎝. 2. In the relation between the menarche and the season, the objects usually have the menarche in the percentage of 36.82 in summer and in the percentage of 20.72 in August. 3. While the age of the menarche has be in being stationary in Norway and Sweden, that has been decreased 1960 to 1987 in Korea. 4. The ages of the menarche and the 3rd class industry has been lowered than that in the 1st and 2nd class industry.

        Background and aims: The Apolipoprotein E is a ligand of both the protein component LDL receptor as well as the apo E LDL-Receptor related protein (LRP). It modulates the receptor binding of lipoproteins, with the apolipoprotein E found on cell surfaces as its component, thus serving an important role in the lipid metabolism by carrying out the intracellular transport of cholesterol in lipoproteins. The gene for apolipoprotein E is the product of three common genotypes as well as many more rare alleles. The common genotypes are ε2, ε3, and ε4, and are expressed in the three phenotype isoforms of E2, E3, and E4. In the event that E4 is the main component, a rise in the cholesterol level, as the result of down-regulation of the LDL receptor, is observed. Therefore, those samples with E4 genotypes are known to be in much higher risk of coronary artery disease than those with ε3/ε3, while those with ε2 are in low risk (with the exception of hypertiglyceremai Ⅲ). The aim of this study is to analyze in patients with ischemic heart disease the role of aplipoprotein E alleles in order to seek its correlation with coronary artery disease, as well as to seek whether the polymorphism of apo E produces any differences in the severity of coronary artery disease according to plasma lipid levels. Methods: The subjects for study were 273 patients admitted to the Internal Cardiology Division of the Soonchunhyang University Hospital form December 1998 to February 1999. The subjects were divided into the two groups of which one was ischemic heart disease (IHD) experiment group totaling 105 (avg.60.1 years of age, male/female ratio = 69/36) and the control group totaling 168 (avg. 59.7 years of age, male/female = 73:95). The coronary angiogram was given to 127 subjects, and of this total, 94 have developed significant stenosis in the coronary artery. The stages of the analyzing of the apo E phenotype was first, the separation of DNA from the blood samples, subjecting it to the PCR from with 228 base pairs of expanded products were obtained. The band was determined by means of the reverse hybridization principle on the nitrocellulose strip. Results: From the 105 patients the distributions of apo E phenotypes were as follows: ε3/2(5.7%), ε4/2(1.9%), ε3/3(70%), ε4/3(20%), ε4/4(1.9%). The relative frequencies of each allele are as follow: ε2 (0.038), ε3 (0.833), ε4 (0.128). The results show as follows: ⅰ) The IHD experiment group to have a higher occurrence of ε4/3 phenotypes as well as ε4 alleles than the control group. ⅱ) Both the control group and IHD group showed the largest distribution of ε3/3 for phenotypes, and ε4 for alleles. ⅲ) The IHD group showed less ε2/3 phenotypes as well as significantly less allele frequency of ε3 in comparison to the control group. ⅳ) the IHD group showed a much lower level of HDL in comparison to the control group, while the LDL was significantly higher; samples including the apo ε2 showed a significantly higher level of HDL than those without. Among the control group, samples including apo ε2 showed a significantly higher level of TG (triglyceride) than samples without. No significant difference was found between the experiment apo ε4 sample and the control plasma lipid sample. ⅴ) No significant correlation was found between an apo E polynorphism and the number of involved arteries of a coronary angiogram. Conclusion: Between the experiment IHD group and control group were found differences in the frequency of alleles. The polymorphism of apo E alleles may contribute as a risk factor to the development of heart disease by involving itself in the metabolism and modulation of plasma lipids.

      • 接觸酸化法을 利用한 LAS 除去效率에 關한 硏究

        This study was performed to know LAS concentration in the small urban stream and estimate the LAS removal efficiency when apply to natural purification method with variable media. When we used gravel and concrete as media, the average LAS concentration of influent was 819.7㎍/ℓ (112.0 ∼ 1671.0 ㎍/ℓ). The average LAS concentration of effluent gravel media reactor was 58.5 ㎍/ℓ (0 ∼ 158.4 ㎍/ℓ) and average removal efficiency was 93.8%. The average LAS concentration of effluent concrete media reactor was 33.1 ㎍/ℓ (0 ∼ 136.8 ㎍/ℓ) and average removal efficiency was 96.1%. And when we used tire and scrubber, the average LAS concentration of influent was 693.7 ㎍/ℓ (241.5 ∼ 1330.3 ㎍/ℓ). The average LAS concentration of effluent tire media reactor was 88.4 ㎍/ℓ (47.8 ∼ 131.8 ㎍/ℓ) and average removal efficiency was 83.9%. The average LAS concentration of effluent scrubber media reactor was 26.1 ㎍/ℓ(0 ∼ 58.9 ㎍/ℓ) and average removal efficiency was 95.3%.
