        Halomonas alkalitolerans sp. nov., a Novel Moderately Halophilic Bacterium Isolated from Soda Meadow Saline Soil in Daqing, China

        A moderately halophilic bacterial strain 15-13^T, which was isolated from soda meadow saline soil in Daqing City, Heilongjiang Province, China, was subjected to a polyphasic taxonomic study. The cells of strain 15-13^T were found to be Gram-negative, rod-shaped, and motile. The required growth conditions for strain 15-13^T were: 1-23% NaCl (optimum, 7%), 10-50°C (optimum, 35°C), and pH 7.0-11.0 (optimum, pH 9.5). The predominant cellular fatty acids were C18:1 ω7c (60.48%) and C16:0 (13.96%). The DNA G+C content was 67.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequence comparisons indicated that strain 15-13^T clustered within a branch comprising species of the genus Halomonas. The closest phylogenetic neighbor of strain 15-13^T was Halomonas pantelleriensis DSM 9661^T (98.9% 16S rRNA gene sequence similarity). The level of DNA-DNA relatedness between the novel isolated strain and H. pantelleriensis DSM 9661^T was 33.8%. On the basis of the phenotypic and phylogenetic data, strain 15-13^T represents a novel species of the genus Halomonas, for which the name Halomonas alkalitolerans sp. nov. is proposed. The type strain for this novel species is 15-13^T (=CGMCC 1.9129^T =NBRC 106539^T).

        The mechanism of enhanced photocatalytic activity of SnO2 through fullerene modification

        Carbon nanomaterials are prominent building blocks in the synthetic van der Waals (vdW) heterostructures with desired properties. Scientific understanding of their interfacial interactions is the premise to design this kind of vdW heterostructures with optimal performance.We here study the mechanism of enhanced photocatalytic activity of SnO2 by fullerene modification at electronic level, to explore the interfacial interaction and its correlation with photocatalytic activity. The results show that the interfacial interaction increases with the number of C atom of fullerene, and leads to some of C atoms be positively/ negatively charged, making the fullerene a highly active co-catalyst in heterostructures. Compared to pristine SnO2, the band gap of the heterostructures is much smaller, leading to their absorption wavelength extending the entire visible region. Interestingly, a staggered type-II band alignment in the C20 (C60)/SnO2 (101) heterostructures results into the robust separation of photoexcited charge carriers between the two constituents, indicating that the fullerene is an effective sensitizer, and thus enhanced photocatalytic activity. These findings can rationalize the available experiment and will be of broad interest in developing the highly efficient semiconductor photocatalysts via fullerene modification.

        Nonlinear Bending of Box Section Beams of Finite Length Under Uniformly Distributed Loading

        In this paper, the bending response of box-section beams of finite length is investigated by using energy methods. The basic assumptions used in the present study are that the total strain energy of a box-section beam subjected to uniformly distributed loading can be simplified into a two-stage analysis process. One is the local bending response of the webs and flanges behaving as plates; the other is the overall bending response of the beam with a deformed cross-section. Analytical solutions for both static and dynamic instabilities of box section beams of finite length subjected to transverse uniformly distributed loading are derived by applying the minimum potential energy principle. To validate the analytical solutions developed, geometric nonlinear finite element analyses are also conducted. Good agreement between the present solutions and the FEA results is demonstrated. Finally, the effects of beam length on the limit critical uniformly distributed load are also discussed.

        MAPK4 silencing in gastric cancer drives liver metastasis by positive feedback between cancer cells and macrophages

        Liver metastasis is a major cause of death in gastric cancer patients, but the underlying mechanisms are poorly understood. Through a combination of in vivo screening and transcriptome profiling followed by quantitative RT-PCR and tissue array analyses, we found that mitogen-activated protein kinase 4 (MAPK4) downregulation in gastric cancer tissues from patients is significantly associated with liver metastasis and poor prognosis. The knockdown of MAPK4 in gastric cancer cells promotes liver metastasis in orthotopic mouse models. MAPK4 depletion in gastric cancer cells induces the secretion of macrophage migration inhibitory factor (MIF) to polarize tumor-associated macrophages (TAMs) in orthotopic xenograft tumors. Moreover, TAMs activate epithelial–mesenchymal transition of gastric cancer cells to suppress MAPK4 expression, which further increases MIF secretion to polarize TAMs. Taken together, our results suggest a previously undescribed positive feedback loop between cancer cells and macrophages mediated by MAPK4 silencing that facilitates gastric cancer liver metastasis.

