        Comparison of Hybridization Behavior between Double and Single Strand of Targets and the Application of Asymmetric PCR Targets in cDNA Microarray

        Double stranded targets on the cDNA microarray contain representatives of both the coding and noncoding strands, which will introduce hybridization competition with probes. Here, the effect of double and single strands of targets on the signal intensity and the ratios of Cy5/Cy3 within the same slide were compared. The results show that single stranded targets can increase the hybridization efficiency without changing the Cy5/Cy3 ratio. Based on these results, a new strategy was established by generating cDNA targets with asymmetric PCR, instead of conventional PCR, to increase the sensitivity of the cDNA microarray. Furthermore, the feasibility of this approach was validated. The results indicate that the cDNA microarray system based on asymmetric PCR is more sensitive, with no decrease in the reliability and reproducibility as compared with that based on conventional symmetric PCR.

      • Trends in Nasopharyngeal Carcinoma Mortality in China, 1973-2005

        Nasopharyngeal carcinoma (NPC) is a disease with distinct ethnic and geographic distribution. The incidence of NPC in Chinese residing in Asia has declined over the last few decades, but NPC mortality trends in the entire Chinese population over time have not been systematically evaluated. In this study, we examined NPC mortality at the national level in China between 1973-2005. Mortality rates were derived from the databases of national retrospective surveys on cancer mortality conducted in the periods of 1973-1975, 1990-1992, and 2004-2005, respectively. NPC was classified according to the International classification of diseases. Age-adjusted mortality rates were calculated by direct standardization according to the world standard population. Trends in rates were evaluated by age, gender, geographic areas, and socioeconomic status. From 1973 to 2005, there was a general trend of decrease in NPC mortality in China, with higher rates in the south on a downward trend in the north. The age-standardized NPC mortality rates were 2.60 per 100,000 in 1973-1975, 1.94 per 100,000 in 1990-1992, and 1.30 per 100,000 in 2004-2005, respectively. The trend was similar in both men and women, in both urban and rural areas, but the declining rates in females were more remarkable than in males. The mortality rates were higher for the age groups above 50 years than those less than 50 years of age, both showing downward trend over 30-year period. In summary, the overall NPC mortality has consistently decreased in China over the past three decades, particularly in women and in old adults.

      • Heating Control Design of a Space Camera

        When a space camera works on the track, it presents in vacuum environment, under the condition of low temperatures, the camera can only maintain its temperature level by absorption of radiation. If stayed in this environment for a long time, mechanical structure of the space camera may become brittle, lose strength, and an exfoliation phenomenon may appear, in addition, abnormal operation and malfunction of electronic equipments may also occur, these may affect imaging quality of the camera. In this paper, a heating control method is designed for a space flight off-axis camera. Firstly, heating control working platform of the camera is introduced, a design method of heating control is determined. After that, heating control treatment procedure is introduced. Finally, the ground temperature monitor is used to validate correctness of the method designed. The comparison diagram before and after thermal analysis and temperature data obtained from experiments are presented, results show that, the heating control method is simple and effective, it has wide applicability and strong generalization performance.

      • Down-regulation of SENP1 Expression Increases Apoptosis of Burkitt Lymphoma Cells

        Objective: To investigate the effect of down-regulation of Sentrin/SUMO-specific protease 1 (SENP1) expression on the apoptosis of human Burkitt lymphoma cells (Daudi cells) and potential mechanisms. Methods: Short hairpin RNA (shRNA) targeting SENP1 was designed and synthesized and then cloned into a lentiviral vector. A lentiviral packaging plasmid was used to transfect Daudi cells (sh-SENP1-Daudi group). Daudi cells without transfection (Daudi group) and Daudi cells transfected with blank plasmid (sh-NC-Daudi group) served as control groups. Flow cytometry was performed to screen GFP positive cells and semiquantitative PCR and Western blot assays were employed to detect the inference efficiency. The morphology of cells was observed under a microscope before and after transfection. Fluorescence quantitative PCR and Western blot assays were conducted to measure the mRNA and protein expression of apoptosis related molecules (caspase-3, 8 and 9). After treatment with $COCl_2$ for 24 h, the mRNA and protein expression of hypoxia inducible factor -$1{\alpha}$ (HIF-$1{\alpha}$) was determined. Results: Sequencing showed the expression vectors of shRNA targeting SENP1 to be successfully constructed. Following screening of GFP positive cells by FCM, semiqualitative PCR showed the interference efficiency was $79.2{\pm}0.026%$. At 48 h after transfection, the Daudi cells became shrunken, had irregular edges and presented apoptotic bodies. Western blot assay revealed increase in expression of caspase-3, 8 and 9 with prolongation of transfection (P<0.05). Following hypoxia treatment, mRNA expression of HIF-$1{\alpha}$ remained unchanged in three groups (P>0.05) but the protein expression of HIF-$1{\alpha}$ markedly increased (P<0.05). However, in the sh-SENP1-Daudi group, the protein expression of HIF-$1{\alpha}$ remained unchanged Conclusion: SENP1-shRNA can efficiently inhibit SENP1 expression in Daudi cells. SENP1 inhibition may promote cell apoptosis. These findings suggest that SENP1 may serve as an important target in the gene therapy of Burkitts lymphoma.

