GnRH-agonist에 의한 인간 과립-황체화 세포의 세포사멸과 PBR 단백질의 발현

Objective: To investigate whether GnRH-agonist (GnRH-Ag) using in IVF-ET affects apoptosis of human granulosa-luteal cells and expression of peripheral benzodiazepine receptor (PBR) protein involved in the apoptosis of the cells. Methods: Granulosa-luteal cells obtained during oocyte retrieval were cultured and treated with $10^{-5}M$ GnRH-Ag. Apoptosis of the cells by the treatment was confirmed using DNA fragmentation analysis 24 h after culture. The presence of PBR protein within the cells was examined by immunofluorescence staining and the expression of the protein was analyzed by Western blotting. In addition, it was measured for progesterone and nitric oxide (NO) produced by granulosa-luteal cells after GnRH-Ag treatment. To evaluate the relationship between NO production and PBR expression, sodium nitroprusside (SNP) as a NO donor was added in media and investigated the expression of PBR protein by Western blotting. Results: Apoptosis increased in the granulosa-luteal cells 24 h after GnRH-Ag treatment, whereas the expression of PBR protein significantly decreased. Furthermore, the production of progesterone and nitric oxide (NO) by the cells significantly fell from 12 h after the treatment. In the results of Western blotting after SNP treatment, the expression of PBR protein increased in the treatment with SNP alone to the granulosa-luteal cells, but was suppressed in the treatment with GnRH-Ag and SNP. Additionally, the staining result of PBR protein in the cells showed the even distribution of it through the cell. Conclusion: These results demonstrate that GnRH-Ag treatment induces apoptosis, decreasing expression of PBR protein and NO production in human granulosa-luteal cells. The present study suggests that one of the apoptosis mechanism of human granulosa-luteal cells by GnRH-Ag might be a signal transduction pathway via NO and PBR.

This research intended to develop youth civic education program proper for smart era based on the research on the actual condition of youth' democratic citizen capability which was conducted during 2011~2012 by National Youth Policy Institute. According to the research on the actual condition, Korean youth showed the best level in cognitive area in respect of democratic citizen, however the ratio to participate in the actual civil activity appeared the lowest in the world. Accordingly, this research intended to develop adolescents' civic education program which can be the base to be connected with experiential․practical program, and related institutes such as public education and youth organization, etc. which can draw the program suitable for smart environment in which present adolescents live, multicultural period, and global period, and youth' spontaneous participation and active behavior, and to provide necessary program manual. In the situation of insufficient social recognition on adolescent civil education and the social structure as college entrance exam focused education, proper democratic civil education and instructional method are insufficient. As the efforts for this, the researcher planned adolescents' civic capability cultivation program who live in smart era, “SMART CITIZEN” and composed the contents. 본 연구는 한국청소년정책연구원 2011~2012년에 실시한 청소년의 민주시민 역량실태조사를 기반으로 스마트 시대에 맞는 청소년 시민 교육 프로그램을 개발한 것이다. 실태조사에 따르면 한국 청소년의 민주시민에 대한 인식적인 영역에서는 최상의 수준이지만 실제 시민활동에 참여하는 비율은 세계에서 가장 낮은 수준으로 나타났다. 이에 따라 현재 청소년이 살아가고 있는 스마트환경, 다문화시대, 글로벌 시대에 맞는 프로그램, 청소년의 자발적 참여와 능동적 행동을 이끌어 낼 수 있는 체험적․실천적 프로그램, 공교육과 청소년단체 등 관련 기관과 연계되고 필요한 프로그램 매뉴얼을 제공하기 위한 기반이 될 수 있는 청소년 시민교육 프로그램을 개발하기 위함이다. 청소년 시민교육에 대한 사회적 인식 부족과 입시위주 교육과 같은 사회구조 속에서 적절한 민주시민교육과 교육방법은 미흡한 상태이다. 이를 위한 시도로서 스마트시대를 살아가는 청소년들의 시민역량 함양 프로그램“SMART CITIZEN”을 기획하고 내용을 구성하였다.

임신과 동반된 Wilson`s disease는 매우 드문 질환으로 알려져 있으며, 병의 진행에 대한 기전과 모체 및 태아의 상호 관계에 대해서도 많은 보고가 되어있지 않다. 더욱이 치료받지 않은 Wilson`s disease 환자에서 성공적인 임신과 분만은 많이 알려진 바 없다. 저자들은 간경화와 용혈성 빈혈을 동반한 치료받지 않은 Wilson`s disease 산모에서 성공적인 임신과 분만을 경험하였기에 문헌 고찰과 함께 보고하는 바이다. Wilson`s disease associated with pregnancy is not common and the underlying mechanism triggering the event is not known at present. There are few reports of successful pregnancies in women with untreated Wilson`s disease. Recently we experienced a 28-year-old pregnant woman with Wilson`s disease who had liver cirrhosis with hemolytic anemia. The patient had a successful pregnancy outcome with no complication.