        Sometimes, people can be exposed to moderateor high doses of radiation accidentally or through the environment. Radiation can cause great harm to several systemswithin organisms, especially the hematopoietic system. Severaltypes of drugs protect the hematopoietic system againstradiation damage in diff erent ways. They can be classifi edas “synthetic drugs” and “natural compounds.” Their cellularmechanisms to protect organisms from radiation damageinclude free radical-scavenging, anti-oxidation, reducinggenotoxicity and apoptosis, and alleviating suppression ofthe bone marrow. These topics have been reviewed to providenew ideas for the development and research of drugsalleviating radiation-induced damage to the hematopoieticsystem.

        Expression profiles of circular RNAs in sheep skeletal muscle

        Objective: Circular RNAs (circRNAs) are a newfound class of non-coding RNA in animals and plants. Recent studies have revealed that circRNAs play important roles in cell proliferation, differentiation, autophagy and apoptosis during development. However, there are few reports about muscle development-related circRNAs in livestock. Methods: RNA sequencing analysis was employed to identify and annotate circRNAs from longissimus dorsi of sheep. Reverse transcription followed by real-time quantitative (q) polymerase chain reaction (PCR) analysis verified the presence of these circRNAs. Targetscan7.0 and miRanda were used to analyse the interaction of circRNA-microRNA (miRNA). To investigate the function of circRNAs, an experiment was conducted to perform enrichment analysis hosting genes of circRNAs using gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) pathways. Results: About 75.5 million sequences were obtained from RNA libraries of sheep skeletal muscle. These sequences were mapped to 729 genes in the sheep reference genome. We identified 886 circRNAs, including numerous circular intronic RNAs and exonic circRNAs. Reverse transcription PCR (RT-PCR) and DNA sequencing analysis confirmed the presence of several circRNAs. Real-Time RT-PCR analysis exhibited resistance of sheep circRNAs to RNase R digestion. We found that many circRNAs interacted with muscle-specific miRNAs involved in growth and development of muscle, especially circ776. The GO and KEGG enrichment analysis showed that hosting genes of circRNAs was involved in muscle cell development and signaling pathway. Conclusion: The study provides comprehensive expression profiles of circRNAs in sheep skeletal muscle. Our study offers a large number of circRNAs to facilitate a better understanding of their roles in muscle growth. Meanwhile, we suggested that circ776 could be analyzed in future study.

        Effects of different culture systems on the culture of buffalo (Bubalus bubalis) spermatogonia stem cell-like cells in vitro

        Currently, the systems for culturing buffalo spermatogonial stem cells (SSCs) in vitro are varied, and their effects are still inconclusive. In this study, we compared the effects of culture systems with undefined (foetal bovine serum) and defined (KnockOut Serum Replacement) materials on the in vitro culture of buffalo SSC-like cells. Significantly more DDX4- and UCHL1-positive cells (cultured for 2 days at passage 2) were observed in the defined materials culture system than in the undefined materials system (p < 0.01), and these cells were maintained for a longer period than those in the culture system with undefined materials (10 days vs. 6 days). Furthermore, NANOS2 (p < 0.05), DDX4 (p < 0.01) and UCHL1 (p < 0.05) were expressed at significantly higher levels in the culture system with defined materials than in that with undefined materials. Induction with retinoic acid was used to verify that the cultured cells maintained SSC characteristics, revealing an SCP3+ subset in the cells cultured in the defined materials system. The expression levels of Stra8 (p < 0.05) and Rec8 (p < 0.01) were significantly increased, and the expression levels of ZBTB16 (p < 0.01) and DDX4 (p < 0.05) were significantly decreased. These findings provided a clearer research platform for exploring the mechanism of buffalo SSCs in vitro.