        Identification and Molecular Evolutionary Analysis of TLR5 Gene from the Tibetan macaque (Macaca thibetana)

        In order to identify the Toll-like receptor 5 (TLR5) as a putative candidate disease-resistance gene in Tibetan macaque (Macaca thibetana), two pairs of primers were designed based on the TLR5 gene sequence of rhesus macaque (Macaca mulatta,NM_001130429). The primers were used to amplify the TLR5 gene from Tibetan macaque, by the polymerase chain reaction (PCR). The compiled sequences were analyzed by bioinformatics. The DNA sequencing and additional combined results showed that the Tibetan macaque TLR5 gene is about 2825 bp and contains an open reading frame of 2577 bp encoding for 858 amino acids. Homology analysis of TLR5 in both species showed that the amino acid and nucleotide identity is about 99.7% and 99.8% and their transmembrane and intracellular domains appeared more conservative than the extracellular domains of proteins. However, re-examining the entire Tibetan macaque TLR5 coding sequence we found that a purifying selection was also acting on the TLR5 gene region encoding for its intracellular domain of the protein. Even though the selection tests indicated that the TLR5 gene experienced a strong purifying selection in the process of evolution,most likely because its potential role in the primate adaptive immune defense, the Tibetan macaque still has the highest re-lationship with the rhesus macaque.

        Robust Unit Commitment with Virtual Synchronous Renewables

        With the increasing penetration of renewable energy sources and the consequent displacement of synchronous generators, the frequency stability of power systems deteriorates since RESs cannot participate in active power and frequency control. By emulating the outer characteristics of SGs and deloading a controllable margin, RESs can also keep headroom and provide frequency responses. This paper focuses on incorporating virtual synchronous renewables into the unit commitment to optimize the joint scheduling of energy production and spinning reserves. First, based on the interval representation of renewable uncertainty, the renewable output is decomposed into the stochastic and dispatchable portions. The dispatchable portion, defi ned as VSRs, can keep deterministic regulation headroom and participate in power balance regulation. Second, a robust security-constrained unit commitment model is proposed to achieve optimal scheduling in face of multiple uncertainties. The fl exibility of the pumped-storage hydro station is also considered. Third, the solution strategies which linearize the nonlinear terms in the objective function and constraints are introduced. Finally, the correctness and eff ectiveness of the robust unit commitment model are tested and verifi ed in the modifi ed 39-bus system.

        Effect of Lactobacillus rhamnosus hsryfm 1301 on the Gut Microbiota and Lipid Metabolism in Rats Fed a High-Fat Diet

        Accumulating evidence indicates that lactic acid bacteria could improve host physiology and lipid metabolism. To investigate the effect of the gut microbiota on host lipid metabolism, a hyperlipidemic rat model was established by feeding rats a high-fat diet for 28 days, and the gut microbiota of the rats was analyzed using real-time PCR before and after administration of Lactobacillus rhamnosus hsryfm 1301 and its fermented milk for 28 days. The findings showed that the Lactobacillus spp., Bifidobacterium spp., Bacteroides spp., and Enterococcus spp. content in the hyperlipidemic rats gut was increased significantly (p < 0.05), while the Clostridium leptum and Enterobacter spp. content was decreased significantly after intervening with L. rhamnosus hrsyfm 1301 and its fermented milk for 28 days (p < 0.05). Furthermore, the lipid levels of the serum and the liver were decreased significantly (p < 0.05) and the fecal water content was increased significantly (p < 0.05) in the hyperlipidemic rats after the intervention, and hepatocyte fatty degeneration of liver tissues was also prevented. A positive correlation was observed between the Clostridium leptum content and the level of serum cholesterol, triglycerides, low-density lipoprotein, and high-density lipoprotein, and a negative correlation was observed between the Enterobacter spp. content and the Lactobacillus spp. and Bifidobacterium spp. content in the hyperlipidemic rats gut. These results suggest that the gut microbiota and lipid metabolism of hyperlipidemic rats could be improved by supplementation with L. rhamnosus hsryfm 1301 and its fermented milk.

        Probiotic supplements alleviate gestational diabetes mellitus by restoring the diversity of gut microbiota: a study based on 16S rRNA sequencing

        Probiotics effectively prevent and improve metabolic diseases such as diabetes by regulating the intestinal microenvironment and gut microbiota. However, the effects of probiotics in gestational diabetes mellitus are not clear. Here, we showed that probiotic supplements significantly improved fasting blood glucose in a gestational diabetes mellitus rat model. To further understand the mechanisms of probiotics in gestational diabetes mellitus, the gut microbiota were analyzed via 16S rRNA sequencing. We found that compared with the normal pregnant group, the gestational diabetes mellitus rats had decreased diversity of gut microbiota. Moreover, probiotic supplementation restored the diversity of the gut microbiota in gestational diabetes mellitus rats, and the gut microbiota structure tended to be similar to that of normal pregnant rats. In particular, compared with gestational diabetes mellitus rats, the abundance of Firmicutes and Actinobacteria was higher after probiotic supplementation. Furthermore, activating carbohydrate metabolism and membrane transport pathways may be involved in the potential mechanisms by which probiotic supplements alleviate gestational diabetes mellitus. Overall, our results suggested that probiotic supplementation might be a novel approach to restore the gut microbiota of gestational diabetes mellitus rats and provided an experimental evidence for the use of probiotic supplements to treat gestational diabetes melitus.