      • RNAi-based Knockdown of Multidrug Resistance-associated Protein 1 is Sufficient to Reverse Multidrug Resistance of Human Lung Cells

        Up-regulation of multidrug resistance-associated protein 1 (MRP1) is regarded as one of the main causes for multidrug resistance (MDR) of tumor cells, leading to failure of chemotherapy-based treatment for a multitude of cancers. However, whether silencing the overexpressed MRP1 is sufficient to reverse MDR has yet to be validated. This study demonstrated that RNAi-based knockdown of MRP1 reversed the increased efflux ability and MDR efficiently. Two different short haipin RNAs (shRNAs) targeting MRP1 were designed and inserted into pSilence-2.1-neo. The shRNA recombinant plasmids were transfected into cis-dichlorodiamineplatinum-resistant A549 lung (A549/DDP) cells, and then shRNA expressing cell clones were collected and maintained. Real time PCR and immunofluorescence staining for MRP1 revealed a high silent efficiency of these two shRNAs. Functionally, shRNA-expressing cells showed increased rhodamine 123 retention in A549/DDP cells, indicating reduced efflux ability of tumor cells in the absence of MRP1. Consistently, MRP1-silent cells exhibited decreased resistance to 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) and DDP, suggesting reversal of MDR in these tumor cells. Specifically, MRP1 knockdown increased the DDP-induced apoptosis of A549/DDP cells by increased trapping of their cell cycling in the G2 stage. Taken together, this study demonstrated that RNAi-based silencing of MRP1 is sufficient to reverse MDR in tumor cells, shedding light on possible novel clinical treatment of cancers.

        Expression and evaluation of porcine circovirus type 2 capsid protein mediated by recombinant adeno-associated virus 8

        Background: Porcine circovirus type 2 (PCV2) is an important infectious pathogen implicated in porcine circovirus-associated diseases (PCVAD), which has caused significant economic losses in the pig industry worldwide. Objectives: A suitable viral vector-mediated gene transfer platform for the expression of the capsid protein (Cap) is an attractive strategy. Methods: In the present study, a recombinant adeno-associated virus 8 (rAAV8) vector was constructed to encode Cap (Cap-rAAV) in vitro and in vivo after gene transfer. Results: The obtained results showed that Cap could be expressed in HEK293T cells and BABL/c mice. The results of lymphocytes proliferative, as well as immunoglobulin G (IgG) 2a and interferon-γ showed strong cellular immune responses induced by Cap-rAAV. The enzyme-linked immunosorbent assay titers obtained and the IgG1 and interleukin-4 levels showed that humoral immune responses were also induced by Cap-rAAV. Altogether, these results demonstrated that the rAAV8 vaccine Cap-rAAV can induce strong cellular and humoral immune responses, indicating a potential rAAV8 vaccine against PCV2. Conclusions: The injection of rAAV8 encoding PCV2 Cap genes into muscle tissue can ensure long-term, continuous, and systemic expression.

        Framework for automated finite element analysis with an ontology-based approach

        To obtain accurate and reliable results of finite element analysis (FEA) requires a high level of expertise and the fullscale physical context information, which are bottlenecks restricting the application of achievements of FEA in industry. This paper proposes an ontology-based framework including a hierarchy transfer approach and a three-stage automated finite element analysis method to solve these problems. The hierarchy transfer approach is proposed to create different transfer formats according to the data, information, and knowledge, to carry out the integration at different levels. The knowledge found in design and FEA theories is presented by ontology in order to uniformly describe the physical phenomenon with the same semantic meanings. This involves the development of a shared design and FEA ontology, as well as, specific application ontologies in the Ontology Web Language (OWL). The three-stage automated finite element analysis method is applied to mark up artifact in problem definition, to reuse domain knowledge in problem formulation, and to enable the automation of the FEM analysis process in the solution routine with the application of AI techniques. The feasibility and effectiveness of the framework and concepts are empirically validated by a case study.
